Purpose: The objective of this study was to determine the responses of normal meniscus to collagenase activity. It was hypothesized that meniscal explants exposed to collagenase would significantly increase release of pro-inflammatory cytokines and degradative enzymes, in a dose-dependent manner, compared to control.Methods: Menisci were harvested from adult dogs (n = 6) euthanized for reasons unrelated to this study. Meniscal explants were created from the central portion of lateral and medial meniscus. Explants were injected with 100 µl collagenase at a concentration of 50 µg/ml, 5 µg/ml, or 0 µg/ml of collagenase. Explants were cultured for 12 days, and media were changed and collected every 3 days for biomarker analyses. Differences among collagenase concentrations were determined by a three factor ANOVA with adjustment for multiple comparisons, with pre-adjustment statistical significance set at p < 0.05.Results: When data from all explants were compared, the 50 µg group released significantly higher IL-6 and PGE2, and the 5 µg group released significantly higher levels of MMP-3 and CTX-II compared to the 0 µg group. Explants from the medial meniscus released significantly more MMP-1, MMP-2, MMP-3, and MMP-13 in response to stimulation with 5 µg/ml of collagenase compared to explants from the lateral meniscus.Discussion: The data from this study indicate that in response to localized degradative enzyme activity, the meniscus increases the release of pro-inflammatory and degradative biomarkers in a dose-dependent manner. Further, these data indicate potential differences in metabolic responses of lateral versus medial menisci to collagenase insult.

Purpose Treatment of meniscus tears is a persistent challenge in orthopedics. Although cell therapies have shown promise in promoting fibrocartilage formation in in vitro and preclinical studies, clinical application has been limited by the paucity of autologous tissue and the need for ex vivo cell expansion. Rapid dissociation of the free edges of the anterior and posterior meniscus with subsequent implantation in a meniscus lesion may overcome these limitations. The purpose of this study was to explore the effect of rapidly dissociated meniscus tissue in enhancing neotissue formation in a radial meniscus tear, as simulated in an in vitro explant model. Materials and methods All experiments in this study, performed at minimum with biological triplicates, utilized meniscal tissues from hind limbs of young cows. The effect of varying collagenase concentration (0.1%, 0.2% and 0.5% w/v) and treatment duration (overnight and 30 minutes) on meniscus cell viability, organization of the extracellular matrix (ECM), and gene expression was assessed through a cell metabolism assay, microscopic examination, and quantitative real-time reverse transcription polymerase chain reaction analysis, respectively. Thereafter, an explant model of a radial meniscus tear was used to evaluate the effect of a fibrin gel seeded with one of the following: (1) fibrin alone, (2) isolated and passaged (P2) meniscus cells, (3) overnight digested tissue, and (4) rapidly dissociated tissue. The quality of in vitro healing was determined through histological analysis and derivation of an adhesion index. Results Rapid dissociation in 0.2% collagenase yielded cells with higher levels of metabolism than either 0.1% or 0.5% collagenase. When seeded in a three-dimensional fibrin hydrogel, both overnight digested and rapidly dissociated cells expressed greater levels of collagens type I and II than P2 meniscal cells at 1 week. At 4 and 8 weeks, collagen type II expression remained elevated only in the rapid dissociation group. Histological examination revealed enhanced healing in all cell-seeded treatment groups over cell-free fibrin controls at weeks 1, 4, and 8, but there were no significant differences across the treatment groups. Conclusions Rapid dissociation of meniscus tissue may provide a single-step approach to augment regenerative healing of meniscus repairs.

Tendinopathy is a common clinical pathology found in athletes and workers with mixed treatment results. Piperine, a major alkaloid found in the black and long pepper, has been demonstrated to have variety of pharmacological properties such as analgesic and anti-inflammatory effects. The present study was designed to investigate the effects of piperine on collagenase-induced Achilles tendon injury. Rats were intratendineously injected with collagenase in the right Achilles tendon, followed by intragastrical administration of piperine (100 mg/kg). Morphological structure and biochemical analysis of glycosaminoglycans, hydroxyproline, collagen III, and the activity of matrix metallopeptidases in the tendon tissues were performed. Our results showed that collagenase injection resulted in clear degenerative changes in the tendon. Administration of piperine improved the morphological structure of tendon, increased glycosaminoglycans and hydroxyproline levels, and inhibited the expression and activities of MMP-2 and MMP-9. Furthermore, piperine inhibited the activation of ERK and p38 signaling pathways in injured tendon. These results indicate a beneficial role of piperine against collagenase-induced tendon injury.

Tendinopathy is a significant clinical problem that can result from repetitive activity. While the precise etiology of this condition remains unclear, the cellular response to cyclical loading is believed to have a contributory role to the pathology of tendinopathy. This study examined the short-term biochemical response of avian flexor digitorum profundus tendon to repetitive cyclic loadings of varying magnitude. An in vitro tendon explant model was utilized to apply four levels of haversine tensile stress (peak stress of 0, 3, 12, and 18 MPa) at 1.0 Hz, 8 hr/day for 3 days. The 12 and 18 MPa levels were known to cause significant mechanical damage based on previous work. Tissue media was recovered and analyzed for prostaglandin E(2) (PGE(2)), lactate dehydrogenase (LDH, measure of cell death), and collagenase levels. Tissue samples were recovered and analyzed for cell viability, total collagen, and sulfated glycosaminoglycan content. Collagenase, LDH, and PGE(2) levels were found to be influenced by loading magnitude (p0.05) with higher levels being present at higher load magnitudes. Varying cyclical load magnitude caused minimal compositional changes as collagen content and glycosaminoglycan did not change. These results indicate that elevated cyclical mechanical loading of tendon quickly results in altered biochemical tissue responses indicative of tissue injury. More sustained cyclical loading over time may be required for these initial responses to induce more dramatic tissue changes as observed in clinical tendinopathy.

