We examined the direct effects of toxaphene and endrin, chlorinated insecticides that are widespread in the environment, on myometrial contractions and on the secretion of hormones involved in regulating these contractions. Granulosa, luteal, endometrial and myometrial cells, and myometrial strips from non-pregnant cows were incubated with both insecticides at environmentally relevant doses. Toxaphene inhibited and endrin stimulated the secretion of testosterone and oestradiol from granulosa cells. Toxaphene also inhibited and endrin stimulated the expression of the mRNA encoding the precursor of oxytocin (OT), as well OT secretion in luteal cell cultures. Moreover, endrin increased OT secretion from granulosa cells. Neither insecticide exerted an effect on progesterone secretion from luteal cells. Only toxaphene decreased the secretion of prostaglandins (PGF2 and PGE2) from endometrial cells. Meanwhile, only endrin decreased basal myometrial contractions, which was accompanied by inhibition of PGF2 secretion from the myometrium. Both endrin and toxaphene also decreased the force of the OT-stimulated myometrial contractions, whereas only toxaphene inhibited the stimulatory effect of OT on the force of myometrial contractions. In contrast to endrin, toxaphene decreased synthesis and secretion of one of the primary stimulators of myometrial contractions (OT) and indirectly inhibited OT signal reception in the myometrium by reducing E2 secretion. Both insecticides decreased OT-stimulated myometrial contractions; therefore, they may inhibit further transmission of the OT signal. Moreover, endrin inhibited basal myometrial contractions, potentially resulting from reduced PGF2 secretion from the myometrium. Abstract: Organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) were determined in sediment samples collected from 34 locations in Lake Qarun, Egypt to elucidate their concentrations, distribution, possible sources and potential ecological risk. A total of 25 OCPs and 29 PCBs were identified and quantified. OCPs and PCBs were found to be ubiquitous pollutants in the aquatic environment of Lake Qarun, particular near urban areas reflecting the local usage and input of these pollutants. Total concentrations of OCPs and PCBs in sediments ranged from 1.01 to 164.8ngg−1 and 1.48 to 137.2ngg−1, respectively. Among OCPs, γ-HCH, heptachlor, aldrin, endrin, p,p′-DDE, oxy-chlordane and endosulfan I and II were the most abundant compounds. The composition of PCB congeners was dominated by the tri-, tetra-, penta- and hexa-chlorinated biphenyls. The ratios of certain metabolites (isomers) to their parent compounds (mixtures) indicated there are still new inputs of lindane, endrin, heptachlor and technical endosulfan to Qarun Lake, while the ratios of (p,p′-DDE+p,p′-DDD)/∑p,p′-DDTs indicate earlier usage of DDT. Contamination levels of PCBs and HCHs can be categorized as moderate to high compared to other urbanized regions worldwide. According to established sediment quality guidelines, γ-HCH, endrin and chlordanes would be more concerned OCP species for the ecotoxicological risk in Lake Qarun. [Copyright &y& Elsevier]

Abstract: Perchlorate is a known endocrine disruptor present in groundwater, vegetables and dairy food products in many regions of the United States. It interferes with the uptake of iodide into the thyrocyte by the sodium–iodide symporter at the basolateral surface, thus potentially disrupting the synthesis of thyroid hormone. Because transport of iodide from the thyroid follicular cells to the follicular lumen is mediated by the protein pendrin at the apical surface, we hypothesized that perchlorate may also interact with this protein. Therefore, HeLa cells were transfected with the human SLC26A4 gene, which encodes pendrin, to generate an in vitro mammalian system expressing the recombinant pendrin protein (HeLa-PDS). The HeLa-PDS cells, along with untransfected cells, were then cultured in presence of iodide and/or perchlorate. Intracellular levels of these two chemicals were measured by ion chromatography tandem mass spectrometry. Results from this study show that iodide