The intensification of aquaculture has led to a rise in fish infections, necessitating the search for alternative antibiotics. In this context, our study investigated the effects of dietary supplementation with Chaetomorpha aerea, a filamentous green algae, on the immune health and resistance to infections in tilapia (Oreochromis mossambicus). Diets containing varying concentrations of C. aerea (0, 1, 2, 5, and 10 g/kg) were prepared and administered to the fish for 30 days, followed by a challenge with Edwardsiella tarda to evaluate survival rates. The results were significant. The diet containing 5 g/kg of C. aerea (group T3) brought about substantial improvements in hematological parameters, including increases in red blood cell count (RBC), hematocrit (Hct), and hemoglobin (Hb). The T3 group exhibited a robust immune response, with higher lysozyme and ceruloplasmin activity in immunological assays. LBP gene expression was significantly elevated in the spleen and thymus of fish in the T3 group, which correlated with higher survival after bacterial challenge compared to the control group. Principal Component Analysis (PCA) and cluster analysis confirmed that the 5 g/kg concentration stood out for maximizing immunological benefits without compromising the overall health of the fish. These findings highlight the robust immune response in the T3 group, a key finding of our study. We conclude that supplementation with C. aerea represents a promising and sustainable alternative in the formulation of diets for tilapia, contributing to improved health and resistance to diseases. α-Melanocyte-stimulating hormone (α-MSH) is a well-studied neuropeptide controlling skin and hair color. Besides, numerous immunomodulation roles of α-MSH were recorded in humans and mice. However, the regulatory effects of α-MSH in teleost immunity haven't been well elucidated. In this study, several precursor molecules of α-MSH (POMCs) and its receptors (MCRs) in Nile tilapia (Oreochromis niloticus) were characterized, and their expression characteristics and specific functions on antibacterial immunity were determined. Overall, POMCs and MCRs were principally detected in the brain, skin, and liver, and were remarkably promoted post Streptococcus agalactiae infection. However, tiny POMCs and MCRs were observed in tilapia immune organs (head kidney and spleen) or lymphocytes, and no evident immunomodulation effect was detected in vitro. Moreover, the in vivo challenge experiments revealed that α-MSH protects tilapia from bacterial infection by regulating responses in the brain and intestine. An 8-week feeding trial was performed to evaluate the effects of dietary leucine (Leu) and valine (Val) levels on growth performance, glycolipid metabolism and immune response in Oreochromis niloticus. Fish (15.23 ± 0.05 g) were randomly fed four diets containing two Leu levels (1.2% and 2.3%) and two Val levels (0.7% and 1.4%) as a 2 × 2 experimental design (LL-LV, LL-HV, HL-LV and HL-HV). Compared with LL-LV group, the growth parameters (final weight, daily growth coefficient (DGC) and growth rate per metabolic body weight (GR MBW )), feed conversion rate (FCR), the activities of intestinal amylase, lipase, creatine kinase (CK) and Na + , K + -ATPase, liver NAD + /NADH ratio, as well as the expression of SIRT1, GK, PK, FBPase, PPARα, CPT IA, ACO and IL10 all increased significantly in the HL-LV group; however, in the high Val group, final weight, DGC, GR MBW , intestinal enzyme activities, as well as the expression of PEPCK, SREBP1, FAS, IL8 and IL10 of the HL-HV group were significantly lower than those of the LL-HV group, while the opposite was true for the remaining indicators. Significant interactions between dietary Leu and Val were observed in final weight, DGC, GR MBW , plasma IL1β and IL6 levels, intestinal amylase and CK activities, liver NAD + /NADH ratio, as well as the expression of SIRT1, PK, PEPCK, FBPase, SREBP1, FAS, PPARα, CPT IA, ACO, NF-κB1, IL1β, IL6 and IL10. The highest values of growth parameters, intestinal enzyme activities and expression of SIRT1, FBPase, PPARα, CPT IA and ACO were observed in the HL-LV group, while the opposite was true for the expression of SREBP1, FAS, PPARα, NF-κB1, IL1β and IL6. Tilapia lake virus (TiLV) is a notable contagious agent that causes massive economic losses in the tilapia industry globally. Evaluations of the histological changes associated with TiLV infection are not only crucial for diagnosis, but also to gain an understanding of the disease. We therefore synthesized a rabbit polyclonal immunoglobulin G antibody against TiLV and developed an immunohistochemical (IHC) procedure to detect TiLV localization in the tissues of infected fish for comparison with in situ hybridization (ISH) testing. A total of four different sample cohorts derived from TiLV-infected fish was used to validate the IHC procedure. The TiLV IHC application was successfully developed and facilitated nuclear and cytoplasmic immunolabelling in the intestines, gills, brain, liver, pancreas, spleen, and kidneys that corresponded with the ISH results. Apart from the ISH results, TiLV-IHC signals were clearly evident in the endothelial cells of various organs, the circulating leukocytes in the blood vessels, and the areas of tissue inflammation. Among the tested sample cohorts, the intestines, gills, and brain had IHC-positive signals, highlighting the possibility of these organs as common TiLV targets. Immunological staining pattern and distribution corresponded with the TiLV viral load but not the inoculation route. The TiLV IHC was also capable of detecting TiLV infection in the experimentally challenged ornamental cichlids, Mozambique tilapia, giant gourami, and naturally infected tilapia, indicating the dynamic range of IHC for TiLV detection. Overall, our study delivers the first IHC platform to detect TiLV infection and provides novel evidence of cellular tropism during TiLV infection. Our findings also reveal the TiLV distribution pattern of infected fish and propose the endotheliotropism and lymphotropism of this virus, which requires further elaboration. Tilapia lake virus (TiLV) is an emerging virus associated with high mortality in cultured tilapia. Since the first report of tilapia lake virus, it has been detected in diseased tilapia in sixteen countries around the world. Thus, there is an urgent need to develop an efficacious vaccine to prevent TiLV disease (TiLVD) and reduce its global economic impact. Understanding the role of the adaptive immune response following exposure of tilapia to TiLV is a critical step in the development of such a vaccine. In this study, we challenged red hybrid tilapia by cohabitation or intraperitoneal injection and demonstrated that surviving fish develop a protective immunity. We also demonstrated that tilapia that survived experimental infections possess significant antibodies against the protein encoded by the TiLV segment 4. We then developed a TiLV indirect ELISA to determine the antibody response in tilapia. The ELISA revealed high antibody levels in survivors of experimental challenges and following outbreaks on farms. The ELISA effectively distinguished TiLV-exposed from unexposed tilapia and was used to monitor anti-TiLV antibody kinetics following infection. During the primary infection, tilapia developed an antibody response as early as 7 days post TiLV challenge (dpc), peaked at 15 dpc, showed a gradual decline up until about 42 dpc, but persisted in some fish up until day 110 dpc. Upon re-infection, an increased antibody response occurred within 7-14 days, demonstrating that tilapia that survive TiLV infections develop humoral memory. In conclusion, our