6. Relationships between Anhydrous and Solvated Species of Dexketoprofen Trometamol: A Solid-State Point of View

Abstract

The three different solid forms of dexktoprofen trometamol (DK-T_A, DK-T_B, and DK-T_2H2O) have been studied by using X-ray diffraction and calorimetric methods. In the molecular and crystal structure of DK-T_A, the thermodynamically stable form used in the tablets production process, all the potential H-bond donors and acceptors of each trometamol cation are involved in strong H-bonds which hold together the cations as well as the dexketoprofen/trometamol ionic pairs. The dihydrate species is quickly formed starting from DK-T_Aapparently without the formation of an amorphous phase, and the process is fully reversible. In addition, the two solids have comparable crystal density (cell parameters and space group of the resulting hydrate were determined by X-ray powder diffraction). We speculate that the anhydrous and hydrate crystal architectures did not suffer upon the hydration/dehydration process and that the hydrate form belongs to the class of the channel hydrates and most probably is a planar hydrate. Finally, the unit cell parameters and space group were determined also for the metastable DK-T_Bsolid form.

7. Solvatomorphs of the Dapsone Structurally Related Molecule 3,3′-Diaminodiphenyl Sulfone with Class 2 Residual Solvents: Insights into Their Crystal Structures, Stability, and Transformations

Abstract

Three new solvates of 3,3′-diaminodiphenyl sulfone (3APS·ACN_m, 3APS·DX_m, and 3APS·2DX_o)and their solid-state analyses are reported. The two monosolvate compounds (3APS·ACN_mand 3APS·DX_m) may be obtained in the pure form by using different synthetic strategies (e.g., slow evaporation of the solvent, slurry, or adsorption of the solvent), while the disolvate one (3APS·2DX_o) was always obtained in the mixture form with 3APS·DX_m. Interestingly, a previously reported acetonitrile solvate (3APS·ACN_o) was never obtained during the different synthesis attempts. The stability (varying times and temperatures) of the investigated compounds and a preliminary selective adsorption test are also reported. An in silicoanalysis of the two solvatomorphs with acetonitrile (ACN) and the two different 1,4-dioxane (DX) solvates provides hints to rationalize the results of the solid-state behavior investigation.

Abstract

A combined X-ray diffraction (XRD),solid state nuclear magneticresonance (SSNMR), differential scanning calorimetry (DSC), and modelingapproach has been applied to study the solid–solid transitionof ibuprofen sodium salt between the hydrated racemic compound (RS-H)and the anhydrous conglomerate (RS-A). For comparison, the dihydrate→ anhydrous transformation of the sodium salt of the pure S-enantiomerof ibuprofen was also investigated by means of SSNMR and DSC. Allthe solid state studies indicate that the RS-H → RS-A transitionis fully reversible under different experimental conditions [temperature,pressure (ambient and vacuum), and type of atmosphere (N2, air, and static dry by P2O5)]. The staticand dynamic disorder affecting the isobutyl fragment in RS-H, alreadyobserved by SSNMR, has been further investigated by single crystalXRD and computational techniques.
