It has been established that administration of teturam (200 mg/kg) per os once a day for 14 days retards reparative regeneration of the liver in rats following partial hepatectomy and exerts an additional damaging action on the population of regenerating hepatocytes. Administration of potassium orotate (100 mg/kg) per os once a day for 14 days speeds up, on the contrary, organ regeneration. The combined use of the drugs potentiates paradoxically the negative hepatotropic effect of teturam. It is concluded that experimental reparative regeneration of the rat liver can be applied to the assessment of hepatotropism of the antialcoholic drugs.

It has been established in mouse experiments that potassium orotate (100 mg/kg) and piracetam (500 mg/kg) given in chronic oral doses have an antidepressant activity according to the "behavioral despair" test. It has been demonstrated that antidepressant activity of potassium orotate (20 and 50 mg/kg) and piracetam (50 and 100 mg/kg) is associated with a psychostimulant effect.

Chronic administration of potassium orotate to rats per os in doses of 5, 10, 20, 50 and 100 mg/kg was followed by different correlations of the training under stress with anxiolytic activity. The doses of the drug were defined, at which the maximally pronounced effects under consideration were in optimal correlations, as were doses which entailed inhibition of the training in the presence of a relatively low tranquilizing activity, low emotional responsiveness and aggressiveness. It is suggested that natural analogs of pyrimidine, whose precursor is orotic acid, are universal endogenous regulators of mnemonic and antianxiety functions.

Experiments on rats have shown the lowered content of ascorbic acid in blood and urine during administration of retabolil, nerobol or potassium orotate. Retabolil and potassium orotate increased the content of ascorbic acid in tissues and the content of reduced glutathion in blood. Nerobol produced a rise in vitamin C blood content in the test organs.

Chronic administration of potassium orotate to rats per os in a dose of 100 mg/kg produces anxiolytic action that is manifested in antiaggressive, antiphobic and anti-conflict effects similar to the characteristic effects of diazepam. Besides, it provokes the changes in the spectral characteristics of the cat brain electrograms, common to tranquilizers. Based on the data obtained and analysis of the reported evidence the authors advance the hypothesis according to which endogenous pyrimidines, particularly orotic acid, possessing structural fragments necessary for the interaction with the sites of specific binding of 1,4-benzodiazepines may realize adaptive anxiolytic function both within the system of metabolic adaptation and when involved into the system of endogenous regulators and modulators of neuropsychic processes.

It has been shown in experiments on 3-4-week-old rats that the use of pyridoxine, riboflavin, potassium orotate, folic and glutamic acids for 30 days accelerates the recovery of the working capacity after maximal exercise, increases the content of ATP in the muscles, and reduces the blood urea content.
