The visual system of teleost fish shows growth and regeneration capacities during the entire animal's life. Thus, the visual system of adult fish serves as a model for studying neurogenesis in the vertebrate central nervous system (CNS). Our study focused on the expression pattern of Sox2 in the fish visual system. Sox2 is a transcription factor known for its function in keeping stem cell properties, and as a regulator of cell fate during development, especially in the visual system. We used two different fish species: Astatotilapia burtoni and Danio rerio. In the visual system of fish, we identified Sox2 positive cells in the stem cell niche in the peripheral retina, in Müller cells and amacrine cells in the differentiated retina, and glial cells in the optic nerve (ON). We did not observe hardly any Sox2 expression in the optic nerve head (ONH). In the ON, Sox2 positive glial cells were lining the fascicles of new axons. Taking together, the broad spectrum of Sox2 expression indicates that this protein has different functions in the CNS of adult vertebrates. The results suggest that Sox2 has functions associated with the pathway of new axons from the retina. To understand the variety of cell types and subtypes and their plasticity potential in the visual system of fish will be essential to comprehend the growing and regenerating CNS in adult vertebrates., (Copyright © 2019 Elsevier B.V. Purpose: To determine whether transplantation of olfactory mucosal cells (OMCs) is able to rescue the loss of optic nerve axons after the intraocular pressure (IOP) is elevated in rats., Methods: The IOP was raised by injection of magnetic microspheres into the anterior chamber of the eye. OMCs cultured from the adult olfactory mucosa were transplanted into the region of the optic disc., Results: We demonstrated that although the raised IOP returned to its normal level at six weeks, there was an irreversible 58% loss of optic nerve axons in the control group. However, the loss of the axons was reduced to 23% in the group with the transplanted OMCs. The Pattern Electroretinograms (pERG) showed that the decrement of the voltage amplitudes in association with the raised IOP was significantly alleviated in the group with transplantation of OMC., Conclusions: Transplantation of OMCs is able to rescue loss of optic nerve axons induced by raised IOP in the rats. The pERG recording suggested that the functional activities of the axons are also protected., Translational Relevance: The results demonstrated the ability of the transplanted OMCs to protect against the loss of the optic nerve axons and the loss of function caused by raised IOPs. The findings provide a basis for future human clinical trials by autografting OMCs from autologous nasal epithelial biopsies to treat or delay glaucoma diseases., (Copyright © 2019 Elsevier B.V. In vertebrates from fishes to mammals, optic nerve injury induces increased expression ofSuppressor of Cytokine Signaling 3(SOCS3) mRNA, a modulator of cytokine signaling that is known to inhibit CNS axon regeneration. Unlike amniotes, however, anamniotes successfully regenerate optic axons, despite this increase. To address this seeming paradox, we examined the SOCS3 response to optic nerve injury in the frog,Xenopus laevis, at both the mRNA and protein levels. Far from being only transiently induced, SOCS3 mRNA expression increased throughout regeneration in retinal ganglion cells, but immunostaining and Western blots indicated that this increase was reflected at the protein level in regenerating optic axons but not in ganglion cell bodies. Polysome profiling provided additional evidence that SOCS3 protein levels were regulated post-translationally by demonstrating that the mRNA was efficiently translated in the injured eye. In tumor cells, another member of theSOCS gene family,SOCS2, is known to mediate SOCS3 degradation by targeting it for proteasomal degradation. Unlike the SOCS2 response in mammalian optic nerve injury, SOCS2 expression increased inXenopusretinal ganglion cells after injury, at both the mRNA and protein levels; it was, however, largely absent from both uninjured and regenerating optic axons. We propose a similar degradation mechanism may be spatially restricted inXenopusto keep SOCS3 protein levels sufficiently in check within ganglion cell bodies, where SOCS3 would otherwise inhibit transcription of genes needed for regeneration, but allow them to rise within the axons, where SOCS3 has pro-regenerative effects., (Copyright © 2018 Elsevier B.V. Diabetic retinopathy is the leading cause of acquired blindness in working-age individuals. Recent work has revealed that neurodegeneration occurs earlier than vascular insult and that distal optic nerve damage precedes retinal degeneration and vascular insult. Since we have shown that optic nerve degeneration is reduced after optic nerve crush in Galectin-3 knockout (Gal-3 -/-) mice, we decided to investigate whether Gal-3 -/- could relieve inflammation and preserve both neurons and the structure of the retina and optic nerve following 8 weeks of diabetes. Diabetes was induced in 2-month-old male C57/bl6 WT or Gal-3 -/- mice by a single injection of streptozotocin (160 mg/kg). Histomorphometric retinal analyses showed no gross difference, except for a reduced number of retinal ganglion cells in WT diabetic mice, correlated to increased apoptosis. In the optic nerve, Gal-3 -/- mice showed reduced neuroinflammation, suggested by the smaller number of Iba1+ cells, particularly the amoeboid profiles in the distal end. Furthermore, iNOS staining was reduced in the optic nerves of Gal-3 -/- mice, as well as GFAP in the distal segment of the optic nerve. Finally, optic nerve histomorphometric analyses revealed that the number of myelinated fibers was higher in the Gal-3 -/- mice and myelin was more rectilinear compared to WT diabetic mice. Therefore, the present study provided evidence that Gal-3 is a central target that stimulates neuroinflammation and impairs neurological outcomes in visual complications of diabetes. Iron deficiency has a critical impact on maturational mechanisms of the brain and the damage related to neuroanatomical parameters is not satisfactorily reversed after iron replacement. However, emerging evidence suggest that enriched early experience may offer great therapeutic efficacy in cases of nutritional disorders postnatally, since the brain is remarkably responsive to its interaction with the environment. Given the fact that tactile stimulation (TS) treatment has been previously shown to be an effective therapeutic approach and with potential application to humans, here we ask whether exposure to TS treatment, from postnatal day (P) 1 to P32 for 3min/day, could also be employed to prevent neuroanatomical changes in the optic nerve of rats maintained on an iron-deficient diet during brain development. We found that iron deficiency changed astrocyte, oligodendrocyte, damaged fiber, and myelinated fiber density, however, TS reversed the iron-deficiency-induced alteration in oligodendrocyte, damaged fiber and myelinated fiber density, but failed to reverse astrocyte density. Our results suggest that early iron deficiency may act by disrupting the timing of key steps in visual system development thereby modifying the normal progression of optic nerve maturation. However, optic nerve development is sensitive to enriching experiences, and in the current study we show that this sensitivity can be used to prevent damage from postnatal iron deficiency during the critical period., (Copyright © 2016 Elsevier B.V. The transcription factor Pax2 actively participates in the development of the vertebrate visual system. In adults, Pax2 expression persists in a subpopulation of Müller cells and/or astrocytes in the retina and optic nerve head (ONH), although its function remains elusive. In a previous work we showed that the pax2 gene expression is modified and the Pax2(+) astrocyte population in the ONH strongly reacted during the regeneration of the retina after a lesion in goldfish. In the present work we have analyzed Pax2 expression in the goldfish ONH