results demonstrated that tilapia mount antibody responses against TiLV that supports protective immunity to subsequent TiLV disease. The persistence of anti-TiLV antibodies in survivors following a single exposure suggests a single vaccination might be adequate to protect tilapia during the entire grow-out period. Red tilapia (Oreochromis sp.) has become one of the most important fish in aquaculture. Bacterial infection caused by Flavobacterium columnare, the causative agent of columnaris disease, has been now identified as one of the most serious infectious diseases in farmed red tilapia and cause major financial damage to the producers. Among the effective prevention and control strategies, vaccination is one of the most effective approach. As the surface of living fish is covered by mucus and directly associated with the mucosal immunity, we therefore hypothesized that better adsorption on mucosal surfaces and more efficient vaccine efficacy could be enhanced biomimetic nanoparticles mimicking the mucoadhesive characteristic of live F. columnare. In this work, we describe an effective approach to targeted antigen delivery by coating the surface of nanoparticles with mucoadhesive chitosan biopolymer to provide "pathogen-like" properties that ensure nanoparticles binding on fish mucosal membrane. The physiochemical properties of nanovaccines were analyzed, and their mucoadhesive characteristics and immune response against pathogens were also evaluated. The prepared vaccines were nano-sized and spherical as confirmed by scanning electron microscope (SEM). The analysis of hydrodynamic diameter and zeta-potential also suggested the successful modification of nanovaccines by chitosan as indicated by positively charged and the overall increased diameter of chitosan-modified nanovaccines. In vivo mucoadhesive study demonstrated the excellent affinity of the chitosan-modified nanovaccines toward fish gills as confirmed by bioluminescence imaging, fluorescent microscopy, and spectrophotometric quantitative measurement. Following vaccination with the prepared nanovaccines by immersion 30 min, the challenge test was then carried out 30 and 60 days post-vaccination and resulted in high mortalities in the control. The relative percent survival (RPS) of vaccinated fish was greater than 60% for mucoadhesive nanovaccine. Our results also suggested that whole-cell vaccines failed to protect fish from columnaris infection, which is consistent with the mucoadhesive assays showing that whole-cell bacteria were unable to bind to mucosal surfaces. In conclusion, we could use this system to deliver antigen preparation to the mucosal membrane of tilapia and obtained a significant increase in survival compared to controls, suggesting that targeting mucoadhesive nanovaccines to the mucosal surface could be exploited as an effective method for immersion vaccination., (Copyright © 2019 Elsevier Ltd. The administration of probiotics during early ontogenetic stages can be an effective way to manipulate the gut microbiota of animals. Specifically, the administration of probiotics can enhance gut-colonization success and regulate the immune response. In this study, the effects of early contact with probiotic Lactococcus lactis subsp. lactis JCM5805 on the gut microbial assembly of larvae Nile tilapia were examined. The effects of JCM5805 on IFNα expression through the TLR7 and TLR9-dependent signal transduction pathway as well as larval disease resistance were studied. Three days postfertilization, embryos were randomly allocated into nine 30 L tanks with a concentration of 20 eggs L -1 . Triplicate tanks were performed for each treatment. Treatments included a control group (C), a low probiotic concentration group (T1), where JCM5805 was added to the water at 1 × 10 4  cfu ml -1 , and a high probiotic concentration group (T2), where JCM5805 was added to the water at 1 × 10 8  cfu ml -1 . Probiotics were administered continuously for 15 days. qPCR was used to analyze transcript levels of the TLR7, TLR9, MyD88, IRF7 and IFNα genes using RNA extracted from whole embryos on day 5 and 10, and from the intestine of larvae on day 15. Transcription of these genes was also measured in the intestine, liver and spleen of larvae one month after the cessation of probiotic administration. The results showed that MyD88 and IRF7 were significantly elevated on days 5 and 10 in the T2 group. TLR9 and IFNα were also significantly elevated on days 5, 10 and 15 during probiotic application of T2 (P < 0.05). One month after the cessation of probiotics administration, no significant difference was observed in the expression of these genes (P > 0.05). The larvae were fed probiotics for 15 days and were infused with Streptococcus agalactiae strain WC1535 at a final concentration of 1 × 10 6  cfu ml -1 . The survival rate of T2 was significantly higher than that of the C group (P < 0.05). Microbial characterization by Illumina HiSeq sequencing of 16S rRNA gene amplicons showed the significantly higher presence of JCM5805 in the guts of T2 after 15 days of probiotic continuous application. Although JCM5805 was below the detection level after the cessation of probiotic for 5 days, the gut microbiota of the exposed tilapia larvae in T2 remained clearly different from that of the control treatment after the cessation of probiotic administration. These data indicated that a high concentration of the probiotic strain JCM5805 upregulated the expression of IFNα via the TLR7/TLR9-Myd88 pathway and enhanced disease resistance of larvae. JCM5805 was only transiently detected and thus was not included in the stable larval microbiota. The early microbial exposure of tilapia larvae affects the gut microbiota at later life stages. Vaccination is the most effective approach for prevention of infectious diseases in aquaculture. Although immersion vaccination is more applicable compared to in-feed/oral administration and injection, this method suffers from low potency as the efficiency of uptake of antigens through mucosal membranes is limited. In this study, we have successfully developed a mucoadhesive vaccine delivery system to enhance the efficacy of direct immersion vaccination against Flavobacterium columnare, the causative agent of columnaris disease in red tilapia. A formalin-killed negatively charged, bacterial cell suspension was used to prepare a mucoadhesive vaccine by electrostatic coating with positively charged chitosan. Our results demonstrate that the chitosan-complexed vaccine greatly increases its mucoadhesiveness, thus increasing the chances of vaccine uptake by the gill mucosa and improving the protection obtained against columnaris infection. The surface charge of the chitosan-complexed vaccine was altered from anionic to cationic after chitosan modification. Tilapia were vaccinated with the prepared chitosan-complexed vaccine by immersion. The challenge test was then carried out 30 and 60 days post vaccination, which resulted in a high level of mortalities in the non-vaccinated and uncomplexed vaccine groups. A high relative percentage survival (RPS) of vaccinated fish was noted with the mucoadhesive vaccine. Our results indicated that the naked vaccine failed to protect the fish from columnaris infection, which is consistent with the mucoadhesive assays performed during the study showing that the naked vaccine was unable to bind to mucosal surfaces. Synbiotics, a synergistic combination of probiotics and prebiotics, are currently regarded as one of the most practical nutritional supplements in tilapia farms. In this study, the effect of supplementing the diet of red tilapia (Oreochromis spp.) with Jerusalem artichoke (Helianthus tuberosus) and