Satellite DNA (satDNA) constitutes an important fraction of repetitive DNA in eukaryotic genomes, but it is barely known in most species. The high-throughput analysis of satDNA in the grasshopper Pyrgomorpha conica revealed 87 satDNA variants grouped into 76 different families, representing 9.4% of the genome. Fluorescent in situ hybridization (FISH) analysis of the 38 most abundant satDNA families revealed four different patterns of chromosome distribution. Homology search between the 76 satDNA families showed the existence of 15 superfamilies, each including two or more families, with the most abundant superfamily representing more than 80% of all satDNA found in this species. This also revealed the presence of two types of higher-order repeats (HORs), one showing internal homologous subrepeats, as conventional HORs, and an additional type showing non-homologous internal subrepeats, the latter arising by the combination of a given satDNA family with a non-annotated sequence, or with telomeric DNA. Interestingly, the heterologous subrepeats included in these HORs showed higher divergence within the HOR than outside it, suggesting that heterologous HORs show poor homogenization, in high contrast with conventional (homologous) HORs. Finally, heterologous HORs can show high differences in divergence between their constituent subrepeats, suggesting the possibility of regional homogenization.

The desert locust (Schistocerca gregaria) has been used as material for numerous cytogenetic studies. Its genome size is estimated to be 8.55 Gb of DNA comprised in 11 autosomes and the X chromosome. Its X0/XX sex chromo- some determinism therefore results in females having 24 chromosomes whereas males have 23. Surprisingly, little is known about the DNA content of this locust' sh uge chromo- somes. Here, we use the Feulgen Image Analysis Densitometry and C-banding techniques to respectively esti- mate the DNA quantity and heterochromatin content of each chromosome.WealsoidentifythreesatelliteDNAsusingboth restriction endonucleases andnext-generationsequencing. We then use fluorescent in situ hybridization to determine the chromosomal location of these satellite DNAs as well as that of six tandem repeat DNA gene families. The combination of the results obtained in this work allows distinguishing be- tween the different chromosomes not only by size, but also by the kind of repetitive DNAs that they contain. The recent publication of the draft genome of the migratory locust (Locusta migratoria), the largest animal genome hitherto se- quenced, invites for sequencing even larger genomes. S. gregaria is a pest that causes high economic losses. It is thus among the primary candidates for genome sequencing. But this species genome is about 50 % larger than that of L. migratoria, and although next-generation sequencing cur- rently allows sequencing large genomes, sequencing it would mean a greater challenge. The chromosome sizes and markers provided here should not only help planning the sequencing project and guide the assembly but would also facilitate assigning assembled linkage groups to actual chromosomes.

Most supernumerary (B) chromosomes are parasitic elements carrying out an evolutionary arms race with the standard (A) chromosomes. A variety of weapons for attack and defense have evolved in both contending elements, the most conspicuous being B chromosome drive and A chromosome drive suppression. Here, we show for the first time that most microspermatids formed during spermiogenesis in two grasshopper species contain expulsed B chromosomes. By using DNA probes for B-specific satellite DNAs in Eumigus monticola and Eyprepocnemis plorans, and also 18S rDNA in the latter species, we were able to count the number of B chromosomes in standard spermatids submitted to fluorescence in situ hybridization, as well as visualizing B chromosomes inside most microspermatids. In E. plorans, the presence of B-carrying microspermatids in 1B males was associated with a significant decrease in the proportion of B-carrying standard spermatids. The fact that this decrease was apparent in elongating spermatids but not in round ones demonstrates that meiosis yields 1:1 proportions of 0B and 1B spermatids and hence that B elimination takes place post-meiotically, i.e., during spermiogenesis, implying a 5-25% decrease in B transmission rate. In E. monticola, the B chromosome is mitotically unstable and B number varies between cells within a same individual. A comparison of B frequency between round and elongating spermatids of a same individual revealed a significant 12.3% decrease. We conclude that B chromosome elimination during spermiogenesis is a defense weapon of the host genome to get rid of parasitic chromosomes.

