Abstract

Research was conducted on the mechanism of caldesmon inhibition of ATPase activity of actinomycin. Targeted mutagenesis and NMR studies show that interaction of two sites of the caldesmon domain 4B with the actin subdomain 1 elicits the inhibitory activity.

Abstract

The regulatory interaction of the 10 kilodalton c-terminal fragment of caldesmon with actin and the influence of mutation of caldesmon residues 691-696 were investigated via nuclear magnetic resonance spectroscopy, binding studies and adrenosine triphosphitase activity assays. The results showed simultaneous dual-sited contacts with F-actin for the segments of the 658C sequence flanking tryptophan residues 692 and 722. The results further revealed that both 658C contacts with actin were substituted by substoichiometric concentrations of troponin-I and caldesmon.

Abstract

Phosphorylation of the cardiac troponin complex by PKA at S22 and S23 of troponin I(Tnl) accelerates Ca(sup 2+) release from troponin C (TnC). The conclusion states that phosphorylation switch binds to TnC through an extended interaction site spanning residues R19 to A34.