Lactobacillus rhamnosus GG (LGG) was evaluated. Growth performance, serum biochemical parameters, intestinal morphology, goblet cell counts, immune parameters and protection against Aeromonas veronii challenge were determined. The results showed that fish fed with synbiotic-supplemented diets had a significantly higher (P < 0.05) feed conversion ratio (FCR), specific growth rate (SGR), and average daily gain (ADG) than fish fed with a control diet. The synbiotic-supplemented diet increased glucose, total protein and the total cholesterol levels. The absorptive area of the proximal and distal intestine of fish fed on the synbiotic diet was significantly higher (P < 0.05) than in those fed with probiotics (LGG), prebiotic-supplemented diets (JA), and the control diet. Goblet cell counts revealed that the numbers of acid mucous cells, neutral mucous cells and double-staining mucous cells of fish fed the synbiotic-supplemented diet (JA + LGG) were significantly higher (P < 0.05) in the proximal and distal intestine. Fish fed the synbiotic-supplemented diets also exhibited significantly higher (P < 0.05) lysozyme activity. The cumulative mortalities of fish fed with a synbiotic-supplemented diet were significantly lower than those of fish fed other diets. The results suggested the beneficial effect of JA and LGG synbiotic diet on growth performance and health status of red tilapia. We investigated the effects of dietary enriched with Shilajit in three dose levels: 2.0, 4.0, and 6.0 g kg -1 in the basal diet on growth rate, non-specific immune response, and disease resistance for 60 days in Oreochromis mossambicus against an opportunist pathogen, Aeromonas hydrophila. The results showed that the Shilajit enriched diets at 4.0 and 6.0 g kg -1 diets significantly (p < 0.05) enhance survival rate, weight gain (WG), protein efficiency ratio (PER), specific growth rate (SGR), and feed efficiency (FE), whereas it significantly decrease the feed conversion ratio (FCR) from weeks 1-4 when compared with control. The innate immune parameters, such as the phagocytic, complement, respiratory burst, and lysosome activities in infected fish fed with all Shilajit enriched diet were significantly (p < 0.05) enhances on weeks 2-4. A very low cumulative mortality, 8.0% and 12.0% were observed in the infected fish fed with 4.0 and 6.0 g kg -1 Shilajit enriched diets, whereas 18% mortality was found with 2 g kg -1 diet. In this study we examined the effect that a Francisella noatunensis (Fno) infection had on hybrid red tilapia (Oreochromis niloticus × Oreochromis mossambicus) subsquently infected with Streptococcus agalactiae. A variety of hemato-immunological parameters (haematocrit, total red blood cell count, mean corpuscular volume, total white blood and differential cell counts, total plasma protein, plasma lysozyme and plasma peroxidase activities, and respiratory burst and phagocytic activities of head-kidney macrophages) were measured in hybrid red tilapia that had been previously exposed to an Fno outbreak in a tilapia grow-out farm. The head-kidneys of these apparently healthy survivors, when checked by PCR were found to be Fno-positive with hemato-immunological parameters that were similar to fish without an a priori infection. The only exception was the percentage lymphocyte count in the peripheral blood, which was slightly, but significantly, lower in the Fno-infected fish, compared to those without the infection. When experimentally infected with S. agalactiae, the Fno-infected fish died more rapidly and at a significantly higher rate than fish without the infection. During the challenge, the hemato-immunological parameters of both groups of fish were very similar, although the Fno-infected fish, challanged with S. agalactiae expressed significantly higher plasma lysozyme and peroxidase activities, and their head kidney macrophages had significantly higher respiratory burst activity compared to non-Fno-infected fish challanged with S. agalactiae. The only two parameters for which Fno-infected fish showed significantly lower expressions than that of their non-infected counterparts were haematocrit and total red blood cell count. The cause of the rapidity and higher rates of mortality observed in the Fno-infected fish when challenged with S. agalactiae is unknown; but it may be due to a reduced erythropoiesis capability within the head-kidney because of the presence of Fno., (Copyright © 2018. Streptococcus agalactiae (S. agalactiae) is a gram-positive pathogen that causes a wide range of infections in fish and other animals including humans. Bacterial ghosts (BGs) are nonliving, empty cell envelopes and are well represented as novel vaccine candidates. In this study, we examined the immunogenicity and protective efficacy of S. agalactiae ghosts (SAG) against a virulent challenge in tilapia. Nonliving SAG was generated by a culture with Penicillin and Streptolysin, and then treated with the MIC of sodium hydroxide. The formation of a transmembrane lysis tunnel structure in SAG was visualized by electron microscopy. To investigate the SAG as a vaccine candidate, fish were divided into three groups, A (SAG immunized), B [Formalin-inactivated S. agalactiae (FSA) immunized] and C (phosphate-buffered saline, PBS-immunized control). The IgM antibody responses were significantly stronger in the SAG-immunized group than in FSA-immunized group, which was higher than in the non-immunized control group (P < 0.05). Moreover, phagocytic activity (percent phagocytes, PP) was significantly higher (p < 0.05) in the SAG-immunized group than in FSA-immunized group, which was higher than in the non-immunized control group (P < 0.05). In addition, non-specific immune immunity, such as lysozyme and superoxide dismutase activities, in the SAG-immunized fish showed significantly higher activities than FSA-immunized fish and the control group fish (P < 0.05). Also, fish immunized with SAG and FSA showed significantly higher (p < 0.05) gene expression of IL-1β, TNF-α, IFN-γ and TGF-β in the head kidney and spleen than fish treated with PBS during the whole observed period. In addition, fish immunized with SAG showed significantly higher gene expression of L-1β, TNF-α, and TGF-β in the spleen than in the FSA-immunized fish. Although there was no significant (P > 0.05) difference of survival rate (SR) or relative percent survival (RPS) between SAG and FSA immunized groups, they were all significantly more protected against the S. agalactiae challenge (SR: 86.67%, RPS: 76.395) and (SR: 80.00%, RPS: 67.50%) respectively, compared to the PBS-treated group (SR: 33.33%). The animal intestine provides a competitive environment for the microbiota. Successful colonization by pathogens requires sensing chemical signals to regulate the expression of virulence genes. Some bacteria rely on a two-component chemical signal transduction system, named FusKR, to regulate virulence genes expression by intestinal fucose. Here we construct the fucP gene deletion strain prove FucP regulation of the T3SS in E. tarda. The result showed that the mutant strain had down-regulated significantly the gene expression of FusKR and T3SS compared to the wild-type strain (P < 0.05). This mutant strain significantly increased LD 50 in zebrafish compared to the wild-type strain (P < 0.05), and significantly decreased penetration and motility in mucin than the wild-type strain (P < 0.05). Meanwhile, tilapia infected with mutant strain show significantly reduced E. tarda adherence and colonization than those infected with the wild-type strain (P < 0.05). Fish infected with EIB202 and ΔfucP showed significantly higher (P < 0.05) gene expression of IL-1β, TNF-α, IFN-γ, TGF-β