and perchlorate uptake increases significantly in HeLa-PDS cells as compared to untransfected cells. Thus, recombinant HeLa cells expressing pendrin protein accumulate iodide and perchlorate intracellularly, indicating that pendrin is involved in the uptake of perchlorate. Additional results from this study suggest that iodide and perchlorate competitively inhibit each other for uptake by pendrin. The ability of perchlorate to compete with iodide for uptake by both basal and apical transporters may increase the potential of perturbation of thyroid homeostasis and therefore the estimated risk posed to susceptible human populations. Abstract: Sixteen organochlorine pesticides (OC) were analyzed in several tissue types (abdominal and subcutaneous fat, liver and brain) from juvenile (n =14), immature (n =9), subadult (n =7) and adult (n =20) razorbill (Alca torda) collected from the southwestern Mediterranean coastline, in the East of Spain (La Marina, Elche, Alicante, Spain). These razorbills had drowned in fishing nets (most probably) while searching for food. The objective was to assess the exposure to organochlorine pesticide residues in this wintering population of marine birds. This paper presents, as far as we are aware, the first published data on OC concentrations in razorbills. The highest levels were found in abdominal fat followed by subcutaneous fat, liver and brain. A significant positive relationship was found between age and OC levels in tissues, and with the highest levels in adults. The group of ∑Drins had the highest concentrations, followed by ∑DDT, ∑Endosulfan, ∑HCH and ∑Heptachlor, with endrin aldehyde being the compound which reached the highest levels. The p,p′-DDE/p,p′-DDT ratio in fatty tissues suggests exposure to non-degraded DDT and thus is present in the environment despite its prohibition. The OC levels detected were higher than those found in other studies on Alcidae, which may be explained by the Mediterranean habitat in which the birds were found. The extent of organochlorine pesticides (OCPs) contamination in coastal waters around LaiZhou Bay and JiaoZhou Bay in Shandong Peninsula, northern China, was investigated. The areas around the two bays are both densely populated, thrived with intensive agriculture and industrial activities. Multi-techniques including GC–MS, GC–μECD coupled with chemical peak confirmation and strict QC procedures were used for the quantitative determination of 15 OCPs including alpha, beta, gamma and delta isomers of hexachlorocyclohexane (HCH), pentachloronitrobenzene (PCNB), heptachlor, aldrin, endosulfan, p,p′-DDE, dieldrin, endrin, p,p′-DDD, o,p′-DDT, p,p′-DDT and methoxychlor. The survey results show that contaminations by OCP residues remain widespread in the areas, but the averaged concentration levels are all below the regulatory limits, e.g., CMC limits (acute criterion values) specified in US Environmental Protection Agency (USEPA) and China national standards. Average concentration of OCPs in water samples were from undetectable to 3. 8ngl−1 in LaiZhou Bay and from 0. 1 to 3. 9ngl−1 in JiaoZhou Bay, respectively. Abstract: Bottom sediment samples from 121 sites of the Lake Albufera of Valencia were analyzed. Dieldrin, endrin, heptachlor and op′-DDT were not detected (<0.01ngg−1) in 88–93% of the sites. Aldrin and HCB concentration ranges were between <0.01 and 0.1ngg−1 in 86% and 94% of the sites, respectively. Heptachlor-epoxide and lindane 95% confidence intervals were 0.2–0.5 and 0.06–0.12, respectively. The greatest average concentration corresponds to pp′-DDE, pp′-DDD and pp′-DDT. The sum of six isomers and derivatives of the DDT average concentration reaches 2.1ngg−1, as opposed to 2.7ngg−1 for the sum of 13 pesticides considered. In the site with a major contamination, 27.0ngg−1 of pp′-DDD and 12.8ngg−1 of pp′-DDT were accumulated. The DDE:DDT proportion average was 0.37, indicating an aged DDT contamination. Concentrations of pesticides in sediments were compared to three sediment quality guidelines, and indicated that a low biological effects level can be expected in either sediments or aquatic organisms.
