In this study, we tested the inhibitory activity of 45 natural products extracted from the plant Toona sinensis on SHP2 protein, and identified four natural product inhibitors. The natural product 1,2,3,6-Tetragalloylglucose (A-1) was first reported as a competitive inhibitor of SHP2, with an IC 50 value of 0.20 ± 0.029 μM and the selectivity of 1.8-fold and 4.35-fold to high homologous proteins SHP1 and PTP1B, respectively. Compound A-1 also showed high inhibitory activity on SHP2-E76K and SHP2-E76A mutants, with IC 50 values of 0.95 ± 0.21 μM and 0.29 ± 0.045 μM, respectively. Cell viability assay showed that compound A-1 could inhibit the proliferation of a variety of cancer cells. Apoptosis assay showed that compound A-1 could effectively induce apoptosis of KRASG12C-mut NCI-H23 and KRASG12S-mut A549 cells. Western blot assay showed that compound A-1 could down regulate the phosphorylation levels of Erk1/2 and Akt in NCI-H23 and A549 cells. Molecular docking showed that compound A-1 could effectively dock to the catalytic active region of SHP2. Molecular dynamics simulation explored the effect of compound A-1 on SHP2, revealing the deep-seated binding mechanism. Pest management technology has been a promising bioconversion method for waste resource utilization. Unlike many pests that consume waste, the larvae of Lucilia sericata , also known as maggots, have many outstanding advantages as following: with their strong adaption to environment and not easily infected and exhibiting a medicinal nutritional value. Herein, the potential efficacies of maggot polysaccharides (MP), as well as their underlying mechanisms, were explored in Dextran sulfate sodium (DSS)-induced colitis mice and TNF-α-elicited Caco-2 cells. We extracted two bioactive polysaccharides from maggots, MP-80 and MP-L, whose molecular weights were 4.25 × 103 and 2.28 × 103 g/mol, respectively. MP-80 and MP-L contained nine sugar residues: 1,4-α-Arap, 1,3-β-Galp, 1,4,6-β-Galp, 1,6-α-Glcp, 1-α-Glcp, 1,4-β-Glcp, 1-β-Xylp, 1,2-α-Manp, and 1-β-Manp. We demonstrated that MP-80 and MP-L significantly ameliorated DSS-induced symptoms and histopathological damage. Immuno-analysis revealed that compared with MP-L, MP-80 could better restore intestinal barrier and reduced inflammation by suppressing NLRP3/NF-κB pathways, which might be attributed to its enriched galactose fraction. Moreover, 16S rRNA sequencing revealed that MP-80 and MP-L both improved the dysbiosis and diversity of gut microbiota and acted on multiple microbial functions. Zinc oxide (ZnO) has attracted a substantial interest in cancer research owing to their promising utility in cancer imaging and therapy. This study aimed to synthesized ZnO nanoflowers coated with albumin to actively target and the inhibit skin melanoma cells. We synthesized bovine serum albumin (BSA)-coated ZnO nanoflowers (BSA@ZnO NFs) and evaluated it's in vitro and in vivo therapeutic efficacy for skin cancer cells. BSA@ZnO NFs were prepared via single-step reduction method in the presence of plant extract (Heliotropium indicum) act as a capping agent, and further the successful fabrication was established by various physico-chemical characterizations, such as scanning electron microscopy (SEM), Fourier transform infra-red (FT-IR) spectroscopy, and x-rays diffraction (XRD) analysis. The fabricated BSA@ZnO NFs appeared flower like with multiple cone-shaped wings and average hydration size of 220.8 ± 12.6 nm. Further, BSA@ZnO NFs showed enhanced cellular uptake and cytocidal effects against skin cancer cells by inhibiting their growth via oxidative stress compared uncoated ZnO NFs. Moreover, BSA@ZnO NFs showed enhance biosafety, blood circulation time, tumor accumulation and in vivo tumor growth inhibition compared to ZnO NFs. Due to its low molecular weight and abundant functional groups, water-soluble lignin (WSL) is considered as a more potent antioxidant than traditional industrial lignin in biofields. However, few studies have been conducted to evaluate its intracellular and endogenous reactive oxygen species (ROS)-scavenging ability, especially for the intervention of ROS-related disease in vivo. In this work, WSL in bamboo autohydrolysate (WSL-BM) and wheat stalk autohydrolysate (WSL-WS) were isolated and characterized to comparably analyze their bioactivities. The composition analysis and NMR characterization