Simple sequence repeats (SSRs), also known as microsatellites, are one of the prominent DNA sequences shaping the repeated fraction of eukaryotic genomes. In spite of their profuse use as molecular markers for a variety of genetic and evolutionary studies, their genomic location, distribution, and function are not yet well understood. Here we report the first thorough joint analysis of microsatellite motifs at both genomic and chromosomal levels in animal species, by a combination of 454 sequencing and fluorescent in situ hybridization (FISH) techniques performed on two grasshopper species. The in silico analysis of the 454 reads suggested that microsatellite expansion is not driving size increase of these genomes, as SSR abundance was higher in the species showing the smallest genome. However, the two species showed the same uneven and nonrandom location of SSRs, with clear predominance of dinucleotide motifs and association with several types of repetitive elements, mostly histone gene spacers, ribosomal DNA intergenic spacers (IGS), and transposable elements (TEs). The FISH analysis showed a dispersed chromosome distribution of microsatellite motifs in euchromatic regions, in coincidence with chromosome location patterns previously observed for many mobile elements in these species. However, some SSR motifs were clustered, especially those located in the histone gene cluster.

In somatic cells colchicine promotes the arrest of cell division at prometaphase, and chromosomes show a sequential loss of sister chromatid arm and centromere cohesion. In this study we used colchicine to analyse possible changes in chromosome structure and sister chromatid cohesion in prometaphase I-arrested bivalents of the katydid Pycnogaster cucullata. After silver staining we observed that in colchicine-arrested prometaphase I bivalents, and in contrast to what was found in control bivalents, sister kinetochores appeared individualised and sister chromatid axes were completely separated all along their length. However, this change in chromosome structure occurred without loss of sister chromatid arm cohesion. We also employed the MPM-2 monoclonal antibody against mitotic phosphoproteins on control and colchicine-treated spermatocytes. In control metaphase I bivalents this antibody labelled the tightly associated sister kinetochores and the interchromatid domain. By contrast, in colchicine-treated prometaphase I bivalents individualised sister kinetochores appeared labelled, but the interchromatid domain did not show labelling. These results support the notion that MPM-2 phosphoproteins, probably DNA topoisomerase IIalpha, located in the interchromatid domain act as "chromosomal staples" associating sister chromatid axes in metaphase I bivalents. The disappearance of these chromosomal staples would induce a change in chromosome structure, as reflected by the separation of sister kinetochores and sister axes, but without a concomitant loss of sister chromatid cohesion.

Sister chromatid cores, kinetochores and the connecting strand between sister kinetochores were differentially silver stained to analyse the behaviour of these structures during meiosis in normal and two spontaneous desynaptic individuals of Chorthippus jucundus (Orthoptera). In these desynaptic individuals most of the chromosomes appear as univalents and orient equationally in the first meiotic division. Despite this abnormal segregation pattern, the changes in chromosome structure follow the same timing as in normal individuals and seem to be strictly phase dependent. Chromosomes in the first prometaphase have associated sister kinetochores and sister chromatid cores that lie in the chromosome midline; we propose that this promotes the initial monopolar orientation of chromosomes. However, the requirements of tension for stable attachment to the spindle force the autosomal univalents to acquire amphitelic orientation. Sister kinetochores behave in a chromosome orientation-dependent manner and, in the first metaphase, they appear to be interconnected by a strand that can be detected by silver impregnation, as seen in the second metaphase of wild-type individuals. The disappearance of the sister kinetochore-connecting strand, needed for equational chromatid segregation, however, can only take place in the second meiotic division. This connecting strand is ultimately responsible for the inability of chromosomes to segregate sister chromatids in the first anaphase.

The cohesin complex plays a key role for the maintenance of sister chromatid cohesion and faithful chromosome segregation in both mitosis and meiosis. This complex is formed by two structural maintenance of chromosomes protein family (SMC) subunits and two non-SMC subunits: an α-kleisin subunit SCC1/RAD21/REC8 and an SCC3-like protein. Several studies carried out in different species have revealed that the distribution of the cohesin subunits along the chromosomes during meiotic prophase I is not regular and that some subunits are distinctly incorporated at different cell stages. However, the accurate distribution of the different cohesin subunits in condensed meiotic chromosomes is still controversial. Here, we describe the dynamics of the cohesin subunits SMC1α, SMC3, RAD21 and SA1 during both meiotic divisions in grasshoppers. Although these subunits show a similar patched labelling at the interchromatid domain of metaphase I bivalents, SMCs and non-SMCs subunits do not always colocalise. Indeed, SA1 is the only cohesin subunit accumulated at the centromeric region of all metaphase I chromosomes. Additionally, non-SMC subunits do not appear at the interchromatid domain in either single X or B chromosomes. These data suggest the existence of several cohesin complexes during metaphase I. The cohesin subunits analysed are released from chromosomes at the beginning of anaphase I, with the exception of SA1 which can be detected at the centromeres until telophase II. These observations indicate that the cohesin components may be differentially loaded and released from meiotic chromosomes during the first and second meiotic divisions. The roles of these cohesin complexes for the maintenance of chromosome structure and their involvement in homologous segregation at first meiotic division are proposed and discussed.

