Neurons of the rostral part of inferior premotor cortex of the monkey discharge during goal-directed hand movements such as grasping, holding, and tearing. We report here that many of these neurons become active also when the monkey observes specific, meaningful hand movements performed by the experimenters. The effective experimenters' movements include among others placing or retrieving a piece of food from a table, grasping food from another experimenter's hand, and manipulating objects. There is always a clear link between the effective observed movement and that executed by the monkey and, often, only movements of the experimenter identical to those controlled by a given neuron are able to activate it. These findings indicate that premotor neurons can retrieve movements not only on the basis of stimulus characteristics, as previously described, but also on the basis of the meaning of the observed actions. Grasping is the most important skilled motor act of primates. It is based on a series of sensorimotor transformations through which the affordances of the objects to be grasped are transformed into appropriate hand movements. It is generally accepted that a circuit formed by inferior parietal areas AIP and PFG and ventral premotor area F5 represents the core circuit for sensorimotor transformations for grasping. However, selection and control of appropriate grip should also depend on higher-order information, such as the meaning of the object to be grasped, and the overarching goal of the action in which grasping is embedded. In this review, we describe recent findings showing that specific sectors of the ventrolateral prefrontal cortex are instrumental in controlling higher-order aspects of grasping. We show that these prefrontal sectors control the premotor cortex through two main gateways: the anterior subdivision of ventral area F5-sub-area F5a-, and the pre-supplementary area (area F6). We then review functional studies showing that both F5a and F6, besides being relay stations of prefrontal information, also play specific roles in grasping. Namely, sub-area F5a is involved in stereoscopic analysis of 3D objects, and in planning cue-dependent grasping activity. As for area F6, this area appears to play a crucial role in determining when to execute the motor program encoded in the parieto-premotor circuit. The recent discovery that area F6 contains a set of neurons encoding specific grip types suggests that this area, besides controlling "when to go", also may control the grip type, i.e., "how to go". We conclude by discussing clinical syndromes affecting grasping actions and their possible mechanisms.

The aim of the present study was to assess the cognitive profiles of children with autistic spectrum disorder and of their healthy siblings (Siblings). With the term cognitive profile, we indicate the relationship extant among the values of verbal and performance subtests of the Wechsler Intelligence Scale. The conducted statistical analyses indicated that, although siblings showed a normal intelligent quotient and did not differ in this aspect from typically developing group, their cognitive profile was amazingly similar to that of their relatives affected by autism. A k-means clustering analysis on the values of single subtests further confirmed this result, showing a clear separation between typically developing children on the one side, and autistics and their siblings on the other. We suggest that the common cognitive profile observed in autistic children and their siblings could represent a marker of liability to autism and, thus, a possible intermediate phenotype of this syndrome.

Does motor mirroring in humans reflect the observed movements or the goal of the observed motor acts? Tools that dissociate the agent/object dynamics from the movements of the body parts used to operate them provide a model for testing resonance to both movements and goals. Here, we describe the temporal relationship of the observer's motor excitability, assessed with transcranial magnetic stimulation (TMS), with the observed goal-directed tool actions, in an ecological setting. Motor-evoked potentials (MEPs) to TMS were recorded from the opponens pollicis (OP, thumb flexor) and the extensor indicis proprius (EIP, index extensor) muscles of participants while they observed a person moving several small objects with a pair of normal pliers (closed by finger flexion) or reverse pliers (opened by finger flexion). The MEPs were a significant predictor of the pliers' kinematics that occurred in a variable time interval between -400 and +300 ms from TMS. Whatever pliers' type was being observed, OP MEPs correlated positively and EIP MEPs correlated negatively with the velocity of pliers' tips closure. This datum was confirmed both at individual and at a group level. Motor simulation can be demonstrated in single observers in a "real-life" ecological setting. The relation of motor resonance to the tool type shows that the observer's motor system codes the distal goal of the observed acts (i.e., grasping and releasing objects) in terms of its own motor vocabulary, irrespective of the actual finger movements that were performed by the observed actor.

