Levans of different structures and molecular weights (MW) can display various techno-functional and health-promoting properties. In the present study, selected levans were produced by the transfructosylation of sucrose catalyzed by levansucrases from Bacillus amyloliquefaciens and Gluconobacter oxydans, and their structural, techno-functional and anti-inflammatory properties were investigated. NMR and methylation/GC analysis confirmed the structure of β-(2, 6) levans. The structural characterization led to the classification of levans as high MW (HMW, ≥100 kDa), low MW (LMW, ≤20 kDa) and mix L/HMW ones. Levan with higher MW had more linear fructosyl units with fewer reducing ends and branching residues. LMW levan showed the highest foaming capacity and stability while HMW levan had the highest emulsion stability. HMW and mix L/HMW levans showed comparable water and oil-holding capacities, which were higher than LMW. HMW and mix L/HMW levans were found to have gelling properties at low concentrations. The rheological behaviour of HMW levan-based gel was a more viscous-like gel, while that of mix L/HMW levan-based one showed more elastic solid like-gel. The temperature also influenced the rheology of levan, showing that the mix L/HMW levan gel network was the most thermal stable as its viscoelasticity remained constant at the highest temperature (75 °C). Cranberries of Stevens variety, mainly used for juice production, were processed into pomace, from which alcohol insoluble solids (AIS) were obtained. The cell wall polysaccharides were sequentially extracted from AIS, and characterized in terms of monosaccharide profile, sugar linkage and molecular weight distribution. Pectic polysaccharides represented more than 90% of the carbohydrates contained in hot buffer (HA), chelating agents (CH) and diluted alkali (DA) extracts. HA extract contained homogalacturonan with 75% being methyl esterified, and pectic arabinan with traces of pectic galactan, type II arabinogalactan and 1,4-β-glucan. CH extract, recovered with the highest yield (11.0% w/w), was composed mainly of homogalacturonan. DA extract included homogalacturonan with 2% methyl esterification, abundant arabinan and galactans and traces of 1,4-β-glucan. Glucomannan, xylan and xyloglucan represented 66% of the carbohydrates present in the last concentrated alkali extract (CA), the rest being pectic arabinan and galactan. A bi-enzymatic system using two multi-enzymatic preparations (Depol 670L and Gamanase 1.5L) was investigated for the production of prebiotic galacto/galacto(arabino)-oligosaccharides and oligomers with well-defined degree of polymerisation (DP) from potato galactan-rich rhamnogalacturonan I. Depending on the reaction condition, yields of low (DP of 2-6) and high-MW oligosaccharides (DP of 7-12) and oligomers (DP of 13-70) varied between 0.1-13.9, 0.0-37.5 and 0.0-75.7%, respectively. Substrate concentration and Depol 670L/Gamanase 1.5L ratio were identified as the most significant linear terms in oligosaccharide and oligomer yield models, respectively. Moreover, interaction between reaction time and substrate concentration had a significant effect on the yield of oligosaccharides, while interaction between reaction time and Depol 670L/Gamanase 1.5L ratio affected significantly the yield of oligomers. Higher yields of both oligosaccharides and oligomers were obtained when equal amount of Depol 670L and Gamanase 1.5L was used in combination. Galactose-rich oligosaccharides/oligomers (oligo-RG I) were produced by the enzymatic treatment of potato galactan-rich rhamnogalacturonan I (RG I) with endo-β-1,4-galactanase and Depol 670L multi-enzymatic preparation. The digestibility study revealed that 81.6 and 79.3% of RG I and its corresponding oligomers remained unhydrolyzed, respectively. The prebiotic properties of RG I and its hydrolysates were investigated using a continuous culture system inoculated with immobilized fecal microbiota. Both RG I and oligo-RG I have stimulated the growth of Bifidobacterium spp. and Lactobacillus spp., with oligo-RG I hydrolysates being more selectively fermented by these beneficial bacteria. Furthermore, none of RG I nor its hydrolysates increased the populations of Bacteroidetes and Clostridium leptum. Total amounts of short chain fatty acids, generated upon the fermentation of oligo-RG I, were higher than those obtained with its parent RG I and the positive control (fructooligosaccharides). Maple syrups with selected degree Brix (°Bx) (15, 30, 60) were investigated as reaction systems for levansucrase from Bacillus amyloliquefaciens. The enzymatic conversion of sucrose present in the maple syrup and the production of the transfructosylation products were assessed over a time course of 48h. At 30°C, the use of maple syrup 30°Bx led to the highest levansucrase activity (427.53μmol/mg protein/min), while maple syrup 66°Bx led to the highest converted sucrose concentration (1.53M). In maple syrup 30°Bx, oligolevans (1080%). In maple syrup 66°Bx, the most abundant products were oligolevans at 30°C and levans (DP≥30) at 8°C. The acceptor specificity study revealed the ability of B. amyloliquefaciens levansucrase to synthesize a variety of hetero-fructooligosaccharides (FOSs) in maple syrups 15°Bx and 30°Bx enriched with various disaccharides, with lactose being the preferred fructosyl acceptor.