showed that both WSL-BM and WSL-WS contained relatively similar components and substructures, but WSL-BM contained higher contents of phenolic OH groups. Both WSL samples exhibited excellent biocompatibility with the concentration below 50 μg/mL, while WSL-BM exhibited superior ROS-scavenging ability and ROS-related ulcerative colitis treatment potential at same concentration. In addition, WSL-BM also showed better performance in ameliorating inflammation and oxidative stress in RAW 264.7 cells and colitis mice by activating Nrf2 and suppressing NFκB signaling, resulting in an overall improvement in both macroscopic and histological parameters. Overall, these results implied that WSL from gramineous biomass can be used as a novel anti-inflammatory and antioxidative agent in the biomedical field. A cellulose-based bio-absorbent with various and plenty of amino groups was successfully prepared from corn stalk to achieve quantitative removal of Congo red from wastewater with wide pH values (5 ≤ pH ≤ 10). The maximum removal amount was 8.0 mmol·g−1 (5572 mg·g−1) under pH = 6.0 and 45 °C, which was obviously higher than reported absorbents. Investigation on dynamic adsorption and recyclability in authentic wastewater found that the removal efficiency of Congo red was >98 % within 180 min and decreased slightly in industrial water after five cycles, denoting this adsorbent with great potential for environmental application. The characterization results proved that 7.58 mmol·g−1 of different amino groups (-NH 2 , -NH- and -NR 2) were introduced on adsorbent surface by two steps of modification and were the major functional groups for adsorption of Congo red. The inferred adsorption mechanism revealed that Congo red could be adsorbed equivalently on the amino groups by strong electrostatic interactions or hydrogen bonds. Different amino groups played different roles in adsorption due to great differences in protonation ability in 5 ≤ pH ≤ 10. The study was expected to high-efficiently remove Congo red from acidic or alkaline wastewater, and offered an alternative strategy for biowaste treatment of corn stalks in a high value-added manner. Platelet-derived growth factor receptor β (PDGFRβ) plays an important role in hepatic fibrosis and is closely associated with hepatic stellate cells (HSCs) activation. Previously, by modeling PDGFRβ affinity chromatography, we found that gomisin D can target PDGFRβ. However, whether gomisin D has anti-fibrosis effects through targeting PDGFRβ remained unclear. In this study, the effect of gomisin D on hepatic fibrosis was evaluated in vivo and vitro. HSC cell lines and primary HSC were cultured and functionally we found that gomisin D promotes HSC apoptosis, inhibits HSCs activation and proliferation. A male BALB/c mouse liver fibrosis model was established to comfirm gomisin D (especially in 50 mg/kg) could improve liver fibrosis by inhibiting HSCs activation. In addition, gomisin D had a good binding ability with PDGFRβ (KD = 3.3e-5 M). Mechanically, gomisin D regulated PDGF-BB/PDGFRβ signaling pathway by targeting PDGFRβ, further more inhibited HSC activation, subsequently inhibited inflammatory factors, ultimately improved CCl 4 -induced liver fibrosis. Overall, gomisin D could inhibit HSC proliferation and activation, promote HSC apoptosis, and alleviate CCl 4 -induced hepatic fibrosis by targeting PDGFRβ and regulating PDGF-BB/PDGFRβ signaling pathway. Reperfusion therapy is the most effective treatment for acute myocardial infarction. However, reperfusion itself can also cause cardiomyocytes damage. Pyroptosis has been shown to be an important mode of myocardial cell death during ischemia-reperfusion. Non-coding RNAs (ncRNAs) play critical roles in regulating pyroptosis. The regulation of pyroptosis by microRNAs, long ncRNAs, and circular RNAs may represent a new mechanism of myocardial ischemia-reperfusion injury. This review summarizes the currently known regulatory roles of ncRNAs in myocardial ischemia-reperfusion injury and interactions between ncRNAs. A composite (ZS–UiO–66–NH 2) zirconium crosslinked sodium alginate gel beads (ZS)–metal–organic skeleton (UiO–66–NH 2) were prepared in this study through in–situ growth under simple, green and mild conditions for removal of the salicylic acid (SA) and sulfosalicylic acid (SSA) from water. The physicochemical properties