In fungi, vegetative incompatibility is a conspecific non-self recognition mechanism that restricts formation of viable heterokaryons when incompatible alleles of specific het loci interact. In Podospora anserina, three non-allelic incompatibility systems have been genetically defined involving interactions between het-c and het-d, het-c and het-e, het-r and het-v. het-d and het-e are paralogues belonging to the HNWD gene family that encode proteins of the STAND class. HET-D and HET-E proteins comprise an N-terminal HET effector domain, a central GTP binding site and a C-terminal WD repeat domain constituted of tandem repeats of highly conserved WD40 repeat units that define the specificity of alleles during incompatibility. The WD40 repeat units of the members of this HNWD family are undergoing concerted evolution. By combining genetic analysis and gain of function experiments, we demonstrate that an additional member of this family, HNWD2, corresponds to the het-r non-allelic incompatibility gene. As for het-d and het-e, allele specificity at the het-r locus is determined by the WD repeat domain. Natural isolates show allelic variation for het-r. [ABSTRACT FROM AUTHOR]

This study considered the systems controlling sexual and self-recognition in Amylostereum areolatum, a homobasidiomycetous symbiont of the Sirex woodwasp. To investigate the structure and organization of these systems in A. areolatum, we identified a portion of a putative homologue (RAB1) of the pheromone receptor genes of Schizophyllum commune and Coprinus cinereus, and a portion of a putative homologue of the S. commune mitochondrial intermediate peptidase ( mip) gene. Diagnostic DNA-based assays for mating-type were developed and their application confirmed that the fungus has a heterothallic tetrapolar mating system. Segregation analysis showed that RAB1 is linked to mating-type B, while mip is linked to mating-type A. The results of sexual and vegetative compatibility tests suggest that sexual recognition in A. areolatum is controlled by two multiallelic mat loci, while self-recognition is controlled by at least two multiallelic het loci. Therefore, despite the association of A. areolatum with the woodwasp and the unique mixture of sexual and clonal reproduction of the fungus, both recognition systems of the fungus appear to be similar in structure and function to those of other homobasidiomycetes. In the filamentous fungi, cell fusion between unlike individuals generally triggers a cell-death reaction known as vegetative incompatibility. In Podospora anserina, it was shown that, during this cell-death reaction, there is a strong increase in proteolytic activity. Here, we report the purification of a 36-kDa protease that is induced during the incompatibility reaction. An internal peptide of this protein displayed a strong similarity with the PEPC subtilisin-like serine protease from Aspergillus niger. This led us to use the pepC gene as a probe to clone the homologous gene from P. anserina, which we called pspA. The expression of pspA is upregulated at the transcript level during the progress of the incompatibility reaction. This induction is diminished in strains bearing suppressors of the vegetative incompatibility reaction. The fact that pspA is homologous to PrB, a vacuolar protease involved in autophagy in yeast, suggests that the incompatibility cell-death reaction and autophagy might be related processes. The diversity within 16 natural progenies of the grapevine dieback fungus, Eutypa lata, was investigated by sampling single-ascospore isolates mainly in France and using random amplified polymorphic DNA (RAPD) markers, vegetative compatibility (VC), and pathogenicity testing. The combination of RAPD and VC data identified each isolate as a unique genotype within each progeny. Only three RAPD haplotypes did not cluster within the expected groups, i.e. the ascospore families. Within each set of clustering haplotypes, Mendelian 1:1 ratios for absence and presence were observed for RAPD markers, indicating that each progeny was the result of a biparental cross. Only one mycelium was obtained when isolation was performed from the discolored wood sustaining the perithecial stroma. This mycelium was identified as a likely parent of the corresponding progeny by RAPD analysis. The level of diversity measured by the average distance between haplotypes calculated from RAPD data, the percentage of vegetatively compatible pairs and the range of pathogenicity appeared similar between all but one progeny, indicating that crosses occurred within a random-mating population. The mating reaction (macrocyst formation) and vegetative compatibility (which is believed to be associated with only the mating type locus in D. discoideum) of asexual, bisexual and homothallic strains of Dictyostelium discoideum were examined. Three asexual strains were vegetatively compatible with a matA tester strain and vegetatively incompatible with a mata tester strain, so we propose that these asexual strains are in fact strains of matA mating type with defective mating capacity. Two bisexual and two homothallic strains were vegetatively incompatible with both matA and mata tester strains, indicating that they either express both mating alleles or that they have a third mating type allele. On the basis of observations on the relative mating capacity with matA and mata tester strains, we propose that bisexual strains are closely related to homothallic strains and are not strains carrying a third mating type allele. Hence we suggest that D. discoideum has a one locus, two allele mating system. The homothallic strains AC4 and ZA3A may express both established mating type alleles ( matA and mata). The het-c locus of the filamentous fungus Podospora anserina controls heterokaryon formation through genetic interaction with alleles of the unlinked loci het-e and het-d. We have isolated four wild-type and two mutant alleles of the het-c locus. A comparison of the predicted proteins encoded by the different wild-type alleles revealed an unusual high level of amino-acid replacements compared to silent polymorphisms but only one amino-acid difference is sufficient to modify the specificity of het-c alleles. Chimeric genes constructed in vitro may exhibit a new specificity different from that of any known wild-type allele. [ABSTRACT FROM AUTHOR]