and HSP-70 in head kidney than fish treated with PBS in the whole observed period; however CPP-3 did not show significant differences (P > 0.05) in all groups. Fish infected with EIB202 showed significantly higher (P < 0.05) gene expression of TGF-β in head kidney than fish treated with ΔfucP in the whole observed period; however other cytokines did not show significant differences (P > 0.05) in the whole observed period. In addition, Fish infected with EIB202 showed significantly higher (P < 0.05) gene expression of IL-1β, TNF-α and TGF-β in spleen than fish treated with ΔfucP in the whole observed period, however IFN-γ, CPP-3, and HSP-70 did not show significant differences (P > 0.05) in the whole observed period. Although the gene expression of cytokines was induced similarly by both strains, all results indicate that the fucP gene deletion down-regulates the key gene expression of FucKR and T3SS, reduces the pathogenicity of E. tarda in fish, particularly decreases inducing the gene expression of TGF-β in the head kidney and IL-1β, TNF-α and TGF-β in the spleen., (Copyright © 2018 Elsevier Ltd. Recently, several immunostimulants such as β-glucan, microbial and plant products have been used as dietary supplements to combat disease outbreaks in aquaculture. The present study investigates the potential of Portunus pelagicus β-1, 3 glucan binding protein based zinc oxide nanoparticles (Ppβ-GBP-ZnO NPs) supplemented diet on growth, immune response and disease resistance in Mozambique tilapia, Oreochromis mossambicus. The immune-related protein β-GBP was purified from the haemolymph of P. pelagicus using Sephadex G-100 affinity column chromatography. Ppβ-GBP-ZnO NPs was physico- chemically characterized and experimental feed was formulated. Fish were separately fed with commercial diet (control-group I) and Ppβ-GBP (group II, III, IV), Ppβ-GBP-ZnO NPs (group V, VI, VII), chem-ZnO NPs (VIII, IX, X) mixed diet at the concentration of 0.001%, 0.002% and 0.004% respectively. Triplicate groups of O. mossambicus were fed with experimental diets twice a day for 30 days. Fish receiving Ppβ-GBP-ZnO NPs supplemented diet showed a significant increase (P < 0.05) in growth performance. Cellular immune responses (myeloperoxidase activity, lysozyme activity and reactive oxygen species activity) and humoral immune responses (complement activity, antiprotease activity and alkaline phosphatase activity) were evaluated at an interval of 15 days during the feeding trial. Results demonstrate that both cellular and humoral immune responses were substantially increased (P < 0.05) in fish fed with 0.004% of Ppβ-GBP-ZnO NPs supplemented diet than others. Antibiofilm potential of Ppβ-GBP-ZnO NPs against Aeromonas hydrophila was visualized through confocal laser scanning microscopy (CLSM), which reveals reduction in the preformed biofilm thickness to 10 μm  at the concentration of 50 μg/ml. Furthermore, after 30 days of feeding trial, fish were challenged with aquatic fish pathogen A. hydrophila (1 × 10 7  cells ml -1 ) through intraperitoneal injection. Challenge study displayed a reduced mortality rate in fish fed with diet containing Ppβ-GBP-ZnO NPs. The impact of Mucuna pruriens (L.) seed meal diet on growth performance, innate immune response, and disease resistance in Oreochromis mossambicus against Aeromonas hydrophila is reported for the first time. Infected O. mossambicus was fed with 2 g kg -1 , 4 g kg -1 , and 6 g kg -1 of M. pruriens seed meal diets significantly increased initial body weight (IBW) and final body weight (FBW) over control, for a period of 4 weeks. At 4 g kg -1 and 6 g kg -1 the enriched diet significantly (P < 0.05) enhanced the survival rate, weight gain (WG), protein efficiency ratio (PER), specific growth rate (SGR), feed conversion ratio (FCR), and feed efficiency (FE) when compared to the control; besides, from weeks 2-4 these diets significantly (P < 0.05) enhanced the complement activity, phagocytic activity, respiratory burst activity, and lysosome activity. On being fed with 4 g kg -1 and 6 g kg -1- enriched diets the cumulative mortality was lower (10% and 12%) than with 2 g kg -1 diet (24%). The present investigation suggests that the enriched diet at 4 g kg -1 and 6 g kg -1 improved the growth performance more than that of the control and negative control; it also positively enhanced the innate immunity and disease resistance against A. hydrophilla in O. mossambicus. The present study evaluated the dietary supplementation of probiotic Bacillus licheniformis Dahb1 on the growth performance, immune parameters and antioxidant enzymes activities in serum and mucus as well as resistance against Aeromonas hydrophila in Mozambique tilapia Oreochromis mossambicus. Fish (24 ± 2.5 g) were fed separately with three diets, 1) commercial diet (control), 2) diet containing probiotic at 10 5  cfu g -1 (D1) and 3) diet containing probiotic at 10 7  cfu g -1 (D2) for 4 weeks. Growth performance in term of final weight (FW) specific growth rate (SGR) and feed conversion ratio (FCR), immune parameters of total protein (TP), alkaline phosphatase (ALP), myeloperoxidase (MPO), lysozyme (LYZ), reactive oxygen species (ROS), reactive nitrogen species (RNS) and antioxidant parameters of superoxide dismutase (SOD) and glutathione peroxidase (GPx) in serum and mucus were evaluated after 2 nd and 4 th weeks. The FW, SGR, and FCR of fish fed with D1 and D2 significantly improved (p < 0.05). The activities of ALP, LYZ and MPO in the mucus were significantly higher (p < 0.05) in fish that fed D1 and D2. The TP, ROS, RNS, SOD and GPx in the serum were significantly higher (p < 0.05) in fish that fed D1 and D2. In addition, the challenge test showed that fish fed D1 and D2 enhanced significantly (p < 0.05) the resistance against A. hydrophila (1 × 10 7  cells ml -1 ). The impact of Cucurbita mixta (L.) seed meal enriched diet on growth performance, innate immune response, and disease resistance in Oreochromis mossambicus against Aeromonas hydrophila was investigated. O. mossambicus was fed with 2 g kg -1 , 4 g kg -1 , and 6 g kg -1  C. mixta seed meal diets for a period of 4 weeks. The results indicated that C. mixta seed meal diets at 4 g kg -1 and 6 g kg -1 significantly (P < 0.05) enhances the survival rate, weight gain (WG), protein efficiency ratio (PER), specific growth rate (SGR), feed conversion ratio (FCR), and feed efficiency (FE) from weeks 1-4 when compared to control. C. mixta seed meals administered as feed supplements significantly (P < 0.05) enhanced the complement activity, phagocytic activity, respiratory burst activity, and lysosome activity in infected fish fed with 4 g kg -1 and 6 g kg -1 of C. mixta seed meal enriched diet from weeks 2-4. The cumulative mortality was lower in the fish fed with 4 g kg -1 and 6 g kg -1 of C. mixta seed meal enriched diets (15% and 18%) than with 2 g kg -1 diet (26%). In this research, we focused on the efficacy of aqueous and ethanol leaf extracts of Psidium guajava L. (guava) based experimental diets on the growth, immune, antioxidant and disease resistance of tilapia, Oreochromis mossambicus following challenge with Aeromonas hydrophila. The experimental diets were prepared by mixing powdered (1, 5 and 10 mg/g) aqueous and ethanol extract of guava leaf with commercial diet. The growth (FW, FCR and SGR), non-specific cellular immune (myeloperoxidase activity, reactive oxygen activity and reactive nitrogen activity) humoral immune (complement activity, antiprotease, alkaline phosphatase activity and lysozyme activity) and antioxidant enzyme responses (SOD, GPX, and CAT) were examined after 30 days of post-feeding. A significant enhancement in the biochemical and