24. Transaminase GOT and GPT activity in extirped sprouts of normal and opaque-2 Maize (Zea maysL.) seedlings

Abstract

The increased activity of GOT (E.C.2.6.1.1.) and GPT (E.C.2.6.1.2.) transaminases in maize seedlings found as a marker of genotype opaque-2, was investigated in extirped sprouts of both genotypes, normal and opaque-2. The enzymatic activity was determined in three maize samples from breeding experiments, each sample consisting of a genotype pair, normal and opaque-2, collected from segregating ears of maize plants in the S1generation. The seedlings were aseptically grown for 7 days in two variants of cultivation, intact seedlings and sprouts extirped after 4 days of germination. In the intact seedlings of genotype opaque-2 an increased activity of GOT and GPT, as compared to the intact normal plants, was observed. The extirpation of the sprouts enhanced GOT and GPT activity in the sprouts of both genotypes. However, in extirped sprouts the GOT and GPT activity was found to be still higher in the genotype opaque-2 as compared with the sprouts of normal genotype. Thus it seems that the increased transaminase activity in the sprouts of genotype opaque-2 is genetically determined. The increase does not result from an induction of enzyme synthesis through the supply of amino acids translocated from the endosperm to the sprouts. The absolute level of transaminases in the different breeding samples is dependent on the parenteral lines, the relative level of GOT and GPT activities is higher in the genotype opaque-2.

Abstract

Changes in the phenylalanine ammonia-lyase (PAL) activity, accumulation of phenolic acids and ionically-bound peroxidase activity in thein vitroselected embryogenic and nonembryogenicMedicago sativacallus cultures resistant to the filtrate ofFusariumspp. were found. The PAL activity in bothin vitroselected cultures during a 4-week cultivation on a medium with phytotoxins was higher than in the control calli grown on a medium without toxin. The filtrate fromFusariumspp. evoked an increase in the contents of all determined phenolic acids in the selected calli. They occurred predominantly bound as esters. The most pronounced portions in the elevated acids level were of ester-bound p-hydroxybenzoic, vanillic, p-coumaric and ferulic acids. The ionic cell wall-bound peroxidase activity in both selected calli cultivated on a medium with a filtrate was twice as high as the activity determined in the control cultures. The activity of soluble peroxidase was not influenced by challenge with a filtrate. No significant differences were found between thein vitroselected embryogenic and nonembryogenic alfalfa callus cultures in the response to the phytotoxic filtrate.

26. Comparison ofin vivoactivity of L-tryptophan synthas in plants with a low and a high content of L-tryptophan

Abstract

The activity of L-tryptophan synthase (TS, E.C.4.2.1.20) was comparedin vivoin seedlings of plants high in L-tryptophan (L-trp) (pea and kohlrabi) and low in this amino acid (maize). In maize the TS was studied both in the normal and in the opaque-2 genotype that forms an endosperm richer in essential amino acids. The activity of TS was determined on the basis of the increase in radioactivity of the chromatographically purified L-trp-14C, synthetized after vacuum infiltration of L-serine-14C-(U) and ineubation for 24 h. As regards the TS activity in seedlings, maize is comparable to pea and kohlrabi; in contrast to this TS is less active in pea seedlings, which can be attributed to the presence of TS inhibitor (CHEN and BOLL 1969). In ripening maize kernels and leaves adjacent to the ear the TS activity is about 20 times lower than in seedlings. The differences in the activity of TS in the genotypes of maize could not be detected, even at the period of seed ripening. Therefore the differences in L-trp content in the investigated plants cannot be explained by a differing activity of TS. TS is probably not the determining regulator of L-trp level in plants, its activity is relatively high even in plants low in L-trp.

Abstract

The metabolism of14C-anthranilic acid (14C-AA) in kohlrabi (Brassica oleraceaL. var.gongylodesL.) and the effect of radiation gamma60Co on this metabolism was investigated. In hypocotylar segmnents of seven days old etiolated seedlings14C-AA was metabolised par, tially to its detoxication product14C-β-glucoside of AA. Simultaneously L-tryptophan was also formed, which in these plants is a precursor of indolic glucosinolates glucobrassicin and neoglucobrassicin.