of ZS–UiO–66–NH 2 were characterized using various analytical methods. The influencing factors in the adsorption process including pH of solution, amount of adsorbent, coexisting ions, adsorption time, reaction temperature and equilibrium concentration of SA/SSA were performed in batch adsorption. The experimental results indicated that ZS–UiO–66–NH 2 had high stability and could achieve efficient adsorption of SA/SSA in broad pH range (2–9) and salinity (0–0.2 mol·L−1). SA and SSA adsorbed on the composite at 293 K reached high values of 193 and 167 mg·g−1 from Langmuir model, respectively. Kinetic and isotherm studies demonstrated that the adsorption processes were mainly multilayer heterogeneous chemisorption. Thermodynamic data manifested that the two processes were exothermic and spontaneous with increasing entropy. ZS–UiO–66–NH 2 can effectively remove SA/SSA from simulated wastewater under different pH and can be reused after elution with a NaHCO 3 solution (5 mmol·L−1). Lignin sulfonate (LS), a waste material from the paper pulping, was modified with benzoic anhydride to obtain benzoylated lignin sulfonates of adjustable hydrophilicity (BLS). When BLS was combined with difenoconazole (Di), a broad-spectrum fungicide, lignin-based, non-crosslinked nanoparticles were obtained either by solvent exchange or solvent evaporation. When a mass ratio of 1:5 LS: benzoic anhydride was used, the Di release from Di@BLS 5 after 1248 h was ca. 74 %, while a commercial difenoconazole microemulsion (Di ME) reached 100 % already after 96 h, proving the sustained release from the lignin nanocarriers. The formulation of Di in lignin-based nanocarriers also improved the UV stability and the foliar retention of Di compared to the commercial formulation of the fungicide. Bioactivity assay showed that Di@BLS 5 exhibited high activities and duration against strawberry anthracnose (Colletotrichum gloeosporioides). Overall, the construction of fungicide delivery nano-platform using BLS via a simple non-crosslinked approach is a novel and promising way to develop new formulations for nanopesticide and the development of sustainable agriculture. In this work, the modified‑sodium alginate gel beads were prepared by sol-gel method. Due to the presence of water channels in the sodium alginate gel bead, amidoxime groups and PO 4 3− were exposed to the surface of the adsorbent to the maximum extent, resulting in the excellent adsorption capacity of modified‑sodium alginate gel beads. The introduction of amidoxime-modified hydroxyapatite significantly improved the adsorption capacity and the adsorption rate of the gel beads. The adsorption capacity increased from 308.7 to 466.0 mg/g and the adsorption equilibrium time was shortened from 300 min to 120 min. The modified‑sodium alginate gel bead possessed the advantages of short adsorption time, high adsorption efficiency and large adsorption capacity, which could be regarded as a potential adsorbent for uranium. Moreover, the uranium removal ability on the modified gel beads was mainly attributed to the Coulomb force between PO 4 3− and uranium and the complexation between uranium and amidoxime groups. Hemorrhage is a potentially life-threatening emergency that can occur at any time or place. Whether traumatic, congenital, surgical, disease-related, or drug-induced, bleeding can lead to severe complications or death. Therefore, the development of efficient hemostatic materials is critical. However, the results and prognosis demonstrated by clinical means of hemostasis do not reach expectations. With the development of technology, novel hemostatic materials have been developed from polysaccharides (chitosan, hyaluronic acid, alginate, cellulose, cyclodextrins, starch, dextran, and carrageenan), peptides (self-assembling peptides), and proteins (silk fibroin, collagen, gelatin, keratin, and thrombin). These new materials exhibit high hemostatic efficacy due to the enhancement or interaction of various hemostatic mechanisms. The main forms include adhesives, sealants, bandages, hemostatic powders, and hemostatic sponges. This article introduces the clotting process and principles of hemostatic methods and reviews the research on polysaccharide-, peptide-, and protein-based hemostatic materials in the last five years. The design ideas and hemostatic principles of polysaccharide-, peptide-, and