Earlier we have shown that certain flavonoids (e.g., quercetin and myricetin) are high-affinity reducing cosubstrates for cyclooxygenase 1 and 2 (COX-1 and COX-2). These compounds can bind inside the peroxidase active site of COX-1 and COX-2 and donate an electron from one of their B-ring hydroxyl groups to hematin. Based on these earlier findings, it is speculated that some of the natural flavonoids that are structural analogs of quercetin but lack the proper B-ring hydroxyl groups might function as novel inhibitors of COX enzymes by blocking the effect of the reducing cosubstrates. This idea is tested in the present study. Computational docking analysis together with quantum chemistry calculation shows that galangin, which shares the same overall structure as quercetin but does not have any hydroxyl group in its B-ring, can bind inside the peroxidase active sites of COX-1 and COX-2 in a similar manner as quercetin, but it has little ability to effectively donate its electrons, thereby blocking the effect of the reducing cosubstrates like quercetin. Further experimental studies confirm that galangin can inhibit, both in vitro and in vivo, quercetin-mediated activation of the peroxidase activity of COX-1 and COX-2. The results of this study demonstrate that galangin is a novel naturally-occurring inhibitor of COX-1 and COX-2, acting by blocking the function of the reducing cosubstrates at the peroxidase sites.

DNA methylation represents an important epigenetic regulation of the genome. Earlier studies have suggested that dietary phenolic compounds including those contained in coffee, tea and soy products may modulate the level of DNA methylation. In this study, we first characterize the effect of caffeic acid phenethyl ester (CAPE) and other dietary phenolic compounds on DNA methylation in vitro. The IC50 values of CAPE, daidzein, isorhamnetin and genistein are 7.6, 6.9, 6.2, and 4.3 μM, respectively, in an in-vitro enzymatic assay system. Computational analysis indicates that CAPE, daidzein, isorhamnetin and genistein can bind inside the DNA substrate-binding site in human DNMT1 with a favorable binding energy. In an animal study, we find that maternal CAPE treatment shifts the coat color distribution of the 21-day-old Avy/a offspring towards the yellow phenotype, indicating that CAPE inhibits the methylation of the agouti gene promoter sequence in vivo. The results from this study may shed light on the potential epigenetic effect in the offspring resulting from maternal intake of certain coffee phenolics during pregnancy.

Animal studies have shown that endogenous estrogens such as 17β-estradiol (E2) can modulate lipid profiles in vivo, and this effect is generally thought to be mediated by the estrogen receptors (ERs). The present study sought to test a hypothesis that some of the endogenous estrogen metabolites that have very weak estrogenic activity may exert some of their modulating effects on lipid metabolism in an ER-independent manner. Using ovariectomized female rats as an in vivo model, we found that 4-hydroxyestradiol (4-OH-E2) has a markedly stronger effect in reducing the adipocyte size and serum cholesterol level in rats compared to E2, despite the weaker estrogenic activity of 4-OH-E2. Moreover, when E2 or 4-OH-E2 is used in combination with ICI-182,780 (an ER antagonist), some of their lipid-modulating effects are not blocked by this antiestrogen. Interestingly, two of the O-methylation metabolites of 4-OH-E2, namely, 4-methoxyestradiol and 4-methoxyestrone, which have much weaker estrogenic activity, were also found to have similar lipid-modulating effects compared to 4-OH-E2. Mechanistically, up-regulation of the expression of leptin, cytochrome P450 7A1 and LXRα genes is observed in the liver of animals treated with E2 or 4-OH-E2, and the up-regulation is essentially not inhibited by co-treatment with ICI-182,780. These results demonstrate that some of the endogenous E2 metabolites are functionally important modulators of lipid metabolic profiles in vivo. In addition, our findings indicate that an ER-independent pathway likely mediates some of the lipid-modulating effects of endogenous estrogens and their metabolic derivatives.

Glutamate is an endogenous excitatory neurotransmitter. At high concentrations, it is neurotoxic and contributes to the development of certain neurodegenerative diseases. There is considerable controversy in the literature with regard to whether glutamate-induced cell death in cultured HT22 cells (an immortalized mouse hippocampal cell line) is apoptosis, necrosis, or a new form of cell death. The present study focused on investigating the mechanism of glutamate-induced cell death. We found that glutamate induced, in a time-dependent manner, both necrosis and apoptosis in HT22 cells. At relatively early time points (8–12 h), glutamate induced mostly necrosis, whereas at late time points (16–24 h), it induced mainly apoptosis. Glutamate-induced mitochondrial oxidative stress and dysfunction were crucial early events required for the induction of apoptosis through the release of the mitochondrial apoptosis-inducing factor (AIF), which catalyzed DNA fragmentation (an ATP-independent process). Glutamate-induced cell death proceeded independently of the Bcl-2 family proteins and caspase activation. The lack of caspase activation likely resulted from the lack of intracellular ATP when the mitochondrial functions were rapidly disrupted by the mitochondrial oxidative stress. In addition, it was observed that activation of JNK, p38, and ERK signaling molecules was also involved in the induction of apoptosis by glutamate. In conclusion, glutamate-induced apoptosis is AIF-dependent but caspase-independent, and is accompanied by DNA ladder formation but not chromatin condensation.

Indoleamine 2,3-dioxygenase is a rate-limiting enzyme involved in the conversion of tryptophan to various endogenous catabolites, such as 3-hydroxyanthranilic acid, 3-hydroxykynurenine, and L-kynurenine. This study aims to provide evidence to support the hypothesis that some of the tryptophan catabolites may exert strong immunosuppression against allograft rejection by inhibiting allogeneic T-cell response. The mixed lymphocyte proliferation assay was used as an in-vitro model and the rat cardiac allograft as an in-vivo model. We found that 3-hydroxyanthranilic acid and 3-hydroxykynurenine inhibited T-cell proliferation in a concentration-dependent manner. Both dendritic cell-stimulated T-cell proliferation and baseline T-cell proliferation (in the absence of dendritic cells) were inhibited by these tryptophan catabolites. Using the rat cardiac allograft model, we showed that a single i.v. administration of 3-hydroxyanthranilic acid+allogeneic dendritic cells seven days before cardiac grafting markedly prolonged the graft survival in the recipient (from seven days to fifteen days). Additional studies showed that this single administration of 3-hydroxyanthranilic acid wiped out a majority of specific T-cell sub-populations that were activated by allogeneic dendritic cells. This strong inhibition by 3-hydroxyanthranilic acid is believed to be an important mechanism contributing to the development of immunotolerance against the allogeneic graft. These observations offer a potential strategy for using tryptophan catabolites together with allogeneic dendritic cells to induce selective immunotolerance against allogeneic grafts in the recipients.