The mode of inheritance of supernumerary segments located on three different chromosome pairs was investigated in controlled crosses with specimens of the grasshopper Chorthippus jacobsi. While extra segments located on chromosomes M5 and M6 showed Mendelian inheritance, that on S8 did not. Thus, the two supernumerary heterochromatic chromosome segments located distally on the S8 chromosome accumulated through non-Mendelian transmission through both sexes. The observed transmission patterns may be explained by gametic selection for spermatozoa carrying segmented S8 chromosomes, in addition to meiotic drive for segmented S8 chromosomes in heterozygous females. The significance of these findings for the maintenance of these polymorphisms in natural populations is discussed.

Kinetochores and chromatid cores of meiotic chromosomes of the grasshopper species Arcyptera fusca and Eyprepocnemis plorans were differentially silver stained to analyse the possible involvement of both structures in chromatid cohesiveness and meiotic chromosome segregation. Special attention was paid to the behaviour of these structures in the univalent sex chromosome, and in B univalents with different orientations during the first meiotic division. It was observed that while sister chromatid of univalents are associated at metaphase I, chromatid cores are individualised independently of their orientation. We think that cohesive proteins on the inner surface of sister chromatids, and not the chromatid cores, are involved in the chromatid cohesiveness that maintains associated sister chromatids of bivalents and univalents until anaphase I. At anaphase I sister chromatids of amphitelically oriented B univalents or spontaneous autosomal univalents separate but do not reach the poles because they remain connected at the centromere by a long strand which can be visualized by silver staining, that joins stretched sister kinetochores. This strand is normally observed between sister kinetochores of half-bivalents at metaphase II and early anaphase II. We suggest that certain centromere proteins that form the silver-stainable strand assure chromosome integrity until metaphase II. These cohesive centromere proteins would be released or modified during anaphase II to allow normal chromatid segregation. Failure of this process during the first meiotic division could lead to the lagging of amphitelically oriented univalents. Based on our results we propose a model of meiotic chromosome segregation. During mitosis the cohesive proteins located at the centromere and chromosome arms are released during the same cellular division. During meiosis those proteins must be sequentially inactivated, i.e. those situated on the inner surface of the chromatids must be eliminated during the first meiotic division while those located at the centromere must be released during the second meiotic division.

The distinctive behaviors of chromosomes in mitosis and meiosis depend upon differences in kinetochore position. Kinetochore position is well established except for a critical transition between meiosis I and meiosis II. We examined kinetochore position during the transition and compared it with the position of kinetochores in mitosis. Immunofluorescence staining using the 3F3/2 antibody showed that in mitosis in grasshopper cells, as in other organisms, kinetochores are positioned on opposite sides of the two sister chromatids. In meiosis I, sister kinetochores are positioned side by side. At nuclear envelope breakdown in meiosis II, sister kinetochores are still side by side, but are separated by the time all chromosomes have fully attached in metaphase II. Micromanipulation experiments reveal that this switch from side-by-side to separated sister kinetochores requires attachment to the spindle. Moreover, it is irreversible, as chromosomes detached from a metaphase II spindle retain separate kinetochores. How this critical separation of sister kinetochores occurs in meiosis is uncertain, but clearly it is not built into the chromosome before nuclear envelope breakdown, as it is in mitosis.

Univalents, that is, chromosomes lacking an attached partner at the first meiotic division, show extremely faulty transmission. Most segregational errors stem from amphitelic (mitotic-like) orientation at metaphase I followed by anaphase I lagging. Our studies in living grasshopper spermatocytes show that amphitelic orientation may provoke spindle collapse: spindle elongation and cytokinesis are impaired and an unreduced restitution nucleus is formed. This does not prevent meiotic progression and eventually leads to the production of diploid gametes. The morphology and characteristics of spindle collapse in our material, as revealed by in vivo observation and tubulin immunostaining, indicate an active role of the chromosomes in the whole process.

