Cyclocarya paliurus (Batal.) leaves, which contain a range of bioactive compounds, have been used as a traditional Chinese medicine homologous food since ancient times. However, there is a paucity of literature on comprehensive studies of alkaloids in the leaves of Cyclocarya paliurus (Batal.). For the first time, this study aimed to discover and identify alkaloids extracted from Cyclocarya paliurus (Batal.) leaves by ultra-high performance liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry (UPLC-QTOF-MS). A total of ten alkaloids have been identified from Cyclocarya paliurus (Batal.) leaves based on accurate mass spectra (mass accuracy, isotopic spacing and distribution) and comparison to fragmentation spectra reported in the literature. In vitro, alkaloids alleviated insulin resistance by increasing glucose consumption and glycogen content in insulin resistance HepG2 cells. The RNA-seq and western blotting results showed that alkaloids could upregulate the expression of phosphatidylinositol 3-kinase (PI3K), and increase the phosphorylation of insulin receptor protein kinase B (AKT). This study not only clarified the chemical constituents and revealed that diverse alkaloids also presented from Cyclocarya paliurus (Batal.) leaves, also, it will provide chemical information on potential compounds for developing new drugs. • Changes in the volatiles of navel orange were investigated during maturation. • Principal component analysis showed good separation of fruits at different maturity. • Changes in the relative content of volatiles were clearly observed during growth. This study investigated the changes in volatile organic compounds (VOCs) of navel orange during fruit maturation. The volatiles in navel orange from July (young fruit period) to November (commercial maturation period) were separated by headspace solid-phase microextraction (HS-SPME) and analyzed using gas chromatography–mass spectrometry (GC–MS). Fifty-seven VOCs were identified and classified into five groups. Through principal component analysis, the samples of different months were discriminated, and one-way analysis of variance showed that 55 volatiles significantly differed during growth. Among the VOCs, the relative concentration of monoterpenes increased with maturity, and D-limonene was the component with the highest content. Similar behavior was exhibited for aldehydes, reaching the maximum content in November. On the contrary, esters decreased its concentration from July to November. The concentration of sesquiterpenes gradually increased, reaching the maximum level between August and November. Moreover, alcohols reached theirs largest content in September, β-Linalool had the highest content among the alcohols. These results provided important theoretical support for the development of navel orange components and comprehensively elucidate the flavor characteristics during the ripening of navel orange. An original way of brewing Cyclocarya paliurus leaves, especially in China, is to steep leaves in hot water before people drink it directly. Recently, infusing tea leaves with cold water, a creative way in Taiwan, has become a popular way of making tea. This study was designed to investigate the differences in metabolites among three brewing methods (cold-brewing, steep-brewing, and boil-brewing) based on UPLC-QTOF-MS metabolomics experiments and the feasibility of cold-brewing methods for C. paliurus leaves. Unsupervised analysis (PCA) explained 54.6% (positive ion mode) and 57.4% (negative ion mode) of the total variance, whereas supervised analysis (OPLS-DA) with cross-validated R2Y and Q2 values > 0.5, could reveal potential metabolites with better discrimination among the three brewing methods. Fifteen potential differential metabolites were chosen and identified, and nine of them were further confirmed with reference standards. This study suggested that the cold-brewing method without an increase in temperature protected the phenol aromatic ring, thereby obtaining more phenolic acid compounds from C. paliurus leaves. The composition and structure of the food matrix can have a major impact on the digestion. The aim of this work was to investigate the effects of endogenous proteins and lipids on starch digestibility in rice flour, with an emphasis on establishing the underlying physicochemical mechanisms involved. Native long-grain indica rice flour and rice flour with the lipids and/or proteins removed were subjected to a simulated digestion in vitro . A significant increase in starch digestibility was observed after removal of proteins, lipids, or both. The starch digestibility of the rice flour without lipids was slightly lower than that without proteins, even though the proteins content was about 10-fold higher than the lipids content. Microstructural analysis suggested that the proteins and lipids were normally attached to the surfaces of the starch granules in the native rice flour, thus inhibiting their contact with digestive enzymes. Moreover, the proteins and lipids restricted the swelling of the starch granules, which may have decreased their digestion by reducing their surface areas. In addition, amylose-lipid complex was detected in the rice flour, which is also known to slow down starch digestion. Abstract: A fungal endophyte MD89 with obvious antioxidant activities was isolated from pigeon pea and identified as Chaetomium globosum by ITS sequence. Different fractions from MD89 culture were compared and evaluated by total phenol (TP) content, total flavonoid (TFL) content, DPPH radical scavenging, reducing power and lipid peroxidation assays, respectively. Results showed that EtOAc extracts had high content of TP and TFL, and strong antioxidant activities (IC50 value was 6.87, 15.19, 16.78μg/mL, respectively). Furthermore, the EtOAc extracts were analyzed by LC–MS/MS and a good antioxidant compound, apigenin, was found. The activities of superoxide dismutase (SOD) and glutathione reductase (GR) in HepG2 cells treated with MD89 EtOAc extracts were also determined. It is revealed that EtOAc extracts increased the activities of SOD and GR in HepG2 cells significantly in a dose-dependent manner. On the basis of those results, we conclude that MD89 represents a valuable natural antioxidant source and may be applicable in health food industry potentially. [Copyright &y& Elsevier]

[Display omitted] • We isolated a novel phage LPSTLL with broadest lytic range infecting Salmonella among the reported phages. • A new genus of Siphoviridae family, LPSTLLvirus , with LPSTLL as the type phage, was proposed. • LPSTLL has ability as a candidate for Salmonella biocontrol agent in the food matrices without impact on the sensory quality. Salmonella is one of the most common foodborne pathogens around the world. Phages are envisioned as a new strategy to control foodborne pathogenic bacteria and food safety. A Salmonella specific lytic phage vB_SalS-LPSTLL (LPSTLL) was selected for food applications on the basis of lytic range, lytic efficiency, functional stability and characteristics. Phage LPSTLL was able to lyse 11 Salmonella serotypes, which represents the broadest range reported Salmonella phages, and was able to suppress the growth of Salmonella enterica in liquid culture over nine hours. LPSTLL exhibited rapid reproductive activity with a short latent period and a large burst size in one-step growth experiment. LPSTLL remained active over a pH range of 3.0 to 12.0, and at incubation temperatures up to 60 °C for 60 min, indicating wide applicability for food processing and storage. Significant reductions of viable Salmonella were observed in diverse foods (milk, apple juice, chicken and lettuce) with reductions up to 2.8 log CFU/mL recorded for milk. Sensory evaluation indicated that treatment with phage LPSTLL did not alter the visual or tactile quality of food matrices. Genome analysis of LPSTLL indicated the absence of any virulence or antimicrobial resistance genes. Genomic comparisons suggest phage LPSTLL constitutes a novel member of a new genus, the LPSTLLvirus with the potential for Salmonella biocontrol in the food industry. [ABSTRACT FROM AUTHOR]