Mirror neurons are a distinct class of neurons that discharge both during the execution of a motor act and during observation of the same or similar motor act performed by another individual. However, the extent to which mirror neurons coding a motor act with a specific goal (e.g., grasping) might also respond to the observation of a motor act having the same goal, but achieved with artificial effectors, is not yet established. In the present study, we addressed this issue by recording mirror neurons from the ventral premotor cortex (area F5) of two monkeys trained to grasp objects with pliers. Neuron activity was recorded during the observation and execution of grasping performed with the hand, with pliers and during observation of an experimenter spearing food with a stick. The results showed that virtually all neurons responding to the observation of hand grasping also responded to the observation of grasping with pliers and, many of them to the observation of spearing with a stick. However, the intensity and pattern of the response differed among conditions. Hand grasping observation determined the earliest and the strongest discharge, while pliers grasping and spearing observation triggered weaker responses at longer latencies. We conclude that F5 grasping mirror neurons respond to the observation of a family of stimuli leading to the same goal. However, the response pattern depends upon the similarity between the observed motor act and the one executed by the hand, the natural motor template.

It has been suggested that the deficit in understanding others' intention in autism depends on a malfunctioning of the mirror system. This malfunction could be due either to a deficit of the basic mirror mechanism or to a disorganization of chained action organization on which the mirror understanding of others' intention is based. Here we tested this last hypothesis investigating the kinematics of intentional actions. Children with autism and typically developing children (TD) were asked to execute two actions consisting each of three motor acts: the first was identical in both actions while the last varied for its difficulty. The result showed that, unlike in TD children, in children with autism the kinematics of the first motor act was not modulated by the task difficulty. This finding strongly supports the notion that children with autism have a deficit in chaining motor acts into a global action.

There are two radically different views on the functional role of the dorsal visual stream. One considers it as a system involved in space perception. The other is of a system that codes visual information for action organization. On the basis of new anatomical data and a reconsideration of previous functional and clinical data, we propose that the dorsal stream and its recipient parietal areas form two distinct functional systems: the dorso-dorsal stream (d-d stream) and the ventro-dorsal stream (v-d stream). The d-d stream is formed by area V6 (main d-d extrastriate visual node) and areas V6A and MIP of the superior parietal lobule. Its major functional role is the control of actions "on line". Its damage leads to optic ataxia. The v-d stream is formed by area MT (main v-d extrastriate visual node) and by the visual areas of the inferior parietal lobule. As the d-d stream, v-d stream is responsible for action organization. It, however, also plays a crucial role in space perception and action understanding. The putative mechanisms linking action and perception in the v-d stream is discussed.

Positron emission tomography imaging of cerebral blood flow was used to localize brain areas involved in the representation of hand grasping movements. Seven normal subjects were scanned under three conditions. In the first, they observed precision grasping of common objects performed by the examiner. In the second, they imagined themselves grasping the objects without actually moving the hand. These two tasks were compared with a control task of object viewing. Grasp observation activated the left rostral superior temporal sulcus, left inferior frontal cortex (area 45), left rostral inferior parietal cortex (area 40), the rostral part of left supplementary motor area (SMA-proper), and the right dorsal premotor cortex. Imagined grasping activated the left inferior frontal (area 44) and middle frontal cortex, left caudal inferior parietal cortex (area 40), a more extensive response in left rostral SMA-proper, and left dorsal premotor cortex. The two conditions activated different areas of the right posterior cerebellar cortex. We propose that the areas active during grasping observation may form a circuit for recognition of hand-object interactions, whereas the areas active during imagined grasping may be a putative human homologue of a circuit for hand grasping movements recently defined in nonhuman primates. The location of responses in SMA-proper confirms the rostrocaudal segregation of this area for imagined and real movement. A similar segregation is also present in the cerebellum, with imagined and observed grasping movements activating different parts of the posterior lobe and real movements activating the anterior lobe.