Plant (Secale cereale, Triticum aestivum) and animal (Eyprepocnemis plorans) meiocytes were analyzed by indirect immunostaining with an antibody recognizing histone H3 phosphorylated at serine 10, to study the relationship between H3 phosphorylation and chromosome condensation at meiosis. To investigate whether the dynamics of histone H3 phosphorylation differs between chromosomes with a different mode of segregation, we included in this study mitotic cells and also meiotic cells of individuals forming bivalents plus three different types of univalents (A chromosomes, B chromosomes and X chromosome). During the first meiotic division, the H3 phosphorylation of the entire chromosomes initiates at the transition from leptotene to zygotene in rye and wheat, whereas in E. plorans it does so at diplotene. In all species analyzed H3 phosphorylation terminates toward interkinesis. The immunosignals at first meiotic division are identical in bivalents and univalents of A and B chromosomes, irrespective of their equational or reductional segregation at anaphase I. The grasshopper X chromosome, which always segregates reductionally, also shows the same pattern. Remarkable differences were found at second meiotic division between plant and animal material. In E. plorans H3 phosphorylation occurred all along the chromosomes, whereas in plants only the pericentromeric regions showed strong immunosignals from prophase II until telophase II. In addition, no immunolabeling was detectable on single chromatids resulting from equational segregation of plant A or B chromosome univalents during the preceding anaphase I. Simultaneous immunostaining with anti-tubulin and anti-phosphorylated H3 antibodies demonstrated that the kinetochores of all chromosomes interact with microtubules, even in the absence of detectable phosphorylated H3 immunosignals. The different pattern of H3 phosphorylation in plant and animal meiocytes suggests that this evolutionarily conserved post-translational chromatin modification might be involved in different roles in both types of organisms. The possibility that in plants H3 phosphorylation is related to sister chromatid cohesion is discussed.

We describe novel nucleolar structures, observed by thin section electron microscopy in oocyte nuclei of the grasshopper Locusta migratoria, which we interpret, based on morphological and compositional criteria, as rDNA transcription units. Morphologically they resemble the condensed and foreshortened "Christmas trees" seen in Miller spreads of nucleolar chromatin prepared from the same biological material. They contain DNA and rRNA as shown by immunocytochemistry and in situ hybridization and are concentrated in several intranucleolar cavities. The presumptive rDNA transcription units extend throughout the interior of these nucleolar pockets or are selectively enriched at their outermost zones in close contact with the surrounding fibrillarin-positive dense component. We suggest that the nucleolar pockets of Locusta oocytes are equivalent to the fibrillar centers of somatic nucleoli and discuss possible implications for the current understanding of the functional organization of nucleoli.

Orientational movements and modes of segregation at anaphase I were analyzed in three different types of univalents in living spermatocytes of the grasshopper species Eyprepocnemis plorans, namely the sex univalent, three types of accessory chromosomes and spontaneous and induced autosomal univalents. When two or more univalents were present in the same spindle, their dynamics were directly compared. Chromosomes may show variable velocity and number of reorientations: the X and the most common B types (B1 and B2) are slow and rarely reorient, a more geographically restricted B (B5) is faster and reorients more often, and autosomal univalents are the fastest and show the highest frequency of reorientations. Nonetheless, the X and the accessories are rigorously reductional at anaphase I whereas autosomal univalents often fail to migrate or divide equationally. This indicates that orientational and segregational behavior are controlled mainly by chromosomal rather than cellular characteristics and that chromosomes may display a great variety of strategies to achieve regular segregation.

The two nucleolus organizing chromosome pairs of the grasshopper Pyrgomorpha conica can carry a proximal supernumerary heterochromatic segment. We employed different cytological techniques to characterize and analyze the possible origin of this segment. The supernumerary segment and the nucleolus organizing regions (NORs) show similar responses after C-banding plus either Giemsa or acridine orange, and chromomycin A3/distamycin A staining to detect GC-rich chromosome regions. Fluorescence in situ hybridization with a biotinylated rDNA probe demonstrated that the segment originated by amplification of the rDNA genes. However, as the silver staining indicates, the ribosomal genes present in the segment are not active since no nucleolus is formed. The use of in situ digestion with the isoschizomeric MspI and HpaII restriction endonucleases and subsequent Giemsa, ethidium bromide or chromomycin A3/distamycin A staining, suggests that the segment has been inactivated by DNA methylation.

