Hill's plots of oxygen binding isotherms reveal the presence of a transition between two different oxygen affinities at the beginning and end of the isotherm. They correspond to the two conformations anticipated by the MWC model, namely, the T and R conformations at the beginning and end of oxygen binding, when the lower affinity of the T form develops into the higher affinity of the R form. The difference between the binding Gibbs free energy changes of the two affinities (Δ G(L)) is the free energy of binding cooperativity. Notably, Δ G(L) is positive in favor of the T form, which moves to a higher energy level upon oxygen release. Osmotic stress reveals a higher volume/surface ratio of deoxyhemoglobin, with a positive Δ G(W) also in favor of the T form. An increasing protein concentration shifts the isotherms to the right, indicating the formation of intermediate polymeric forms. The enthalpy of the intermediates shows a strong absorption of heat at the third oxygenation step because of polymer formation with quinary, and higher-order, structures. The disassembly of intermediate polymers releases energy with a negative Δ G that compensates and allows the positive values of Δ G(L). High-energy polymers are the barrier preventing the relaxation of the T and R conformations into one another. The MWC allosteric model is the best justification of oxygen binding cooperativity.

Cross-linked human hemoglobin (HbA) is obtained by reaction with bis(3,5-dibromosalicyl) sebacate. Peptide maps and crystallographic analyses confirm the presence of the 10 carbon atom long sebacyl residue cross-linking the two beta82 lysines of the beta-cleft (DecHb). The Adair's constants, obtained from the oxygen binding isotherms, show that at the first step of oxygenation normal hemoglobin and DecHb have a very similar oxygen affinity. In DecHb negative binding cooperativity is present at the second step of oxygenation, which has an affinity 27 times lower than at the first step. Positive cooperativity is present at the third binding step, whose affinity is 380 times that of the second step. The fourth binding step shows a weak negative cooperativity with an affinity one-half that of the third step. Crystals of deoxy-DecHb diffracted to 1.9 angstroms resolution. The resulting atomic coordinates are very similar to those of Fermi et al. [(1984) J. Mol.Biol. 175, 159-174] and Fronticelli et al. [(1994) J. Biol Chem. 269, 23965-23969] for deoxy-HbA. The electron density map of deoxy-DecHb indicates the presence of the 10 carbon bridge between the beta82 lysines. Molecular modeling confirms that insertion of the linker into the T structure requires only slight displacement of the two beta82 lysines. Instead, insertion of the linker into the R and R2 structures [Shaanan (1983) J. Mol. Biol. 171, 31-59; Silva et al. (1992) J. Biol. Chem. 267, 17248-17256] is hindered by serious sterical restrictions. The linker primarily affects the partially and fully liganded states of hemoglobin. The data suggest in DecHb concerted conformational changes at each step of oxygenation.

We have reported [Bucci, E., Fronticelli, C., & Grycznski, Z. (1991) Biochemistry 30, 3195-3199] that in human and bovine hemoglobins the release of heat at the subsequent steps of oxygenation is not constant. This is especially evident in the binding of the third O2 molecule, which is an endothermic event. This phenomenon was attributed to peculiar conformations of the intermediates of oxygenation, not included in the fundamental R/T transition of the system. To test this hypothesis, we have explored the effect of conformational constraints on the thermodynamics of the intermediates of oxygenation. The assumption was that intramolecular constraints would stabilize the intermediates into conformations similar to the R and T forms reducing the variability of their enthalpies. We have analyzed the temperature dependence of the oxygen binding isotherms of human hemoglobin cross-linked either between the beta 82 or between the alpha 99 lysines by bis(3,5-dibromosalicyl)fumarate. The measurements were perfomred at pH 9.0 in 0.1 M borate buffer in order to avoid thermal effects due to oxygen-linked binding of anions and protons. The data were analyzed singularly by local procedures and simultaneously using global procedures. The two cross-links had opposite effects. The cross-link between the beta-subunits decreased while that between the alpha-subunits increased the endothermic behavior of the third step of oxygenation. Also, the cross-link between the beta-subunits increased the fractional amount of the triligated species at intermediate stages of oxygenation, while that between the alpha-subunits decreased this quantity to hardly detectable values.(ABSTRACT TRUNCATED AT 250 WORDS)