protein-based hemostatic materials are mainly introduced. Periodontitis is a chronic inflammation of the periodontium caused by a persistent bacterial infection, resulting in destruction of the supporting structures of teeth. Analysis of microbial composition in saliva can inform periodontal status. Actinobacillus actinomycetemcomitans (Aa), Porphyromonas gingivalis (Pg), and Streptococcus mutans (Sm) are among reported periodontal pathogens, and were used as model systems in this study. Our atomic force microscopic (AFM) study revealed that these pathogens are biological nanorods with dimensions of 0.6–1.1 μm in length and 500–700 nm in width. Current bacterial detection methods often involve complex preparation steps and require labeled reporting motifs. Employing surface-enhanced Raman spectroscopy (SERS), we revealed cell-type specific Raman signatures of these pathogens for label-free detection. It overcame the complexity associated with spectral overlaps among different bacterial species, relying on high signal-to-noise ratio (SNR) spectra carefully collected from pure species samples. To enable simple, rapid, and multiplexed detection, we harnessed advanced machine learning techniques to establish predictive models based on a large set of raw spectra of each bacterial species and their mixtures. Using these models, given a raw spectrum collected from a bacterial suspension, simultaneous identification of all three species in the test sample was achieved at 95.6 % accuracy. This sensing modality can be applied to multiplex detection of a broader range and a larger set of periodontal pathogens, paving the way for hassle-free detection of oral bacteria in saliva with little to no sample preparation. In this study, a novel magnetic macroporous chitin microsphere (MMCM) was developed for enzyme immobilization. Chitin nanofibers were prepared and subsequently subjected to self-assembly with magnetic nanoparticles and PMMA (polymethyl methacrylate). Following this, microspheres were formed through spray drying, achieving a porous structure through etching. The MMCM serves as an effective support for immobilizing enzymes, allowing for their covalent immobilization both on the microsphere's surface and within its pores. The substantial surface area resulting from the porous structure leads to a 2.1-fold increase in enzyme loading capacity compared to non-porous microspheres. The MMCM enhances stability of the immobilized enzymes under various pH and temperature conditions. Furthermore, after 20 days of storage at 4 °C, the residual activity of the immobilized enzyme was 2.93 times that of the free enzyme. Even after being recycled 10 times, the immobilized enzyme retained 56.7 % of its initial activity. It's noteworthy that the active sites of the enzymes remained unchanged after immobilization using the MMCM, and kinetic analysis revealed that the affinity of the immobilized enzymes rivals that of the free enzymes. Trophoblast cell surface antigen 2 (Trop2) has emerged as a potential target for effective cancer therapy. In this study, we report a novel anti-Trop2 antibody IMB1636, developed using hybridoma technology. It exhibited high affinity and specificity (K D = 0.483 nM) in binding both antigens and cancer cells, as well as human tumor tissues. hIMB1636 could induce endocytosis, and enabled targeted delivery to the tumor site with an in vivo retention time of 264 h. The humanized antibody hIMB1636, acquired using CDR grafting, exhibited the potential to directly inhibit cancer cell proliferation and migration, and to induce ADCC effects. Moreover, hIMB1636 significantly inhibited the growth of MDA-MB-468 xenograft tumors in vivo. Mechanistically, hIMB1636 induced cell cycle arrest and apoptosis by regulating cyclin-related proteins and the caspase cascade. In comparison to commercialized sacituzumab, hIMB1636 recognized a conformational epitope instead of a linear one, bound to antigen and cancer cells with similar binding affinity, induced significantly more potent ADCC effects against cancer cells, and displayed superior antitumor activities both in vitro and in vivo. Trauma-induced articular cartilage damages are common in clinical practice. Hydrogels have been used to fill the cartilage defects and act as extracellular matrices for cell migration and tissue regeneration. Lubrication and stability