Despite the enormous importance of insoluble proteins in biological processes, their structural investigation remains a challenging task. The development of artificial enzyme-like catalysts would greatly facilitate the elucidation of their structure since currently used enzymes in proteomics largely lose activity in the presence of surfactants, which are necessary to solubilize insoluble proteins. In this study, the hydrolysis of a fully insoluble protein by polyoxometalate complexes as artificial proteases in surfactant solutions is reported for the first time. The hydrolysis of zein as a model protein was investigated in the presence of Zr(IV) and Hf(IV) substituted Keggin-type polyoxometalates (POMs), (Et2 NH2 )10 [M(α-PW11 O39 )2 ] (M = Zr or Hf), and different concentrations of the anionic surfactant sodium dodecyl sulfate (SDS). Selective hydrolysis of the protein upon incubation with the catalyst was observed, and the results indicate that the hydrolytic selectivity and activity of the POM catalysts strongly depends on the concentration of surfactant. The molecular interactions between the POM catalyst and zein in the presence of SDS were explored using a combination of spectroscopic techniques which indicated competitive binding between POM and SDS towards the protein. The reactivity of a range of Keggin and Wells-Dawson type heteropolyacids (HPAs): H3 PW12 O40 H4 SiW12 O40 , H3 PMo12 O40 , K6 P2 W18 O62 , and NaH2 W12 O4 , towards the heavily glycosylated α-1-acid glycoprotein (AGP) is reported. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD) show that after incubation of the protein with HPAs at 80 °C and pH 2.8 complete hydrolysis of terminal glycosidic bond has been achieved, resulting in the removal of sialic acids with no observed destruction of the protein core or the residual glycan chains. The 1 H NMR spectroscopy confirmed that the released sialic acids preserve intact structure upon their excision from the protein, which makes the reported method suitable for the analysis of sialic acid modifications which play an important role in numerous biological processes. The presence of other sugars was not detected by 1 H NMR and HPAEC-PAD, suggesting that HPAs hydrolyze only the terminal glycosidic bond in the glycoprotein, resulting in the selective release of sialic acid from AGP. The kinetic results have shown that under equal temperature and pH conditions, the hydrolysis of the terminal glucosidic bond occurred faster in the presence of HPAs compared to conventional mineral acids. The observed rate constants were in the range 6,7×10-2 -11,9×10-2 min-1 and the complete and selective excision of sialic acids could be achieved within 60 min of incubation. The Trp fluorescence and CD spectroscopy show that non-covalent interaction between HPA and protein takes place in solution which could lead to stabilization of the sialosyl cation that is formed during the glycosidic bond hydrolysis by anionic HPA cluster.

A new class of hexameric Ln12 -containing 60-tungstogermanates, [Na(H2 O)6 ⊂Eu12 (OH)12 (H2 O)18 Ge2 (GeW10 O38 )6 ](39-) (Eu12 ), [Na(H2 O)6 ⊂Gd12 (OH)6 (H2 O)24 Ge(GeW10 O38 )6 ](37-) (Gd12 ), and [(H2 O)6 ⊂Dy12 (H2 O)24 (GeW10 O38 )6 ](36-) (Dy12 ), comprising six di-Ln-embedded {β(4,11)-GeW10 } subunits was prepared by reaction of [α-GeW9 O34 ](10-) with Ln(III) ions in weakly acidic (pH 5) aqueous medium. Depending on the size of the Ln(III) ion, the assemblies feature selective capture of two (for Eu12 ), one (for Gd12 ), or zero (for Dy12 ) extra Ge(IV) ions. The selective encapsulation of a cationic sodium hexaaqua complex [Na(H2 O)6 ](+) was observed for Eu12 and Gd12 , whereas Dy12 incorporates a neutral, distorted-octahedral (H2 O)6 cluster. The three compounds were characterized by single-crystal XRD, ESI-MS, photoluminescence, and magnetic studies. Dy12 was shown to be a single-molecule magnet.