after optic nerve (ON) crush. At one week post-injury, when the regenerating axons arrive at the ONH, the pax2 gene expression level increases as well as the number of Pax2(+) astrocytes in this region. These Pax2(+) astrocytes show a higher number of Cytokeratin (Ck)(+)/GFAP(+) processes compared with control animals. In contrast, a different S100(+) astrocyte population is not modified and persists similar to that of controls. Furthermore, we find a ring that surrounds the posterior ONH that is formed by highly reactive astrocytes, positive to Pax2, GFAP, Ck, S100, GS and ZO1. In this region we also find a source of new astrocytes Pax2(+)/PCNA(+) that is activated after the injury. We conclude that Pax2(+) astrocytes constitute a subpopulation of ONH astrocytes that strongly reacts after ON crush and supports our previous results obtained after retina regeneration. Altogether, this suggests that pax2 gene expression and Pax2(+) astrocytes are probably directly involved in the process of axonal regeneration., (Copyright © 2012 Elsevier B.V. Mammals do not regenerate axons in their central nervous system (CNS) spontaneously. This phenomenon is the cause of numerous medical conditions after damage to nerve fibers in the CNS of humans. The study of the mechanisms of nerve regeneration in other vertebrate animals able to spontaneously regenerate axons in their CNS is essential for understanding nerve regeneration from a scientific point of view, and for developing therapeutic approaches to enhance nerve regeneration in the CNS of humans. RICH proteins are a novel group of proteins implicated in nerve regeneration in the CNS of teleost fish, yet their mechanisms of action are not well understood. A number of mutant versions of the zebrafish RICH (zRICH) protein were generated and characterized at biochemical and cellular levels in our laboratory. With the aim of understanding the effects of RICH proteins in neuronal axon outgrowth, stable transfectants derived from the neuronal model PC12 cell line expressing zRICH Wild-Type or mutant versions of zRICH were studied. Results from differentiation experiments suggest that RICH proteins enhance neuronal plasticity by facilitating neurite branching. Biochemical co-purification results have demonstrated that zRICH binds to the cytoskeletal protein tubulin. The central domain of the protein is sufficient for tubulin binding, but a mutant version of the protein lacking the terminal domains, which cannot bind to the plasma membrane, was not able to enhance neurite branching. We examined the histologic findings of optic nerve axons and changes in kinesin-1, which is involved in axonal flow, in N-methyl-d-aspartate (NMDA)-induced neurotoxicity in rats. Substantial degenerative changes visualized as black profiles and pale large axons were observed 72h after NMDA injection, but those degenerative changes were not apparent in axons 12 and 24h after injection. Morphometric analysis showed a significant, approximately 40% reduction in the number of axons 72h after NMDA injection. Immunohistochemical study showed that there was a recognizable loss of neurofilament-immunopositive dots, but myelin basic protein immunostaining was unchanged 72h after NMDA injection. Western blot analysis showed early elevation of kinesin-1 (KIF5B) protein levels in the retina 24 and 72h after NMDA injection. Conversely, significant decreases in KIF5B protein levels in the optic nerve were seen during the same time course. Immunohistochemical study also showed that there was a reduction in KIF5B immunoreactivity in axons, but neurofilament immunostaining was unchanged 24h after NMDA injection. These findings suggest that the intravitreal injection of NMDA causes neurofilament loss without myelin alteration in the early stage. Multiple sclerosis (MS) patients may suffer from optic disturbances. Toxin-induced demyelinations have frequently been developed to investigate the cellular and structural aspects of demyelination and remyelination processes, separately. The present study describes functional consequence of lysolecithin (LPC)-induced lesion in the adult rat optic nerves and chiasm by recording the visual evoked potentials (VEPs) from the visual cortex and its correlation with myelin basic protein (MBP) expression in lesion site. Records of VEP were obtained at 2, 7, 14 and 28 days post-injection. We observed that the VEPs generated by light stimuli progressively changed in both amplitude and latency after the lesion as well as in comparison with those generated in control animals. These observations were confirmed through measurement of mRNA expression level for MBP which is one of the important genes expressed in mature oligodendrocytes and Schwann cells. The level of MBP mRNAs in demyelinated chiasm and optic nerves decreased following lysolecithin injection with its least value on day 7, and then it increased to the control level 14 days post-lesion. However, it continued to increase even after that and reached a maximum level 28 days post lesion. Results of the present paper show that, LPC injection in the chiasm share functional and molecular alterations which are found in demyelinating disorders in both the optic nerves and chiasm and also these alterations were coming back to level of control animal on 28 days post lesion, which is typically seen in myelin repair process. The present paper provides new insights into the experimental toxin-induced models that may be useful for evaluating the functional recovery of demyelinated optic nerves and chiasm following various repairing strategies. It also seems to be useful for studying the protective or remyelinating effects of different therapies in e.g. optic apparatus which is more affected by MS., (2010 Elsevier B.V. Protocadherin-19 has been implicated in some neurological diseases, but even the basic properties of this protocadherin have not yet been characterized well. Hence, various basic properties of chicken protocadherin-19 were examined to elucidate its biological role. The protocadherin-19 expressed in L cells was localized at the intercellular contact sites and showed Ca(2+)-dependent homophilic cell aggregation activity that was relatively weak but showed stringent specificity. The results of a pull-down assay using fusion proteins of the cytoplasmic domain and glutathione S-transferase yielded specifically bound proteins. In the bound fractions, liquid chromatography-mass spectrometry identified Nck-associated protein 1 and cytoplasmic FMP1 interacting protein 2, which have been reported to bind to glutathione S-transferase fused with the cytoplasmic domain of OL-protocadherin, suggesting that these proteins generally have affinity for delta protocadherins. Protocadherin-19 was mainly expressed in the central nervous system. In the chicken retina, protocadherin-19 was expressed as early as embryonic day 5 and was localized in the ganglion cell layer, inner plexiform layer, and optic nerve layer. Chicken protocadherin-19 was co-localized with syntaxin 1 in inner plexiform layer and was also expressed in the optic nerve and in specific layers of optic tectum. These results suggest that protocadherin-19 plays a role as an adhesion protein in optic nerve fiber bundling, optic nerve targeting, and/or synapse formation., (Crown Copyright (c) 2010. A centrifugal visual system showing FMRF-amide-like immunoreactivity has been demonstrated in Lampetra fluviatilis by using immunocytochemical and hodological techniques. From 50 to 60 immunoreactive neurons, labelled after contralateral intraocular injection of rhodamine beta-isothiocyanate, form a small, clearly defined, nucleus in the lateral neural plate of the magnocellular preoptic nucleus. These cells give rise to immunoreactive axons which have been observed at the base of the nucleus, in the optic chiasma and in the optic nerve, to project into the intermediate plexiform layer of the retina, which separates the layer of internal horizontal cells from the layer of external horizontal cells. This FMRF-amide-like immunoreactive centrifugal visual system is compared to that described in Gnathostomes.

