Korean pine is a native tree species in Northeast China. In order to meet the needs of germplasm resource evaluation and molecular marker-assisted breeding of Korean pine, we collected Korean pine clones from 7 populations in Northeast China, analyzed the genetic diversity and genetic structure by SSR molecular marker technology and clustered them to revealed the inter- and intrapopulation differentiation characteristics of each clone. The fingerprint profiles of 161 Korean pine clones were also constructed. 77 alleles were detected for 11 markers, and 18 genotypes were identified on average for each marker. The PIC of the different markers ranged from 0.155-0.855, and the combination of PI and PIsibs for the 11 markers was 3.1 × 10-8 and 1.14 × 10-3, respectively. MANOVA showed that genetic variation existed mainly within populations, accounting for 98% of the total variation. The level of genetic differentiation among populations was low, with an average Nm between populations of 11.036. Genetic diversity is lower in the Lushuihe population and higher in the Tieli population. The 161 Korean pine clones were divided into 4 or 7 populations, and the 7 populations were not clearly distinguished from each other, with only the Lushuihe population showing partial differentiation. There is no significant correlation between the genetic distance of Korean pine populations and the geographical distance of their superior tree sources. This result can provide recommendations for future Korean pine breeding programs. The combination of 11 markers could completely distinguish 161 clones and establish the fingerprint. Genetic diversity of Korean pine clones from the 7 populations was abundant, and the genetic distances of individuals and populations were evenly dispersed. The latest trends in hazelnut production are moving in the direction of selection and breeding of more productive cultivars, isolation of native clones, and more intensive clonal selection of rootstocks aimed at enhancing the agronomic performance of plants. Serbia stands out in the production of quality planting material by grafting on Turkish filbert (Corylus colurna L.), which does not form shoots and develops in the form of a tree. The aim of this research was to investigate the success achieved by grafting leading Italian cultivars (Tonda gentile romana, Tonda di Giffoni, and Tonda Gentile della Langhe) and their clones on Turkish filbert seedlings using technology developed at the University of Novi Sad, Faculty of Agriculture, Serbia, as well as determine possible differences in the quality and variability of the obtained planting material. For this purpose, from the end of March to the end of April, two-year-old C. colurna generative rootstocks (seedlings) were grafted by the whip and tongue method. At the beginning of September, the grafted plants were counted, and after the plants entered the dormant period (autumn in the year of grafting), they were taken out of the soil and classified. The obtained results revealed that the chosen hazel cultivars and clones exhibited excellent grafting success rate. In both analyzed years, as well as throughout the entire study period, greater grafting success was achieved using clones relative to the main cultivars. Over the two-year study period, the highest grafting success was achieved by clone AD17. Class I grafted plants were obtained in 80% of the cases, especially with Tombesi and AD17 clones, while significantly fewer Class I grafted plants were produced by grafting basic cultivars. Clones AD17 and Tombesi also produced grafted plants of greatest height and graft union diameter. Valeriana jatamansi Jones (Syn. V. wallichii DC.) is an aromatic, medicinal herb used as a tranquilizer and in treating sleep disorders. Rhizome is mainly used to extract essential oil (EO) and valepotriates. High quality and economic yield of rhizomes are available in the third year of growth. Therefore, the cultivation of V. jatamansi is not picking up, and over-exploitation of this plant from wild habitats to meet the increasing demand of the pharmaceutical industry is the cause of threat to the genetic diversity of the species. Further, collections from the wild are heterogeneous, resulting in variable produce. The development of clonal lines can ensure uniform quality and yield of rhizome biomass. An effective clonal propagation method was standardized using different hormonal concentrations of naphthalene acetic acid (NAA) on apical shoot cuttings from the selected clone CSIR-IHBT-VJ-05 for different time durations and raised over various planting media. NAA treatment of 50 ppm concentration for 30 min was found optimum for root induction in apical shoots of V. jatamansi. Variations for EO composition within the clone were non-significant, while samples of the control population were variable. The best quality EO (patchouli alcohol ∼62%) was available during the third year of plant growth. A propagation technique for large-scale quality plant material (QPM) production has been standardized to reduce the stress over natural resources and promote V. jatamansi for use in the aromatic and pharmaceutical industry.