Novel organic-inorganic hybrids of various sizes were generated by reaction of 1,8-octanediphosphonic acid (ODP) and (NH4)6Mo7O24 in aqueous solution. The formation of rodlike hybrids with variable numbers of covalently bound ODP and polyoxomolybdate (POM) units can be tuned as a function of increasing (NH4)6Mo7O24 concentration at fixed ODP concentration. The chemical structure of the ODP/POM hybrids was characterized by (1)H, (31)P, and (95)Mo NMR spectroscopy. Heteronuclear (31)P DOSY (diffusion- ordered NMR spectroscopy) and molecular mechanics (MM) calculations were applied to determine the size and shape of the nanosized hybrids generated at various ODP/POM ratios. For this purpose, the structures of ODP/POM hybrids with variable numbers of ODP and POM units were optimized by MM and then approximated as cylinder-shaped objects by using a recently described mathematical algorithm. The thus-obtained cylinder length and diameter were further used to calculate the expected diffusion coefficients of the ODP/POM hybrids. Comparison of the calculated and experimentally determined diffusion coefficients led to the most probable ODP/POM hybrid length for each sample composition. The (31)P DOSY results show that the length of the hybrids increases with increasing POM concentration and reaches a maximum corresponding to an average of 8 ODP/7 POM units per chain at a sample composition of 20 mM ODP and 14 mM POM. With excess POM, above the latter concentration, the formation of shorter-chain hybrids terminated by Mo7 clusters at one or both ends was evidenced on further increasing the POM concentration. The results demonstrate that the combination of (31)P DOSY and MM, although virtually unexplored in POM chemistry, is a powerful innovative strategy for the detailed characterization of nanosized organic-inorganic POM-based hybrids in solution.

Six diethylene triamine pentaacetic acid (DTPA) bisamide derivatives functionalized with p-toluidine (DTPA-BTolA), 6-aminocoumarin (DTPA-BCoumA), 1-naphthalene methylamine (DTPA-BNaphA), 4-ethynylaniline (DTPA-BEthA), p-dodecylaniline (DTPA-BC12PheA) and p-tetradecyl-aniline (DTPA-BC14PheA) were coordinated to dysprosium(III) and the magnetic and optical properties of the complexes were examined in detail. The complexes consisting of amphiphilic ligands (DTPA-BC12PheA and DTPA-BC14PheA) were further assembled into mixed micelles. Upon excitation into the ligand levels, the complexes display characteristic Dy(III) emission with quantum yields of 0.3-0.5% despite the presence of one water molecule in the first coordination sphere. A deeper insight into the energy-transfer processes has been obtained by studying the photophysical properties of the corresponding Gd(III) complexes. Since the luminescence quenching effect is decreased by the intervention of non-ionic surfactant, quantum yields up to 1% are obtained for the micelles. The transverse relaxivity r2 per Dy(III) ion at 500 MHz and 310 K reaches a maximum value of 27.4 s(-1) mM(-1) for Dy-DTPA-BEthA and 36.0 s(-1) mM(-1) for the Dy-DTPA-BC12PheA assemblies compared with a value of 0.8 s(-1) mM(-1) for Dy-DTPA. The efficient T2 relaxation, especially at high magnetic field strengths, is sustained by the high magnetic moment of the dysprosium ion, the coordination of water molecules with slow water exchange kinetics and long rotational correlation times. These findings open the way to the further development of bimodal optical and magnetic resonance imaging probes starting from single lanthanide compounds.