In this study, optical coherence tomography (OCT) with an axial resolution of 15 mum was used to investigate the depth and degeneration dependences of the refractive index (RI) of articular cartilage collected from bovine patellae in vitro. Eighteen disks of articular cartilage with a diameter of 6.35 mm harvested from different patellae were successfully prepared. Each disk was cut into two halves and three horizontal cartilage slices (n = 18 x 2 x 3) with an approximately equal thickness of 0.5 mm were further prepared from each half disk. The cartilage slices were digested by two different enzymes, collagenase and trypsin, to disturb collagen fibrils and proteoglycans, respectively. The samples were submerged in the physiological saline and tested using OCT before and after the enzyme digestion and the RI for each specimen was calculated. The RI of articular cartilage from the superficial to deep regions was 1.361 +/- 0.032 (mean +/- SD), 1.338 +/- 0.036, and 1.371 +/- 0.041 for normal specimens; 1.357 +/- 0.036, 1.331 +/- 0.030, and 1.392 +/- 0.037 for trypsin digested specimens; and 1.361 +/- 0.032, 1.336 +/- 0.048, and 1.376 +/- 0.043 for those treated by collagenase, respectively. Two-factor repeated measure ANOVA revealed that for all the three groups of specimens, the RI in different depths was significantly different (p0.05). However, we found that the trypsin and collagenase treatments did not exert a significant effect on the RI (p0.05). The results suggested that the depth dependence of articular cartilage should be taken into account when OCT is used for related measurement.

Matrix metalloproteinases (MMPs) and their specific tissue inhibitors (TIMPs) participate into extracellular matrix degradation in physiological and pathological conditions. We hypothesized that MMP expression in pulp tissue changes in response to caries attack and investigated the gene expression profiles of MMPs and TIMPs in pulp tissue of sound and carious teeth with cDNA microarray. cDNA microarray demonstrated an extremely high MMP-13 (collagenase-3) mRNA expression in pooled pulp samples of sound and carious teeth, with less pronounced expression of MMP-16 (MT3-MMP) and TIMP-1. Real-time quantitative polymerase chain reaction of individual pulp samples revealed a wide range of the MMP-13 expression level between pulp samples with possible downregulation of MMP-13 expression during caries progression. Western blot and immunohistochemical staining confirmed the presence of MMP-13 with no observable differences between sound and carious teeth pulp tissues. The results reveal that MMP-13 is expressed and synthesized in pulp tissue, an interesting feature considering the very limited expression of MMP-13 in normal adult tissues. Further studies with a larger sample size are needed to clarify the changes in MMP-13 expression during caries progression.

The substrate spectrum of the tandem collagen-binding domain (CBD) of Clostridium histolyticum class I collagenase (Co1G) was examined both in vitro and in vivo. CBD bound to insoluble type I, II, III and IV collagens in vitro, and to skin, aorta, tendon, kidney. trachea and corneal tissues containing various types of collagen fibrils or sheets. CBD hound to all kinds of collagen fibrils regardless of their diameters and also bound to sheet-forming collagen in the glomerular basal lamina or Descemet's membrane of the cornea. This wide substrate spectrum expands possible applications of the drug delivery system we proposed previously (PNAS 95:7018-7023, 1998). Therapeutic agents fused with CBD will hind not only to subcutaneous tissues, but also to other tissues containing non-type I collagen.

Based on the similarity of fibril diameters in healing and grafted ligaments, it has been speculated that all small fibrils represent newly synthesized collagen. Alternatively, small fibrils in grafts could be due to enzymatic degradation of endogenous large fibrils. This study examined the effect of collagenase on collagen fibril diameters in normal NZW rabbit MCLs. Midsubstance MCL slivers were incubated in buffer for 72 or 144 h for comparison with slivers incubated in buffer containing 4 units/ml bacterial collagenase. The samples were examined under TEM for fibril diameter analysis. Mean fibril diameters of 3-day and 6-day collagenase-treated MCLs were significantly reduced, resembling 40-week scar values. These results suggest that collagenase treatment can alter collagen fibril diameter and shape in normal rabbit MCL, thus it is possible that despite their similarity to ligament scars, that at least some small fibrils in ligament grafts may be enzymatically reduced endogenous fibrils.

The Achilles tendon is a frequent site for degeneration, and advanced understanding of this pathology requires an animal model that replicates the human condition. The aim of this study was to explore whether intratendinous collagenase injection combined with treadmill running created a pathology in the rat Achilles tendon consistent with human Achilles tendinosis. Collagenase was injected into one Achilles tendon of 88 high-capacity running (HCR) rats, which were randomized into treadmill running and cage control groups. Running animals ran at speeds up to 30 m/min on a treadmill at a 15° incline for up to 1 h/d, 5 d/week for 4 or 10 weeks. Cage control animals maintained cage activity. Collagenase induced molecular, histopathological and mechanical changes within the Achilles tendon at 4 weeks. The mechanical changes persisted at 10 weeks; however, the histopathological and majority of the molecular changes were no longer present at 10 weeks. Treadmill running had minimal effect and did not exacerbate the collagenase-induced changes as there were no statistical interactions between the interventions. These data suggest combined intratendinous collagenase injection and treadmill running does not create pathology within the Achilles tendon of rats selectively bred for HCR that is consistent with human Achilles tendinosis.

The levels of collagenase inhibitor, both free and bound to metalloproteinases, were evaluated at 7 days [deposit phase (DP)] and 14 days [resorptive phase (RP)] of evolution of the subcutaneous carrageenin-induced granuloma in the guinea pig. The level of free collagenase inhibitor was considerably higher in the supernatant of DP granulomas (7.95 +/- 1.53 U/mg protein) as compared to that of RP granulomas (2.53 +/- 0.41 U/mg protein). When the samples were heated at acid pH to release the inhibitor from metalloproteinase-inhibitor complexes, free inhibitor was recovered in both phases. However, the units of recovered collagenase inhibitor were several fold higher in all RP granulomas in comparison with DP granulomas (6.88 +/- 2.46 vs 1.5 +/- 0.53). Therefore, DP and RP tissues exhibited similar total amount of tissue inhibitor. By HPLC, collagenase inhibitor activity was localized in a fraction consistent with the size of TIMP. These results suggest a different balance of collagenase and collagenase inhibitor during the evolution of the granuloma; an excess of inhibitor over metalloproteinases appears to predominate during the phase of collagen accumulation contrasting with an inverse situation when the granuloma is healing.

