The purpose of this study was to prepare spiky titanium dioxide nanoparticles-loaded Plantaginis Semen polysaccharide (SN-TiO 2 -PSP), and the structural characterization and immune response of infectious laryngotracheitis (ILT) vaccine in Hetian chickens were investigated. The structural characterization of SN-TiO 2 -PSP was analyzed by FT-IR, TEM, and TGA analysis. And the immune organs indexes, lymphocytes proliferation, specific antibody levels, and ratios of CD4+ and CD8+ T lymphocytes were studied. Structural characterization results showed that SN-TiO 2 -PSP has a typical polysaccharide absorption peak and good stability. The SN-TiO 2 -PSP's shape was similar to sea urchin, and its zeta potential and particle size were 27.56 mV and 976.11 nm, respectively. In vivo results showed that SN-TiO 2 -PSP could enhance the proliferation of peripheral lymphocytes, specific antibody levels, CD4+ and CD8+ T lymphocytes ratios, IL-4 and INF-γ levels in Hetian chickens vaccinated with ILT vaccine on D 7 , D 14 , D 21 , and D 28. In addition, SN-TiO 2 -PSP not only enhanced the indexes of immune organs but also promoted the development of immune organs. The aim of this study was to explore the indirect immunomodulatory activities and its mechanism of enzymatic hydrolysis of Hericium erinaceus polysaccharides (EHEP) in the MODE-K/DCs co-culture model. According to the TEER value, transmission of phenol red and AKP activity of MODE-K cells, single model was established in order to evaluate the eligibility of MODE-K cells monolayer. Then the MODE-K/DCs co-culture model was set up and HEP and EHEP were added into the apical chamber, DCs were obtained for the expression of key surface markers, the ability of phagocytosis, the morphology, the secretion of cytokines and the production of target proteins. We found that after 21 d of culture, the MODE-K cells monolayer became intact and dense, which can be used for the MODE-K/DCs co-culture model. Under the treatment of HEP and EHEP, immature DCs become into mature DCs with the high expression of CD86 and MHCII, the low antigens up-taking, the typical morphology, the more content of IL-12 and TNF-α and the high level of TLR4, MyD88 and NF-κB proteins. However, compared with HEP, EHEP showed the better immunomodulatory activities. In recent years, the utilization of CS-MWCNT as targeted drug carriers has attracted considerable attention. Hericium erinaceus polysaccharide (HEP) has been reported as an immunostimulant to improve immune responses. This study was focussed on developing CS-MWCNT encapsulating HEP (CS-MWCNT-HEP). Using in mice peritoneal macrophages, we found the immune response could be effectively regulated by CS-MWCNT-HEP, promoted the expression of the MHCII, CD86, F4/80 and gp38. Moreover, the mice immunized with CS-MWCNT-HEP nanoparticles significantly extended PCV 2 -specific IgG immune response and the levels of cytokines. In this study, the enzyme degradation of Hericium erinaceus polysaccharide (HEP) was successfully modified with endo -rhamnosidase to obtain the enzymatic hydrolysis of Hericium erinaceus polysaccharide product (EHEP). The gas chromatography–mass spectrometry (GC–MS), high performance gel permeation chromatography (HPGPC), Fourier transformed infrared spectrometry (FT-IR), scanning electron microscopy (SEM), atomic particle microscopy (AFM), nuclear magnetic resonance (NMR) and particle size distribution were used to characterize polysaccharides. In vitro, EHEP significantly enhanced the phagocytosis, NO, CD40 and CD86 by macrophage than HEP. In vivo, female Balb/c mice were injected respectively with EHEP and HEP after administrated with cyclophosphamide, once a day for 7 days. On days 11, the morphology and structure of jejunal sections, immunofluorescence of spleen and peritoneal macrophages were determined. These results indicated that the enzymatic hydrolysis product could enhance the activation of peritoneal macrophages, and enhance the immunomodulation function of HEP. This study demonstrated that enzymatic modification was an effective method to improve the activities of HEP, and could be developed as a potential technology for use in pharmaceutical and cosmeceutical industry. In present study, the optimal condition of prepared drug was determined by response surface methodology. In addition, their physicochemical properties, drug release and uptake ability of CS-MWCNT-HEP were studied, and the distribution of the drug in ICR mice and the sites of action were further evaluated. Under the optimal condition, the mean experimental loaded efficiency 68.55 ± 1.47% was corresponded well with the predicted value of 68.28%. The results of in vitro experiments proved that a release of the drug in a pH-dependent behavior. Flow cytometry and inverted microscope showed that the uptake of CS-MWCNT-HEP in Raw264.7 cells increased