In the present study, the effects of a purified fraction of polysaccharides from the fruits of Lycium barbarum L. (LBPs), named LBPs-4, on the dextran sodium sulfate (DSS)-induced colitis in mice were evaluated. The results showed that LBPs-4 decreased disease activity index score, prevented colon shortening and reduced plasma levels of pro-inflammatory cytokines (tumor necrosis factor alpha (TNF-α), interferon gamma (IFN-γ), monocyte chemoattractant protein-1 (MCP-1) and prostaglandin E2) in mice with colitis. LBPs-4 could increase the relative abundances of Akkermansia and Bifidobacterium in gut microbiota, and it also mitigated the intestinal barrier damage by upregulating the level of tight junction protein ZO-1 and the number of goblet cells in colon. Moreover, the results of in vitro culture indicated that the growth of Bifidobacterium longum subsp. infantis CCX 19042 was promoted by LBPs-4, whereas the culture media of LBPs-4 by Bacteroides ovatus with or without addition of mucin could enhance the growth of Akkermansia muciniphila. Collectively, these results suggested that LBPs-4 should be potential prebiotics for the treatment of colitis.

Trichinellosis is a serious food-borne zoonotic parasitic disease with global distribution, causing serious harm to public health and food safety. Molting is prerequisite for intestinal larval development in the life cycle of T. spiralis. Metalloproteinases play an important role in the molting process of T. spiralis intestinal infective larvae (IIL). In this study, the metalloproteinase Tsdpy31 was cloned, expressed and characterized. The results revealed that the Tsdpy31 was expressed at various T. spiralis stages and it was principally located in cuticle, hypodermis and embryos of the nematode. Recombinant Tsdpy31 (rTsdpy31) had the catalytic activity of natural metalloproteinase. Silencing of Tsdpy31 increased the permeability of larval new cuticle. When the mice were orally challenged with dsRNA treated- muscle larvae, the burden of intestinal adult and muscle larvae in Tsdpy31 dsRNA treatment group was significantly reduced, compared with the control green fluorescent protein (GFP) dsRNA and PBS groups (P 0.05). Tsdpy31 may play a major role in the new cuticle synthesis and old cuticle shedding. Tsdpy31 also participates in T. spiralis embryonic development. We conclude that Tsdpy31 could be a candidate vaccine target molecule against intestinal T. spiralis ecdysis and development.

To illustrate the correlations between molecular structures and the film-forming properties of thermoplastic starch from various botanical sources, starches from cereal, tuber and legume were modified by thermoplastic extrusion and the corresponding thermoplastic starch films were prepared including thermoplastic corn starch (TCS), thermoplastic rice starch (TRS), thermoplastic sweet potato starch (TSPS), thermoplastic cassava starch (TCAS) and thermoplastic pea starch (TPES) films. TPES film displayed a higher tensile strength (6.28 MPa) and stronger water resistance, such as lower water solubility (15.70 %), water absorption (42.35 %), and water vapor permeability (0.285 g·mm·h

Natural products (+)-nootkatone is an important sesquiterpene compound and is widely used in pharmaceutical, cosmetic, agricultural and food industries. The aim of this study was to analyze the differentially expressed proteins (DEPs) during citrus aroma compound (+)-valencene biotransformation to (+)-nootkatone by Yarrowia lipolyticaby with high-throughput LC-MS/MS. A total of 778 proteins were differentially expressed, 385 DEPs were significantly up-regulated and 393 DEPs were markedly down-regulated. It was found that the enzymes transformed (+)-valencene to (+)-nootkatone were mainly existed in yeast intracellular and precipitated under the condition of 30-40 % ammonium sulfate. Most DEPs involved in amino acid and fatty acid metabolism were down-regulated during (+)-valencene biotransformation. The DEPs related to the carbohydrate metabolism, energy metabolism and most of transporter proteins were significantly up-regulated. Furthermore, the key enzymes involved in (+)-valencene transformation might be related to cytochrome P450s (gene2215 and gene2911) and dehydrogenases (gene6493). This is the first time that proteomics was used to investigate the metabolism mechanism of Yarrowia lipolytica during (+)-valencene biotransformation. The proteomic analysis of Yarrowia lipolytica provided a foundation for the molecular regulatory mechanism in the biotransformation to (+)-nootkatone from (+)-valencene.