Astrocytes maintain an intimate relationship with central nervous system (CNS) neurons and play a crucial role in regulating their biochemical environment. A rise in neural tissue pressure in the CNS is known to lead to axonal degeneration however the response of astrocytes during the early stages of neural injury has not been studied in great detail. The optic nerve is a readily accessible model in which to study CNS axonal injury. Previous work from our laboratory has shown that an acute increase in intraocular pressure (IOP) results in axonal cytoskeleton changes and axonal transport retardation within the optic nerve head. Axonal changes occurred in a time-dependent manner with the magnitude of change being proportional to the duration of the IOP rise. Using glial fibrillary acidic protein (GFAP) as a marker of astrocytes we have now studied pressure induced changes in astrocyte structure in the optic nerve head. Using confocal microscopy we found that an increase in IOP resulted in morphological changes in the astrocytes that were consistent with previous reports of swelling. In addition there was also a decrease in GFAP intensity within these astrocytes. These changes occurred in a time-dependent manner with the chronology of change coinciding with that of axonal change. There was no evidence of apoptosis in regions where astrocyte changes were found. The present results provide evidence that in the early stages of neural tissue pressure rise there are both astrocyte and axonal injury.

The development of glial precursor cells in the mammalian optic nerve depends on retinal ganglion cell (RGC) axons, but the signals that mediate this neuron-to-glia interaction have not been fully characterized. Sonic hedgehog (Shh) is expressed by RGCs, and we showed previously that it is required for the specification of astrocyte lineage cells at the optic disc. To study the role of RGC-derived Shh on astrocyte development at later developmental stages, we generated mice with a conditional ablation of Shh in the peripheral retina and analyzed gene expression and glial cell development in the optic nerve. Astrocyte development was initiated in the optic nerves of these mutant mice; however, the expression of Hedgehog (Hh) target genes, Gli1 and Ptch1 and cell cycle genes, Ccnd1 and Cdc25b in the optic nerves were downregulated. Astrocyte proliferation was markedly reduced. Oligodendrocyte precursor cells were fewer in the optic nerves of mutant mice, possibly as a consequence of reduced secretion of growth factors by astrocytes. At a later developmental stage, optic nerve axons displayed signs of Wallerian degeneration, including reduction of astrocyte processes, degenerating glial cells and formation of distended axons. We also demonstrate that the Hh pathway can be activated in optic nerve-derived astrocytes in vitro, but fails to induce cell cycle gene expression and proliferation. RGC-derived Shh signalling isthus necessary in vivo for maintenance of astrocyte proliferation, affecting both axo-glial and normal glial cell development in the optic nerve.

In spite of the numerous reports on the optic fiber distribution in the optic nerve and tract of vertebrates, there have been few studies of the visual pathway in reptiles. The arrangement of fibers in the optic nerve and tract of the turtle Mauremys leprosa was studied by placing a small granule of carbocyanine dye (DiI or DiA) in one of the four quadrants of the retina. The labeled fibers were traced through transverse sections of the retinofugal pathway with confocal microscopy. Retinal axons displayed a quadrant-specific order along the optic nerve. However, retinal ganglion cell axons were re-organized as they passed through the chiasmatic region of the optic pathway. In the optic tract, the nasal and temporal fibers remained intermingled, but there was segregation of dorsal from ventral fibers. This re-ordering is similar to that described in other vertebrates, suggesting the existence of an evolutionarily conserved mechanism.

This work represents a comprehensive pathological description of the retina and visual pathways in naturally affected Scrapie sheep. Twenty naturally affected Scrapie sheep and 6 matched controls were used. Eyes, optic nerves and brain from each animal were fixed and histologically processed using hematoxylin-eosin, followed by immunohistochemical staining for prion protein (PrPsc) and glial fibrillar acidic protein (GFAP). Retinal histopathological changes were observed in only 7 clinically affected animals and mainly consisted of loss of outer limitant layer definition, outer plexiform layer atrophy, disorganization and loss of nuclei in both nuclear layers, and Müller glia hypertrophy. PrPsc was detected in the retina of 19 of the 20 sheep and characterized by a disseminated granular deposit across layers and intraneuronally in ganglion cells. The inner plexiform and the ganglion cell layers were the structures most severely affected by PrPsc deposits. PrPsc exhibited a tendency to spread from these two layers to the others. A marked increase in the number and intensity of GFAP-expressing Müller cells was observed in the clinical stage, especially at the terminal stage of the disease. Spongiosis and PrPsc were detected within the visual pathways at the preclinical stage, their values increasing during the course of the disease but varying between the areas examined. PrPsc was detected in only 3 optic nerves. The results suggest that the presence of PrPsc in the retina correlates with disease progression during the preclinical and clinical stages, perhaps using the inner plexiform layer as a first entry site and diffusing from the brain using a centrifugal model.

The purpose of this study is to evaluate the changes in neurofilament light (NF-L) protein in the optic nerve in rat kainate and monkey ocular hypertension models. In the rat model, optic nerve damage was induced by kainate injection into the vitreous body. In the monkey model, photocoagulation of the trabecular meshwork led to laser-induced ocular hypertension. NF-L in optic nerve extract was quantified by quantitative immunoblot using an imaging analyzer. The amount of NF-L in optic nerve was compared between normal and kainate-treated groups at 7 days after kainate injection. Specimens from rat optic nerves and retinas were evaluated histologically to examine the correlations between damage and amount of NF-L in the optic nerve. In monkeys, the amount of NF-L in the optic nerve was compared between control fellow and ocular hypertensive eyes. Injection of kainate induced morphological optic nerve and retinal damage in rats. The amount of NF-L in the optic nerve was significantly reduced in kainate-treated eyes (vs. normal eyes). The amount of NF-L correlated with the cell count in the ganglion cell layer and the axon number in the optic nerve at 7 days after kainate injection. Further, 6-cyano-7-nitro-quinoxaline-2,3-dione, a non N-methyl-D-aspartate receptor antagonist, suppressed the kainate-induced reduction in NF-L and retinal damage. In the monkey model, ocular hypertension and morphological optic nerve damage were shown by laser-treated eyes. The amount of NF-L in the optic nerve was reduced in laser-treated eyes. In conclusion, in rat kainate and monkey ocular hypertension models, immunoblot evaluation of NF-L shows the reduction of the amount of NF-L in the optic nerves of treated-eyes. The amount of NF-L correlated with the morphological retinal and optic nerve damage in rats. These findings indicate that immunoblot evaluation of NF-L in the optic nerve may provide a quantitative index of optic nerve damage.