In this study, we have identified and characterized metalloproteinases and tissue inhibitors of metalloproteinases (TIMPs) in human plasma. Treatment of plasma with trypsin or aminophenylmercuric acetate resulted in activation of latent gelatinolytic activity. Fractionation of plasma by gelatin Sepharose chromatography resulted in the isolation of 72 kDa and 92 kDa gelatinases/type IV collagenases. The 72 kDa gelatinase was purified by gel filtration chromatography. Stromelysin-1 was isolated from plasma by Matrex green A affinity chromatography. Immunoblotting of plasma fractions with antibodies to unique peptide regions of human gelatinases differentiated the 72 kDa gelatinase from the 92 kDa gelatinase. Antibodies to the amino terminal peptides of each enzyme were used to determine that plasma gelatinases circulate as latent proenzymes. Immunoblotting with antibodies directed against human stromelysin identified a 57 kDa stromelysin. TIMP-1 (28 kDa) and TIMP-2 (21 kDa) were also identified by immunoblotting of gelatin Sepharose bound plasma proteins using non-crossreacting antibodies to each protein.

Our laboratories have previously measured the capacity for collagenase production by normal and immobilized rabbit periarticular connective tissues (PCT), i.e., anterior cruciate ligaments (ACL), medial collateral ligament (MCL) and patellar tendon (PT). Rabbit limbs were immobilized for 4 weeks and then the tissues were cultured for 4 days. Collagenase and inhibitor (TIMP) were measured in the medium. Collagenase activity was significantly decreased in the medium from all experimental tissues, relative to the levels in contralateral control tissues. On the other hand there was no significant change in inhibitor levels in the three experimental tissues. These results support the idea that collagenase is decreased due to decreased enzyme expression and not to increased inhibitor production.

Acupuncture, a traditional Chinese therapeutic technique, has been put into practice for more than 4000 years and widely used for pain management since 1958. However, what is the mechanism underlying the acupuncture for analgesia effects by stimulation of acupoints, what substances receive the original mechanical acupuncture signals from the acupoints, or what transforms these signals into effective biological signals are not well understood. In this work, the role of collagen fibers at acupoints during acupuncture analgesia on rats was investigated. When the structure of the collagen fibers at Zusanli (ST36) was destroyed by injection of type I collagenase, the needle force caused by the acupuncture declined and the analgesic effects of rotation or lift-thrusting manipulations was attenuated accompanying the restraint of the degranulation ratios of mast cells. We propose that collagen fibers play an important role in acupuncture-induced analgesia, and they participate in signal transmission and transform processes.

Activation of procollagenase constitutes a crucial event in collagenolytic activity regulation. In this study we have purified by DEAE-cellulose, Ultrogel AcA-44, and zinc chelate sepharose chromatographies, a procollagenase-activator from the culture medium of the guinea pig carrageenin granuloma model. On SDS-PAGE, the activator migrates as a principal band of Mr approximately 44,000. The molecule activates procollagenase from human lung fibroblasts in a concentration dependent manner and an enhancement of collagenase activity of trypsin-treated crude culture medium was observed. A loss of about 50% of its activity occurs after heating. In addition, this activator degrades gelatin and casein. All these data suggest that this procollagenase-activator might be stromelysin. The activator was found in both phases of the granuloma, at 7 days when collagen is actively deposited and an important proportion of the collagenolytic activity remains in latent form; and at 14 days, when this enzymatic activity is fully expressed.

Metalloproteinases that degrade extracellular matrix molecules play important roles in development and progression of various diseases. Among them, collagenases are unique as they have an ability to degrade triple helical interstitial collagens into 3/4 and 1/4 fragments, a crucial step for collagenolysis in the tissue. Collagenases, consisting of a catalytic domain and a hemopexin domain, requires both domains for collagenolysis. The enzymes unwind triple helical collagen before they hydrolyze the peptide bonds. Aggrecanases are also multidomain metalloproteinases belonging to the ADAMTS family, and the noncatalytic ancillary domains also play an important role in recognition of aggrecan and their activities. Attenuation of collagenase and aggrecanase activities will be achieved by inhibitors or antibodies that interact directly with those noncatalytic ancillary domains (exosite inhibitors). Such molecules will be attractive for therapy as they will be highly selective because they are based on the unique mechanism of each proteinase.

This study examines the similarities and differences in the noncollagenous domain (NC1) of type IV collagen from human glomerular basement membrane (hGBM), alveolar basement membrane (hABM), and placenta (hPBM). Following collagenase digestion, NC1 domain was isolated on Bio-Gel A-0.5m or by cation exchange chromatography on S-Sepharose. NC1 from each source was characterized by SDS PAGE, and two dimension NEPHGE/SDS PAGE. Immunoblotting and ELISA inhibition was performed using antibody probes specific for M28 , M28+, M26 and M24 monomer subunits of human NC1. It was observed that all NC1 subunits were present in hGBM and hABM derived material, however M28 and M28+ monomers were absent in hPBM NC1. These findings indicate that while alpha 1(IV) and alpha 2(IV) collagen chains are present in hGBM, hABM and hPBM, alpha 3(IV) and alpha 4(IV) collagen chains are only found in hGBM and hABM but are absent in hPBM. It can now be appreciated that heterogeneity of alpha (IV) chain composition exists in basement membranes from various organs.