We studied the orientation and segregation of a particular quadrivalent in living grasshopper spermatocytes. Quadrivalents were detached from the spindle by micromanipulation, then placed and bent as desired. The detached quadrivalents reattach and orient on the spindle. Their orientation is determined by the same principles that apply to ordinary chromosomes in mitosis and meiosis, but the outcome is different. Certain characteristics of the quadrivalent lead to a variety of orientations rather than the single one typical of ordinary chromosomes. Two kinetochores in the quadrivalent are linked to the others by unusually long, flexible chromosome arms. These kinetochores may face either the same pole or opposite poles and tend to orient initially to the pole toward which they face. Consequently, the initial orientation of the flexibly linked kinetochores is variable, and, moreover, they frequently reorient. In contrast, the other two kinetochores are as rigidly connected as those in a small bivalent and so display the typical back-to-back arrangement. Usually, this arrangement leads quickly to a stable orientation of the two kinetochores to opposite poles. Sometimes, however, the back-to-back arrangement changes to a side-by-side arrangement so that the orientation of both kinetochores to the same pole is favored. The combined effect of this diverse behavior is that the quadrivalent has four stable orientations, each leading to a different distribution of chromosomes in anaphase. The result is genetic chaos. Ironically, this chaos is produced by the same mechanisms that, in ordinary bivalents and mitotic chromosomes, produce a single stable orientation and genetically appropriate chromosome distribution.

The hypothesis, proposed by Nolte, that all locusts undergo an increase in their chiasma frequency when swarming is examined in the Migratory Locust, Locusta migratoria, using (1) animals which have been isolated in the laboratory at hatching, (2) crowded and isolated animals supplied by an independent laboratory and (3) field solitaria material. Animals isolated in the laboratory at hatching did not show a reduced chiasma frequency when compared with crowded controls despite the fact that they did develop a solitaria phenotype. Likewise, crowded and isolated animals supplied by workers at the University of Paris showed no difference in their chiasma frequency. Finally, animals living at very low density on the flood plains of the River Niger in Mali, West Africa, and exhibiting extreme phase solitaria characteristics proved to have very high chiasma frequencies which is the exact antithesis of the hypothesis. — Thus there is no evidence that there is any relationship between phase status or density and chiasma frequency in Locusta migratoria. This is in direct contradiction to Nolte's claims but in agreement with a previous study by the present author on Schistocerca gregaria.

Individual living cells in metaphase were exposed to a steep temperature gradient by placing a microheater near one spindle pole. The cells were then fixed and the spindle was examined by electron microscopy. The structure of the warmer half-spindle differed from the cooler half-spindle in several ways. Kinetochore microtubules were nearly parallel in the warmer half-spindle but were divergent in the cooler. The total length of microtubules in the warmer half-spindle was 52 per cent greater and the number of kinetochore microtubules per kinetochore averaged 16 per cent higher than in the cooler half-spindle. The warmer half-spindle was longer than the cooler. These observations clearly demonstrate a locally enhanced assembly of microtubules in the warmer half-spindle. The electron microscope study makes still clearer the unusual character of chromosome movement in the differentially heated cells: the structure of the warmer half-spindle is hard to distinguish from that in normal cells, yet chromosome movement there is far slower than normal (Nicklas, 1979).

Using a modified technique which allowed observation of chromosome orientation in the primary oocyte of grasshoppers at the onset of anaphase, it has been possible to establish that the B-chromosomes is distributed preferentially on the egg side of the metaphase plate rather than the polar body side. The frequency of this preferential orientation matches very closely the level of preferential transmission determined from breeding experiments using individuals from the same population. The spindle is asymmetrical in the primary oocyte of this species, and a possible explanation of the meiotic drive is proposed as a result of the conical shape of nucleoplasm surrounding this spindle. The autosomal chiasma frequency of these females is generally lower than comparable males and is increased by the presence of B chromosomes; but the ciasma frequencies of the sexes respond differently to the addition of 1 and 2 B-chromosomes.

Two mosaic sibling embryos of the Australian plague locust, Chortoicetes terminifera are reported with haploid and diploid cell lines in widely differing proportions. One small chromosome pair involved in the two cases has alternative morphology and a B-chromosome is present in one. In addition, G-banding identifies two medium-sized chromosome pairs and alternative states of a second small pair. Using these markers it is clear that both diploid cell lines are homozygous for the chromosomes of the corresponding haploid line. These embryos have thus developed by accidental parthenogenesis from haploid cells, some of which were duplicated by endomitosis after development began.

DNAs from three grasshopper species and three locust species (Family Acrididae, Order Orthoptera) have been characterised by a variety of biochemical techniques as part of a general investigation of their molecular evolution. Base compositions were inferred from thermal denaturation profiles and analytical CsCl and Cs2SO4 pcynography. Repeated sequence content of the genomes was studied by renaturation kinetics incorporating absorption measurements. Relative genome size was measured by microdensitometry of nuclear preparations.