immunological parameters of fish were observed fed with experimental diets compared to control. The dietary supplementation of P. guajava leaf extract powder for 30 days significantly reduced the mortality and increased the disease resistance of O. mossambicus following challenge with A. hydrophila at 50 μl (1 × 10 7  cells ml -1 ) compared to control after post-infection. A novel cell line THK, derived from the tilapia head kidney, was developed and characterized. The THK cell line comprised fibroblastoid cells that markedly proliferated in Leibovitz L-15 medium containing 2%-15% fetal bovine serum (FBS) at 20 °C-35 °C. Cell proliferation was dependent on the FBS concentration, and the optimal temperature for proliferation ranged between 25 °C and 30 °C. THK cells were characterized for the presence of phagocytic activity, acid phosphatase, alkaline phosphatase, α-naphthyl acetate esterase, lipofuscin, and tyrosinase. Transcripts of CD33, CD53, CD82, CD205, macrophage colony stimulating factor receptor, GATA2, and GATA3 that are specific for leucocytes or monocytes/macrophages or both were detected in the THK cells through PCR. However, THK cells lacked for CD83, a specific marker for dendritic cells. The results indicated that the fibroblastoid THK cells were melanomacrophage-related progenitors. PCR revealed that the THK cells exhibited the transcripts of toll-like receptor 1 (TLR1), TLR2, TLR3, and CD200, of which concern with immunity as well as the transcripts of vascular endothelial growth factor receptor 3, angiomotin, and angiopoietin-like protein 2 that associate with angiogenesis regulation and macrophage proliferation. The immunostimulatory effect of phospholipopeptide biosurfactant from Staphylococcus hominis (GenBank Accession No: KJ564272) was assessed with Oreochromis mossambicus. The non-specific (serum lysozyme activity, serum antiprotease activity, serum peroxidase activity and serum bactericidal activity), specific (bacterial agglutination assay) immune responses and disease resistance activity against Aeromonas hydrophila were examined. Fish were intraperitonially injected with water soluble secondary metabolite (biosurfactant) of S. hominis at a dose of 2 mg, 20 mg and 200 mg kg(-1) body weight. Commercial surfactant surfactin (sigma) at 20 mg kg(-1) was used as standard and saline as negative control. All the doses of water soluble biosurfactant tested, significantly enhanced the specific, nonspecific immunity and disease resistance from the day of post administration of phospholipopeptide biosurfactant till the tail of the experimental period. Tilapia is a group of cultured teleost fishes whose production is threatened by some diseases. Identification of DNA markers associated with disease resistance in candidate genes may facilitate to accelerate the selection of disease resistance. The gene encoding a duodenase, which can trigger immune response, has not been studied in fish. We characterized the cDNA of duodenase-1 gene of hybrid tilapia. Its ORF is 759 bp, encoding a serine protease of 252 amino acids. This gene consisted of five exons and four introns. Its expression was detected in all 10 tissues examined, and it was highly expressed in the intestine and kidney. After a challenge with the bacterial pathogen, Streptococcus agalactiae, its expression was up-regulated significantly in the intestine, liver and spleen. We identified seven SNPs in the gene and found that four of them were significantly associated with the resistance to S. agalactiae (P < 0.05). The CGTCC haplotype, CAGTC/CGGTC and CGTCC/CGTCC diplotype were significantly associated with the resistance to S. agalactiae (P = 0.00, 0.04 and < 0.0001, respectively). In addition, one SNP was associated significantly with growth traits (P < 0.05). These findings suggest that the duodenase-1 gene plays an important role in the resistance to S. agalactiae in tilapia. The SNP markers in the duodenase-1 gene associated with resistance to the bacterial pathogen, may facilitate the selection of tilapia resistant to the bacterial disease., (Copyright © 2015 Elsevier Ltd. This study investigated effects of dietary Aloe vera on growth performance, some haemato-biochemical parameters and disease resistance against Streptococcus iniae in tilapia (GIFT). Five groups were designed including a basal diet (control) and 100% A. vera powder incorporated in fish feed at 0.5% 1%, 2%, and 4%/kg feed, which were administered for 8 weeks. Fish fed 0.5%, 1%, and 2% A. vera supplemented diet significantly improved (p < 0.05) weight gain, absolute growth rate and specific growth rate. Feed intake significantly increased in fish fed with A. vera diet at 1% and 2%/kg feed. Feed efficiency ratio, feed conversion ratio, and hepatosomatic index were significantly enhanced in 4% A. vera supplemented fish over unsupplemented ones (p < 0.05). Several haemato-biochemical indices were examined before and after fish were challenged with S. iniae pathogen containing 7.7 × 10(6) CFU cells mL(-1). A. vera supplemented fish showed a significant increase (p < 0.05) in red blood cells, hematocrits (Hb), hemoglobin (Hb), white blood cells (WBC), neutrophils, monocytes, eosinophils, serum total protein, glucose and cortisol after challenge when compared to unsupplemented ones. Meanwhile, 4% A. vera supplemented fish showed a decrease (p < 0.05) in RBC, Hb, Ht, WBC, and mean corpuscular hemoglobin (MCH) after challenge compared to unsupplemented ones and other supplemented ones. In addition, lower mean corpuscular volume values (MCV) (p < 0.05) were observed in fish fed with A. vera diet at 2% and 4% A. vera/kg feed than those fed unsupplemented diet. Unchallenged fish fed 0.5%, 1%, and 2% A. vera showed significantly higher values (p < 0.05) of mean corpuscular hemoglobin concentration (MCHC) than those fed unsupplemented diet and 4% A. vera supplemented diet. There was a significant increase (p < 0.05) in the neutrophil to lymphocyte ratio (N/L) within experimental groups after challenge; N/L ratio in A. vera unsupplemented fish and those supplemented with A. vera diet at 1%/kg feed increased significantly (p < 0.05) throughout challenge period; while those fed 4% A. vera supplemented diet maintained higher values at all experimental stages among groups. There was a significant correlation (p < 0.05, r = 0.53) between N/L ratio and glucose concentration, 96 h after challenge. Aloe had no significant effect (p > 0.05) on the survival of the fish when compared to the control; no mortality was recorded in challenge trial. This study compares the effects of two Lactobacillus strains, highly adhesive Lactobacillus brevis JCM 1170 (HALB) and less-adhesive Lactobacillus acidophilus JCM 1132 (LALB), on the survival and growth, adhesive gut bacterial communities, immunity, and protection against pathogenic bacterial infection in juvenile hybrid tilapia. During a 5-week feeding trial the fish were fed a diet containing 0 to 10(9) cells/g feed of the two Lactobacillus strains. Samples of intestine, kidney, and spleen were taken at the start and at 10, 20, and 35 days for analysis of stress tolerance and cytokine gene mRNA levels and to assess the diversity of adhesive gut bacterial communities. A 14-day immersion challenge with Aeromonas hydrophila NJ-1 was also performed following the feeding trial. The results showed no significant differences in survival rate, weight gain, or feed conversion in the different dietary treatments. The adhesive gut bacterial communities were strikingly altered in the fish fed either the HALB or the LALB, but the response was more rapid and substantial with the adhesive strain. The two strains induced similar changes in the patterns (upregulation or downregulation) of intestinal, splenic or kidney cytokine expression, but they differed in the degree of response for these genes. Changes in intestinal HSP70 expression levels coincided with changes in the similarity coefficient of the adhesive gut bacterial communities between the probiotic treatments. The highest dose of the HALB appeared to protect against the toxic effects of immersion in A. hydrophila (P < 0.05). Recently, tilapia hepcidin (TH)1-5 was characterized, and its antimicrobial functions against several pathogens were reported. The antimicrobial functions of another shrimp antimicrobial peptide (AMP), chelonianin, were also characterized using a recombinant chelonianin protein (rcf) that was expressed by a stably transfected Chinese hamster ovary (CHO) cell line against pathogen infections in fish. The function of the overexpression of both AMPs in zebrafish muscles was not examined in previous studies. Herein, we investigated the antimicrobial functions of TH1-5 and chelonianin against Vibrio vulnificus (204) and Streptococcus agalactiae (SA48) in transgenic TH1-5 zebrafish and transgenic chelonianin zebrafish. The presence of TH1-5 and chelonianin enhanced the inhibitory ability in transgenic AMP zebrafish against the two different bacterial infections. The bacterial number of either V. vulnificus (204) or S. agalactiae (SA48) had decreased at 96 h after injection into transgenic AMP zebrafish muscle compared to non-transgenic zebrafish muscle. Additionally, immune-related gene expressions analyzed by real-time PCR studies showed the modulation of several genes including interleukin (IL)-10, IL-22, IL-26, MyD88, Toll-like receptor (TLR)-1, TLR-3, TLR-4, nuclear factor (NF)-κB, tumor necrosis factor (TNF)-α, and lysozyme, and significant differences were found between transgenic AMP zebrafish and wild-type zebrafish injected with PBS at 1-24 h. These results suggest that several immune-related gene expressions were induced in transgenic TH1-5 and chelonianin zebrafish which effectively inhibited bacterial growth. The survival rate dropped to 86.6% in transgenic chelonianin zebrafish after 28 days of infection compared of the 50% survival rate in transgenic TH1-5 zebrafish after 28 days of infection. Almost all metabolic processes in an organism alternate through high and low activity phases with a regular periodicity of nearly 24h. These daily/diel variations are governed by factors such as light, weather conditions, availability of food or predator activity. The immune system in fish is expected to follow the same routine based on external cues from the environment which it lives. The present study was carried out to investigate such daily/diel variations in selected immune parameters such as serum lysozyme and peroxidases activity, total serum globulin level and peripheral blood leukocyte count in Oreochromis mossambicus. The fish were maintained in semi natural condition (i.e.12L:12D). The results showed significant rise in serum peroxidases and lysozyme between 0200 h and 0600 h of the day and serum cortisol exhibited elevated level between 2200 h and 0600 h. The present paper describes the effect of water-soluble fraction of the leaves of the Indian medicinal plant, Tinospora cordifolia (Miers) on the non-specific immunity and disease resistance in Oreochromis mossambicus (Peters). Fish were intraperitoneally injected with 0, 6, 60 or 600 mg kg(-1) body weight, of the water soluble fraction. The non-specific humoral (lysozyme, antiprotease and complement) and cellular (production of reactive oxygen and nitrogen species and myeloperoxidase) responses and disease resistance against Aeromonas hydrophila were tested. All the doses of water-soluble fraction tested, significantly enhanced the serum lysozyme, antiprotease and natural haemolytic complement activities on most of the days tested. Similarly, all the doses of water-soluble fraction used, enhanced the cellular myeloperoxidase activity on all the days tested. The enhancement in the ROS and RNI production by peripheral blood leucocytes was observed on almost all the days tested, in most of the treated groups. All the doses of water-soluble fraction when administered as a single or double dose gave protection in terms of reduced percent mortality which is reflected in the increased Relative Percent Survival (RPS) values. The results clearly indicate the immunostimulatory and disease resistance properties of T. cordifolia leaf fraction and so its potential to be used as an immunoprophylactic in finfish aquaculture., (Copyright 2010 Elsevier Ltd. Respiratory burst, lysozyme and phagocytic activities, and immunoglobulin levels in response to the pathogen Aeromonas hydrophila were examined in tilapia (Oreochromis mossanbicus, 10.65 +/- 2.5 g) injected individually with hot-water extract of Toona sinensis at 4 or 8 microg g(-1). Tilapia receiving the hot-water extract of T. sinensis at either dose had significantly increased respiratory burst, phagocytic activity and lysozyme activity towards A. hydrophila by 1 and 2 days post injection. No significant differences in total immunoglobulin levels were observed among the tilapia that received the two different doses of hot-water extract of T. sinensis at 4 and 8 microg g(-1). In another experiment, a Tilapia that had been injected with hot-water extract of T. sinensis was challenged with A. hydrophila at 5 x 10(7) colony-forming units (cfu) fish(-1). The survival of tilapia that received the hot-water extract of T. sinensis at 8 microg g(-1) was significantly higher than fish that received phosphate buffered saline and the control fish after 2 days, and at the termination of the experiment (7 days after the challenge). The objective of the study was to analyze the effect of serum from freshwater (FW) exposed tilapia or from 25 ppt seawater (SW) exposed tilapia on the ability to mediate the phagocytic activity of tilapia phagocytes. To analyze the phagocytic activity, head kidney (HK) and spleen leukocytes were tested in 300 or 500 mOsm medium using three different treatment groups (a) control, (b) addition of 25% serum from freshwater (FW) exposed tilapia, and (c) addition of 25% of serum from 25 ppt seawater (SW) exposed tilapia. HK leukocytes cultured in 300 and 500 mOsm media for 4 h showed an increase of phagocytic ability in the control group as compared to the addition of serum from either FW or SW exposed tilapia. HK leukocytes exposed to 500 mOsm medium showed a higher phagocytic ability than those leukocytes exposed to 300 mOsm medium in each corresponding group. Concurrently, spleen leukocytes in the control group showed a higher phagocytic ability than those leukocytes with the addition of serum from FW or SW exposed tilapia. As compared to spleen leukocytes cultured in 300 mOsm medium, leukocytes cultured in 500 mOsm medium showed an increase of phagocytic ability within their respective group. To further investigate the observed phenomenon, 2D-gel electrophoresis was performed for analyzing the differentially expressed proteins in serum that was thought to influence the phagocytic ability. Up-regulated serum proteins in SW exposed tilapia contained complement C3 protein, NADH dehydrogenase (Ubiquinone) Fe-S protein 3, Mg(2+)-dependent neutral sphingomyelinase, Semaphorins, and Caspase 3. Taken together these results suggest that addition of serum decreased the phagocytic activity in HK and spleen leukocytes in vitro, furthermore, induced proteins semaphorin, complement C3, Mg(2+)-dependent neutral sphingomyelinase, and Caspase 3 are up-regulated in the serum, which might have decreased the phagocytic activity upon exposure to hyperosmotic solutions.