We demonstrated previously that growth promoting factors in general could induce the secretion of interstitial collagenase into the medium of human fibroblast cells (HF). In this study, the effect of tumor necrosis factor-alpha (TNF-alpha) on the induction of collagenase and tissue inhibitor of metalloproteinases (TIMP) was examined. Stimulation of quiescent HF cells with 10 ng/ml TNF-alpha induced the secretion of Mr 57,000, 52,000 procollagenases into the medium. The collagenase activity was elevated 2.8-fold after TNF-alpha treatment. Northern blot analysis of the steady-state mRNA indicated a tenfold elevation of collagenase transcript after 24 h treatment with 10 ng/ml TNF-alpha. The increase in collagenase mRNA was due to transcriptional activation of collagenase gene activity. TIMP mRNA level increased three-fold after TNF-alpha treatment. The activity of TNF-alpha on collagenase and TIMP induction may play an important role in tissue inflammatory, repair and remodeling processes after wound and injury.

We investigated whether different protocols for the digestion of adult human articular cartilage influence the cell yield and capacity to attach and proliferate in culture dishes. Chondrocyte yields were expressed as a percentage of the total number of cells in the tissue, determined both histologically (using the dissector method) and biochemically (measuring the DNA content of tissue digests). Human cartilage specimens (n = 79) were digested using different protocols based on combinations of collagenase II (CGN), trypsin/EDTA, hyaluronidase, and tosyllysylchloromethane (TLCM). Yields of viable chondrocytes were the highest within a specific range of CGN concentrations and digestion times, but always22% of the total available cells. The combination of CGN with trypsin/EDTA or TLCM accelerated the digestion process but did not significantly increase cell yields. The percentage of viable cells that attached to culture dishes ranged 75-85% (19% of the total) and was reduced by TLCM. Doubling times of attached cells were comparable in all experimental groups. Our results indicate that chondrocyte yields and capacity to attach and proliferate are not highly sensitive to the specific isolation protocol used. However, typically used cartilage digestion protocols yield only a small fraction of the total available cells, possibly introducing an uncontrolled selection of certain chondrocyte subpopulations.

The matrix metalloproteinases collagenase (MMP-1) and stromelysin (MMP-3) are often coordinately expressed, and their promoters contain similar regulatory elements, including an AP-1 site at about -70. There are, however, additional sequences including an adjacent ETS site at about -90 in both promoters, and a NIP (nuclear inhibitory protein) binding site in the stromelysin promoter. In this paper, we have investigated the role of these elements in transcriptional activation by phorbol myristate acetate (PMA). Using mobility shift assays, we demonstrate that in the collagenase promoter, PMA induction requires the binding of nuclear proteins to the ETS site as well as to the adjacent AP-1 element. In the stromelysin promoter, we used mutational analysis and DNA/protein interactions to illustrate a role for a single ETS site and for the NIP element in phorbol induction. These data suggest that ETS elements interact with other cis-acting sequences in these promoters to elicit transcriptional activation, and that the placement of the ETS sites in these promoters may influence transcriptional activity.

Since hyaluronan anchors the proteoglycan-rich pericellular matrix to chondrocytes, hyaluronan-cell interactions may direct cartilage matrix assembly. To test this hypothesis, the competitive binding of hyaluronan hexasaccharides for native hyaluronan during matrix assembly, accumulation and retention in embryonic cartilage was studied. Chondrocytes released from explants with collagenase P retained pericellular matrices, but chondrocytes appeared "matrix-free" when released from hexasaccharide-treated explants. Decreased safranin O staining was also observed in the hexasaccharide-treated explants. This loss of proteoglycan retention was demonstrated quantitatively in the cartilage extracts and recovered in the media. The continual presence of hexasaccharides in micromass cultures resulted in decreased proteoglycan deposition. Increased proteoglycan retention, indicative of matrix repair, occurred following hexasaccharide wash-out. Thus, native hyaluronan-chondrocyte interactions are important for the assembly and maintenance of cartilage matrix.

Glomerular hypertension has been considered to play an important role in the development of glomerulosclerosis. Mesangial expansion, a precursor of glomerulosclerosis, may be dependent not only on synthesis of matrix but also on degradation of the deposited matrix. In the present study we investigated the effect of simulated glomerular pressure on 72 kDa metalloproteinase activity (measured as degradation of gelatin) of mesangial cells. To simulate glomerular pressure conditions we have used an in vitro mechanical stretch/relaxation system as well as an alternative system which allowed direct application of pressure on mesangial cells. Mesangial cells were grown either under control (no added pressure), stretch/relaxation, or direct mean pressure of 45-50 mm Hg (physiologic glomerular pressure) conditions. Mesangial cells grown under stretch/relaxation showed a higher (P0.02) level of 72 kDa metalloproteinase activity when compared with control cells (control, 6.9 +/- 0.8 vs. stretch, 9.9 +/- 0.5 ng gelatin degraded/micrograms protein). Similarly, direct stimulated glomerular pressure increased (P0.01) mesangial cell 72 kDa metalloproteinase activity when compared with control cells. The effect of mechanical strain on mesangial cell 72 kDa metalloproteinase activity peaked at the end of the third day (control, 4.5 +/- 1.1 vs. stretch 15.3 +/- 1.1 ng gelatin degraded/micrograms protein, P0.001). Both mechanical strain and direct pressure to mesangial cells modulated 72 kDa metalloproteinase activity in a dose dependent manner. Mechanical stretch equivalent to normal simulated glomerular pressure increased mesangial 72 kDa metalloproteinase activity by 88%; whereas mesangial cell 72 kDa metalloproteinase activity declined to basal levels at higher simulated glomerular pressure (equivalent to 100 mm Hg). Likewise, direct higher pressure (100 mm Hg) also lowered 72 kDa metalloproteinase activity to basal levels. These results indicate that higher glomerular pressure inhibits degradation of type IV collagen. This may cause an accumulation of extracellular matrix and may thus be contributing to the expansion of the mesangium.