The genus Caledia acontains two species. C. species nova 1 is restricted to the Oriomo Plateau of S. W. Papua and has a complement of twelve telocentric chromosomes. The second species C. captiva has a much wider distribution pattern--from S.W. Papua in the North, down the entire Eastern seaboard of Australia to Southern Victoria. It is also found in the Northern Territory. Although the chromosome number is the same as C. species nova 1, four major and distinct chromosomal races can be distinguished in C. captiva.--The basic "ancestral" race is found in Tropical North Queensland at the base of the Cape York Peninsula. All twelve chromosomes are telocentric and the karyotypic organization is similar to that found in C. species nova 1 and in other Acridines. A second, "general purpose" karyotypic race has a wide distribution between S. W. Papua, Arnhem Land and the East Australian coast as far South as Brisbane. It is considered a derivative form of the "ancestral" type and is fixed for small pericentric inversions on seven pairs of chromosomes. In the South-Eastern Queensland region there exists a further race which carries large pericentric inversions on all the autosomes and the X chromosome. The situation here is confounded since the basic chromosomes can be represented as either acro- or telocentrics. Various levels of polymorphism for the inversions exist between different chromosomes in different populations indicating considerable differentiation within this zone. This race is almost completely surrounded by the "general purpose" karyotype where the races are contiguous in certain parts of the range.--The South-Eastern corner of Australia is characterised by a chromosome race quite different from those found further North. Here a complex pericentric inversion system exists involving a series of seven small inversions and larger inversions on chromosomes 1, 2, 4 and 10. Chromosomes 2 and 4, in particular, are highly polymorphic.--The presence and persistence of these 4 chromosomal races can be accounted for in terms of the known climatic changes which have occured in this region in the recent past.

The formation of chiasmata in six full sib male partially asynaptic individuals of Locusta migratoria has been studied. The mean chiasma frequency per cell was 2.3 both at diplotene and metaphase I. Chiasmata tended to be distributed evenly among the bivalents. The frequency and distribution of the chiasmata in each type of bivalent (L, M, or S) depended on the level of asynapsis and on interference between the bivalents. Short bivalents were the most affected by interference, while M bivalents associated independently of L and S bivalent behaviour.

Additional samples from a discrete population of the pygmy grasshopper Tettigidea lateralis, at the periphery of the species range, have shown that the frequency of the B-chromosomes has remained stable over a two year period (30-35%), and that there is not significant difference for this metric in the two sexes. The intensity of the preferential movement of the B with the X at male first meiotic division has also remained constant in time and homogenous in different individuals. Hence it is possible that this distortional effect plays a role in the equilibrium frequency of the B in the population. The B's may possess special adaptive properties under ecologically marginal conditions, since in a number of more 'central' demes they occur at much lowere frequencies (7-9%). -Unique morphological and/or behavioral variants of the standard B were encountered in addition to distinct cases of spontaneous fragmentation of A elements. The meiotic behavior and chromatic expression of these centric fragments provide evidence on the possible origin and evolution of supernumeraries. A modified. Giemsa staining technique has been used to identify regions of centromeric (C-) hererochromatin in mitotic and meiotic chromosomes. The C-banding pattern of the X and the allocyclically similar B is compared. It suggested that the B may have originated from the X by deletion of centromeric heterochromatin. This may have affected the centromere "strength" of the nascent B leading to its preferential movement with the X at anaphase I.

Chromosome distribution has been investigated at metaphase and C-metaphase in neuroblast cells of Locusta migratoria embryos. There is no evidence for generalised somatic association of homologues or nucleolus organising chromosomes at either stage. Homologous chromosomes 9 are found closer together than expected in C-metaphase but this seems to be due to their tendency to lie in the centre of the C-metaphase squash. (Summary see p. 332.)

The basis for stable versus unstable kinetochore orientation was investigated by a correlated living-cell/ultrastructural study of grasshopper spermatocytes. Mal-oriented bivalents having both kinetochores oriented to one spindle pole were induced by micromanipulation. Such mal-orientations are stable while the bivalent is subject to tension applied by micromanipulation but unstable after tension is released. Unstable bivalents always reorient with movement of one kinetochore toward the opposite pole. Microtubules associated with stably oriented bivalents, whether they are mal-oriented or in normal bipolar orientation, are arranged in orderly parallel bundles running from each kinetochore toward the pole. Similar orderly kinetochore microtubule arrangements characterized mal-oriented bivalents fixed just after release of tension. A significantly different microtubule arrangement is found only some time after tension release, when kinetochore movement is evident. The microtubules of a reorienting kinetochore always include a small number of microtubules running toward the pole toward which the kinetochore was moving at the time of fixation. All other microtubules associated with such a moving kinetochore appear to have lost their anchorage to the original pole and to be dragged passively as the kinetochore proceeds to the other pole. Thus, the stable anchorage of kinetochore microtubules to the spindle is associated with tension force and unstable anchorage with the absence of tension. The effect of tension is readily explained if force production and anchorage are both produced by mitotic motors, which link microtubules to the spindle as they generate tension forces.