Using the mutagenesis and a gene expression system previously described [Fronticelli et al. (1991) J. Protein Chem. 10, 495-501], we have replaced Val67E11 in the distal heme pocket of the beta-chains of hemoglobin with Thr. The valine to threonine substitution is isosteric and only modifies the polarity of the beta-heme environment. The absorption and CD spectra of the resultant mutant hemoglobin were essentially the same as that of wild-type protein, indicating that the mutation did not cause any large conformational changes and that a water molecule was not coordinated to the ferrous iron atom. Equilibrium measurements of oxygen binding to the mutant indicate a 2-fold decrease in overall affinity relative to native or wild-type human hemoglobin. Thermodynamic analyses of O2 binding curves, based either on the sequential Adair model or on the MWC two-state model, indicated that the overall decrease of O2 affinity in the system was due to a lower association equilibrium constant for the intermediates of oxygenation, particularly those involved at the third ligation step. The functional characteristics of the mutant hemoglobin in either the T- or R-state were not modified greatly by the mutation; however, the Bohr effect and sensitivity to C1- were increased, suggesting a role of the intermediates of oxygenation in the modulation of these parameters.(ABSTRACT TRUNCATED AT 250 WORDS)

We have measured the temperature dependence of the oxygen-binding isotherms of human and bovine hemoglobin at pH 9.0 in 0.1 M borate buffer. In both hemoglobins the ionization of the Bohr protons is finished at this pH; therefore, their heat does not interfere with the measurements. Two sets of curves have been obtained, which have been analyzed by either singular or global procedures for estimating the enthalpy changes of subsequent steps of oxygenation. The data indicate that in human hemoglobin the reaction with oxygen is enthalpy driven for steps 1, 2, and 4 while it is entropy driven for step 3. In bovine hemoglobin this phenomenon is even more evident: steps 2 and 4 are enthalpy driven while steps 1 and 3 are entropy driven. The discontinuous distribution of heat at subsequent steps of oxygenation suggests that the T to R transition in hemoglobin is not a monotonic process and involves conformations with novel characteristics.

Band 3 was labeled with N-[[(iodoacetyl)amino]ethyl]-5-naphthylamine-1-sulfonate either exofacially in the intact washed erythrocytes or endofacially by treating inside-out vesicles. Exo labeling resulted in the labeling of several other proteins, besides band 3, which could not be removed from the membrane. Therefore, the exo-labeled band 3 was extracted and purified by chromatography on DEAE-cellulose in Triton X-100. The endo labeling also resulted in the labeling of several other proteins. In this case, washing with NaOH removed all labeled material except band 3 from the vesicles. The lifetime of bound N-[(acetylamino)ethyl]-5-naphthylamine-1-sulfonate was heterogeneous, suggesting the positioning of the label in different environments either because different sites were labeled or because of positional freedom of the label at the same point of attachment. The main fraction of emission intensity had a lifetime near 20 ns, as expected for a hydrophobic environment. The rest showed a lifetime of about 3 ns in the exo-labeled band 3 and 9 ns in the endo-labeled band 3. Both lifetimes appeared to be independent of temperature between 5 and 25 degrees C, suggesting shielding of the probe from the solvent. Quenching phenomena must be responsible for both the 3- and 9-ns lifetimes, not due to residual heme, as proven by the persistence of such quenching in the Triton X-100 extracted protein. The correlation times indicated the presence of a short component, between 2 and 4 ns in the different systems, probably due to the presence of a flexible portion in the structure of the protein.(ABSTRACT TRUNCATED AT 250 WORDS)