One of the most consistent observations in abdominal aortic aneurysm (AAA) disease is the disorganization and disruption of elastin and other matrix components of the aortic wall. The enzymatic basis for the biochemical features of AAA has been investigated beginning with the demonstration on substrate gel enzymography of a typical "profile" of proteinase activities in AAA tissue extracts which degrade gelatin, casein and elastin. A recombinant TIMP-1 affinity column was developed and three of the elastolytic/caseinolytic activities with approximate molecular weights of approximately 80 kDa, approximately 50 kDa and approximately 32 kDa were partially purified from these extracts. Affinity for rTIMP-1 suggests that these enzymes are members of the matrix metalloproteinase (MMP) family. High molecular weight forms of two MMPs, collagenase (MMP-1) and stromelysin-1 (MMP-3), were also isolated from the AAA tissue on this column; active forms of MMP-1 could be demonstrated by immunoblotting techniques in this preparation under reducing conditions. Infiltrating inflammatory cells are known sources of these proteolytic activities; analysis of these cell populations in the aneurysmal aortic wall using fluorescence-activated cell counting revealed a fifty-fold increase in macrophages (a well-known source of matrix-degrading enzymes) as well as a significant increase in lymphocytes.

The cleavage rates of insoluble and acid-soluble bovine collagens by tissue collagenases from the cultures of C3H mouse mammary adenocarcinoma and from human rheumatoid synovial fibroblasts at 37 degrees C were compared by measuring dansyl (Dns)-IIe and Dns-Leu, amino-terminal residues at the cleaved sites after dansylation. The kinetic parameters, Kcat and Km, defined functionally, were as follows: for insoluble Achilles tendon collagen, Km values were 0.89 and 0.83 microM; Kcat/Km 39.2 and 317 microM-1 h-1. For acid-soluble skin collagen the values were 0.83 and 0.82 microM; 44.6 and 354 microM, respectively. The supernatants from the insoluble and acid-soluble collagens contained 28% and 92% of total Dns-IIe and 1.3% and 43% of total Hyp, respectively. Results showed that both tissue collagenases cleaved insoluble collagen almost as rapidly as they did reconstituted collagen fibrils. The cleavage of collagen was proportional to enzyme concentration. The presence of Dns-IIe and Dns-Leu in the supernatant arises from a second cleavage of the collagen molecule.

Extracts of cartilage have been reported to inhibit many serine proteinases and metalloenzymes. Such inhibition may be important in protecting cartilage against degradation by chondrocytic proteinases such as collagenase, stromelysin and by leukocytic proteases, such as elastase. We report here isolation and partial characterization of a 17-kD elastase inhibitor from 0.5 M NaCl extracts of both nasal septum cartilage and articular cartilage, which inhibits elastase and represents 0.08% of the weight of nasal cartilage and 0.002% of the weight of articular cartilage. The protein was highly specific for elastase and did not inhibit cartilage metalloproteinases, suggesting that it may be mainly directed toward protecting cartilage against leukocytic proteases. The inhibitor had a blocked amino-terminus, was high in serine and glycine and lacked carbohydrate. The ease with which the inhibitor was extracted from cartilage suggests that it may function in vivo as a highly abundant elastase inhibitor which is secreted into synovial fluid from cartilage. The inhibitor was shown to be synthesized by bovine articular cartilage in explant culture and nearly all of the metabolically labeled material was secreted into the culture media. The inhibitor cross-reacted with polyclonal antibodies to bovine neck ligament alpha-elastin and antibodies to the inhibitor reacted with bovine neck ligament elastin. The properties of this inhibitor are different than those of any other reported cartilage derived inhibitor.

Collagenolytic enzymes were quantitated by a method based on spectrophotometry of suspended reconstituted collagen fibrils. To obtain optically stable suspensions it was necessary to perform a short sonication of the aggregated fibrils at 10 degrees C. When fibrils were cleaved with mammalian fibroblast collagenase at 35 degrees C the triple helical collagen fragments (TCA and TCB) would uncoil spontaneously and the decreasing turbidity was used as an estimate of enzyme activity. The method is a specific collagenase assay since a possible cleavage in the non-helical parts of the collagen molecule with contaminating proteinases is without effect on the turbidity of the suspension and the collagen substrate is not converted to gelatin at 35 degrees C. After 1 h of incubation 0.2 U (equivalent to 0.2 micrograms) of fibroblast collagenase could be detected. In purification procedures with microbial collagenases many fractions were tested by overnight incubations in disposable cuvettes. Sealing of cuvettes with square silicone stoppers allowed rotation of enzyme-substrate mixtures directly in the cuvettes. Only standard laboratory equipment is required for this assay, which is not dependent on radiolabeling or preparation of specific immunologic reagents.

We demonstrated the cells producing collagenase and the time course of collagenase-production at early stages of wound healing, using histology and two immunohistochemical procedures on cross sections of rat skin harvested 0, 3, 5, 7 and 12 days after full-thickness incisions. A monospecific rabbit polyclonal antibody to neutral collagenase purified from rat myometrial cells was used to demonstrate collagenase production. Specificity of this reaction was confirmed by blocking the reaction with excess homogeneously purified antigen. Macrophages were simultaneously labelled using a mouse anti-rat monoclonal antibody recognizing exclusively mature macrophages. Intracellular collagenase was not reliably detectable at day 0, but was prominent at days 3 and 5 and thereafter declined. Double labeling technique showed occasional macrophages producing collagenase in the developing granulation tissue, but most cells labeled as macrophages were negative for collagenase. Most activity was found in fibroblasts adjacent to granulation tissue elements. Since the granulation tissue parallels revascularization in a dendritic pattern, a cross section at three days typically shows an annulus of collagenase-positive cells surrounding a branch of the active granulation tissue. At days 5, 7 and 12 after wounding the pattern of collagenase expression became indistinct as more tissue was involved in the granulation process. However, double-labelling for macrophages and collagenase showed the dichotomy between collagenase expression and presence of macrophages to persist. The finding that collagenase is produced in connective tissue adjacent to granulation tissue suggests an inductive process, possibly due to diffusion of cytokines produced by granulation tissue elements.

