Escherichia coli can perform at least two modes of anaerobic hydrogen metabolism and expresses at least two types of hydrogenase activity. Respiratory hydrogen oxidation is catalysed by two 'uptake' hydrogenase isoenzymes, hydrogenase -1 and -2 (Hyd-1 and -2), and fermentative hydrogen production is catalysed by Hyd-3. Harnessing and enhancing the metabolic capability of E. coli to perform anaerobic mixed-acid fermentation is therefore an attractive approach for bio-hydrogen production from sugars. In this work, the effects of genetic modification of the genes encoding the uptake hydrogenases, as well as the importance of preculture conditions, on hydrogen production and fermentation balance were examined. In suspensions of resting cells pregrown aerobically with formate, deletions in Hyd-3 abolished hydrogen production, whereas the deletion of both uptake hydrogenases improved hydrogen production by 37% over the parent strain. Under fermentative conditions, respiratory H2 uptake activity was absent in strains lacking Hyd-2. The effect of a deletion in hycA on H2 production was found to be dependent upon environmental conditions, but H2 uptake was not significantly affected by this mutation.

The biodegradation of tributyl phosphate by a mixed culture of Pseudomonads was demonstrated. Growth and the rate of tributyl phosphate consumption were variable and divisible into rapid and slow rates. Rapidly growing, rapidly tributyl phosphate-utilising cultures contained a 22–24 kb DNA fragment isolated by two methods, which was not visible in the cultures growing slowly. The mixed culture gave five periods of rapid growth interspersed with periods of poor growth during 7 months of weekly subculture, with the 22–24 kb DNA fragment detectable during the rapidly growing periods only. Seven Pseudomonads isolated from the culture grew at the expense of tributyl phosphate as the sole phosphorus source but spontaneously and irreversibly lost this ability after eight serial subcultures.

The acid phosphatase of an atypical Citrobacter sp. was purified in two isoforms, designated CPI and CPII, which had different Km values for glycerol 1-phosphate and glycerol 2-phosphate The enzyme was not inhibited by the end-product glycerol. Enzyme activity was increased in the presence of phosphate acceptor molecules having free hydroxyl groups (glycerol, methanol, ethanol). 31P-nuclear magnetic resonance spectroscopy indicated transfer of the liberated phosphate onto the alcohol, with the de novo production of (e.g.) glycerol 1-phosphate by enzyme supplemented with phosphomonoester substrate and glycerol.

The effect of deleting the genes encoding the twin-arginine translocation (Tat) system on H2 production by Escherichia coli strain MC4100 and its formate hydrogenlyase upregulated mutant (DeltahycA) was investigated. H2 evolution tests using two mutant strains defective in Tat transport (DeltatatC and DeltatatA-E) showed that the rate doubled from 0.88+/-0.28 mL H2 mg dry weight-1 L culture-1 in the parental strain, to 1.70+/-0.15 and 1.75+/-0.18 mL H2 mg dry weight-1 L culture-1, respectively, in the DeltatatC and DeltatatA-E strains. This increase was comparable to that of a previously characterized hydrogen over-producing E. coli strain carrying a DeltahycA allele. Construction of a tatC, DeltahycA double deletion strain did not increase hydrogen production further. Inactivation of the Tat system prevents correct assembly of the uptake hydrogenases and formate dehydrogenases in the cytoplasmic membrane and it is postulated that the subsequent loss of basal levels of respiratory-linked hydrogen and formate oxidation accounts for the observed increases in formate-dependent hydrogen evolution.

An atypical Citrobacter sp. isolated from a heavy metal-polluted site has a PhoN-type acid phosphatase that is responsible for the precipitation of heavy metals as cell-bound metal phosphates via liberated inorganic phosphate, supplied via enzymic cleavage of a phosphomonoester substrate. Phosphatase activity, comprising two isoenzymes designated CPI and CPII, was resistant to cadmium (II), zinc (II) and lead (II), but sensitive to uranyl and vanadyl oxycations and cupric ion. The monovalent cations of mercury (I) and silver (I), and trivalent yttrium (III) were inhibitory, particularly to CPII. Theanionic counterion did not influence metal toxicity. The Km of CPII towards some phosphomonoester substrates was higher than than that of CPI, suggesting that although the two isoenzymes were shown previously to be closely related, subtle differences exist between them that justify their classification as separate isoenzymes, for which the physiological function is still obscure.

A Citrobacter sp. was reported previously to accumulate heavy metals as cell-bound heavy metal phosphates. Metal uptake is mediated by the activity of a periplasmic acid-type phosphatase that liberates inorganic phosphate to provide the precipitant ligand for heavy metals presented to the cells. Amino acid sequencing of peptide fragments of the purified enzyme revealed significant homology to the phoN product (acid phosphatase) of some other enterobacteria. These organisms, together with Klebsiella pneumoniae, previously reported to produce acid phosphatase, were tested for their ability to remove uranium and lanthanum from challenge solutions supplemented with phosphatase substrate. The coupling of phosphate liberation to metal bioaccumulation was limited to the metal accumulating Citrobacter sp.; therefore the participation of species-specific additional factors in metal bioaccumulation was suggested.

The effect of 500 nM NaCl on the growth, and phosphatase production of a Citrobacter sp. was investigated. Although growth was retarded, phosphatase production was enhanced by 50%. Relief from osmotic stress using the osmoprotectant glycine betaine gave normal growth, but phosphatase activity was reduced. The Citrobacter sp. ceased to grow following a shift to anaerobic conditions, but anaerobically-incubated cells continued to produce phosphatase after a transient lag.

Fungal volatilization of antimony and other group Vb elements has been proposed to have a causal role in the sudden infant death syndrome (SIDS; cot death). The ability of fungi to produce volatile arsenic and antimony compounds in pure culture was examined using Scopulariopsis brevicaulis, reported as an inhabitant of PVC cot mattress covers, and Phaeolus schweinitzii, a wood decay fungus known to be a good volatilizer of arsenic. Volatile arsenic compounds were detected from all cultures grown on arsenic-supplemented media, but antimony volatilization was not reliably detected. Although antimony levels above the baseline sensitivity of the analytical technique were detected in four (out of 24) of the samples analyzed, the concentrations recorded were too low to be reliably interpreted as evidence for volatilization. Our results are discussed in relation to hypotheses regarding the causes of SIDS.

A phosphatase-overproducing mutant of a Citrobacter sp. was isolated by a novel method. This exploited the proposed role for the enzyme in metal detoxification by precipitation of heavy metals with enzymically-liberated inorganic phosphate at the cell surface. The mutant, tolerant to an otherwise growth-inhibitory concentration of copper, overproduced phosphatase approx. 3-fold as compared to the parent strain. With a comparable enhancement in uranium uptake, the industrial potential of the mutant in metal waste detoxification is apparent, and is discussed.