of the filler materials are essential to achieve a satisfying healing effect in cartilage regeneration. However, conventional hydrogels failed to provide a lubricous effect, or could not anchor to the wound to maintain a stable curing effect. Herein, we fabricated dually cross-linked hydrogels using oxidized hyaluronic acid (OHA) and N -(2-hydroxypropyl)-3-trimethylammonium chitosan chloride (HTCC) methacrylate (HTCCMA). The OHA/HTCCMA hydrogels, which were dynamically cross-linked and then covalently cross-linked by photo-irradiation, showed appropriate rheological properties and self-healing capability. The hydrogels exhibited moderate and stable tissue adhesion property due to formation of dynamic covalent bonds with the cartilage surface. The coefficient of friction values were 0.065 and 0.078 for the dynamically cross-linked and double-cross-linked hydrogels, respectively, demonstrating superior lubrication. In vitro studies showed that the hydrogels had good antibacterial ability and promoted cell proliferation. In vivo studies confirmed that the hydrogels were biocompatible and biodegradable, and exhibited a robust regenerating ability for articular cartilage. New antibacterial agents are needed to overcome the challenges of microbial food contamination. In this study, we investigated the potential of Elaeagnus mollis polysaccharide (EMP) to modify black phosphorus (BP) for use as a bactericide for foodborne pathogenic bacteria. The resulting compound (EMP-BP) displayed enhanced stability and activity compared with BP. EMP-BP exhibited an increased antibacterial activity (bactericidal efficiency of 99.999 % after 60 min of light exposure) compared to EMP and BP. Further studies revealed that photocatalytically generated reactive oxygen species (ROS) and active polysaccharides acted collectively on the cell membrane, leading to cell deformation and death. New antibacterial agents are needed to overcome the challenges of microbial food contamination. In this study, we investigated the potential of Elaeagnus mollis polysaccharide (EMP) to modify black phosphorus (BP) for use as a bactericide for foodborne pathogenic bacteria. The resulting compound (EMP-BP) displayed enhanced stability and activity compared with BP. EMP-BP exhibited an increased antibacterial activity (bactericidal efficiency of 99.999 % after 60 min of light exposure) compared to EMP and BP. Further studies revealed that photocatalytically generated reactive oxygen species (ROS) and active polysaccharides acted collectively on the cell membrane, leading to cell deformation and death. The raw and processed Polygonum multiflorum Thunb (PM) are used to treat different diseases, and PM has also been reported to have hepatotoxic effects. Moreover, mounting evidence indicates that processed PM is less toxic than raw PM. The changes in efficacy and toxicity of PM during the processing are closely related to the changes in chemical composition. Previous studies have mainly focused on the changes of anthraquinone and stilbene glycosides during process. Polysaccharides, as main components of PM, showed many pharmacological effects, but its changes in the processing has been neglected for a long time. In this study, the polysaccharides of PM in the raw (RPMPs) and processed products (PPMPs) were determined and the liver injury model induced by acetaminophen was utilized to evaluate the impact of polysaccharides on the liver. Results showed that the heteropolysaccharides RPMPs and PPMPs both comprised Man, Rha, GlcA, GalA, Glc, Ara and Xyl, but markedly differed in polysaccharide yield, molar ratio of monosaccharide composition and M w. In vivo analysis, results showed that demonstrated that RPMPs and PPMPs both exerted hepatoprotective effects by upregulating antioxidant enzymes and repressing lipid peroxidation. It is noteworthy that the polysaccharide yield of processed PM was seven-fold higher than that of raw PM, so it is speculated that processed PM has better hepatoprotective effects at the same dose of decoction. The present work provides an important foundation for studying the polysaccharide activity of PM and further revealing the processing mechanism of PM. This study also proposed a new hypothesis that the significant increase of polysaccharide content in processed PM may be another reason that the product PM causes less liver injury.
