Yersinia polynucleotide phosphorylase ( PNPase), a 3′-5′ exoribonuclease, has been shown to affect growth during several stress responses. In Escherichia coli, PNPase is one of the subunits of a multiprotein complex known as the degradosome, but also has degradosome-independent functions. The carboxy-terminus of E. coli ribonuclease E ( RNase E) serves as the scaffold upon which PNPase, enolase (a glycolytic enzyme), and Rhl B helicase all have been shown to bind. In the yersiniae, only PNPase has thus far been shown to physically interact with RNase E. We show by bacterial two-hybrid and co-immunoprecipitation assays that Rhl B and enolase also interact with RNase E. Interestingly, although PNPase is required for normal growth at cold temperatures, assembly of the yersiniae degradosome was not required. However, degradosome assembly was required for growth in the presence of reactive oxygen species. These data suggest that while the Yersinia pseudotuberculosis PNPase plays a role in the oxidative stress response through a degradosome-dependent mechanism, PNPase's role during cold stress is degradosome-independent. A whole-genome DNA microarray was constructed to dissect expression profiles of Vibrio parahaemolyticus in response to a sudden temperature downshift from 37 to 10 °C. The mRNA level of each gene at each of three time points (20, 40 and 60 min after temperature downshift) was compared with that just before cold treatment. Clustering analysis of time-course data revealed nine gene clusters with different time-dependent expression patterns. Downregulation of metabolism-related genes was obviously dominant over upregulation at all time points. The distinct negative regulation of metabolism-related genes would account for a generally reduced cellular protein pool resulting from the sudden temperature downshift. In contrast, cold shock had a ‘neutral and balanced’ regulatory action on nonmetabolic cellular pathways, which likely brought about the remodelling of cell envelope structures and transport/binding functions. We identified a 171-bp 5′-untranslated region in the cspA transcript. The cspA gene encoded cold shock protein A (CspA), and CspA was shown to be the major cold shock protein in V. parahaemolyticus. Evident regulatory motifs were conserved within the cspA promoter regions of Escherichia coli and V. parahaemolyticus. Changes in gene expression play a critical role in enhancing the ability of cyanobacteria to survive under cold conditions. In the present study, Spirulina platensis cultures were grown at the optimal growth temperature, in the light, before being transferred to dark conditions at 22 °C. Two dimensional-differential gel electrophoresis was then performed to separate differentially expressed proteins that were subsequently identified by MS. Among all differentiated proteins identified, a protein involved in fatty acid biosynthesis, (3R)-hydroxymyristoyl-[acyl-carrier-protein]-dehydratase encoded by fabZ, was the most up-regulated protein. However, the fatty-acid desaturation proteins were not significantly differentiated. This raised the question of how the unsaturated fatty acid, especially γ-linolenic acid, content in the cells in the cold–dark shift remained stable compared with that of the cold shift. Thus, a study at the transcriptional level of these desaturase genes, desC, desA and desD, and also of the fabZ gene was conducted. The results indicated that in the dark, where energy is limited, mRNA stability was enhanced by exposure to low temperatures. The data demonstrate that when the cells encounter cold stress with energy limitation, they can maintain their homeoviscous adaptation ability via mRNA stability. [ABSTRACT FROM AUTHOR]

The Bacillus subtilis laboratory strain JH642 shows a cold-sensitive phenotype after a temperature shift from 37 to 15°C in comparison to wild type strain MR168. A mutation in the acetolactate synthase complex IlvBH was found to be partially responsible for this growth defect after cold shock. Via DNA sequencing, genetic and biochemical studies, this defect was characterized, which entails a substitution of two adenines to guanines in the ilvB gene. This results in an amino acid substitution from lysine at position 176 to glycine. As a consequence, the acetolactate synthase efficiency in strain JH642 was found to be reduced by 51-fold. One csp-like gene, called cspA, from the lactic acid bacterium Lactobacillus casei was identified by an inverse polymerase chain reaction approach based on degenerate primers. cspA encodes a protein of 66 amino acid residues, which displays at least 74% identity with Csp proteins of Lactobacillus genera. Northern blot analysis revealed that cspA is transcribed monocistronically and that its expression is induced after a temperature downshift from 37°C to 20°C. The transcriptional start site has been determined and is situated 98 bp upstream of the initiation codon. A cspA mutant strain was constructed and it showed reduced growth rate compared with the wild type at both optimal and low temperatures, demonstrating that CspA plays an important role in the physiology of L. casei. A decrease in environmental temperature leads to the synthesis of Δ5-unsaturated fatty acids in Bacillus subtilis by the fatty acid desaturase Des. Des is regulated by the two-component system DesKR. To understand the mechanism of cold signal perception and transduction by the membrane domain and the cytosolic domain of DesK, we expressed the cytosolic domain of DesK in trans under the control of a xylose-inducible promoter without the membrane domain. We performed growth experiments and a Northern blot analysis. Our results show that the kinase function of the cytosolic domain of DesK is temperature-independent, leading to a constitutive expression of the des gene. These findings support the conclusion that the membrane domain of DesK is the temperature-sensing element of the two-component system. [Copyright &y& Elsevier]

The two csp-like genes from the lactic acid bacterium Lactobacillus delbrueckii ssp. bulgaricus were characterized and designated cspA and cspB. The gene cspA has been identified using a polymerase chain reaction (PCR)-based approach with degenerated primers and further characterized using an inverse PCR strategy. cspA encodes a protein of 65 amino acid residues which displays between 81 and 77% identity with proteins CspL and CspP of Lactobacillus plantarum. cspB has been identified as a cspA ortholog using the partial sequence of the L. bulgaricus ATCC11842. cspB encodes a protein of 69 amino acids which has 42% identity with CspA. Northern blot analyses showed that cspA is transcribed as a single gene and that its transcription increased after a temperature downshift from 42 to 25°C. In contrast, cspB is part of an operon transcribed at constant level irrespective of the temperature. These results indicate that cspA encodes the only Csp-like protein of L. bulgaricus induced by a downshift of temperature. [Copyright &y& Elsevier]
