Medicinal chemistry has been benefited by combinatorial chemistry and high-throughput parallel synthesis. Ionic liquids reduce the materials and energy intensity of chemical processes and products, minimize or eliminate the dispersion of harmful chemicals in the environment, maximize the use of renewable resources and extend the durability and recyclability of products. It is possible to tune the physical and chemical properties by varying the nature of the cations and anions. Ionic liquids can be easily recovered, cleaned up, and reused repeatedly.

The synergy of the combined use of DMSO and an ionic liquid viz. (bbim)+Br− has brought about a rapid and efficient esterification of sodium carboxylates with acyl and alkyl halides under ambient conditions in excellent isolated yields (90–95%) in short reaction times (12–40 min).

The nuclear as well as side-chain bromination of activated aromatic substrates has been achieved in high yields and substantial regioselectivity with N-Bromosuccinimide (NBS) over HZSM-5.

The synergy of the combined use of DMSO and an ionic liquid viz. (bbim)+Br− has brought about a mild, convenient, efficient, and rapid protocol for the synthesis of 1-substituted-1H-1,2,3,4-tetrazoles via the condensation of amines, triethyl orthoformate, and sodium azide at ambient temperature in excellent isolated yields (85–90%). The inherent Bronsted acidity of ionic liquid and high polarity of both IL and DMSO resulted in a significant enhancement in the reaction rate.

Proline-catalyzed synthesis of 1,8-dioxo-decahydroacridines is achieved via one-pot, three-component condensation of aromatic aldehydes, cyclic diketone, and aryl amines in aqueous ethanol medium. This method offers the advantages of proceeding in neutral and mild conditions, giving high to excellent yields of acridines with easy workup procedure.

Quinoxaline derivatives have been synthesized in excellent yields using an ionic liquid (IL) (viz., 1-n-butylimidazolium tetrafluoroborate) as a reaction medium as well as promoter from various 1,2-diketones and aryl-1,2-diamines. The process is general for the synthesis of quinoxaline derivatives from aromatic as well as aliphatic-1,2-diketones. The advantages of the present method are ambient reaction temperature, simplicity of operation, high yields of products, the recyclability of the IL, and ecofriendly nature of the reaction medium.

An efficient regioselective approach to the synthesis of tetrasubstituted pyrimidines was developed by sequential functionalization of easily available Biginelli 3,4-dihydropyrimidine-2(1H)-ones via dehydrogenation, chlorination followed by palladium catalyzed C–C Suzuki/Sonogashira coupling reaction. All the synthesized compounds were evaluated in vitro for their antifungal activities against Candida albicans, Cryptococcus neoformans, Benjaminiella poitrasii, Yarrowia lipolytica, and Fusarium oxysporum, and antibacterial activities against Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus.

A convenient method for the syntheses of non‐1‐ynitols 8a–8d, by chemoselective addition of (trimethylsilyl)‐propargylmagnesium bromide at the anomeric center of a 1‐unprotected sugar, in the presence of 3‐C′‐halide and 3‐C′‐silyl functions in the side chain is described. In addition, an efficient method for the synthesis of 3‐C′‐hydroxymethyl sugar 3 via the addition of C1 silyl Grignard reagents to ulose 1 and subsequent oxidation by the Fleming‐Tamao method in excellent yields is reported. Also, a suitable acid‐catalyzed isomerization of the 1,2‐O‐isopropylidene group to the 2,3‐O‐isopropylidene group (5a–5f), to get access to the anomeric center, in good to excellent yields has been depicted.

A highly efficient and facile method has been described for the synthesis of substituted 2-aminothiazoles in water without any added catalyst or co-organic solvent. The reaction was carried out at ambient temperature and the products were obtained in excellent isolated yields. The developed protocol is successfully applied for the preparation of an anti-inflammatory drug, fanetizole.

A simple, efficient, and environmentally friendly one‐pot, three‐component synthesis of β‐enaminones via Sonogashira coupling–Michael addition sequences under solvent‐free conditions has been reported. Also the synthesis of β‐enaminones has been achieved in high yields by the direct reaction of amines with ynones under solvent‐free conditions.

A highly efficient and rapid synthesis of 2-amino-4-arylthiazoles and 2-methyl-4-arylthiazole from α-bromoketone and thiourea/thioamide is described using room temperature ionic liquid at ambient conditions. The method is simple, rapid and practical, generating thiazole derivatives in excellent isolated yields. This protocol is utilized for a commercially feasible synthesis of an anti-inflammatory agent, Fanetizole.

An efficient synthesis leading directly to 1-substituted-1H-1,2,3,4-tetrazoles from easily available amines and sodium azide in stoichiometric proportions using a room-temperature ionic liquid, namely, 1-n-butylimidazolium tetrafluoroborate in excellent yields is described. The inherent Bronsted acidity and high polarity of the IL results in a significant enhancement in the reaction rate.

A highly efficient one-pot methodology has been developed to synthesize a class of substituted l-pyridylimidazo[1,5-a]pyridines, using Bronsted acidic ionic liquid 1-butylimidazolium tetrafluoroborate, [Hbim]BF 4 in excellent isolated yields in the absence of any catalyst. The methodology is characterized by simple work-up procedures along with efficient recovery and recycling of the ionic liquid, which acts as a reaction medium as well as a promoter.

A general and efficient copper-, ligand- and solvent-free synthesis of ynones by coupling of a wide range of acid chlorides with terminal alkynes catalyzed by palladium(II) acetate at room temperature is reported.

Results of the optimized conditions for the one-pot synthesis of 2-substituted indoles via palladium acetate catalyzed tandem Sonogashira coupling 5-endo-dig cyclization at room temperature under ultrasonic irradiation and standard stirred conditions are described. Electron-donating and electron-withdrawing groups present in both coupling partners were well tolerated under these mild conditions. A copper-, ligand- and amine-free condition is an important feature of this protocol. Significant enhancement of reaction rates was observed for the reactions employing ultrasonic irradiation.

The Sonogashira reaction proceeds at ambient temperature (30 degrees C) in acetone or room-temperature ionic liquid, 1,3-di-n-butylimidazolium tetrafluoroborate ([bbim]BF4), as solvent under ultrasound irradiation to give enhanced reaction rates, excellent chemoselectivity, and high yields in the absence of a copper cocatalyst and a phosphine ligand. TEM analysis showed the formation of stable, crystalline, and polydispersed Pd(0) nanoparticles as catalyst for the reaction.

Several room-temperature ionic liquids (ILs) based on 1-butylimidazolium salts with varying anions were synthesized and evaluated for the preparation of biologically active substituted quinolines and fused polycyclic quinolines using the Friedlander heteroannulation reaction. On screening, 1-butylimidazolium tetrafluoroborate [Hbim]BF4 was found to be the best ionic liquid for the heteroannulation reaction, and the reasons to this effect are well explained. The reactions proceed very well under relatively mild conditions without any added catalyst. The IL acts as a promoter for this regiospecific synthesis and can be recycled. By this green approach, various quinolines were prepared in excellent yields and purity and well-characterized.