Human apohemoglobin was labeled at Val-beta 1 with pyridoxamine 5'-phosphate. Correlation times were evaluated from steady-state fluorescence anisotropy and lifetime measurements upon quenching with KI. Multiple correlation times were present in the system with a major component of 23.3 ns and a minor component of less than 0.1 ns. The initial depolarization produced by these fast motions occurred in a cone with a semiangle near 33 degrees. The sedimentation velocity and circular dichroism spectra of pyridoxal 5'-phosphate labeled apohemoglobin were very similar to those of unlabeled apohemoglobin. Addition of 8-anilino-1-naphthalenesulfonate did not modify these parameters. Energy transfer of fluorescence was measured between the label, pyridoxamine 5'-phosphate, positioned at Val-beta 1, as donor, and 8-anilino-1-naphthalenesulfonate, bound inside the heme pocket, as acceptor. A quantum yield of 0.21 was measured for labeled apohemoglobin with a standard of pyridoxamine 5'-phosphate. Quenching of the lifetime and of the emission of the donor in the presence of acceptor was measured at 390 nm upon excitation at 313 nm. From these parameters, and on assumption of a random orientation of the fluorophores, an average distance of about 25 A was estimated between the two probes. Numerical correction for 85% saturation of the donor with acceptor produced distances near 23 A for the quenching of emission intensity and near 19 A for the quenching of lifetimes. In the tridimensional model of deoxyhemoglobin, the distance between Val-beta 1 and the nearest iron is about 22 A. Transfer to acceptors positioned in the alpha subunits was negligible. Taking into account the dimensions of the probes, it appears that removal of heme and the consequent loss of helical structure of the system did not produce an expansion of the beta subunits.

We have investigated the denaturation by guanidine hydrochloride (Gdn X HCl), temperature, and pH of hemoglobin beta subunits and of the peptides beta (1-146), beta (56-146), and beta (1-55). The last peptide was insensitive to all of the three agents. In the other polypeptides denaturation by Gdn X HCl and temperature showed the presence of several structural domains characterized by different stabilities. Analyses of the data obtained in Gdn X HCl indicated the presence in beta subunits of an alpha-helical domain involving some 40 amino acids whose free energy of denaturation is only 2000 cal. This domain is heme dependent, and removal of heme in apo-beta (1-146) abolishes the presence of the domain as a structural entity. Acid denaturation reveals in beta subunits, apo-beta (1-146), and beta (56-146) the presence of buried histidines with very similar characteristics, indicating a similar tertiary structure in the three polypeptides. This suggests that removal of the heme produces an unfolding of beta subunits, involving preferentially the 1-55 portion of the chain. This portion of the polypeptide contributes substantially to the formation of the alpha 1 beta 2 interface in hemoglobin. The low stability of this domain implies a very small contribution to the stability of the system as a whole. Instead it makes it very sensitive to conformational attitudes of the heme, suggesting a role in the mechanism of ligand binding cooperativity and subunits interactions in hemoglobin.

The interaction of carbon monoxide beta chains with two allosteric effectors, namely inositol hexaphosphate and benzenehexacarboxylate, was studied. The sedimentation coefficient (s20,w) of the liganded beta chains was measured to be the same both in the presence and absence of the two effectors suggesting that the protein exists as a tetramer under the conditions of our titration and optical studies. The binding of benzenehexacarboxylate to the liganded beta chains was investigated by potentiometric titration in the pH range 6.7-8.0. The results at pH 7.4 showed a binding of 2 mol of benzenehexacarboxylate per tetramer, with an association constant of 1.26 X 10(4) 1. mol-1 at 20 degrees C. The Hill coefficient for the binding was determined to be 0.73. Similar experiments on the interaction of inositol hexaphosphate with the beta chains showed a binding of 2 mol of the effector per tetramer with identical Hill coefficient (0.737) and comparable association constants (0.88 X 10(4)1. mol-1). The value below unity of the Hill coefficient, found for the binding of the two effectors to the protein, probably reflected an anticooperativity produced by the different net electric charges of the free protein and the protein-effector complex. The difference in protons bound per mole of heme by the beta subunits in the presence and absence of benzenehexacarboxylate appeared consistent with the proposal that two groups per chain changed their pK from 6.6 to 7.4 upon the interaction. In the presence of benzenehexacarboxylate, the protonation of these groups appeared to be cooperative, suggesting a conformational change of the protein upon the binding. The absorption spectra of carbon monoxide beta chains in the Soret region was markedly altered by benzenehexacarboxylate and inositol hexaphosphate. The features in the difference spectra of the protein obtained with the two effectors were qualitatively identical and indicated changes in the heme environment produced by the interaction of the effectors with the beta chains. Concomitant changes in circular dichroism and optical rotatory dispersion of the liganded beta chains caused by the addition of the two effectors provided supporting evidence for the conformational change in the protein produced by the binding of the effectors.