The X chromosome can be identified with the light microscope throughout all stages of the gonial cell cycle (including interphase) in the grasshopper Brachystola magna. At gonial mitotic stages the X chromosome gives the appearance of being undercondensed or negatively heteropycnotic. At interphase the X projects out from the body of the nucleus.--Examination with the electron microscope reveals that the X is compartmentalized at least two gonial cell cycles prior to the entry of the cells into meiotic prophase. The membrane layers that envelope the X chromatin at interphase remain associated with the X chromosome throughout gonial mitotic stages providing the ultrastructural basis for the apparent negative heteropycnosis observed with the light microscope.--The X chromosome is inactive in RNA synthesis during gonial mitotic stages but is hyperactive in RNA synthesis when compared to autosomes at gonial interphase.--X-chromosome condensation which reaches its maximum at premieotic interphase is initiated at or prior to the pre-pentultimate gonial division.

In 8 out of 20 Tasmanian populations of Phaulacridium vittatum from 0.3–11.0 percent of the males carried a single supernumerary chromosome. In such males the X and B univalents are both heteropycnotic at first prophase of male meiosis and associate with another in a non-homologous manner in about two-thirds of the diplotene cells examined. In all 56 B-containing individuals studied, however, these associations lapse by first metaphase and the X and the B move at random with respect to one another at first anaphase. The supernumerary in this species is stable and only 5 of the 56 individuals with supernumeraries showed evidence of B-chromosome non-disjunction in the pre-meiotic mitoses. Since there was no other evidence for the loss or gain of supernumeraries in the male line it is clear that B-transmission is regular in the males of this species. There were significant differences between populations with respect to mean cell chiasma frequency but there was no significant effect of the B-chromosome on this metric. Additionally a comparison of mean cell chiasma frequency in follicles with and without supernumeraries from a mosaic individual shows no significant difference.

In most animals, including grasshoppers, the X chromosome is heterochromatic (heteropycnotic) during prophase I and metaphase I of spermatogenesis. This report describes one grasshopper male in which at these states some of the X chromosomes contained an euchromatic (E) segment. In grasshoppers, the heteropycnotic state of the X is apparently established prior to the formation of the cysts. The spermatocytes containing the E segments, however, did not comprise whole cysts. It was concluded, therefore, that the E segments resulted from a localized euchromatinization rather than a failure to become heteropycnotic. The cytology of this male was unusual in two other respects. In most of the spermatocytes the chromosomes were longer and thinner than those of other males. In addition, in some of the cells undergoing meiosis, the cytoplasm failed to divide during both meiotic divisions and the resulting spermatids failed to differentiate into sperm. Because in this species both the presence of Xs with E segments and undercondensation are very rare and both involve condensation, it is likely that they are in some way related. Evidence for and against the possibility that the E segments were genetically active and that this activity led to the arrest of some of the spermatids is discussed.

1. In a sample of 27 males and 15 females from an inbred culture of Pyrgomorpha kraussi four classes of individuals could be distinguished. Those: a) With a standard chromosome complement, 2n = 18 + XX/XY, b) Where the standard complement was supplemented by a single B chromosome, c) Presumably polysomic for structurally altered or unaltered members of the A complement, and d) As in c) but with a B chromosome. 2. Abnormal chromosome behaviour indicative of genotypic unbalance was found in twelve males. 3. Abnormality consists in the occurrence of: a) Polyploid cells, b) Aneuploid cells, c) Chromosome breakage, and d) Unstained regions. The last three anomalies affect only the A chromosomes. 4. There is considerably variation both within and between individuals in respect of these anomalies. But the number of stable males in the B chromosome class is significantly higher than in other classes and all the males with S + B are stable. 5. It is argued that metabolic DNA, and consequently heterochromatic B chromosomes, are important in relation to the mechanical aspects of chromosome behaviour. 6. The B chromosomes are of unknown origin but the extra A chromosomes in aneuploid cells rarely form multivalents and thus one of the conditions of independent evolution is partially fulfilled.