The large agglomeration of starch paste in hot water, and fast retrogradation tendency and low transparency of starch gel restrict widespread application of kudzu starch. To improve the above defects, kudzu starch was modified with sequentially α-amylase (AA) and transglucosidase (TG), the latter for varying times. The results indicated that, compared to kudzu starch, amylose content and molecular weight of AA/TG-treated starches reduced by 20.07% and 69.50%, respectively. The proportion of A chain increased by 68.68%, whereas B1, B2 and B3 chains decreased by 14.28%, 48.29% and 23.44%, respectively. The degree of branching dramatically increased by 128.3%. After AA→TG treatment, the changes of starch structure enhanced the functional properties of kudzu starch. The solubility, paste clarity and gelatinization temperature increased, whereas the relative crystallinity, viscosity, storage and loss moduli decreased. Overall, the AA→TG modification would be desirable to improve the functional properties of kudzu starch to expand more large-scale application.

To explore an effective and economic method to prepare higher contents of resistant starch (RS), different enzyme treatments including single pullulanase (PUL), commercial α-amylase (AA) or/and β-amylase (BA) with PUL, and malt endogenous amylase (MA) with PUL were used and the structural, physicochemical properties and digestibility of all modified starches (MS) were compared. All the enzyme-treated starches displayed a mixture of B and V-type diffraction patterns. The MA/PUL-MS showed higher V-type diffraction peak intensity as compared to other modified starches. Compared to the combination of commercial enzyme treatment, the combination of malt enzyme treatment led to higher apparent amylose contents (45.56%), RS content (53.93%) and thermal stability (302 °C), whereas it possessed lower solubility indices and predicted glycaemic index. The apparent viscosity and shear resistance of MA/PUL-MS were lower than that of AA/PUL-MS, whereas that of MA/PUL-MS was higher than that of BA/PUL-MS and BA/AA/PUL-MS. These findings would provide a theoretical and applicative basis to produce foods with lower GI in industrial production.

“Green” solvent ionic liquid (IL) 1-ethyl-3-methylimidazolium acetate ([Emim]Ac) was used as a solvent for maize starch (MS) and potato starch (PS). Different scanning calorimetry (DSC), microscopy, rapid visco analysis, X-ray diffractometry and scanning electron microscopy were carried out to systematically investigate the phase transition, viscosity, crystalline structure and morphology of the two typical starches. The effect of [Emim]Ac/H2O ratio on the behaviors of starch was explored and the difference between native MS and PS in [Emim]Ac/H2O mixtures was compared. The effects of IL were closely related to the starch source and structure. With the increase of [Emim]Ac/H2O ratio, the gelatinization and dissolution of starch occurred competitively and synergistically. DSC results demonstrated that the transition of starch was from a single endotherm to an exotherm/endotherm, and then to a single exotherm with the increase of IL. The gelatinization temperature of MS was as low as 44.4 °C in IL/H2O mixture, which provided a wonderful solvent for MS. RVA, microscopy, XRD and SEM results provided forceful evidence for the effect of [Emim]Ac. This study provided experimental supports and theoretical foundation for understanding the starch behaviors in “green” solvents and expanding industrial applications of starch-based degradable materials in more appropriate solvents.

Gut microbiota and intestinal permeability have been demonstrated to be the key players in the gut-liver cross talk in nonalcoholic fatty liver disease (NAFLD). Lycium barbarum polysaccharides (LBPs), which seem to be a potential prebiotic, and aerobic exercise (AE) have shown protective effects on NAFLD. However, their combined effects on intestinal microecology remain unclear. This study evaluated the effects of LBP, AE, and its combination (LBP + AE) on gut microbiota composition, intestinal barrier, and hepatic inflammation in NAFLD. LBP + AE showed high abundance and diversity of gut microbiota, restored the gut microbiota composition, increased some Bacteroidetes, short chain fatty acids, but decreased Proteobacteria and the ratio of Firmicutes/Bacteroidetes. Simultaneously, LBP, AE, and LBP + AE could restore the colonic and ileum tight junctions by increasing the expression of zonula occludens-1 and occludin. They also downregulated gut-derived lipopolysaccharides (LPSs), hepatic LPS-binding proteins, inflammatory factors, and related indicators of the LPS/TLR4/NF-κB signaling pathway for the liver. Our results implied that LBP could be considered a prebiotic agent, and LBP + AE might be a promising treatment for NAFLD because it could maintain gut microbiota balance, thereby restoring intestinal barrier and exerting hepatic benefits.