Purpose: To study the histology and the physiological function of the retina in the neurological myelin mutant, taiep rats during the postnatal developmental period (P20-P360)., Methods: Electroretinography (ERG) was applied to evaluate intensity dependence and spectral sensitivity of the responses to light. Retinal histology, morphometry, and immunocytochemistry were used to characterize the structure of the retina, with particular emphasis on the Müller (glial) cells., Results: In the taiep rats of all ages studied, the scotopic ERG showed normal a- and b-wave amplitudes and latencies; likewise, the scotopic spectral sensitivity function was the same for control and taiep animals, with a maximal sensitivity (lambda(max)) at 500 nm. However, in adult taiep rats (P90 to P360) a secondary cornea-positive wave ('b(2)') was observed in response to high stimulus intensities, which never occurred in controls. This correlated with the observation that in the photopic ERG responses of the taiep rats, the b-wave was reduced in amplitude, and was followed by a rapid cornea-negative after-potential. After 1 year of life, in taiep rats the outer plexiform layer (OPL) became slightly thinner and the inner plexiform/ganglion cell layers (IPL/GCL) appeared to be swollen, and increased in thickness; in addition, the number of retinal neurons (particularly, of photoreceptor cells) slightly decreased. Increased GFAP immunoreactivity revealed a hypertrophy and reactivity of the Müller cells in 1-year-old taiep rats., Conclusions: The present results suggest the occurrence of a relatively mild and slowly progressing neural retinal alteration in taiep rats, which becomes histologically and functionally evident at the end of the first year of life, and mainly affects the circuit(s) of the photopic ON-response. It is speculated that this alteration is due to missing/altered signals from demyelinated optic nerve.

Gonadotropin-releasing hormone (GnRH) projections from the terminal nerve to the retina are common in fish, but have not been reported in mammals. However, GnRH fibers have been seen previously in the optic nerves (but not retinas) of rats and monkeys. Using prairie voles, we tested the hypotheses that (1) GnRH-immunoreactive (-ir) neurons project into the optic nerve and (2) the retina expresses GnRH receptor mRNA as determined by reverse transcription-polymerase chain reaction (RT-PCR) combined with Southern blotting. In both adult and postnatal-day-2 voles, GnRH-ir fibers were observed within the optic nerve. In adult voles, GnRH-ir fibers projected only a short distance into the optic nerve compared with the much longer length of projections in neonates. Fibers immunoreactive for GnRH were not seen in the retinas of neonates or adults. However, RT-PCR-Southern blotting demonstrated GnRH receptor expression in the retina of adult voles. This study supports the hypothesis that GnRH has the potential of modulating visual processing in the retina of mammals.

We have investigated the localisation of endothelin-1 (ET-1) mRNA and ET-1-like immunoreactivity in retina and anterior portion of optic nerve from human and porcine eyes. In situ hybridisation method revealed expression of ET-1 mRNA mainly in the innermost layers of the retinas, in the retinal pigment epithelium cells as well as in the astrocytes of the optic nerve. Immunohistochemical studies showed that ET-1-like immunoreactivity appeared in the same regions where ET-1 mRNA was expressed as well as in the inner nuclear layer and in the inner segments of photoreceptors. In the nerve fibre and ganglion cell layers, astrocytes expressed both glial fibrillary acidic protein and ET-1 proteins suggesting that these cells may secrete ET-1. Expression of ETA and ETB receptors in human retina were demonstrated by reverse transcription-polymerase chain reaction. Our results demonstrated expression of ET-1 in glial, neural and vascular components of retina and optic nerve from human and porcine eyes.

We have analyzed the immunolabeling with the antibody RT97, a good marker for ganglion cell axons in several species, in the normal and regenerating visual pathways of teleosts. We have demonstrated that RT97 antibody recognizes several proteins in the tench visual system tissues (105, 115, 160, 200, 325 and 335 kDa approximately). By using immunoprecipitation and Western blot we have found that after crushing the optic nerve the immunoreactivity to anti RT97 increased markedly in the optic nerve. In immunohistochemical analysis we also found a different pattern of labeling in normal and regenerating visual pathways. In normal tench RT97 is a good marker for the horizontal cells in the retina, for growing ganglion cell axons which run along the optic nerve from the retina to the optic tectum and of the axon terminals in the stratum opticum and stratum fibrosum and griseum superficiale in the optic tectum. After optic nerve crush, no immunohistochemistry modifications were observed in the retina. However, in accordance with Western blot experiments, in the optic nerve intensely stained groups of regenerating axons appeared progressively throughout the optic nerve as far as the optic tectum. We conclude that the antibody RT97 is an excellent marker of growing and regenerating axons of the optic nerve of fish.

Since calcium activated neutral proteinase (calpain) is present in the central nervous system (CNS) and degrades myelin proteins, this endopeptidase has been suggested to play a role in myelin destruction in demyelinating diseases such as multiple sclerosis (MS). In the present study, calpain immunocytochemical expression was examined in Lewis rats with acute experimental allergic encephalomyelitis (EAE), an animal model for MS and optic neuritis. To identify cells expressing calpain, we labeled rat optic nerve sections for calpain with a polyclonal myelin calpain antibody and with monoclonal antibodies for glial (GFAP, OX42) and inflammatory (CD2, ED2, ED1, IFN-gamma) cell-specific markers. The results showed increased calpain expression in microglia (OX42) and infiltrating macrophages (ED1,2) in EAE compared to normal controls. Astrocytes constitutively expressed calpain in controls and acute EAE. Reactive astrocytes in EAE located in or near inflammatory foci, exhibited markedly increased calpain expression. Most T cells in acute EAE showed low level calpain expression while activated IFN-gamma-producing lymphocytes in inflammatory foci exhibited elevated levels of calpain expression. Thus, our results demonstrate increased calpain expression (at transcriptional and/or translational levels) in a rat model of optic neuritis. A role for calpain in myelin destruction during optic neuritis may be relevant to the pathogenesis of this disorder., (Copyright 1998 Elsevier Science B.V.)

