Compounds that act on mitochondrial functions are considered as promising drugs for the treatment of neurodegenerative diseases and age-related dementias. As a basis for the creation of such potential drugs, bioisosteric cinnamic acid analogs and polymethoxybenzene derivatives were selected. Derivatives of cinnamic acid have a wide range of biological activities, which can be important for drugs aimed at the treatment of neurodegenerative diseases, in particular Alzheimer′s disease. In this work, the neuroprotective activity of bioisosteric cinnamic acid analogs and polymethoxybenzene derivatives was studied. Among the compounds studied, lead substances 3, 4, and 7 have been identified. These compounds show no intrinsic toxicity and have a neuroprotective effect on the cellular model of neurodegeneration associated with calcium stress. The mechanism of their cytoprotective activity is probably due to the influence on mitochondrial functions, because these compounds effectively suppress the calcium-induced process of mitochondrial permeability jump. In addition, one of the substances investigated (7) has antioxidant properties, showing the ability to inhibit lipid peroxidation (LPO) of rat brain homogenate, which may be an additional mechanism of neuroprotective effect. The data obtained make it possible to recommend the investigated substances as a basis for the creation of effective neuroprotective drugs capable of influencing early stages of the development of neurodegenerative diseases., Соединения, направленно действующие на митохондриальные функции, рассматриваются как перспективные лекарственные препараты для лечения нейродегенеративных заболеваний и возрастных деменций. В качестве основы для создания таких потенциальных лекарственных средств были выбраны биоизостерные аналоги коричной кислоты и производные полиметоксибензолов. Производные коричной кислоты имеют широкий спектр биологических активностей, который может иметь значение для лекарственных препаратов, направленных на лечение нейродегенеративных заболеваний, в частности болезни Альцгеймера. В данной работе исследована нейропротекторная активность биоизостерных аналогов коричной кислоты и производных полиметоксибензолов. Среди исследованных соединений выявлены вещества-лидеры 3, 4 и 7. Эти соединения не проявляют собственной токсичности и оказывают нейропротекторный эффект на клеточной модели нейродегенерации, связанной с кальциевым стрессом. Механизм их цитопротекторной активности, возможно, обусловлен влиянием на функции митохондрий, поскольку эти соединения эффективно подавляют кальций-индуцированный процесс скачка митохондриальной проницаемости. Кроме того, одно из исследованных веществ (7) обладает антиоксидантными свойствами, проявляя способность к ингибированию перекисного окисления липидов (ПОЛ) гомогената мозга крыс, что может быть дополнительным механизмом нейропротекторного эффекта. The review and summarizes own and literature data about the role of mitochondria as the important target in the search for drugs for the treatment of neurodegenerative diseases. Aging is a major risk factor for sporadic forms of various neurodegenerative diseases, including Alzheimer′s disease. One of the most argued and currently accepted theories is the Mitochondrial Free Radical Theory of Aging. Mitochondrial hypotheses of the development of sporadic forms of neurodegenerative diseases particularly Alzheimer′s disease, are closely connected with it. Impairments of mitochondrial functions lead to a decrease in their ability to regulate calcium homeostasis in the cell and to a decrease in the threshold for the induction of mitochondrial permeability transition (MPT) pores. MPT inhibitors can be considered as a promising approach to the treatment of neurodegenerative diseases, since these drugs can not only exhibit the properties of neuroprotectors, but also can provide normalization of synaptic activity due to increased calcium capacity of mitochondria. The review presents data on the number of MPT inhibitors, including endogenous compounds melatonin and N-acetylserotonin, their bioisosteric analogue Dimebon and a number of other compounds. The use of mitochondria as a basis for the formation of screening strategy for the search for compounds for the treatment of neurodegenerative diseases is of particular interest – both as a test of their potential toxicity, and as a basis for the creation of metabolic stimulants and drugs with neuroprotective and cognitive-stimulating effect., Обобщены собственные и литературные данные, обосновывающие роль митохондрий как важнейшей мишени при поиске препаратов для лечения нейродегенеративных заболеваний. Старение является основным фактором риска спорадических форм различных нейродегенеративных заболеваний, в том числе и болезни Альцгеймера (БА). Одной из наиболее аргументированных и принятых в настоящее время является свободнорадикальная митохондриальная теория старения. Именно с ней тесно связаны и митохондриальные гипотезы развития спорадических форм нейродегенеративных заболеваний и, в частности, БА. Нарушение митохондриальных функций приводит к снижению их способности регулировать гомеостаз кальция в клетке и снижению порога для индукции поры митохондриальной проницаемости (МРТ). Ингибиторы МРТ можно рассматривать как перспективный подход к терапии нейродегенеративных заболеваний, так как эти препараты могут не только проявлять свойства нейропротекторов, но и обеспечивать нормализацию синаптической активности благодаря увеличенной кальциевой ёмкости митохондрий. В обзоре представлены данные о ряде ингибиторов МРТ, включая эндогенные соединения – мелатонин, N-ацетилсеротонин, их биоизостерный аналог димебон и ряд других соединений. Использование митохондрий как основы для формирования скрининговой стратегии поиска соединений для лечения нейродегенеративных заболеваний представляет особый интерес и как тестирование их потенциальной токсичности, и как основа для создания метаболических стимуляторов и препаратов, обладающих нейропротекторным и когнитивно-стимулирующим действием.

The main pharmacokinetic parameters of a new stimulator of cognitive brain functions, OSPL – 502 have been determined: area under the concentration-time curve, elimination rate constant, half-elimination period, time to reach the maximum concentration, maximum concentration, volume distribution, total clearance and bioavailability of the dosage form. The main metabolites of the active substance of the dosage form of the new stimulator of cognitive functions OSPL – 502 have been analyzed. The data obtained predict the effects of the drug in humans relevant for further clinical investigation., Проведенное исследование фармакокинетических параметров лекарственной формы нового стимулятора когнитивных функций мозга OSPL-502 позволило определить основные фармакокинетические параметры (площадь под кривой “концентрация-время”; константа скорости элиминации; период полуэлиминации; время достижения максимальной концентрации; максимальная концентрация; объём распределения; общий клиренс). Определена биодоступность лекарственной формы. Проанализированы основные метаболиты действующего вещества лекарственной формы нового стимулятора когнитивных функций мозга OSPL-502. Полученные данные позволят спрогнозировать действие препарата у человека для дальнейшего клинического исследования.

An the present study we evaluated the effect of natural sesquiterpene lactones of plants of the genus Inula on the cells of neuroblastoma SK-N-MC under conditions of toxic stress induced by H2O2 and glutamate. Lactones increased cell survival under these conditions and the leader compounds were identified in this series. A putative mechanism for the protective action of lactones on cells under condition toxic stress has been proposed., Исследовано действие природных сесквитерпеновых лактонов растений рода Inula на клетки нейробластомы SK-N-MC в условиях токсического стресса, индуцированного H2O2 и глутаматом. Показана способность лактонов повышать выживаемость клеток в этих условиях и выявлены соединения лидеры в этом ряду. Предложен механизм защитного действия лактонов на клетки при индукции токсического стресса.

The results of in vitro studies demonstrate cytotoxic activity alantolactone and isoalantolactone, presented in the elecampane plant (Inula helenium) of the natural sesquiterpene lactones. These compounds show toxicity against human cancer cell lines: MS, A549, HCT116, MCF7, RD and K562. The involvement of the p53 signaling pathway in the death of tumor cells, as well as the contribution of reactive oxygen species (ROS) formation during cell death, was studied.
