Rett syndrome (RTT) is a neurodevelopmental disorder caused by mutations in the MECP2 gene. In the absence of MeCP2, expression of FXYD domain-containing transport regulator 1 (FXYD1) is deregulated in the frontal cortex (FC) of mice and humans. Because Fxyd1 is a membrane protein that controls cell excitability by modulating Na + , K + -ATPase activity (NKA), an excess of Fxyd1 may reduce NKA activity and contribute to the neuronal phenotype of Mecp2 deficient (KO) mice. To determine if Fxyd1 can rescue these RTT deficits, we studied the male progeny of Fxyd1 null males bred to heterozygous Mecp2 female mice. Maximal NKA enzymatic activity was not altered by the loss of MeCP2, but it increased in mice lacking one Fxyd1 allele, suggesting that NKA activity is under Fxyd1 inhibitory control. Deletion of one Fxyd1 allele also prevented the increased extracellular potassium (K + ) accumulation observed in cerebro-cortical neurons from Mecp2 KO animals in response to the NKA inhibitor ouabain, and rescued the loss of dendritic arborization observed in FC neurons of Mecp2 KO mice. These effects were gene-dose dependent, because the absence of Fxyd1 failed to rescue the MeCP2-dependent deficits, and mimicked the effect of MeCP2 deficiency in wild-type animals. These results indicate that excess of Fxyd1 in the absence of MeCP2 results in deregulation of endogenous K + conductances functionally associated with NKA and leads to stunted neuronal growth., (Copyright © 2018 Elsevier B.V. Rett syndrome (RTT) is an X-linked neurodevelopmental disorder caused by mutations in the MECP2. Several genes have been shown to be MECP2 targets. We previously identified FXYD1 (encoding phospholemman; a protein containing the motif phenylalanine-X-tyrosine-aspartate), a gene encoding a transmembrane modulator of the Na, K-ATPase (NKA) enzyme, as one of them. In the absence of MECP2, FXYD1 expression is increased in the frontal cortex (FC) of both RTT patients and Mecp2(Bird) null mice. Here, we show that Fxyd1 mRNA levels are also increased in the FC and hippocampus (HC) of male mice carrying a truncating mutation of the Mecp2 gene (Mecp2(308)). To test the hypothesis that some of the behavioral phenotypes seen in these Mecp2 mutants could be ameliorated by genetically preventing the Fxyd1 response to MECP2 deficiency, we crossed Fxyd1 null male mice with Mecp2(308) heterozygous females and behaviorally tested the adult male offspring. Mecp2(308) mice had impaired HC-dependent novel location recognition, and this impairment was rescued by deletion of both Fxyd1 alleles. No other behavioral or sensorimotor impairments were rescued. The initiation of mammalian puberty requires a sustained increase in pulsatile release of gonadotrophin releasing hormone (GnRH) from the hypothalamus. This increase is brought about by coordinated changes in transsynaptic and glial-neuronal communication, consisting of an increase in neuronal and glial stimulatory inputs to the GnRH neuronal network and the loss of transsynaptic inhibitory influences. GnRH secretion is stimulated by transsynaptic inputs provided by excitatory amino acids (glutamate) and at least one peptide (kisspeptin), and by glial inputs provided by growth factors and small bioactive molecules. The inhibitory input to GnRH neurons is mostly transsynaptic and provided by GABAergic and opiatergic neurons; however, GABA has also been shown to directly excite GnRH neurons. There are many genes involved in the control of these cellular networks, and hence in the control of the pubertal process as a whole. Our laboratory has proposed the concept that these genes are arranged in overlapping networks internally organized in a hierarchical fashion. According to this concept, the highest level of intra-network control is provided by transcriptional regulators that, by directing expression of key subordinate genes, impose genetic coordination to the neuronal and glial subsets involved in initiating the pubertal process. More recently, we have begun to explore the concept that a more dynamic and encompassing level of integrative coordination is provided by epigenetic mechanisms., (Copyright © 2010 Elsevier B.V. Gamma aminobutyric acid (GABA), the dominant inhibitory neurotransmitter in brain, is involved in the developmental regulation of LHRH secretion. Morphological studies in rodents have demonstrated that LHRH neurons are innervated by GABA-containing processes, suggesting that LHRH secretion is under direct transsynaptic GABAergic control. While GABA acts through two different receptors, GABAA and GABAB, to exert its effects, it appears that GABAA receptors are able to mediate both inhibitory and stimulatory effects of GABA on LHRH neurons. GABAA receptors are heterooligomeric ligand-gated anion channels that exhibit a diverse array of functional and pharmacological properties. This diversity is determined by the structural heterogeneity of the receptors, which are assembled from the combination of different classes of subunits with multiple isoforms. Although several studies have described the effect of GABAA receptor stimulation on LHRH and/or gonadotropin release in prepubertal animals, nothing is known about the receptor subunits that may be expressed in LHRH neurons at this phase in development. Double immunohistofluorescence followed by confocal laser microscopy revealed that subsets of prepubertal LHRH neurons are endowed with alpha 1, alpha 2, beta 2/3, and gamma 2 GABAA receptor subunits. Combined immunohistochemistry for LHRH neurons and in situ hybridization for GABAA subunit mRNAs confirmed that the genes encoding the alpha 1, alpha 2, beta 3 and gamma 2 subunits, but not the gamma 1 subunit, are expressed in LHRH neurons. Notwithstanding the relative insensitivity of these methods, both the immunohistochemical and hybridization histochemical approaches employed indicate that only a fraction of LHRH neurons are endowed with GABAA receptors. This arrangement suggests that those LHRH neurons bearing the appropriate GABAA receptors are responsible for either the entire secretory response to direct GABAergic inputs or for its initiation.

