Abstract

A facile and efficient synthesis of polyhydroquinolines has been developed via four-component condensation reactions of aldehydes, dimedone, ethyl acetoacetate, and ammonium acetate in the presence of Novel SO3H-functionalized ionic liquid catalysts through Hantzsch reaction under ultrasound irradiation without solvent. Simple work-up procedure, environmentally friendly, inexpensive, and non-toxic catalyst, shorter reaction times along with excellent product yields are the significant features of this practical method.

Abstract

Novel SO 3 H-functionalized ionic liquids were successfully applied as catalysts for one-pot Fischer indole synthesis under microwave irradiation and in a water medium. Various types of indoles were prepared using single-carbonyl ketones/aldehydes or cyclohexandiones with aryhydrazine hydrochlorides in 86–96% yields in water under microwave irradiation. The indole products could be conveniently separated from the reaction mixture through filtration, and the catalytic system of [(HSO 3 -p) 2 im][CF 3 SO 3 ]/H 2 O could be reused directly without any treatment. The entire process is simple, time saving, and environmentally friendly.

Abstract

The kinetic resolution of a carbon nucleophile is realized for the first time via Pd-catalyzed asymmetric allylic alkylation with “unstabilized” ketone enolates as the nucleophile, providing both allylated 2,3-disubstituted 2,3-dihydro-4-quinolones and recovered substrates in high yields and high ee (S-factor is 40−145). The application of the methodology in organic synthesis is demonstrated by the ready transformation of an allylated adduct into pyrrolo[3,2-c]quinoline, which features a core structure of biologically active Martinella alkaloids.