Nine individuals of Phaulacridium vittatum in a single population sample of 1250 males collected at Crookwell, N. S.W. contained a single supernumerary chromosome. One further individual had two B-chromosomes. The supernumerary in question was larger than any of the standard set and was distinguishable from them at all stages of mitosis and meiosis. Like the X-univalent the B-chromosome was heteropycnotic at the onset of meiosis but differed from it in size, structure and behaviour. During first prophase single supernumeraries were frequently associated with the X in a non-homologous fashion as a consequence of their precocity. All such associations lapsed before first metaphase and the X and the B moved at random with respect to one another at first anaphase. In the one individual with two supernumeraries the two B-chromosomes showed regular pairing and segregated in a conventional manner. In all these respects the Crookwell supernumerary differed markedly from a morphologically identical B-chromosome present in a population of the same species from Hobart, Tasmania studied by Jackson and Cheung in 1967. Whether these differences in the behaviour of the B-chromosome in the two populations determines the higher frequency of the supernumerary in Hobart (11.3%) as compared with Crookwell (0.8%) remains to be resolved.

Microdissection studies of the dividing neuroblast of the grasshopper,Chortophaga viridifasciata, lead to the following conclusions: 1. The spindle at metaphase is a semisolid body situated in a fluid cytoplasm; it consists largely of longitudinally oriented components; it appears to be attached at its poles to the adjacent region of the plasma membrane by weak astral fibers. From prometaphase through early anaphase the chromosomes are securely attached to the spindle at their centromeres. The spindle develops during prometaphase from the fluid karyolymph; during anaphase it undergoes liquefaction, which begins in the equatorial region and spreads poleward with or slightly in advance of the chromosome centromeres. By late anaphase the chromosomes at each pole are held firmly in a circle at their proximal ends. 2. The interzonal region of the anaphase cell consists of fluid protoplasm traversed by fine, invisible interzonal connections joining the distal ends of sister chromosomes. Its elongation during anaphase is an intrinsic process that is independent of any attachment of the spindle to the adjacent cell membrane by astral rays and appears to be responsible for much of the anaphase separation of sister chromatids. 3. Early anaphase separation of sister chromatids begins at the centromere and progresses distally. 4. Polarization of the neuroblast is largely independent of the spindle and chromosomes. 5. Formation of the cleavage furrow is independent of the spindle and chromosomes, but its position depends to a limited extent on their position at late anaphase.

When Locusta migratoria spermatocytes are surface-spread on various salines, the axial element of leptotene and zygotene chromosomes, and the synaptonemal complex of pachytene chromosomes are well-preserved, although, in most instances, virtually denuded of chromatin. A complex association of chromosome ends with the nuclear membrane is apparent as early as leptotene, and, as pairing is initiated, the nuclear attachment points of the partner half-bivalents fuse, apparently incorporating additional membrane material between them. The meiotic kinetochore originates in association with the axial element during early prophase, and prior to synaptonemal complex formation and chromosome condensation.

1. Eyprepocnemis plorans meridionalis and E. p. ornatipes have a standard acridoid complement of 2n=22+XO acrocentric chromosomes in the male. Meiosis in them is normal and the former has a higher chiasma frequency (mean Xta per cell 13.95 v. 13.33). 2. Reciprocal first generation hybrids between them are described as are hybrid-derivatives produced by the back-cross (E.p.m.xE.p.o.) ♀ x E. p. o ♂. Meiosis in these is characterised by the formation of univalents, asymmetrical bivalents, multiple associations and occasionally, bridges and fragments. 3. The unequal bivalents and some of the multiple associations provide clear evidence for structural hybridity of the interchange type. At least 4–6 such differences appear to obtain between the parental “sub-species” which are thus sibling-species. 4. The number of different end-associations in which a given chromosome can be involved is too high to be accounted for on the basis of interchange alone. If these associations, which very frequently involve the extremely short arms, are chiasmate, then they indicate a multiplicity of end homologies in the parental forms. We, however, incline to the view that they are essentially non-homologous and non-chiasmate, owing their existence to the peculiar genotypic conditions of the hybrids coupled with their structural hybridity. 5. The role of interchange hybridity in the origins of species is discussed. It is argued that where interchanges involve acrocentric chromosomes in particular, then the establishment of the change in the homozygous condition cannot be preceded by an interim balanced polymorphism. 6. The structural differences described here need not have been directly involved in the differentiation of the two subspecies between which they were found. They may have been concerned in the creation of the 2 other sub-species of the species, or the 18 other species which are currently recognised in the genus.