GABA transporters (GATs) play a critical role in the translemmal transport of GABA in neurons and glial cells. Two major brain GATs, GAT-1 and GAT-3, are found in astrocytes in the adult brain. Astroglia demonstrate morphological and molecular changes in response to brain injury and deafferentation. The present study was designed to determine whether the expression of GATs changes after nerve deafferentation using the rat superior colliculus (SC) as a model. The immunoreactivity for GAT-1 and GAT-3, as well as GABA and glutamic acid decarboxylase (GAD)-65 and GAD-67, was studied in the SC of control rats and rats with unilateral optic nerve transections. Immunolabeling for both GAT-1 and GAT-3 was increased in the neuropil of the denervated SC as compared to that for the SC of control rats or for the unaffected SC of experimental rats. In contrast, immunoreactivity for GABA, GAD-65 and GAD-67 was not altered. The change in the immunolabeling of GAT-1 and GAT-3 was detectable at 1 day postlesion and became more evident between 3-30 days postlesion. At the electron microscopic level, immunoreactivity for both GAT-1 and GAT-3 in the unaffected SC was localized to astrocytic processes, whereas GAT-1 immunolabeling was also present in synaptic terminals. In the deafferented SC, immunolabeling for both GATs was elevated in the somata and processes of hypertrophied astrocytes as compared to that in the unaffected SC, whereas GAT-1 labeling in neuronal profiles was largely unchanged. A substantial increase of GAT-1 and GAT-3 in astrocytes following optic nerve transection suggests that these cells play a role in modulating GABA's action in the deafferented SC.

We have recently demonstrated that the formation of myelin sheaths in the optic nerve of young postnatal mice deficient in the myelin-associated glycoprotein (MAG) is retarded when compared to age-matched wild-type mice. In the present study, we analyzed whether impaired myelination of retinal ganglion cell axons is detectable in adult MAG mutants. In optic nerves of 2- and 9-month-old MAG-deficient mice, we observed a significantly increased number of unmyelinated axons compared to age-matched wild-type mice. At both ages, unmyelinated axons in optic nerves of MAG mutants were of small caliber. The number of unmyelinated axons decreased significantly in 9-month-old MAG mutants when compared to 2-month-old MAG mutants, indicative of a slow and long-lasting myelination of axons in the mutant. Our observations support the view that MAG is involved in the initiation of myelination in the CNS.

The localizations of taurine, gamma-aminobutyric acid (GABA) and glial fibrillary acidic protein (GFAP) within the developing rat optic nerve were determined using immunocytochemical techniques on tissues from animals ranging in age from embryonic day 20 to postnatal 28 days. Mature nerves from 3-4-month-old adults were also examined. At the younger ages, taurine immunoreactivity was intense and localized specifically to the optic nerve axons, but by postnatal day 15 and thereafter its predominant localization was in macroglia. Some of these glia were astrocytes as indicated by the specific marker, GFAP. GABA immunoreactivity was present at the same time as taurine but was found only in macroglia. In mature nerves the patterns of taurine, GABA and GFAP distribution (within glia) were highly similar.

A novel CNS specific keratan sulfate proteoglycan is identified by a monoclonal antibody (mAb) TED15. The antibody recognizes a carbohydrate epitope that is removed from the core protein following keratanase digestion. The relative molecular weight of the glycoprotein is not altered following digestion with heparinase, heparitinase or chondroitinase ABC. By immunoblot analysis, the TED15 antigen is differentially distributed in diencephalic derivatives, being present in the optic nerve but absent from the retina. Within the optic nerve, the TED15 antigen is associated with astrocytes, while in the retina no labeling of astrocytes of Müller cells is observed.

In the mammalian peripheral nervous system (PNS), expression of the neural adhesion molecule L1 on Schwann cells and neurons has been correlated with axonal growth during development and regeneration. The present study was undertaken to examine whether a similar correlation exists between a lesion-induced increase of L1 expression and regenerative capacity in the central nervous system (CNS). The fish optic nerve was used as a model for a successfully regenerating region of the CNS. Immunochemical and immunohistological experiments carried out with immunoaffinity purified polyclonal antibodies, generated against L1 from mouse brain, showed that carp optic nerve and brain, but not liver, contained L1 immunoreactivity. Western blot analysis of brain tissue yielded one distinct band at 200 kDa, while a double band at 200 kDa and two low-molecular weight bands at 120 and 100 kDa, possibly degradation products, were seen in the optic nerve. Immunohistological examination of normal optic nerves revealed L1 immunoreactivity, predominantly associated with connective tissue boundaries of nerve fascicles and with blood vessels, as well as inside axonal fascicles. L1 immunoreactivity was increased by 25%, 8 days after crushing of the optic nerve, as determined by radioimmunoassay on a nerve segment distal to the site of injury and compared with untreated control nerves. Increased levels of L1 were also seen by immunohistology and found to be predominantly associated, as in the normal nerve, with connective tissue boundaries and blood vessels. These observations suggest that a lesion-induced increase in L1 expression in the fish optic nerve is associated with axonal regrowth in the CNS.

The distribution of 5-HT-like immunoreactivity in paraformaldehyde-fixed sections of retina, optic nerve and the optic lobe of Octopus vulgaris was studied by both immunofluorescence and avidin-biotin complex (ABC) immunohistochemical methods utilizing polyclonal antibodies to 5-HT. Some immunoreactive serotonin-containing cells were demonstrated in the retinal plexus, optic nerve, the ciliary body and the lens-generating tissue by both methods. An analysis of dissected retina and optic nerve of Octopus vulgaris by high pressure liquid chromatography (HPLC) with an electrochemical detector (ECD) also showed the presence of 5-HT. In the optic lobe, three 5-HT-immunoreactive bands in the plexiform layer of the cortex were clearly immunostained, and in the medulla both the cell islands and the neuropil contained some cells immunostained by both fluorescein isothiocyanate (FITC) and ABC methods. This is the first report on the systemic immunocytochemical visualization of 5-HT-containing cells and/or fibers in the cephalopod visual system.