Potato starch displayed high viscosity, low hydroscopicity and dispersity, and acid susceptibility leading to the limited application of potato starch. To expand the potato starch utility with appropriate processing characteristics, potato starch granules were modified with branching enzyme (BE) and transglucosidase (TG). The results indicated that the susceptibility of potato starch granules to TG was higher than BE. Moreover, the two enzymes showed the synergistic effect in enzymatic modification of potato starch granules. They cooperatively attacked the external and interior of potato starch granules. The crystal forms of potato starch changed from B to C-type after double enzyme treatments, and enzyme-treated starches exhibited homogeneous crystal distribution. Compared to BE or TG alone, the combined action of BE and TG increased significantly the ratio of α-1,6-glycosidic linkage and the amounts of short chains of potato starch, which led to the significant reduction in degree of crystallinity, viscosity, gelatinization temperature and enthalpy, and a remarkable increase in solubility. Especially, the physicochemical characteristics of modified starch largely depended on the treatment time of TG. Thus, through the combination of BE and TG, the appropriate treatment time of TG may be chosen to improve the physicochemical properties of potato starch in processed starch-based products.

To obtain porous starch granules with higher absorption capacities, three types of enzyme combinations were adopted to modify wheat and maize starches: (1) sequential α-amylase (AA) → glucoamylase (GA); (2) sequential branching enzyme (BE) → GA; and (3) sequential AA→BE→GA. The results indicated that AA→BE→GA treatment had a most optimal influence on porous starches. Compared to AA→GA and BE→GA, the mesopores in wheat starch granules treated with AA→BE→GA decreased by 52.82 and 48.70%, respectively. Conversely, the macropores increased by 216.68 and 138.18%, respectively. While for maize starch, the percentages of mesopores and macropores hardly changed after three enzyme combinations. Comparing the three enzyme treatments showed that pore volume (0.005 and 0.007 cm3/g) and pore size (36.35 and 26.54 nm) were largest in the AA→BE→GA treated wheat and maize starches, respectively. Compared to the AA→GA and BE→GA, the adsorption capacities for oil, dye and heavy metal ions, wheat starch treated with AA→BE→GA increased by 46.61 and 242.33%, and 44.52 and 134.41%, and 28.83 and 271.72%, respectively. Correspondingly, that of maize starch increased by 29.71 and 133.29%, and 42.92 and 79.93%, and 28.16 and 161.43%, respectively. These results may provide a new and valuable enzyme combination for optimising porous starch granules with higher absorption capacities.

In this research, a novel polysaccharide (PCP) was extracted from Pleurotus citrinopileatus and purified by Sephadex G-150 gel column, and its antitumor activity was investigated using the model H22 tumor-bearing mice. PCP was found to be composed of arabinose, galactose, glucose, xylose, mannose and glucuronic acid in a proportion of 0.66: 14.59: 10.77: 1: 0.69: 0.23 with average molecular weight of 7.30 × 105 Da. Further analysis suggested that PCP was a pyranose with α-type and β-type glycosidic residues. The antitumor assays in vivo indicated that PCP could effectively suppress H22 solid tumor growth, protect immune organs and improve inflammation and anemia. Besides, Annexin V-FITC/PI double staining and JC-1 staining demonstrated that PCP could induce apoptosis of H22 hepatoma cells. The PI staining assay revealed that PCP induced H22 hepatoma cells apoptosis by arresting cell cycle in S phase. These results suggest that the polysaccharide from Pleurotus citrinopileatus possesses potential value in the treatment of liver cancer.