NAC transcription factors are one of the largest transcription factor families having functions in a variety of stress responses. Few NACs have been reported for interactions between wheat and the wheat rust fungus Puccinia triticina (Pt). In this study, based on analysis of RNA-seq data from wheat line TcLr19 inoculated by Pt, the NAC transcription factor TaNAC069 was cloned from wheat, and its transcriptional activity and homologous dimer formation were verified. Quantitative real-time PCR analysis showed that the expression of TaNAC069 was induced by Pt and associated signaling molecules. To further characterize the function of the TaNAC069 gene in wheat resistance to Pt, virus-induced gene silencing (VIGS) was utilized, and it revealed that Pt resistance in TaNAC069-silenced plants was significantly reduced. Potential interaction targets of TaNAC069 from wheat and Pt were screened and identified by yeast two-hybrid technology. Eukaryotic elongation factor eEF1A, CBSX3 protein, and cold acclimation protein WCOR410c were screened by yeast one-hybrid technology. The results indicate that the TaNAC069 gene plays a positive regulatory role in wheat resistance to Pt, laying a good foundation to analyze the molecular mechanisms of TaNAC069 and its functional role in wheat resistance to Pt.

The expression of Esca disease symptoms depends on several factors, including the grapevine cultivar. In this context, a possible clone-dependent expression of the Esca disease was studied. Two clones of ‘Chardonnay’ grown in the same plot were compared according to their developmental and physiological traits, metabolome, and foliar symptom expression. Analysis of their leaf metabolome highlighted differences related to symptom expression. Interestingly, the content of a few specific metabolites exhibited opposite variations in leaves of symptomatic shoots of clones 76 and 95. Altogether this study showed a clone-dependent expression of Esca disease in ‘Chardonnay’ and the relevance of GC-MS and 3D fluorescence methods to analyze the impact of the disease on the leaf metabolome.

Knowledge on gene action and trait expression are important for effective breeding. The objective of this study was to determine the general combining ability (GCA), specific combining ability (SCA), maternal effects and heritability of drought tolerance, yield and yield components of candidate sweetpotato clones. Twelve genotypes selected for their high yield, dry matter content or drought tolerance were crossed using a full diallel mating design. Families were field evaluated at Masoro, Karama, and Rubona Research Stations of Rwanda Agriculture Board. Success rate of crosses varied from 1.8 to 62.5% with a mean of 18.8%. Family by site interaction had significant effect (P < 0.01) on storage root and vine yields, total biomass and dry matter content of storage roots. The family effects were significant (P < 0.01) for all parameters measured. Broad sense heritability estimates were 0.95, 0.84, 0.68, 0.47, 0.74, 0.75, 0.50, and 0.58 for canopy temperature (CT), canopy wilting (CW), root yield, skin color, flesh color, dry matter content, vine yield and total biomass, respectively. The GCA effects of parents and SCA effects of crosses were significant (P < 0.01) for CT, CW, storage root, vine and biomass yields, and dry matter content of storage root. The ratio of GCA/SCA effects for CT, CW, yield of storage roots and dry matter content of storage roots were higher than 50%, suggesting the preponderance of additive over non-additive gene action in the expression of these traits. Maternal effects were significant (P < 0.05) among families for CT, CW, flesh color and dry matter content, vine yield and total biomass. Across sites, the best five selected families with significant SCA effects for storage root yield were, Nsasagatebo × Otada 24, Otada 24 × Ukerewe, 4-160 × Nsasagatebo, K513261 × 2005-034 and Ukerewe K513261 with 11.0, 9.7, 9.3, 9.2, 8.6 t/ha, respectively.
