Advances in molecular pharmacology and an improved understanding of the mechanism of most diseases have created the need to specifically target the cells involved in the initiation and progression of diseases. This is especially true for most life-threatening diseases requiring therapeutic agents which have numerous side effects, thus requiring accurate tissue targeting to minimize systemic exposure. Recent drug delivery systems (DDS) are formulated using advanced technology to accelerate systemic drug delivery to the specific target site, maximizing therapeutic efficacy and minimizing off-target accumulation in the body. As a result, they play an important role in disease management and treatment. Recent DDS offer greater advantages when compared to conventional drug delivery systems due to their enhanced performance, automation, precision, and efficacy. They are made of nanomaterials or miniaturized devices with multifunctional components that are biocompatible, biodegradable, and have high viscoelasticity with an extended circulating half-life. This review, therefore, provides a comprehensive insight into the history and technological advancement of drug delivery systems. It updates the most recent drug delivery systems, their therapeutic applications, challenges associated with their use, and future directions for improved performance and use.

In this work, CuO nanoparticles (NPs) were prepared from the aqueous extracts of Solanum macrocarpon fruit by using the conventional heating (CuO(h) NPs) and microwave irradiation (CuO(m) NPs) methods. The synthesized nanoparticles were characterized using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and X-ray diffraction (XRD). The SEM analysis revealed that the CuO NPs from both routes contained essentially smooth surfaces, and displayed some degree of agglomeration. The TEM analysis confirmed some spherical morphology with mean particle sizes of 35.60 ± 6.24 nm and 47.14 ± 6.18 nm for the CuO(h) and CuO(m) NPs respectively. While the CuO(m) NPs possessed a single-phase consistent with the face cantered cubic structure of copper oxide, the CuO(h) NPs showed some extra peaks attributed to Cu2O NPs as secondary phase. Electrochemical studies were conducted in order to evaluate the electrochemical properties of the NPs. The responses of a gold screen-printed electrode surface treated with both NPs showed that their redox behaviours on (Fe(CN)6)3−/4− probe and KCl electrolytes vary significantly. In (Fe(CN)6)3−/4− probe, the SPAuE/CuO(h) showed enhanced electrochemical response relative to the bare, while the SPAuE/CuO(m) showed a lower current response than the bare. However, in the KCl electrolye, the SPAuE/CuO(h) and SPAuE/CuO(m) were highly electroactive and demonstrated peak current magnitude that was about 26.5 and 83.38 times higher than that of the bare. In this KCl medium, the magnitude of the oxidation peak current of Cu2+ for SPAuE/CuO(m) was about 3 times higher than that of SPAuE/CuO(h). The percentage contraction in redox coordinates between the 1st and 10th scans in both electrodes were 3.88 and 19.28% for SPAuE/CuO(h) and SPAuE/CuO(m) respectively. Thus, the choice of green synthesis route could be exploited in different fields where green NPs is desired.

Despite advances in managing human immunodeficiency virus (HIV) infection and success in the treatment prognosis using highly active antiretroviral therapy (HAART). The clinical efficacy of this regimen has been associated with increased adverse effects such as metabolic derangements and reproductive dysfunctions. These adverse effects necessitate a nanoparticle delivery vehicle like silver nanoparticles (AgNPs), a multi-functional drug delivery system, to transport the HAART to the viral reservoir site like testis.This study was therefore designed to evaluate the effects of HAART loaded AgNPs (HAART-AgNPs) on testicular oxidative stress markers, an inflammatory biomarker, and histomorphology in a rat model of diabetes.Thirty-six adult male Sprague-Dawley rats were randomly divided into two groups (n = 18) non-diabetic and fructose-streptozotocin (Frt-STZ) induced type 2 diabetes (T2DM). Thereafter, both groups were subdivided into three (n = 6) and treated with distilled water, HAART and HAART-AgNPs.HAART-AgNPs caused a significant increase (p < 0.05) in catalase (23.43 ± 0.92) level vs diabetic control (16.95 ± 1.04). Also, HAART-AgNP caused a significant reduction (p < 0.05) in malondialdehyde, interleukin-6 and blood glucose levels (1.94 ± 0.06, 93.65 ± 3.6, 287.33 ± 22.85 respectively), compared to their respective diabetic control values (2.18 ± 0.12, 143.4 ± 9.2, 372.16 ± 23.16). Furthermore, HAART-AgNPs mitigated tubular atrophy, basement membrane thickening, interstitial distension, fibrous elemental distortion and peri-interstitial tissue alterations in the testis of diabetic rats.The results from this study showed that administration of HAART-AgNPs to diabetic rats reduced testicular inflammation, improved glycaemic control, antioxidant status, and testicular histology. Therefore, conjugation of AgNP with HAART may cater for the reproductive dysfunction during the management of HIV infection.

This study was aimed at investigating the relationship of trace metals with prevalence of bacteria and fungi in polluted farmland of Egbema Kingdom, Delta state. This may contribute to the knowledge on the influence of the microorganism to the arability of agricultural soil from an ecotoxicological viewpoint. Soil samples were analyzed for physico-chemical parameters and trace metals using atomic absorption spectrophotometer (Buck Scientific, 210VGP). The samples were also screened for fungi and bacteria isolates. Trace metals (Fe, Zn, Cd, Ni and Cu) determined in the agricultural soils of Egbema kingdom were below WHO limits. The fungi species with the highest number of occurrence was Aspergillus niger, while the species with the lowest occurrence were Fusarium oxsporium, Penicillium dirty white, and Penicillium sp. The order of abundance of fungi across the stations was Station 2> Station 4> Station 3> Station 1> Station 7> Station 5> Station 6> Station 9> Station 8. The bacteria species with the highest number of occurrence was Proteus sp I, while the species with the lowest occurrence was Alcaligenes sp. which occurred only once at Station 5. The order of abundance of bacteria in the soil samples across the stations was Station 4> Station 3> Stations 5 and 9> Station 2> Station 8> Station 7> Station 6. All trace metals in the agricultural soils of Egbema kingdom were below WHO limits. Fe, Zn, Cd and Ni showed no relationship with the microbial contents of the soil. Various strains of fungi and bacteria (gram positive and gram negative) were observed in the soil samples. An antagonistic relationship was observed between bacteria and fungi. Significant positive correlation occurred between the concentration of copper and bacteria counts indicating that the concentration of copper in the soil might have enhanced the abundance of bacteria in the soil, vice versa. The bacteria might be a promising tool in regulation of soil copper concentration.