Using human heart fibroblasts (HHF), we studied the effect of basic fibroblast growth factor (bFGF) and transforming growth factor-beta (TGF-beta) on the gene expression of type I collagen, collagenase and tissue inhibitor of metalloproteinases (TIMP). Initially, treatment of HHF with bFGF alone (10 ng/ml) resulted in elevated secretion of collagenase into the culture medium. Subsequent treatment of HHF with TGF-beta in combination with bFGF suppressed collagenase secretion. Northern blot analysis reinforced this observation by revealing an enhancement of the steady-state mRNA level of collagenase in response to bFGF. In order to examine if the collagenase gene was affected by bFGF at the transcriptional level, transfection experiments were carried out with a plasmid containing collagenase promoter linked to chloramphenicol acetyltransferase gene (CAT). Basic FGF stimulated CAT activity by four-fold, indicating increased promoter activity whereas the combination of TGF-beta and bFGF resulted in decreased CAT activity. TGF-beta was shown to increase type I collagen and TIMP mRNA levels by 2.5- and 2.1-fold, respectively. These results suggest that TGF-beta and bFGF may play a pivotal role in regulating collagen metabolism in HHF.

Interactions between macrophages and articular surfaces were studied in an in vitro model which has been described before. Either stimulated peritoneal macrophages or a purified population of bone marrow macrophages were incubated with mice femoral heads which were either untreated or were digested with collagenase, trypsin or hyaluronidase prior to incubation. Scanning electron microscopy (EM) examination showed that macrophages attached to the surface and in their vicinity tags and fibers were visible. Transmission EM was used after labeling the surfaces with cationized ferritin employed as a sensitive marker to define the integrity of the articular surface. Alterations of the surface of various degrees of intensity were seen in all the sections examined. No adhering macrophages were found, due probably to detachment of cells during tissue processing for transmission EM. Attachment of macrophages to the surface and alterations of the latter were seen also when hyaluronic acid was added to the incubation medium or when the surfaces had been treated with hyaluronidase before incubation.

Commercial clostridial collagenase was chromatographed on DEAE cellulose and separated into two fractions, A and B. When each of these fractions was further resolved by isoelectric focusing, the collagenase activity was found in both cases to be in protein peaks, designated A-3 and B-2, centered around pH 5.5. Fractions A-3 and B-2 differed, however, in their ability to degrade a soluble form of collagen or to solubilize bovine tendon collagen. Although none of the purified fractions or the original commercial preparation released significant amounts of acid soluble peptides from casein, polyacrylamide gel electrophoresis in sodium dodecyl sulfate of the digest mixtures revealed that all except fraction B-2 partially degraded the casein. When the digestion was carried out in 0.1 % sodium dodecyl sulfate, all fractions but B-2 extensively degraded cytochrome cand catalase. To study the glycoproteins and transglutaminase substrates involved in healing, wounds were made in the skin of anesthetized rats with a biopsy punch. Explants made 1-5 days later were incubated with [3H]-labelled putrescine, fucose or proline. As compared with unwounded skin there was an increased incorporation of label which was greatest at 3 days. Incubation for various times suggests that the incorporation of fucose and proline is dependent on protein synthesis, whereas putrescine is incorporated into preformed proteins. Putrescine and fucose label polypeptides with an Mr of about 45,000 before and 14,000 after reduction. These correspond in size with the aminopropeptide of type III collagen. Other labelled material of higher molecular weight is partly degraded to similar polypeptides on collagenase digestion. Much of the [3H]putrescine in the polypeptides is in the form of gamma-glutamyl putrescine. It is hypothesized that isopeptide linkage of the aminopropeptide III occurs in wound healing.

A monospecific, polyclonal antibody to neutral collagenase purified from rat myometrial cells was used to examine decalcified sections of normal and rachitic rat long bones, including epiphyses, for presence of immunoreactive collagenase in situ. Reactive antigen was uniformly present in hypertrophic chondrocytes of articular and epiphyseal plate cartilage. Preincubation of antibody with excess homogeneously purified antigen blocked the staining, indicating specificity of the staining for the antigen. Reaction was present in the borders of the enlarging lacunae, suggesting functional importance of this enzyme in the process of lacunar enlargement prior to mineralization. No detectable enzyme was observed in osteoblasts, osteocytes, or osteoclasts. No difference between expression of collagenase by bone cells in rachitic or normal bone was seen. Thus, at the level of sensitivity of this procedure, neutral collagenase appears to function in mature rat bone only in enlarging chondrocyte lacunae.

Trypsin-activated collagenase activity was analyzed in media from keloid biopsies as well as keloid fibroblasts using radiolabeled guinea pig skin collagen as a substrate for collagenase digestion. Collagenase activity per tissue dry weight and DNA was similar in keloid and normal dermal biopsies; however, it was significantly elevated in keloid compared to normal dermal fibroblast media. In two paired cell lines where keloid and normal dermal fibroblasts were isolated from the same patient, relative collagen synthesis and trypsin-activated collagenase were increased to a similar extent in the keloid cell lines compared to normals. The biopsy data are in agreement with other findings, and further confirm the hypothesis that increased collagen deposition in keloids does not result from decreased collagen degradation. In vitro, however, the data presented here indicate that in some cell lines increased collagen synthesis may be associated with increased trypsin-activated collagenase.

