3. Phthalates and Phenols, Leukocyte Telomere Length, and Breast Cancer Risk and Mortality in the Long Island Breast Cancer Study Project.

Abstract

Background: Phthalates and phenols from the environment have been inconsistently associated with breast cancer risk or mortality. Studies on the potential modifying role of leukocyte telomere length (LTL), a biomarker of biological aging, on these associations are lacking. Methods: We included 1,268 women from the Long Island Breast Cancer Study Project with available data on phthalate and phenol analytes and LTL measurements. Twenty-two phthalate and phenol analytes were measured in spot urines and LTL was measured in blood. The modifying effect of LTL on the associations of individual analyte with breast cancer risk as well as mortalities was estimated using interaction terms between LTL and urinary concentrations of analyte in logistic regression and Cox regression models, respectively. ORs, HRs, and corresponding 95% confidence intervals for a one-unit (ln μg/g creatinine) increase of urinary phthalate/phenol level were estimated at 10th, 50th, and 90th percentiles of LTL. Results: LTL significantly (P < 0.05) modified associations between 11 of 22 of urinary phthalate/phenols analytes and breast cancer risk. An inverse association between phthalate/phenols analytes and breast cancer risk at shorter LTL and a positive association at longer LTL was generally suggested. No modifying effect was found for LTL on the association between these phthalate/phenols analytes and breast cancer mortalities. Conclusions: LTL may modify the associations between phthalate and phenol exposures and breast cancer risk. Impact: This study is the first study that determined the modifying effect of biological aging in the association between environmental chemical exposure and breast cancer risk. The article discusses a study which examined whether genome-wide dysregulation of circulatinng microRNAs (miRNA) can differentiate hepatocellular carcinoma cases from controls. Particular focus is given to the use of real-time polymerase chain reaction (PCR) in validating candidate miRNAs. Also discussed are clinicopathologic characteristics of subjects, overexpression of miRNAs in hepatocellular carcinoma cases, and the correlation between Taq-Man Low Density Arrays and PCR assays.

The study shows that the degree of change in peripheral blood mononuclear cells (PBMC) DNA methylation is inversely associated with methylation levels at baseline. It concludes that DNA methylation changes associated with baseline values of the assay and vary by assay type.

Abstract

The article offers information on the Long Island Breast Cancer Study Project that explored the role of breast cancer risk and genetic variants in telomere pathway genes. There are 1,067 breast cancer cases and 1,110 control subjects included in the study, with 52 variants of the telomere genes. The results found four single nucleotide polymorphisms related to breast cancer risk under the recessive or dominant model.

The article presents a case-control study which evaluates the role of aflatoxin B1 exposure on the risk of hepatocellular carcinoma (HCC). In this study, baseline blood and urine samples were used to determine the level of aflatoxin-albumin adducts and urinary aflatoxin metabolites. Also, researchers used conditional logistic regression analysis to calculate odds ratios and 95% confidence intervals to examine the effect of aflatoxin exposure on risk of HCC. Results of the study are offered.

A description of the experimental set-up and measurement method is provided. The study concluded that one-carbon metabolism may be an important pathway that could be targeted to improve breast cancer survival.

Abstract

The article focuses on a study which explored whether there was selective expression of pChk2 in precancerous lesions but not in nonneoplastic tissue of oral mucosa. The study used a retrospective cohort design wherein 96 biopsied clinical leukoplakias and erythroplakias with known subsequent progression to squamous cell carcinoma (SCC) were identified. Results of the study showed that pChk2 merits further investigation as a promising biomarker that can discriminate those lesions at risk for developing SCC, regardless of histologic evidence for atypia.