The window method of radiolabeled protein analysis was used to study the transport kinetics of axonally transported cytomatrix proteins as they move through segments of mouse optic axons. Three slow component b (SCb) proteins--actin, a 30 kDa protein, and clathrin--were radiolabeled in the eye and were followed for up to 119 days by quantitative one-dimensional gel electrophoresis. These proteins appeared first in the optic nerve, next in the tract, and last in the superior colliculus. All of the radiolabeled proteins had passed through the optic axons and had been effectively removed from the terminals by 119 days. Two different axonal segments ('windows') were examined in detail: a segment of the axon shaft region in the optic tract, and a segment of axon terminal region in the midbrain superior colliculus. The median transit times of the 3 proteins were 53-100% longer in the colliculus than in the tract, and the pulse transients (the total area under the transport curve in each window) were 180-350% larger in the colliculus than in the tract. These results indicate that at least certain cytomatrix and cytoskeletal proteins have longer residence times in the terminal regions than in the axon proper.

The distribution of glial fibrillary acidic protein (GFAP) and of glial hyaluronate-binding protein (GHAP) was studied by indirect immunofluorescence with monoclonal and polyclonal antibodies in dog, rat and rabbit optic nerve. In dog and rabbit, myelination extends into the optic nerve head inside the eye, while in the rat myelination of the optic nerve ceases abruptly at its entry into the eye. Outside the eye the distribution of the two proteins was similar. Both antigens formed a delicate mesh surrounding myelinated optic nerve axons. In all 3 species GFAP immunoreactivity continued uninterrupted into the optic nerve head inside the eye. Conversely, in both dog and rat, GHAP immunoreactivity ceased abruptly in the region of the lamina cribrosa, a sieve-like structure continuous with the sclera through which bundles of optic nerve axons pass. No staining was observed in the myelinated optic nerve head of the dog nor in the non-myelinated optic nerve head of the rat. In the rabbit lacking a lamina cribrosa, GHAP immunoreactivity did not cease abruptly at the optic nerve entry into the eye, but the staining intensity was reduced in the optic nerve head.

Double mutant mice, which are of the genotype shimld/shimld*jpmsd/Y and therefore express both the shimld and jpmsd mutations, have a CNS myelin protein composition which resembles shimld/shimld alone but not jpmsd/Y alone. The double mutant CNS white matter morphology shows much less myelin and major dense line than either shimld/shimld or jpmsd/Y, but has other features which resemble jpmsd/Y but not shimld/shimld. In contrast, the parallel double mutant shi/shi*jp/Y, which expresses the alleles shi and jp rather than shimld and jpmsd, has already been shown to have biochemical and morphological phenotypes which are consistent with each other, both being intermediate between shi/shi and jp/Y and therefore suggesting partial reciprocal intergenic suppression (Brain Research, 374 (1986) 45-53 and 54-62). To assist in explaining the apparent inconsistencies between the biochemical and morphological phenotypes of the shimld/shimld*jpmsd/Y double mutant and between interactions of allelic mutations at the shi and jp loci, a hypothesis of multiple primary gene functions at these two loci is proposed.

Adult rabbit retina can express regeneration-associated characteristics after optic nerve injury, provided it is supplied with appropriate diffusible substances originating from media conditioned by regenerating fish optic nerves or by optic nerves of a newborn rabbit [Hadani et al., Proc. Natl. Acad. Sci. U.S.A., 81 (1984) 7965; Schwartz et al., Science, 228 (1985) 600]. This was shown by applying the active substances to the injured axons in the form of 'wrap-around' implants, consisting of collagen-coated silicone tubes which had been soaked in the conditioned media (CM). The regeneration-associated response was manifested biochemically and by sprouting of nerve fibers in culture. The present work provides morphological evidence that the implantation prolongs survival of ganglion cells and optic nerve fibers and induces new growth. Light microscopic analysis (using horseradish peroxidase (HRP) for labeling the fibers) revealed, 1 week following optic nerve injury, labeled fibers and ganglion cells in both the implanted and control (injured only or injured and implanted with collagen-coated silicone tubes free of CM) nerves. However, from the second week after the injury, distinct differences in the appearance of viable ganglion cells and labeled fibers, were seen between experimental and control preparations. In sections taken through the optic nerve, at the region distal to the site of injury, HRP-labeled fibers were seen in the experimental nerves 1 week, 2 weeks and to a significantly lesser extent 1 month after injury.(ABSTRACT TRUNCATED AT 250 WORDS)

The permeability of Xenopus tadpole optic nerves to macromolecules was studied in order to evaluate the usefulness of this system for studying mechanisms of serum-induced CNS demyelination in vivo. Single injections of either horseradish peroxidase (HRP), ferritin or fluorescein-conjugated human IgG were injected around the right optic nerve and tadpoles were then sacrificed between 15 min and 48 h. Each of the tracers had penetrated the nerve parenchyma by 30 min. Entry of HRP and ferritin occurred mainly via extracellular clefts between adjacent astrocytic endfeet in the glia limitans region. A similar mode of passage was suggested for IgG. Once within the nerve, the tracers became rapidly associated with myelinated axons. HRP was also seen in the periaxonal space but did not directly penetrate the myelin sheath. By 24 h, extracellular localization of tracer was virtually absent with nearly all of the tracer now being concentrated in vesicles within astrocytic processes and perikarya. The distribution of the tracers was not confined to the optic nerve on the injected side; some was seen in adjacent cranial peripheral nerves and surrounding extraocular musculature. Also, tracers eventually penetrated the pial sheath of the contralateral optic nerve. The results of this study indicate that tadpole optic nerves are permeable to a wide range of macromolecules. Furthermore, the distribution of these tracers to nearby cranial peripheral nerves may provide an important opportunity for testing the differential effect of various substances on central and peripheral myelin sheaths.

The distribution of Fink-Heimer positive degeneration and neurofibrillar proliferation has been examined in the dorsal lateral geniculate nucleus (dLGN) and superior colliculus of albino and pigmented rats following enucleation between post-natal days 12 and 30 and as adults. With survival times of 6 h to 5 days following enucleation, the location of maximum degenerative reaction stained by the Fink-Heimer method changes with increasing survival time in both regions. In the dLGN contralateral to the eye removal the earliest degeneration appears in a lamina occupying the medioventral extent of the nucleus and is rapidly removed; later degeneration fills a central lamina of the nucleus with a patch of degeneration extending to the outer surface of the nucleus at the mediodorsal margin. The latest occurring degeneration fills the outermost lamina and is still obvious when degeneration is largely dispersed from the inner and central laminae; this outer lamina shows an early filamentous degenerative reaction in the adult. The uncrossed optic pathway occupies a portion of the central lamina of the nucleus, and in albino animals it shows a rapid degeneration and dispersion similar to the innermost lamina on the crossed side; in the pigmented animals degeneration of the uncrossed projection starts as early as that of the albino but persists as long as the degeneration of the central lamina on the crossed side. The degeneration time of the sprouted, uncrossed pathway resulting from unilateral enucleation at birth is similar in albino and pigmented rats and resembles in timing the normal uncrossed pathway of pigmented rats. These results suggest that there are at least 3 different fiber populations in the rat optic nerve with different distribution in the dLGN. The uncrossed optic pathway of albino and pigmented rats appears to differ in fiber composition; this may relate to aberrations in mapping of uncrossed projections in the albino.