significantly as the time increased. In vivo experiment proved that the HEP and CS-MWCNT-HEP were mainly accumulated in the kidney, shown the characteristics of kidney metabolism. On the other hand, the extended retention of CS-MWCNT-HEP in the mice could enhance the immune function. CS-MWCNT-HEP has high loaded efficiency and pH-responsive drug released, which could significantly improved the body's immunity and enhance the body's ability to absorbed drugs. These findings proposed a well characterized novel CS-MWCNT-HEP formulation as drug delivery system, and its mechanism and application will be further investigated in our undergoing studies. In this study, the structure characteristic of the Hericium erinaceus polysaccharide (HEP) was investigated using Fourier transformed infrared spectrometry (FT-IR), gas chromatography-mass spectrometry (GC–MS), gel permeation chromatography (GPC), methylation and nuclear magnetic resonance (NMR). The results showed that HEP, with a molecular weight of 43 KDa, was mainly composed of glucose and rhamnose. The linkages of the sugar residues of HEP were → 6) β- d -Glcp-(1 → and → 2) -α- l -Rhap-(1 → residue at the end of the branches. The Fusarium toxin deoxynivalenol (DON)-induced cellular injury model for IPEC-J2 cells was established and used to investigate the protective effects of HEP against the oxidative stress. The results suggest that HEP could significantly protect IPEC-J2 cells from DON-induced oxidative stress, inhibit DON-induced apoptosis and reduce the production of reactive oxygen species (ROS). In previous researches, the results showed that selenium Hericium erinaceus polysaccharide and Hericium erinaceus polysaccharide-loaded poly (lactic- co -glycolic acid) nanoparticles enhanced immune responses. In order to further enhance the immune adjuvant activity and phagocytosis of the nanoparticles, two way of combination (selenium-HEP loaded PLGA nanoparticles and selenium modified HEP-PLGA nanoparticles) were prepared to investigate the effects on macrophages in vitro. After treatment with the nanoparticles, the effects of phagocytosis, co-stimulatory molecules expression, nitric oxide (NO), and cytokines secretion were evaluated. The results showed that the particle size, PDI and zeta potential of the selenium-HEP loaded PLGA nanoparticles (Se-HEP-PLGA) and selenium modifified HEP-PLGA nanoparticles (HEP-PLGA-Se) were presented. Se-HEP-PLGA and HEP-PLGA-Se nanoparticles significantly stimulated phagocytic activity, CD40 and CD86 expression of macrophages. In addition, the levels of NO, TNF-α, IL-1β and IL-6 were enhanced in the peritoneal macrophages by stimulation with Se-HEP-PLGA and HEP-PLGA-Se nanoparticles. Among them, Se-HEP-PLGA showed the best effects on the expression of co-stimulatory molecules, secretions of NO and cytokines. These results indicated that Se-HEP-PLGA could enhance the activation of macrophages, and it could be potentially used as an HEP delivery system for the induction of strong immune responses. The formation of three dimensional network structure is critical in determining mechanical properties of natural rubber (NR). Consequently, it is vital to regulate crosslinking network of NR by controlling vulcanization process. Inspired by our previous studies on contribution of non-rubber components (NRCs) to the excellent properties of NR, we find octylamine in NRCs decreases the activation energy (E a) of vulcanization from 82.73 kJ/mol to 44.34 kJ/mol, thereby reducing vulcanization time from 18.67 min to 2.71 min. From microscopic perspective, octylamine tends to coordinate with zinc ions to improve dispersion of ZnO in NR. And octylamine promotes ring-opening reaction of S 8 to favor formation of polysulfide intermediates. Therefore, the incorporation of octylamine remarkably improves vulcanization efficiency, which contributes to the formation of a more homogeneous network with higher crosslinking density, enhancing remarkably the strength and toughness of NR. As a result, the tensile strength and fracture energy of samples are as high as 31.15 MPa and 68.88 kJ/m2, respectively. In addition, even with a 60 % reduction in ZnO content, the NR samples still maintain high vulcanization efficiency and excellent mechanical properties after the addition of octylamine, which provides a green and feasible way to alleviate the environmental pollution caused by ZnO. • Octylamine in NRCs can remarkably reduce activation energy and characteristic time of vulcanization of NR. • Octylamine promotes the formation of a more homogeneous crosslinking network with higher crosslinking density in NR. • After adding 2% octylamine, the strength and toughness of NR samples are as high as 31.15 MPa and 68.88 kJ/m2, respectively. • When ZnO is reduced by 60%, NR with octylamine still maintains excellent vulcanization efficiency and mechanical properties.