Hen-egg-white lysozyme (HEWL) is specifically cleaved at the Trp28-Val29 and Asn44-Arg45 peptide bonds in the presence of a Keggin-type [Ce(α-PW(11)O(39))(2)](10-) polyoxometalate (POM; 1) at pH 7.4 and 37 °C. The reactivity of 1 towards a range of dipeptides was also examined and the calculated reaction rates were comparable to those observed for the hydrolysis of HEWL. Experiments with α-lactalbumin (α-LA), a protein that is structurally highly homologous to HEWL but has a different surface potential, showed no evidence of hydrolysis, which indicates the importance of electrostatic interactions between 1 and the protein surface for the hydrolytic reaction to occur. A combination of spectroscopic techniques was used to reveal the molecular interactions between HEWL and 1 that lead to hydrolysis. NMR spectroscopy titration experiments showed that on protein addition the intensity of the (31)P NMR signal of 1 gradually decreased due to the formation of a large protein/polyoxometalate complex and completely disappeared when the HEWL/1 ratio reached 1:2. Circular dichroism (CD) measurements of HEWL indicate that addition of 1 results in a clear decrease in the signal at λ=208 nm, which is attributed to changes in the α-helical content of the protein. (15)N-(1)H heteronuclear single quantum coherence (HSQC) NMR measurements of HEWL in the presence of 1 reveal that the interaction is mainly observed for residues that are located in close proximity to the first site in the α-helical part of the structure (Trp28-Val29). The less pronounced NMR spectroscopic shifts around the second cleavage site (Asn44-Arg45), which is found in the β-strand region of the protein, might be caused by weaker metal-directed binding, compared with strong POM-directed binding at the first site.

The molecular steps involved in the self-assembly of Cu(3)(BTC)(2) (BTC=1,3,5-benzenetricarboxylic acid) metal-organic frameworks that enclose Keggin-type H(3)PW(12)O(40) heteropolyacid molecules were unraveled by using solution (17)O, (31)P, and (183)W NMR spectroscopy, small-angle X-ray scattering, near-IR spectroscopy, and dynamic light scattering. In aqueous solution, complexation of Cu(2+) ions with Keggin-type heteropolyacids was observed. Cu(2+) ions are arranged around the Keggin structure so that linking through benzenetricarboxylate groups results in the formation of the Cu(3)(BTC)(2) MOF structure HKUST-1. This is a unique instance in which a templating mechanism that relies on specific molecular-level matching and leads to explicit nanoscale building units can be observed in situ during formation of the synthetic nanoporous material.

A dinuclear gadolinium(III) complex of an amphiphilic chelating ligand, containing two diethylenetriamine-N,N,N',N'',N''-pentaacetate (DTPA) moieties bridged by a bisindole derivative with three methoxy groups, has been synthesized and evaluated as a potential magnetic resonance imaging (MRI) contrast agent. Nuclear magnetic relaxation dispersion (NMRD) measurements indicate that at 20 MHz and 37 degrees C the dinuclear gadolinium(III) complex has a much higher relaxivity than [Gd(DTPA)] (6.8 vs 3.9 s(-1) mmol(-1)). The higher relaxivity of the dinuclear gadolinium(III) complex can be related to its reduced motion and larger rotational correlation time relative to [Gd(DTPA)]. In the presence of human serum albumin (HSA) the relaxivity value of the noncovalently bound dinuclear complex increases to 15.2 s(-1) per mmol of Gd3+, due to its relatively strong interaction with this protein. The fitted value of the binding constant to HSA (Ka) was found to be 10(4) M(-1). Because of its interaction with HSA, the dinuclear complex exhibits a longer elimination half-life from the plasma, and a better confinement to the vascular space compared to the commercially available [Gd(DTPA)] contrast agent. Transmetalation of the dinuclear gadolinium(III) complex by zinc(II) has been investigated. Biodistribution studies suggest that the complex is excreted by the renal pathway, and possibly by the hepatobiliary route. In vivo studies indicated that half of the normal dose of the gadolinium(III) complex enhanced the contrast in hepatic tissues around 40 % more effectively than [Gd(DTPA)]. The dinuclear gadolinium(III) complex was tested as a potential necrosis avid contrast agent (NACA), but despite the binding to HSA, it did not exhibit necrosis avidity, implying that binding to albumin is not a key parameter for necrosis-targeting properties.