Neurons and processes in the stingray retina can be stained using PAP immunohistochemistry and an antibody to serotonin, without pharmacological pretreatment. Most of the cell bodies are in the inner nuclear layer while the processes ramify in the inner plexiform layer suggesting the presence of a population of serotonin containing amacrine cells in this species. Scattered immunopositive axons were observed in the optic nerve from the optic chiasm to the optic nerve head.

The analysis of proteolysis in the nervous system is complicated by the heterogeneity of cell types, extensive reutilization of liberated amino acids, and artifacts that may arise when the integrity of the tissue is disrupted during experimentation. For these reasons, changes in proteolytic activity that are observed during brain development and in neuropathological states may often be difficult to interpret. To minimize these problems, we have developed a technique that permits protein degradation to be investigated specifically within axons of the mouse retinal ganglion cells (RGC). In the present study, the method has been used to examine the degradation of proteins conveyed in the slow phases of axoplasmic transport. When adult C57Bl/6J mice were injected intravitreally with L-[3H]proline, labeled proteins within the primary optic pathway (optic nerve and tract) after 5 days were almost exclusively the slow phase axonal proteins. The rate of degradation of these proteins was then determined within the excised, but otherwise intact, optic pathway by measuring the release of acid soluble radioactivity at 37 degrees C in vitro. At physiological pH, the amino acids released by proteolysis were extensively reutilized. Unless amino acid reutilization was prevented, protein degradative rates were artifactually lowered 3-fold. At least two proteolytic systems within RGC axons actively degraded the slowly transported axonal proteins. A 'neutral' system, stimulated by exogenous calcium ions, was optimally active within the physiological pH range (pH 7.0--7.8). The rate of protein degradation at pH 7.4 was uniform along the RGC axon. An 'acidic' system was optimally active with the incubation was carried out at pH 3.8. This proteolytic activity was calcium-independent and exhibited a proximodistal gradient within the RCG axon with higher activity proximally. Similar proteolytic activities were present in isolated intact retinas but in different proportions. The half-lives of axonal and retinal proteins were comparable to CNS protein half-lives estimated in vivo by methods that take amino acid reutilization into account. These and other recent findings demonstrate the utility of this neuron-specific approach in characterizing proteolytic processes within one cell type that may otherwise be obscured by proteolytic events in other cells when brain tissue is analyzed by conventional methods.

The involvement of the axonal smooth endoplasmic reticulum as a channel for the retrograde axonal transport of horseradish peroxidase (HRP) has been tested by analysing serial sections of 52 HRP-positive organelles in chick optic nerves. The enzyme marker was injected in the posterior, contralateral optic tectum 10 h before fixation of young chicks. The two optic nerves, retinas and optic tecta were incubated for electron microscopic demonstration of HRP. Thin sections of the retinas and tecta and serial thin sections of the optic nerves were studied in some cases with the aid of a goniometer. Of the 52 organelles, 42% had a tubular shape, 46% were oval and 12% were multivesicular bodies. None of the organelles was found to have continuities with other membranous structures, including tubules or cisternae of the smooth endoplasmic reticulum. In 10 cases, the smooth endoplasmic reticulum was followed in serial sections over a length of up to 4 micrometer. In every case, the reticulum appeared to form a continuous system although some tubular extensions apparently ended blindly near other organelles. In neither the 10 series of serial sections nor in any other individual micrographs did any recognizable profile of the smooth endoplasmic reticulum contain HRP. Measurements of the thickness of the membranes of HRP-containing organelles, of the smooth endoplasmic reticulum and of plasmalemma were made, since these membranes have been distinguished on the basis of their thickness in other cells. The plasmalemma in the axons was about 20% thicker than that of the smooth endoplasmic reticulum, and about 9% thicker than that of HRP-labeled organelles. The membrane of the smooth endoplasmic reticulum and HRP-organelles could also be distinguished by this means. It is concluded that in chick retinal ganglion cell axons, HRP is not transported in a retrograde direction via a continuous channel of smooth endoplasmic reticulum.

Gliogenesis in the rat optic nerve is disrupted by neonatal treatment with the mitotic inhibitor 5-azacytidine (5-AZ). The rate of myelination and number of glial cells are markedly reduced in treated animals. We analyzed the physiological consequences of these chemically induced morphological alterations in terms of activity-dependent K+ accumulation in brain extracellular space and characteristics of the compound action potential (CAP). Nerves from 5-AZ-treated animals older than 5 days of age showed significantly higher activity-dependent 'ceiling levels' of extracellular K+ concentration ( [K+]o) than controls. This result is consistent with the hypothesis that glial cells are involved in K+ homeostasis at a cellular level and play a role in helping to set the ceiling level of activity-dependent K+ accumulation. The CAPs of 5-AZ-treated nerves older than 5 days of age were larger and generally of simpler configuration than those observed in control animals, perhaps due, among other factors, to the retained uniformity of axonal conduction velocity caused by inhibition of myelination.

Beta,beta'-iminodipropionitrile (IDPN) is an agent that produces a marked impairment in the transport of neurofilaments. Its effect on other slowly transported cytoskeletal components such as tubulin and actin is variable. Previous studies have evaluated transport of neurofilaments after IDPN intoxication in a neurofilament-rich system (sciatic motor nerves) and in a system devoid of neurofilaments (axons of the dorsal motor nucleus of the vagus). In the former, IDPN impairs the transport of tubulin and actin but to a lesser degree than it does neurofilament proteins. In the latter, tubulin and actin transport were not impaired, and neurofilament proteins were not present. In this study we evaluated the transport of the cytoskeletal components in a system with an intermediate amount of neurofilaments (the visual system). In the visual system, there is a selective and marked (50%) impairment in the transport of neurofilaments with no impairment in transport of tubulin or microtubule-associated proteins (tau group). We conclude that these different patterns of impairment in transport reflect the differences in pre-intoxication neurofilament content of the nerves examined, the effect of IDPN on the transport of the other components of slow transport being secondary to the presence of stagnated neurofilaments. This model also suggests that transport of neurofilaments can be selectively impaired without producing an effect on other major slow transport components.

