In this study, corn stover was first pretreated with formic acid under mild conditions, and more than 70 % of xylan and lignin were removed within the first hour. 173.0 g/L glucose was achieved from total 30 % solid of the pretreated corn stover via fed-batch mode. Moreover, the glucose concentration rose to 194.5 g/L and the hydrolysis time was significantly reduced by 42.9 % with the addition of di-rhamnolipid. On this basis, 89.1 g/L ethanol was obtained by fermentation, and the presence of di-rhamnolipid had no negative effect on fermentation. High-solids enzymatic hydrolysis for biomass has currently received considerable interest. However, the solid effect during the process limits its economic feasibility. This work presented an ordered polyethylene glycol (PEG) pre-incubated strategy for enhancing the auxiliary effect of PEG in a high-solids enzymatic hydrolysis system. The substrate and enzyme were separately pre-incubated with PEG in this strategy. The ordered PEG pre-incubated strategies yielded a maximum glucose concentration of 166.6 g/L from 32 % (w/v) pretreated corncob with an enzymatic yield of 94.1 % by 72 h hydrolysis. Using this method, PEG not only lessened the lignin adsorption to cellulase but also altered particle rheological characteristics in the high-solids enzymatic hydrolysis system as a viscosity modifier. This study offered a new insight into the mechanism behind the PEG synergistic effect and would make it possible to achieve efficient high-solids loading hydrolysis in the commercial manufacture of cellulosic ethanol. Formic acid pretreatment is a promising approach for fractionating biomass, and it has the advantages of efficient recycling and removal of hemicellulose and lignin. Biomass is one of the most plentiful resources on earth, yet its chemical structure differs significantly between woody and herbaceous biomass. The influence of formic acid pretreatment on the fractionation of woody and herbaceous biomasses, as well as changes in physical–chemical properties, was investigated in this study. The results indicated that formic acid is universal in the biorefinery of different biomass, however, herbaceous biomass had greater xylan and lignin removal than woody biomass (especially softwood). Formic acid pretreatment not only considerably improved the enzymatic efficiency of herbaceous biomass, but also had a good effect on the enzymatic efficiency of poplar. Chlorella vulgaris , a marine microalgae strain adaptable to 0–50 g L −1 of salinity, was selected for studying the coupling system of saline wastewater treatment and lipid accumulation. The effect of total nitrogen (TN) concentration was investigated on algal growth, nutrients removal as well as lipid accumulation. The removal efficiencies of TN and total phosphorus (TP) were found to be 92.2–96.6% and over 99%, respectively, after a batch cultivation of 20 days. To illustrate the response of lipid accumulation to nutrients removal, C. vulgaris was further cultivated in the recycling experiment of tidal saline water within the photobioreactor. The lipid accumulation was triggered upon the almost depletion of nitrate (<5 mg L −1 ), till the final highest lipid content of 40%. The nitrogen conversion in the sequence of nitrate, nitrite, and then to ammonium in the effluents was finally integrated with previous discussions on metabolic pathways of algal cell under nitrogen deficiency. The combination of tertiary wastewater treatment and microalgal lipid production is considered to be a promising approach to water eutrophication as well as energy crisis. To intensify wastewater treatment and microalgal biofuel production, the effect of organic and inorganic carbon on algal growth and nutrient removal of Scenedesmus obliquus were examined by varying TOC (total organic carbon) concentrations of 20–120 mg L −1 in wastewater and feeding CO 2 concentrations in the range of 0.03–15%, respectively. The results showed that the maximal biomass and average lipid productivity were 577.6 and 16.7 mg L −1 d −1 with 5% CO 2 aeration. The total nitrogen, total phosphorus and TOC removal efficiencies were 97.8%, 95.6% and 59.1% respectively within 6 days when cultured with real secondary municipal wastewater. • DSA-SCP exhibited a high efficiency for overcoming the recalcitrance of CFS. • Lignin is fatal to the enzymatic hydrolysis of CFS. • Delignification efficiency is blocked by hemicellulose. • A high reliability model was established to predicted enzymatic hydrolysis. The enzymatic digestibility of softwood is hindered for its highly recalcitrant nature to enzymatic attack. In this study, the effects of dilute sulfuric acid pretreatment (DSAP), acidic sodium chlorite pretreatment (SCP), and their combined pretreatments (DSA-SCP and SC-DSAP) on Chinese fir sawdust were investigated, respectively. Results demonstrated that lignin was the most important obstacle, and digestibility increased linearly with lignin removal yield. Furthermore, the results revealed that the order of sequential pretreatment significantly affected the delignification, and hemicellulose should be removed first. Compared to SC-DSAP, DSA-SCP involving the hemicellulose-removal-first strategy exhibited higher delignification efficiency. DSA-SCP caused lignin removal of 92.3% and the enzymatic hydrolysis was high of 97.9%. Finally, a regression model with high reliability was established to quickly evaluate pretreatment process. • A robust Bacillus coagulans CC17A was obtained by adaptive evolution. • CC17A exhibited high tolerance to toxic hydrolysate. • A efficient one-pot process was developed to produced LA from wheat straw. • 9 candidate genes were recommended as the key genes for detoxification in CC17A. In this study, Bacillus coagulans CC17A with highly tolerant to hydrolysate was obtained through adaptive evolution. After 63 generations, the strain CC17A was stably in 45% (v/v) hydrolysate media and could digest multiple inhibitors in the hydrolysate. Based on its promising features, a one-pot process was developed to produce lactic acid (LA) from wheat straw. After dilute acid pretreatment of wheat straw, simultaneous saccharification and co-fermentation was conducted using CC17A without any solid-liquid separation and pre-detoxification. Total 35.50 g LA was produced from 80 g raw substrate and the production yield was as high as 70.9% of theoretical. To elucidate the tolerance mechanism, transcriptomic profiling of CC17A was studied. The highly up-regulated oxidoreductases and phenolic acid decarboxylase are considered to be involved with the inhibitors-tolerance of B. coagulans CC17A. [ABSTRACT FROM AUTHOR]
