Abstract Ultrasound pretreatment was proved to be effective in accelerating waste activated sludge (WAS) disintegration and promoting anaerobic fermentative short-chain fatty acids (SCFA) production. However, due to the high activities of SCFA consumers, SCFA yield is limited. Herein we reported an efficient strategy, i.e., using free ammonia (FA) to aid ultrasound pretreatment, to enhance SCFA accumulation. Experimental results showed that the greatest SCFA yield of 316.7 ± 14.9 mg COD/g VSS was obtained at 2 w/mL ultrasound pretreatment for 15 min followed by initial 60 mg/L FA pretreatment for 2 d, which was 2.2 times (1.7 times) of that from sole ultrasound (FA) pretreatment. The mechanism study revealed that this combination method accelerated sludge solubilization, enhanced the release of biodegradable organics, and caused severe inhibitions to SCFA consumption. Moreover, this method has inhibitory effects on specific activities of hydrolytic microorganisms, SCFA-producing bacteria and methanogens, but the inhibitions to methanogens were severe. This study reported a new low-cost and high-efficient combined method of CaO 2  + free ammonia (FA) pretreatment for sludge anaerobic fermentation. Experimental results showed that the optimal short-chain fatty acids (SCFA) yield of 338.6 mg COD/g VSS was achieved when waste activated sludge (WAS) was pretreated with 0.05 g/g VSS of CaO 2  + 180 mg/L of FA for 3 d, which was 2.5-fold of that from CaO 2 pretreatment and 1.5-fold of that from FA pretreatment. The mechanism investigations exhibited that the CaO 2  + FA could provided more biodegradable substrates, this combination accelerated the disintegration of sludge cells, which thereby providing more organics for subsequent SCFA production. It was also found that the combination of CaO 2 and FA inhibited the specific activities of hydrolytic microbes, SCFA producers, and methanogens to some extents, but its inhibition to methanogens was much severer than that to the other two types of microbes. Abstract It was found in this study that the presence of sophorolipid (SL) enhanced the production of short-chain fatty acid (SCFA) from anaerobic fermentation of waste activated sludge (WAS). Experimental results showed that with an increase of SL addition from 0 to 0.1 g/g TSS, the maximal SCFA yield increased from 50.5 ± 4.9 to 246.2 ± 7.5 mg COD/g VSS. The presence of SL reduced the surface tension between hydrophobic organics and fermentation liquid, which thereby accelerated the disintegration of WAS and improved the biodegradability of the released organics. SL promoted the carbon/nitrogen ratio of the fermentation system, enhancing the conversion of proteins in WAS. Moreover, SL suppressed severely the activities of methanogens, probably due to the drop of pH caused by SL addition. Amplicon sequencing analyses revealed that SL increased the abundance of hydrolytic microbes such as Bacteroides sp. and Macellibacteroides sp., and SCFA producers (e.g., Acinetobacter sp.). Improving the anaerobic treatment performance of waste activated sludge (WAS) to achieve resource recovery is an indispensable requirement to reduce carbon emissions, minimize and stabilize biosolids. In this study, a novel strategy by using urea hydrogen peroxide (UHP) to enhance SCFAs production through accelerating WAS disintegration, degrading recalcitrant substances and alleviating competitive suppression of methanogens. The SCFAs production and acetate proportion rose from 436.9 mg COD/L and 31.3% to 3102.6 mg COD/L and 54.1%, respectively, when UHP grew from 0 to 80 mg/g TSS. Mechanism investigation revealed that OH, O 2 and urea were the major contributors to accelerate WAS disintegration with the sequence of OH> O 2 > urea. Function microbes related to acidification and genes associated with acetate production ([EC:2.3.1.8] and [EC:2.7.2.1]) were upregulated while genes encoding propionic acid production ([EC:6.4.1.3] and [EC:6.2.1.1]) were downregulated. Short-chain fatty acids (SCFA), raw substrates for biodegradable plastic production and preferred carbon source for biological nutrients removal, can be produced from anaerobic fermentation of waste activated sludge (WAS). This paper reports a new, high-efficient and eco-friendly strategy, i.e., using free nitrous acid (FNA) pretreatment combined with Tea saponin (TS), to enhance SCFA production. Experimental