A computer-assisted method is described for optimization of multi-component, mobile phase selection for separation of five phosphoamidothioate enantiomers with a series of silica and chiral columns in normal phase HPLC. The method is based on the triangular solvent selection concept using a statistical scanning method. The optimization of the separation over the experimental region is based on a special polynomial estimation from seven experimental runs, and resolution (Rs) is used as the selection criterion. Excellent agreement was obtained between predicted and experimental data.

The enantiomers of a series of fourteen O-ethyl O-(substituted) phenyl N-isopropyl-phosphoroamidothioates have been separated by high performance liquid chromatography (HPLC) on the Pirkle-type chiral stationary phase. Seven molecular descriptors were calculated and four significant descriptors were chosen to correlate against the experimental lnk' values in order to form the quantitative structure-enantioselectivity retention relationships (QSERRs). Through the QSERRs, the retention and enantioselectivity mechanism were examined.

Three computer-assisted statistical scanning methods, the monofactor optimization system (MOS), multicomponent solvent optimization system (MSOS), and difactor optimization system (DOS) for optimizing HPLC separations, have been integrated to produce the optimization system of HPLC (OS-L). The method proposes the complementary use of these techniques, maximizing their strengths while compensating for their weaknesses. The result of OS-L is a method development strategy which obtains simple solutions for simple separation problems and reserves the more complex solutions for difficult separations.

The separation of enantiomers of a series of eighteen novel nitrogen mustard linked phosphoryl diamide derivatives was investigated on the prepared phenyl carbamate derivative β-cyclodextrin bonded phase in normal-phase HPLC. Some of the enantiomers could be separated in baseline. The chiral recognition mechanism was also suggested for the separation of chiral phosphorus organic compounds.

Five chiral stationary phases (CSPs) were used to separate the enantiomers of a series of O,O-diethyl (p-methyl-benzenesulfonamindo)-aryl(alkyl)-methylphosphonates. A chiral recognition mechanism was presented to explain the resolution of these compounds. Results show that CSP with strong π-acceptor 3,5-dinitrobenzoyl group and high steric hindrance has the best resolution ability in chiral separation of O,O-diethyl (p-methylbenzenesulfonamindo)-aryl(alkyl)-methylphosphonates. When a CSP has just a strong π-acceptor 3,5-dinitrobenzoyl or high steric hindrance it does not have good chiral resolution ability. The chiral recognition is more difficult when the CSP has more than one asymmetric center.

A partially substituted β-cyclodextrin chiral stationary phase was prepared by the reaction of phenyl isocyanate. The enantiomers of a series of O,O-diethyl(p-methylbenzenesulfonamido)-aryl(or alkyl)-methylphosphonates were studied on the prepared phenyl carbamate derivative β-cyclodextrin bonded phase and a commercial (S)-(+)-1–(1-naphthyl) ethylcarbamate derivative β-cyclodextrin bonded phase on normal phase chromatographic condition. Results show that the prepared phenyl carbamate derivative β-cyclodextrin bonded phase has better enantiomeric selectivity to the series of compounds. A chiral recognition mechanism was suggested for the separation of these novel organic phosphorus enantiomers.

A novel pressurized capillary electrochromatography (PCEC) was developed to separate basic drugs on strong cation exchange (SCX) column. The separation result by using PCEC was better than that by using micro-HPLC. The effects of electrical field and pressure on plate height and resolution were investigated. Influence of organic modifier, ionic strength and pH value of buffer on retention behavior were evaluated, and the separation mechanism was also discussed.