Many Chinese herbs are well known for their neuroprotective and anti-oxidant properties. Extracts of Salvia miltiorrhiza and Anemarrhenae asphodeloides, tanshinone IIA (tanIIA), salvianolic acid B (Sal B) and sarsasapogenin (ML-1), were selected to study their dissociation potential towards Aβ42 peptide fibrils and neuroprotective effect on cells. Moreover, derivatives of sarsasapogenin (ML-2, ML-3 and ML-4) have been prepared by the addition of modified carbamate moiety. TanIIA and Sal B have shown to possess a strong ability to dissociate Aβ42 fibrils. The dissociation potential of ML-1 increased upon the introduction of carbamate moiety with N-heterocycles. In silico data revealed that derivatives ML-4 and Sal B interact with Aβ42 regions responsible for fibril stabilization through hydrogen bonds. Contrary, tanIIA binds close to a central hydrophobic region, which may lead to destabilization of fibrils. Sarsasapogenin derivative ML-2 decreased nitride oxide production, and derivative ML-4 enhanced the growth of neurites. The reported data highlight the possibility of using active compounds to design novel treatment agents for Alzheimer's disease.

The structure of extracted condensed tannin (CT) from the fruit of Sour jujube (Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chow) and the molecular mechanisms by which CT inhibits the activity of mushroom tyrosinase were investigated. The structure of CT was characterized by high performance liquid chromatography electrospray ionization mass spectrometry, and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The kinetic assays were used to detect inhibition effect, type and mechanism. UV scanning, fluorescence quenching, copper interacting, o-quinone interaction and molecular docking assays were also used to reveal the molecular mechanisms by which CT inhibit tyrosinase. The results showed the structural units of CT containing afzelechin/epiafzelechin, catechin/epicatechin, and gallocatechin/epigallocatechin. Kinetic analysis showed that CT inhibits both the monophenolase and diphenolase activities of tyrosinase and exhibits reversible, mixed type mechanism. The fruit CT interacts primarily with the copper ions and specific amino acid residue (Asn191, Thr203, Ala202, Ser206, Met201, His194, His54, Glu182 and Ile42) in the active site of tyrosinase to disturb oxidation of substrates by tyrosinase. These results suggested the sour jujube fruit is a potential natural source of tyrosinase inhibitors, and has a potential to be used in food preservation, whitening cosmetics.

Many studies have shown that pollen and its preparation are ideal herbal remedies for the treatment of prostate diseases. Our previous study found that pollen polysaccharides from Chinese wolfberry (WPPs) can induce the apoptosis of prostate cancer DU145 cells. But the antitumor mechanism of WPPs was not clearly understood. Therefore, in the present study, we further investigated the antitumor mechanism of WPPs in DU145 cells and a xenograft mice model. In the present study, the digestion under simulated saliva, gastric and small intestinal conditions and in vitro fermentation by human gut microbiota of Lycium barbarum polysaccharides (LBPS) were investigated. As a result, LBPS contained carbohydrates, uronic acids, protein and polyphenols of 60.96 ± 5.58, 20.98 ± 2.49, 5.30 ± 0.12 and 1.04 ± 0.06%, respectively. Furthermore, LBPS was mainly consisted of glucuronic acid (5.12 mol%), galacturonic acid (12.40 mol%), glucose (2.15 mol%), galactose (39.67 mol%) and arabinose (40.66 mol%). Under simulated saliva, gastric and small intestinal conditions, LBPS was not affected. Whereas, LBPS could be utilized by gut microbiota based on the consumption of monosaccharides, decreased molecular weight and lower content of total carbohydrates after fermentation in vitro. With the proceeding of fermentation, LBPS significantly promoted the production of short-chain fatty acids. Meanwhile, LBPS could alter the microbial community after 24 h fermentation, especially enhancing the relative abundances of genera Bacteroides, Bifidobacterium, Phascolarctobacterium, Clostridium XlVb, Prevotella and Collinsella. All the results suggested that LBPS could be developed as special ingredients for modulating gut microbiota composition and promoting the growth of beneficial bacteria like Bifidobacterium.