results showed 0.90 mg/L FNA pretreatment and 0.05 g/g total suspended solids TS addition (FNA + TS) not only significantly increased SCFA production to 315.3 ± 8.8 mg COD/g VSS (5.52, 1.76 and 1.93 times higher than that from blank, solo FNA and solo TS, respectively) but also shortened fermentation time to 4 days. Mechanism investigations revealed that FNA pretreatment combined with TS cause a positive synergetic effect on sludge solubilization, resulting in more release of organics. In this study, waste activated sludge (WAS) was used as feedstock to generate hydrochars at different temperatures (220 °C and 260 °C) and their effect on sludge anaerobic digestion was evaluated. Experimental results showed that the maximum yield of short-chain fatty acid (SCFA) enhanced by hydrochar (220 °C) and hydrochar (260 °C) were 507.33 and 270.80 mg chemical oxygen demand (COD)/L respectively, which were much higher than that in blank (141.49 mg COD/L). Mechanism investigation confirmed that hydrochar remarkably accelerated the solubilization and hydrolysis of organic matters, enhanced the acidification of hydrolyzed products, and inhibited the activity of methanogenic bacteria as well as promoted the activities of key enzymes. Numerous effective chemical strategies have been explored for short-chain fatty acids (SCFAs) production from waste activated sludge (WAS), but many technologies have been questioned due to the chemical residues. This study proposed a citric acid (CA) treatment strategy for improving SCFAs production from WAS. The optimum SCFAs yield reached 384.4 mg COD/g VSS with 0.08 g CA/g TSS addition. Meanwhile, CA biodegradation occurred and its contribution to the yield of total SCFAs, especially acetic acid, cannot be ignored. Intensive exploration indicated the sludge decomposition, the biodegradability of fermentation substrates, as well as the abundance of fermenting microorganisms were definitely enhanced in the existence of CA. The optimization of SCFAs production techniques based on this study deserved further study. • First study using K 2 FeO 4 as additive to improve microalage anaerobic fermentation. • K 2 FeO 4 addition enhanced HAc-enriched SCFAs production from microalage. • K 2 FeO 4 facilitated microalgae disintegration and organics solubilization. • K 2 FeO4 enriched hydrolytic and SCFA-forming microbes. • K 2 FeO 4 severely inhibited methanogenesis process. Anaerobic fermentation of microalgae was always hindered by its rigid cell wall structure. This paper reports a novel technique, i.e., adding potassium ferrate (K 2 FeO 4) into microalgae fermentation systems to enhance short-chain fatty acids (SCFAs) production. The results showed that the maximum SCFAs production and acetic acid proportion were 732.6 mg COD/g VS and 54.6% at a dosage of 112.8 mg Fe(VI)/g VS, which were 168% and 208% of those in the control, respectively. Mechanism studies revealed that K 2 FeO 4 effectively destroyed surface morphology and cell structure, and thus facilitated microalgae solubilization, providing a large number of biodegradable substrates for subsequent SCFA production. Although K 2 FeO 4 inhibited all the microbial activities relevant to hydrolysis, acidification and methanogenesis processes to some degree, its inhibition to methanogens was much severer than that to other microbes. Deterioration of anaerobic fermentation can occur with the presence of grease in food waste, but little information on eliminating this deterioration is currently available. In this study, it was found that the presence of 10 g/L grease decreased SCFAs production from 16.97 to 13.32 g COD/L and prolonged the optimal fermentation time to 7 days, but could be respectively recovered to 39.10 g COD/L and 4 days with 0.02 mg/g VS (volatile solids) calcium peroxide addition. Mechanism investigations indicated that calcium peroxide facilitated biodegradable organics release and improved grease degradation, thereby providing enough nutrients and better growth environments to microbes for SCFAs-producing, which could be further supported by the elevated enzymes activities responding to hydrolysis and acidification process. Further investigations revealed that among the main derivates of calcium peroxide, OH– and Ca2+ played vital role in SCFAs production promotion, O 2 – and OH radicals were the main contributors to grease degradation.