Most of the tilapia studies are focused on its osmoregulatory mechanism. Meanwhile, less information is available about its innate immune response on fish faced with hyperosmolality. In the present study, in vivo analyses were carried out to investigate the innate immune response of Oreochromis mossambicus, transferred from freshwater to 25 ppt seawater (SW). In vivo, lysozyme activities of plasma and head kidney (HK) were increased at 1h and at 24h after transfer to SW but decreased at 8h after SW transfer. Surprisingly, the alternative complement pathway in plasma increased 8h after SW transfer. The phagocytic capacity of spleen and HK immune cells increased modestly at 1h and at 4h, after SW transfer, but the respiratory burst activity of immune cells in both HK and spleen shows an increase in superoxide release at 8h after SW transfer. Our results reveal that the transfer of fish from conditions of hypoosmolarity to hyperosmolality significantly enhances plasma lysozyme, ACP activity, and both phagocytic and respiratory burst activity. Taken together, the results indicate that exposure of tilapia to hyperosmotic conditions has immunostimulatory effects on its cellular immune reactions (phagocytosis and respiratory burst activity) and humoral reactions (lysozyme activity and complement activity).

Immunostimulatory effects of the oral administration of the medicinal plant, Eclipta alba leaf extracts was studied in tilapia, Oreochromis mossambicus. For this purpose, fish were fed for 1, 2 or 3 weeks with diets containing E. alba leaf aqueous extract at 0, 0.01, 0.1 or 1% levels. After each week, non-specific humoral (lysozyme, antiprotease and complement) and cellular (myeloperoxidase content, production of reactive oxygen and nitrogen species) responses and disease resistance against Aeromonas hydrophila were determined. The results indicated that E. alba aqueous extract administered as feed supplement significantly enhanced most of the non-specific immune parameters tested. Among the humoral responses, lysozyme activity significantly increased after feeding with aqueous extract for 1, 2 or 3 weeks. No significant modulation was noticed in all the cellular responses tested after 3 weeks of feeding, while reactive oxygen species production and myeloperoxidase content showed significant enhancement after 1 week of feeding with aqueous extract. When challenged with A. hydrophila after 1, 2 or 3 weeks of feeding, the percentage mortality was significantly reduced in the treated fish. The highest dose of 1% gave better protection than the other doses with the relative percentage survival (RPS) values of 64, 75 and 32 after feeding for 1, 2 and 3 weeks respectively. The results indicate that dietary intake of E. alba aqueous leaf extract enhances the non-specific immune responses and disease resistance of O. mossambicus against A. hydrophila.

The purpose of this study was to determine the effect of water and hexane soluble fractions of the Indian medicinal plant, Solanum trilobatum on the nonspecific immune mechanisms and disease resistance in Oreochromis mossambicus. Fish were intraperitoneally injected with different doses of 0, 4, 40 or 400 mg kg(-1) body weight of water or hexane soluble fraction. The nonspecific immune mechanisms were assessed in terms of serum lysozyme activity, reactive oxygen species production and reactive nitrogen species production by peripheral blood leucocytes. The functional immunity in terms of percentage mortality and Relative Percent Survival (RPS) was assessed by a challenge with live Aeromonas hydrophila. Almost all the doses of both water and hexane soluble fractions enhanced the serum lysozyme activity. All the doses of water soluble fraction significantly enhanced the ROS production on most of the days tested. In hexane soluble fraction treated groups, the enhancement in the ROS production was observed at least on 2 days. All the doses of water soluble fraction significantly enhanced the production of RNS only on one day. The RNS production was enhanced significantly only in the group treated with 40 mg kg(-1) of hexane fraction. The leaf fraction administration preceding the challenge with live A. hydrophila, decreased the percentage mortality in the experimental group with the consequent increase in RPS values. This preliminary study indicates that S. trilobatum could be used to promote the health status of fish in intensive finfish aquaculture.

The present study was to determine the in vitro effects of singular or combined anti-oxidative vitamins (A, C, and E) and/or minerals (Se, Zn, Cu, Mn, and Fe) on the immune functions of tilapia, Oreochromis hybrids, peripheral blood monocyte-derived, anterior kidney-derived, and spleen-derived macrophages. An optimal dose of vitamins and minerals increased cell viability and lysozyme activity. On the other hand, the above activities decreased at the high doses of combined vitamins (A+C+E group, each 300 microg mL(-1)) or single mineral (Se, Zn, Cu, Mn, and Fe groups, each 200, 800 or 1000 microg mL(-1)). Combining two of the aforementioned vitamins (A+C, A+E, and C+E groups, each 100 microg mL(-1)) was able to prolong cell viable time up to 72 h compared with singular vitamin addition. Before or after adding vitamins or minerals during infection, addition of vitamins decreased the percentage of dead cells and a greater effect was observed for mineral (each 40 or 80 microg mL(-1)) and vitamin (each 100 microg mL(-1)) combinations. A low dose of vitamins increased nitric oxide production and decreased superoxide production, but high dose of vitamins decreased superoxide and nitric oxide productions. Furthermore, minerals also decreased nitric oxide production at concentrations of 40, 80, 200, 800 or 1000 microg mL(-1). The threshold concentrations for cell death by necrosis and/or apoptosis were >1000 and >800 microg mL(-1) for vitamins and minerals, respectively. In conclusion, appropriate concentration of vitamins or minerals can increase tilapia macrophage immunity; nevertheless, extreme concentrations of vitamins or minerals are lethal to cells.