The pH dependence of oxygen affinity of hemoglobin (Bohr effect) is due to ligand-linked pK shifts of ionizable groups. Attempt to identify these groups has produced controversial data and interpretations. In a further attempt to clarify the situation, we noticed that hemoglobin alkylated in its liganded form lost the Bohr effect while hemoglobin alkylated in its unliganded form showed the presence of a practically unmodified Bohr effect. In spite of this difference, analyses of the extent of alkylation of the two compounds failed to identify the presence of specific preferential alkylations. In particular, the alpha 1 valines and beta 146 histidines appeared to be alkylated to the same extent in the two proteins. Focusing our attention on the effect of the anions on the functional properties of hemoglobin, we measured the Bohr effect of untreated hemoglobin in buffers made with HEPES [N-(2-hydroxyethyl)piperazine-N'-2-ethanesulfonic acid], MES [2-(N-morpholino)ethanesulfonic acid], and MOPS [3-(N-morpholino)propanesulfonic acid], which being zwitterions do not need addition of chlorides or other anions for reaching the desired pH. The shape acquired by the Bohr effect curves, either as pH dependence of oxygen affinity or as pH dependence of protons exchanged with the solution, was irreconcilable with that of the Bohr effect curves in usual buffers. This indicated the relevance of solvent components in determining the functional properties of hemoglobin. A new thermodynamic model is proposed for the Bohr effect that includes the interaction of hemoglobin with solvent components. The classic proton Bohr effect is a special case of the new theory.

The fluorescence characteristics of 8-anilino-naphthalene-1-sulfonic acid (ANS) coupled to apohemoglobin and to apohemoglobin labeled with fluorescein iodoacetamide (FIA) at beta-93 have been compared. The quenching of emission of ANS produced by FIA was measured both with steady-state and with time-resolved techniques. In this system the emission of ANS in the beta-heme pockets was totally quenched by FIA at beta-93. Steady-state measurements indicated a 57% efficiency of energy transfer between ANS in the alpha-heme pockets and FIA at beta-93. Time resolution showed that the initial (unquenched) lifetime of ANS was 18.2 ns. In the presence of FIA two new components were generated with lifetimes of 2.0 and 6.6 ns. Assuming a random orientation of the probes, the distances inferred from these measurements were near 4.6 and 3.6 nm for the time-resolved and near 48 A for the steady-state measurements. In the tridimensional model of hemoglobin the distance between the iron atom of the alpha 1 chains and the SH group of the beta 1 chains at position 93 is 3.6 nm in oxyhemoglobin and 4.1 nm in deoxyhemoglobin. To these distances 0.5-1.0 nm may be added to allow for the dimensions of the probes. Thus it appears that removal of the heme fails to produce any important enlargement of the molecule. On the contrary, the data suggest a slight shrinking of apohemoglobin, which may be consistent with a collapse of the heme pocket when heme is removed. The rest of the molecule does not seem to be greatly affected.(ABSTRACT TRUNCATED AT 250 WORDS)