Collagenolytic activity has been demonstrated in the early phase of chemical carcinogenesis of mouse skin following 3-methylcholanthrene application dropwise in acetone or painted on the skin in benzene. In addition very high levels of collagenase could be detected in mouse skin papillomas and carcinomas. In all the tissues investigated, collagenase activity was extracted from the 6000 X g sediment of tissue homogenates with 5 M urea in 50 mM Tris-HCl buffer, pH 7.5. After dialyzing the extract, the enzyme was precipitated with ammonium sulfate and the activity determined against 14C-collagen substrate in solution. This procedure was found suitable for the detection and estimation of collagenase activity in skin tissues with high turnover of collagen and thus offers an attractive alternative to tissue culture methods.

A small molecular weight structural glycopeptide was solubilized after collagenase digestion of the connective tissue capsule surrounding the 5-day sponge-implant of the rat. The major amino acids are one residue each of aspartic and glutamic acids, proline, hydroxyproline and alanine and two residues of glycine, and the carbohydrates are one residue each of glucose, xylose and hexosamine and two residues of mannose. The sum of the amino acid and carbohydrate residues gives a molecular weight of 1635. Dansylation of the glycopeptide produces a single strongly fluorescent yellow-orange amino-terminal spot, not positively identified. The solubilization of the granuloma glycopeptide by collagenase and its composition are suggestive of its association with an immature form of collagen in early granulation tissue.

A monospecific rabbit anti-rat uterus collagenase antibody has been prepared and used to study the distribution of the enzyme in the rat uterus during postpartum involution. Cryostat sections of rat uteri from 24 to 240 hours postpartum were stained by the indirect immunofluorescent method. Nonpregnant rat uterus revealed positive staining in basement membranes, in endometrial stroma, in perimuscular and in vascular connective tissue. During postpartum involution of the uterus two types of changes in uterine collagenase were observed: (1) variations in the distribution of the enzyme, which became selectively localized in the epithelial basement membrane and in the wall of small blood vessels, and (2) variations in the overall intensity of fluorescent staining, which decreased immediately after delivery and slowly increased back to nonpregnant levels in 5 days. The significance of these findings is discussed in relation to the mechanisms of control of collagenase activity in vivo.

98% of the collagen in mature connective tissue is in the form of insoluble collagen fibers, consisting of bundles of polymeric collagen (PC) fibrils. The enzymes concerned in connective tissue remodeling degrade PC rather than tropocollagen (TC). TC is the most usual substrate for collagenase assays, and we believe it is essential to employ PC in any study of the activity of collagenolytic enzymes. In order to facilitate the study of enzymic degradation of PC we have labelled PC with fluorescein iso-thiocyanate to produce F-PC fibrils, containing 5 fluorescein labelled epilson-NH2 groups of lysine per TC molecule within the PC. The fluorescent F-PC is degraded at the same rate as PC with the release of hydroxyprolyl peptides but has the great advantage that the solubilised F-peptides can be quantitated by their fluorescent emission. The technique is described in detail employing bacterial collagenase and mammalian collagenase preparations to illustrate the methodology. The advantages of the fluorescent technique over the collagenolytic assay methods currently in use are outlined.

Ruthenium red was used to stain microfibrils in rat aorta after incubation of the tissues with or without one of the enzymes trypsin, collagenase, phospholipase C, chondroitinase ABC, hyaluronidase or neuraminidase, or the reducing agent dithiothreitol. Microfibrils exhibiting periodicity of ruthenium red binding were associated with elastic laminae and collagen fibrils and appeared to attach these structures to each other as well as to basal lamina. Microfibrils in rat and human aorta demonstrated fibronectinlike immunoreactivity, therefore fibronectin may be a component of aorta microfibrils and important in the architecture of blood vessels.

Chondrosia reniformis sponge collagen, insoluble in its native form, was solubilized by chemical modification of lysyl residues. The solubilized sponge collagen had the same amino acid composition as insoluble collagen and the helicoidal tertiary structure was found by negative Cotton effect to be the same as in native vertebrate collagens. Achromobacter iophagus collagenase, a collagen specific protease, hydrolyzed the soluble sponge collagen. These experiments confirmed that the protein had the same structure as collagen.

The action of commercially available proteolytic enzymes on skeletal matrices was assessed by autoradiographic analysis of grain counts produced by H3-glycine and H3-histidine labeled tissues. In addition, the influence of formalin in fixing solutions on the digestibility of tissue proteins following collagenase and trypsin treatment was also assessed. The presence of formalin is not recommended for either collagenase or trypsin digestion of tissues, whereas Carnoy's fixative is suitable. The sensitivity of the autoradiographic method and standardization of experimental design and procedures allow for the determination of the optimal time required for digestion, as well as, the rate at which a tissue digestive reaction proceeds. Moreover, the method allows for an evaluation and comparison of the potency of enzymes intended for histologic and histochemical studies, using skeletal tissues as the testing model.

Protein accounted for an average of 8.7% w/w of the hyaluronic acid obtained from rooster comb dermis extracts and three types of peptide constituents appeared to be present. A few collagen-like fibers were closely associated with the hyaluronic acid when samples were examined in the electron microscope and collagenase treatment decreased the intrinsic viscosity from 7000-5000 ml/g to 3900-2700 ml/g. The quantities of collagen present, however, were too small to detect chemically with the methods employed. The major peptide consituent was readily separated from the hyaluronic acid by fractionation in a cesium chloride gradient or by treatment with pronase. The viscosity was decreased by the density gradient procedure but not by the pronase digestion. Repeated fractionation in a cesium chloride gradient decreased the intrinsic viscosity still further and a small peptide constituent with a high glycine and serine content remained associated with a hyaluronic acid. The data suggest that an interaction or entanglement with collagen fibers is responsible for the high viscosity of hyaluronic acid in this tissue extract and that the viscosity of purified hyaluronic acid preparations is dependent upon interactions between adjacent polysaccharide chains. Interactions between the major peptide constituent and polysaccharide chains or the small residual peptide component remaining with hyaluronic acid after extensive purification procedures, however, appear to be involved in some organized structure because the presence of the major peptide constituent minimized the decrease in viscosity that occurred when hyaluronic acid samples were lyophilized.