Mozambique tilapia Oreochromis mossambicus acclimated to 27 degrees C were then held at 19, 23, 27 (control), 31 and 35 degrees C, and were examined for non-specific cellular and humoral responses after 12-96 h. Total leucocyte count decreased significantly when fish were transferred to 19 and 23 degrees C after 48 and 96 h, and when transferred to 35 degrees C over 12-96 h, respectively. Respiratory burst decreased significantly when fish were transferred to 19, 31 and 35 degrees C over 24-96 h, whereas phagocytic activity and phagocytic index decreased significantly when fish were transferred to low temperatures (19 and 23 degrees C) and high temperatures (31 and 35 degrees C) over 12-96 h. Lysozyme activity decreased significantly when fish were transferred to 19 degrees C after 12-96 h, but increased significantly when transferred to 31 and 35 degrees C over 48-96 h. Alternative complement pathway (ACH(50)) also decreased significantly when transferred to 19 and 23 degrees C after 12h, but increased significantly when transferred to 31 and 35 degrees C after 24h. In another experiment, tilapia reared at 27 degrees C were injected intraperitoneally with Streptococcus iniae at a dose of 1 x 10(7)colony-forming units (cfu)fish(-1), and then reared onward at water temperatures of 19, 23, 27 (control), 31 and 35 degrees C. Over 48-168 h, the cumulative mortality of S. iniae-injected fish held in 19 and 35 degrees C was significantly higher than that of injected-fish held in 23, 27 and 31 degrees C. It is concluded that transfer of tilapia O. mossambicus from 27 degrees C to low temperatures (19 and 23 degrees C) after 12h, and transfer of fish from 27 degrees C to high temperatures (31 and 35 degrees C) reduced their immune capability. Moreover, tilapia under temperature stress at 19 and 35 degrees C from 27 degrees C decreased its resistance against S. iniae.

The role of FasL/FasR pathways of immunoregulation of programmed cell death in teleost cytotoxic innate immunity has not been previously examined. In the present study, constitutive cytosolic soluble FasL (sFasL) was detected in anterior kidney (AK), peripheral blood (PBL) and liver NCC obtained from tilapia. Ligation of NCC by tumour cells caused the release of sFasL that was associated with lysis of HL-60 targets in 14 h killing assays. Evidence that sFasL mediated this activity was that anti-(human) FasL inhibited tilapia and catfish (cf.) NCC lysis of FasR+ HL-60 tumour cells. Inhibition was concentration dependent. Lysis of IM-9 targets (12% positive for FasR) by (cf.) anterior kidney and PBL NCC was only partially inhibited by anti-FasL mab. Activated NCC from both species were negative for the expression of membrane FasL and FasR. These data confirmed that NCC lyse sensitive tumour cells by multiple effector pathways. Pretreatment of (FasR+) HL-60 cells with anti-FasR mab completely inhibited cf. cytotoxicity at low (100:1) E:T ratios. Anti-FasR mab did not inhibit the lysis of IM-9 targets by cf. NCC. This study demonstrated that for catfish and tilapia, initial target cell conjugate formation was required; however, the terminal killing mechanism depended on at least two different pathways of cytotoxicity. One pathway depended on the release of preformed soluble FasL by activated NCC in the presence of FasR positive target cells. A second pathway has yet to be determined.

The objective of this study was to investigate the effect of trivalent (chromic chloride) and hexavalent (potassium dichromate) forms of chromium in the African mouth breeder Oreochromis mossambicus (Peters), with reference to the humoral immune response and lymphoid cells/organs. The 96 h LD50 for hexavalent and trivalent chromium was found to be 75 and 1,000microg fish(-1), respectively. Groups of fishes were injected intraperitoneally with 10, 1, 0.1 and 0.01% LD50 hexavalent and trivalent forms of chromium and subsequently immunised with bovine serum albumin (5 mg in 0.2 ml physiological saline). Both forms of chromium suppressed the antibody response, with hexavalent chromium being more suppressive than trivalent chromium. Reduction in spleen weight, splenocyte number and the percentage of blood lymphocytes was observed following administration of both forms of chromium. The possible immunological mechanisms behind the differential suppression of the antibody response and the reduction in spleen weight, splenocyte and lymphocyte counts are discussed.

An important component of immediate innate responses of tilapia to stress is the release within minutes of soluble cytokine-like substances into the peripheral circulation. These cytokine-like stress factors bind nonspecific cytotoxic cells (NCC) and produce 3-4-fold increased cytotoxicity. In the present study, the in vivo responses of tilapia NCC following injection with different isolates of intact killed Streptococcus iniae was investigated. Activated cytotoxicity of NCC in the peripheral blood (PB) was produced by increased specific activity of resident cells rather than increased numbers. Tilapia injected intravenously (i.v.) with killed S. iniae produced different cytotoxicity responses compared to fish injected intraperitoneally (i.p.). In the spleen (S) and anterior kidney (AK), there was no correlation between S. iniae isolate and cytotoxicity response at 4, 8 or 24 h following i.p. injection. The NCC response following i.v. injection of killed bacteria was different. Within minutes following i.v. injection, NCC cytotoxicity from the PB increased 100% compared to naive controls. The existence of subsets of differentiated NCC in the PB was suggested because i.v. injection had no amplification effects on NCC from the AK or S. Likewise, NCC from the PB only appeared to exhibit a degree of antigen specificity. S. iniae strain #173 produced activation of cytotoxicity compared to isolates #164 and ATCC. Evidence for soluble factor (cytokine?) involvement in increased cytotoxicity was obtained by passive activation of NCC with serum from #173 (i.v.) injected fish. Incubation of this serum with control (naïve) NCC produced large increases in the cytotoxicity of labelled HL-60 target cells. Similarly obtained serum from fish injected with ATCC and #164 isolates had no amplification activity. Studies were also performed to study the mechanism(s) of passive activation. Flow cytometric analysis revealed that NCC from the S, AK and PB constitutively expressed cytosolic (not membrane) FasL. Stress serum treated NCC obtained from the peripheral blood produced an increase in the expression of FasL, CAS and FADD by Western blot examination. These data indicated that cytokine like factors in the serum of stressed tilapia activate increased NCC cytotoxicity (possibly) by stimulating the expression of proteins involved in activation of programmed cell death.
