In recent years, we have witnessed the explosion of large-scale data in various fields. Classical statistical methodologies, such as linear regression or generalized linear regression, often show inadequate performance on heterogeneous data because the key homogeneity assumption fails. In this paper, we present a flexible framework to handle heterogeneous populations that can be naturally grouped into several ordered subtypes. A local model technique utilizing ordinal class labels during the training stage is proposed. We define a new “progression score” that captures the progression of ordinal classes, and use a truncated Gaussian kernel to construct the weight function in a local regression framework. Furthermore, given the weights, we apply sparse shrinkage on the local fitting to handle high dimensionality. In this way, our local model is able to conduct variable selection on each query point. Numerical studies show the superiority of our proposed method over several existing ones. Our method is also applied to the Alzheimer’s Disease Neuroimaging Initiative data to make predictions on the longitudinal clinical scores based on different modalities of baseline brain image features. Robust and effective shape prior modeling from a set of training data remains a challenging task, since the shape variation is complicated, and shape models should preserve local details as well as handle shape noises. To address these challenges, a novel robust projective dictionary learning (RPDL) scheme is proposed in this paper. Specifically, the RPDL method integrates the dimension reduction and dictionary learning into a unified framework for shape prior modeling, which can not only learn a robust and representative dictionary with the energy preservation of the training data, but also reduce the dimensionality and computational cost via the subspace learning. In addition, the proposed RPDL algorithm is regularized by using the \ell 1 norm to handle the outliers and noises, and is embedded in an online framework so that of memory and time efficiency. The proposed method is employed to model prostate shape prior for the application of magnetic resonance transrectal ultrasound registration. The experimental results demonstrate that our method provides more accurate and robust shape modeling than the state-of-the-art methods do. Quantitative histomorphometry (QH) refers to the process of computationally modeling disease appearance on digital pathology images by extracting hundreds of image features and using them to predict disease presence or outcome. Since constructing a robust and interpretable classifier is challenging in a high dimensional feature space, dimensionality reduction (DR) is often implemented prior to classifier construction. However, when DR is performed it can be challenging to quantify the contribution of each of the original features to the final classification result. We have previously presented a method for scoring features based on their importance for classification on an embedding derived via principal components analysis (PCA). However, nonlinear DR involves the eigen-decomposition of a kernel matrix rather than the data itself, compounding the issue of classifier interpretability. In this paper we present feature importance in nonlinear embeddings (FINE), an extension of our PCA-based feature scoring method to kernel PCA (KPCA), as well as several NLDR algorithms that can be cast as variants of KPCA. FINE is applied to four digital pathology datasets to identify key QH features for predicting the risk of breast and prostate cancer recurrence. Measures of nuclear and glandular architecture and clusteredness were found to play an important role in predicting the likelihood of recurrence of both breast and prostate cancers. Compared to the t-test, Fisher score, and Gini index, FINE was able to identify a stable set of features that provide good classification accuracy on four publicly available datasets from the NIPS 2003 Feature Selection Challenge. Diagnosing and localizing myocardial infarct is crucial for early patient management and therapy planning. We propose a new method for predicting the location of myocardial infarct from local wall deformation, which has value for risk stratification from routine examinations such as (3D) echocardiography. The pipeline combines non-linear dimensionality reduction of deformation patterns and two multi-scale kernel regressions. Confidence in the diagnosis is assessed by a map of local uncertainties, which integrates plausible infarct locations generated from the space of reduced dimensionality. These concepts were tested on 500 synthetic cases generated from a realistic cardiac electromechanical model, and 108 pairs of 3D echocardiographic sequences and delayed-enhancement magnetic resonance images from real cases. Infarct prediction is made at a spatial resolution around 4 mm, more than 10 times smaller than the current diagnosis, made regionally. Our method is accurate, and significantly outperforms the clinically-used thresholding of the deformation patterns (on real data: sensitivity/specificity of 0.828/0.804, area under the curve: 0.909 versus 0.742 for the most predictive strain component). Large medical image datasets form a rich source of anatomical descriptions for research into pathology and clinical biomarkers. Many features may be extracted from data such as MR images to provide, through manifold learning methods, new representations of the population's anatomy. However, the ability of any individual feature to fully capture all aspects morphology is limited. We propose a framework for deriving a representation from multiple features or measures which can be chosen to suit the application and are processed using separate manifold-learning steps. The results are then combined to give a single set of embedding coordinates for the data. We illustrate the framework in a population study of neonatal brain MR images and show how consistent representations, correlating well with clinical data, are given by measures of shape and of appearance. These particular measures were chosen as the developing neonatal brain undergoes rapid changes in shape and MR appearance and were derived from extracted cortical surfaces, nonrigid deformations, and image similarities. Combined single embeddings show improved correlations demonstrating their benefit for further studies such as identifying patterns in the trajectories of brain development. The results also suggest a lasting effect of age at birth on brain morphology, coinciding with previous clinical studies. [ABSTRACT FROM PUBLISHER]