A neutral proteinase, distinct from collagenase, was isolated by gel-filtration from medium that had been conditioned by the culture of explanted porcine gingiva. This enzyme was shown to cleave the alpha 1 (I) chain carboxyterminal telopeptide in native collagen proximal to (i.e. nearer to the helix than) the lysyl residue at position 17 which is known to be important in the formation of covalent intermolecular cross-links. We refer to this activity as 'telopeptidase'. The enzyme had an apparent Mr of 70,000, as determined by gel-filtration. It was inhibited by ethylene diamine tetraacetic acid but not by N-ethyl-maleimide nor by phenylmethylsulphonyl fluoride. It is therefore probably a metalloproteinase. The pH optimum for this activity was 7.0-7.5. Incubation of the enzyme with fibrillar (acid-insoluble) calf-skin collagen resulted in solubilization of collagen in which shortening of the carboxy-terminal telopeptides could be demonstrated. It is suggested that the telopeptidase, acting within a region of the Type I collagen molecule which is known to be essential for the stability of collagen fibrils, could potentially play an important role in collagen degradation in vivo.

Quiescent cultures of normal fetal rat calvarial bone cell populations (RC I and RC IV) and human fibroblasts were incubated with 1.0 ng/ml TGF-beta and the conditioned culture media were processed individually to separate collagenase and 72 kDa-progelatinase from TIMP, the tissue inhibitor of matrix metalloendoproteinases, using mini-columns of heparin- and gelatin-Sepharose. Collagenase synthesis was decreased progressively by TGF-beta in fibroblasts despite a 1.6-fold increase in secreted protein levels and a approximately 1.8-fold increase in 72 kDa-progelatinase synthesis. The human fibroblasts and the osteoblast-enriched RC IV cells showed a greater TGF-beta-induced stimulation in 72 kDa-progelatinase levels over controls compared with the RC I cells. In contrast to RC IV cells, in which TIMP mRNA levels were increased 2.9-fold by TGF-beta, the constitutive level of TIMP transcripts in the RC I cells was greater than 20-fold over that of the RC IV cells, but was not elevated by TGF-beta. TGF-beta also increased TIMP expression in fibroblasts approximately 1.7-fold and PAI-1 levels approximately 5-fold in RC IV cells, and greater than 10-fold in fibroblasts.

The site of attack of purified rheumatoid synovial collagenase on bovine and human tropocollagens has been determined by electron microscopic examination of dimeric and polymeric Segment-Long-Spacing (SLS) fragments produced after collagenase treatment at 27°C. No differences in the mode of attack of rheumatoid synovial collagenase on bovine or human tropocollagens were observed. The enzyme cleaved tropocollagen molecules in the apolar region between bands 41 and 42. (For line nomenclature, see Bruns and Gross1). Examination of junctions of both TC¾ and TC½ fragments of SLS showed this cleavage to be unique. The purified collagenase preparations also removed peptides from both N- and C-terminal ends of the tropocollagen molecule, although whether these cleavages were due to the collagenase itself or to trace amounts of contaminating peptidases is not certain. Autoradiographs obtained from 13-day chick sternal chondrocytes indicate that the incorporation of 3H-proline into collagen, and of Na235SO4 into chondroitin sulfates, are concurrent. Matrix products produced by these cells were identified by their susceptibility to degradation by collagenase and testicular hyaluronidase. Incorporation of the precursors was time dependent, and it was blocked by the addition of puromycin to the culture medium. The labeling patterns of individual chondrocytes show that synthesis of matrix components differs from cell to cell within the sternal population.

The susceptibility of various soluble collagens to rabbit corneal collagenase activity has been tested. The cleavage of the collagens has been assessed by gel electrophoresis and by electron microscopy of SLS crystallites precipitated by ATP. Collagens of both (α1)2α2 and (α1)3 chain types are cleaved at points 23.5 (± 0.5%) of the length of the molecule from the carboxy-terminus. Fractionation studies indicate that the same enzyme is responsible for the attack on both classes of collagen. These observations suggest that the amino-acid sequence or the structure defining the point of collagenase attack in the collagen molecule has been retained through phylogenetic development from the agnatha to primate species, and that more than one of the different classes of collagens distinguished in various vertebrate tissues can be degraded by the same enzyme.Evidence was obtained for the presence of an additional protease activity that was partly resolved from the collagenase by gel filtration. This report attempts to summarize our present knowledge of rheumatoid synovial collagenase and its natural serum inhibitors, beta1-anticollagenase and alpha2-macroglobulin, in relation to cartilage collagen resorption in the rheumatoid joint. Immunolocalization of collagenase across the cartilage/pannus junction is described, and in the light of the finding of the specific, small molecular weight beta1-anticollagenase we propose a model of cartilage erosion based on the interaction between collagenase and its natural inhibitors.

Cultured rabbit aortic smooth muscle cells produce elastic fibers and elastin in their extracellular matrix. Morphological analyses of elastic fibers indicate that they consist of two distinct components which play a major role in elastic early fiber formation namely, the amorphous component and the "microfibrillar" component. During elastogenesis, the early fiber consists of small bundles of filaments. Later, clearly distinguishable small conglomerates of amorphous material are found distributed among the bundles of filaments. The mature elastic fibers have large conglomerates of amorphous material with the filaments present within the interstices. Long term cultures of these cells in the second passage continue to accumulate elastin. The crosslinking amino acid isomers desmosine and isodesmosine, which are unique to insoluble elastin, increase with time in culture. Twenty-four hour pulses with 14C-proline followed by measurements of 14C-hydroxyproline in the collagenase resistant protein of the cell layer show that the synthesis of insoluble elastin is constant up to the 14 week period studied from the time the amorphous material becomes evident ultrastructurally.