If [3H]uridine is injected into the eyes of goldfish during optic nerve regeneration, then the return of fibers to the optic tectum is accompanied by the appearance of [3H]RNA in the tectum. The amount of [3H]RNA arriving in the tectum is consistently greater than in non-regenerating controls and reaches maximum levels (more than 10 times controls) 24 days after optic nerve crush. When [14C]uridine is injected subarachnoidally 1 day prior to sacrificing, the amount of [14C]RNA in the tectum is approximately doubled throughout the regeneration period. In order to characterize the radioactive tectal RNA in these experiments, we have crushed the optic nerves of 15 fish, and 18 days later injected [3H]uridine into both eyes. Five days later [14C]uridine was injected subarachnoidally and all fish were sacrificed a day later. RNA was extracted and fractionated in 2.0% polyacrylamide gels. The amounts of 3H- and 14C-labeled ribosomal as well as small molecular weight RNAs were increased during regeneration. Analysis of the area under the 28S, 18S and 4-7S RNA peaks indicated a small increase in 14C radioactivity in each peak (1.2, 1.5, and 1.5 times control, respectively). On the other hand, 3H radioactivity showed the greatest increase in the 4-7S fraction (8.0 times control) whereas large molecular weight ribosomal fractions were approximately 3 times control. Electrophoresis of the RNA on 10% polyacrylamide gels demonstrated that all of the small molecular weight RNA was confined to the 4S (tRNA) peak. These results suggest that when optic nerves of goldfish regenerate, they may enter the tectum carrying 4S (transfer) RNA.

Following injection of the acylating reagent N-succinimidyl [3H]propionate into the optic nerve of goldfish, labeled protein appeared in the ipsilateral retina and contralateral tectum in a time-dependent manner. Autoradiography indicated the presence of the labeled material in the neuroplasm of the retinal ganglion cells and their projections. While most of the recoverable injected radioactivity was confined to the injection site even after 1 week, labeled proteins arriving in the retina by retrograde flow or in the tectum by anterograde flow had distinctly different patterns, a result suggesting specific transport processes rather than diffusion. In contrast to reported studies with the labeling agent in other species, a prominent 68,000 molecular weight component was not seen. The results are discussed in relation to the role of retrograde transport in regeneration.

Fast and slow axonal transport of protein have been studied in the optic nerves of mice infected with Semliki Forest Virus (SFV) that causes patchy demyelination throughout the CNS. Intravitreal injections of [3H]proline were given at regular intervals after virus inoculation, the labelled protein in the superior colliculi was then measured after survival periods of 18 h or 10 days, for fast and slow axonal transport studies, respectively. Fast transport studies showed an enhanced amount of protein arriving at the optic nerve terminals (superior colliculus) of the SFV-infected mice prior to the onset of demyelination. In contrast, the slow transport studies showed an enhanced amount of protein at the superior colliculus of the SFV-infected mice during the demyelination period. There was no concomitant increase in labelled protein in the retina at any time after the SFV infection. It is proposed that alteration in the transport of the protein constituents other than major myelin specific components may cause disruption of myelin maintenance in SFV infection.

Following an intraocular (i.o.) injection of [N-3H]acetylmannosamine, low molecular soluble radioactive label is distributed rapidly along the optic pathway. At the same time glycoproteins and gangliosides were found radioactively labelled in the distal parts of the optic pathway. Negligible amounts of radioactivity were incorporated into the same structures via the blood. The present data confirm, that glycoproteins are transferred in the retinal ganglion cells by a rapid axonal transport mechanism towards the nerve endings. Unlike glycoproteins labelled gangliosides accumulated in the optic nerves up to 4 days after the precursor injection. Simultaneously radioactive material, soluble in TCA/PTA, disappeared from the optic nerves. The specific radioactivities (disint./min/mug NeuNAc) of the single ganglioside fractions of the optic lobes, which contain the nerve endings of the retinal ganglion cells, differed considerably. Especially one fraction, moving on TLC plates identically to 'GD1a', was up to 20 times higher labelled than the other gangliosides. After an intracerebral injection of the same precursor, radioactivity was incorporated into the ganglioside fractions of the optic lobes to the same extent.

The hazards of using optic nerve (as opposed to optic tract and more distal components of the optic system) to study axonal transport were highlighted by observing the fate of [14C]serine and [3H]glycerol injected into the rabbit eye. Despite prior blockage of axonal transport with colchicine, appreciable radioactivity rapidly appeared in the optic nerve adjacent to the injected eye. Radioactivity decreased exponentially along the entire optic chiasm. Counts were distributed among the lipid, protein, and acid-soluble fractions. Separation of optic nerve lipids revealed appreciable labeling of most lipid classes including those characteristic of myelin; a markedly different labeling pattern was observed for axonally transported lipids. The data are consistent with a mechanism involving extra-axonal diffusion of precursor into the surrounding glia followed by incorporation into lipids and proteins of those cells and ultimately myelin. The phenomenon is discussed in relation to possible errors that were made in interpreting earlier experiments.

Glycosaminoglycans (GAGs) derived from axonally transported proteoglycans were evaluated in regenerating goldfish optic tracts. Fish were administered unilateral optic nerve crushes and stored for 21 days at 21 degrees C. Retinas were labeled by bilateral injection of 35SO4 and sulfated macromolecules axonally transported to the optic tracts were analyzed 8 h postinjection. Results show regenerating tracts contain 6.7-fold more transported 35SO4 in GAGs than their contralateral intact counterparts. Further analysis revealed that regenerating tract GAGs were comprised of 60% chondroitin (4 or 6) sulfate (CS) and 40% heparan sulfate (HS), while unoperated tract GAGs contained 26% CS and 74% HS. These results indicate that there is a large regeneration related increase in the axonal transport of proteoglycans and particular enrichment of transported molecules containing CS chains. The findings can be viewed in the context of recent implication of axonal proteoglycans in processes of fiber outgrowth, adhesion and induction of glial mitosis.

Young chickens and rats were injected intravitreally with [3H]proline and sacrificed at short intervals thereafter. Regression lines calculated for the plotted points (survival time, transport distance) revealed a transport rate of 329 mm/day in the chick and 350 mm/day in the rat. Both rates are close to those reported for peripheral axons (410 +/- 50 mm/day).

The CNS of the mutant mld has a severe myelin deficit. In contrast, myelination is normal in the PNS. Previous studies showed that myelin basic proteins (MBP) are practically missing in mld CNS and PNS tissues. Using an in vitro system and immunoprecipitation, we present evidence that MBP synthesis is repressed in the PNS and in the CNS of mld mutants.