Although the synthesis of nerve growth factor (NGF) in brain regions innervated by magnocellular cholinergic neurons of the basal forebrain is well documented, the cell type(s) able to produce NGF in the central nervous system (CNS) remain only partially characterized. Moreover, little is known regarding the ability of brain areas not innervated by magnocellular cholinergic neurons to express NGF protein. The hypothalamus, which controls the endocrine system, is one of such regions. Primary culture of mixed populations of cells from the fetal hypothalamus were used to identify the presence of NGF in this brain area. Immunocytochemistry revealed that hypothalamic oligodendrocytes and a subpopulation of neurons expressed the NGF protein. In contrast, astrocytes were either immunonegative or equivocally stained. To define whether synthesis of NGF is restricted to a particular cell type, cultures of purified astrocytes, oligodendrocyte progenitor (oligoP) cells and neurons were utilized. They were obtained from the neonatal cerebral cortex to ensure an adequate yield of glial cells. Virtually the entire population of cerebral oligoP cells were found to express NGF protein. In contrast, and similar to hypothalamic astrocytes, cerebral type I astrocytes isolated at the same time as oligoP cells exhibited little or no NGF staining. When type I astrocytes were induced to differentiate in the presence of a serumless, chemically defined medium, a subpopulation of the culture became more robustly positive for the NGF protein. Contrasting with these differences in NGF immunoreactivity, Northern analysis of RNA isolated from purified cerebral type I astrocytes, oligoP cells and neurons demonstrated that NGF mRNA was expressed in each of these cell types at approximately the same levels. The results indicate that: (a) when placed in culture, each of the major cell types within the CNS has the capability of transcribing the NGF gene, and (b) despite similar NGF mRNA levels the cellular content of NGF protein is greater in a subpopulation of neurons and in oligodendrocytes than in astrocytes, suggesting differences in NGF post-transcriptional regulation between these cell types. In addition, the presence of NGF in hypothalamic cells suggests that NGF may be involved in the regulation of specific hypothalamic neuronal systems.

Prostaglandin E2 (PGE2) has been implicated as a mediator of norepinephrine-induced luteinizing hormone-releasing hormone (LHRH) release from the hypothalamus. The present experiments were undertaken to examine the hypothesis that mobilization of Ca2+ from intracellular stores and cyclic AMP (cAMP) formation are involved in this effect. Incubation of median eminence (ME) nerve terminals from juvenile rats in Ca2+-free Krebs-Ringer bicarbonate buffer reduced, but failed to prevent the stimulatory effect of PGE2 on LHRH release. None of 5 calmodulin antagonists or a blocker of calmodulin-dependent kinase affected the LHRH response to PGE2. In contrast, inhibition of intracellular Ca2+ mobilization with TMB-8 or dantrolene in Ca2+-free medium prevented the LHRH releasing effect of PGE2. Similarly to PGE2, the stimulatory effect of the Ca2+ ionophore A 23187 on LHRH release was not affected by inhibition of calmodulin activity. This, however, blocked the increase in PGE2 formation induced by the ionophore. PGE2 evoked a dose-related increase in cAMP accumulation in Ca2+-containing medium and this effect was inhibited both by blockers of intracellular Ca2+ mobilization and by calmodulin antagonists. Surprisingly, removal of extracellular Ca2+ increased basal cAMP levels in the incubation medium without affecting LHRH release; PGE2 induced a further increase in cAMP which was prevented by inhibition of intracellular Ca2+ translocation. Stimulation of adenylate cyclase activity with forskolin (F) resulted in similar increases in cAMP levels both in the presence and absence of extracellular Ca2+. However, F failed to evoke LHRH release in Ca2+-free medium. The results indicate that: (a) the stimulatory effect of PGE2 on LHRH release involves mobilization of intracellular Ca2+ but not the participation of calmodulin; (b) the formation of PGE2 itself is calmodulin-dependent; (c) PGE2 stimulates cAMP formation through a calmodulin-dependent mechanism that requires translocation of intracellular (membrane?) Ca2+; (d) the cAMP system of a population of nerve terminals different from that responsive to PGE2 is normally subjected to a Ca2+-dependent inhibitory control; and (e) although PGE2 is a potent stimulator of cAMP formation in the ME and endogenously produced cAMP can induce LHRH release, in the absence of extracellular Ca2+ PGE2 stimulates LHRH release in a cAMP-independent manner.
