This review extensively surveys the biomedical applications of hydrogels containing dextran. Dextran has gained much attention as a biomaterial due to its distinctive properties such as biocompatibility, non-toxicity, water solubility and biodegradability. It has emerged as a critical constituent of hydrogels for biomedical applications including drug delivery devices, tissue engineering scaffolds and biosensor materials. The benefits, challenges and potential prospects of dextran-based hydrogels as biomaterials are highlighted in this review. • Dextran is a water soluble polysaccharide that can be utilized as a parent material for the preparation of hydrogels. • Hydroxyl is the only functional group present in the dextran molecule. • Other functional groups can easily be achieved through modification methods. • Dextran can be combined with other polymer materials to form hydrogels with desirable properties. • In the presence of dextranase, dextran undergoes degradation under physiological conditions. The tightly bonded shielding coating on biomatrix significantly enhances the functionality of medical devices, bioprostheses in particular. In our work we have obtained a polyelectrolyte coating on a biomatrix by sequentially depositing chitosan and hyaluronic acid (HA) from solutions in carbonic acid under pressure. This approach makes it possible to obtain hybrid biomatrix with a firmly bonded polymer screen due to the electrostatic bonding of polyions. High-precision analysis using a tritium label shows a 3-fold increase in quantity of HA in carbonic acid under pressure compared to the conventional method. The presence of the chitosan layer increases the HA adsorption by 15–20 % due to electrostatic interaction of differently charged polymers. Antimicrobial results show the possibility of implementing an induced antimicrobial response, due to the lysis of the upper layer of the coating (HA) and the release of antimicrobial agents in the case of growth of pathogens on the bioprosthesis. Bone is an alive and dynamic organ that is well-differentiated and originated from mesenchymal tissues. Bone undergoes continuous remodeling during the lifetime of an individual. Although knowledge regarding bones and their disorders has been constantly growing, much attention has been devoted to effective treatments that can be used, both from materials and medical performance points of view. Polymers derived from natural sources, for example polysaccharides, are generally biocompatible and are therefore considered excellent candidates for various biomedical applications. This review outlines the development of chitosan-based biomaterials for the treatment of bone disorders including bone fracture, osteoporosis, osteoarthritis, arthritis rheumatoid, and osteosarcoma. Different examples of chitosan-based formulations in the form of gels, micro/nanoparticles, and films are discussed herein. The work also reviews recent patents and important developments related to the use of chitosan in the treatment of bone disorders. Although most of the cited research was accomplished before reaching the clinical application level, this manuscript summarizes the latest achievements within chitosan-based biomaterials used for the treatment of bone disorders and provides perspectives for future scientific activities. With the development in the field of biomaterials, research on alternative biocompatible materials has been initiated, and alginate in polysaccharides has become one of the research hotspots due to its advantages of biocompatibility, biodegradability and low cost. In recent years, with the further understanding of microscopic molecular structure and properties of alginate, various physicochemical methods of cross-linking strategies, as well as organic and inorganic materials, have led to the development of different properties of alginate hydrogels for greatly expanded applications. In view of the potential application prospects of alginate-based hydrogels, this paper reviews the properties and preparation of alginate-based hydrogels and their major achievements in delivery carrier, dressings, tissue engineering and other applications are also summarized. The research study aimed to maximize the important medical role of star anise extract (SAE) through its loading on a widely available natural polymer (chitosan, Cs). Thus, SAE loaded chitosan nanoparticles (CsNPs) was prepared. The finding illustrated the formation of spherical particles of SAE loaded CsNPs as proved by transmission electron microscope (TEM). In addition, the average particle size of CsNPs and SAE loaded CsNPs are 131.8 ± 24.63 and 318.5 ± 73.94 nm, respectively. Scanning electron microscope (SEM) showed the presence of many spherical particles deposited on the surface of CsNPs owing to the deposition of SAE on the surface and encapsulated into pores of CsNPs. It also showed the presence of elements such as sodium, potassium, copper, magnesium, zinc, calcium, and iron, as well as the elements that accompanied with CsNPs: carbon, oxygen, nitrogen, and phosphorus. The extract was rich in bioactive components, such as anethole, shikimic acid, and different flavonoids, contributing to its medicinal qualities. The bioactive molecules in SAE were assessed by chromatographic analysis. Using the agar well diffusion test, the antibacterial qualities of CsNPs and SAE loaded CsNPs were evaluated against pathogenic bacteria linked to lung illnesses. The most significant inhibition zones showed that the SAE loaded CsNPs had the most antibacterial activity. The anticancer activity using MTT assay was used in the biological assessments to determine the cytotoxicity against the NCl-H460 lung cancer cell line. The results showed that CsNPs loaded with SAE considerably decreased cell viability in a dose-dependent manner, with the most significant anticancer impact by SAE loaded CsNPs. Furthermore, in vivo tests on lung cancer therapy revealed that when compared to other treatment groups, the SAE loaded CsNPs group showed the greatest reduction in tumor biomarkers and inflammation, as seen by decreased levels of Plasma malondialdehyde (MDA), tumor protein 53 (p53), Tumor necrosis factor-alpha (TNF- alpha), and fibronectin. Results concluded that these thorough characterizations, biological assessments, and antibacterial tests have confirmed the effective integration of SAE into CsNPs. Antibiotic resistance (ABR) poses a universal concern owing to the widespread use of antibiotics in various sectors. Nanotechnology emerges as a promising solution to combat ABR, offering targeted drug delivery, enhanced bioavailability, reduced toxicity, and stability. This comprehensive review explores concepts of antibiotic resistance, its mechanisms, and multifaceted approaches to combat ABR. The review provides an in-depth exploration of polymeric nanoparticles as advanced drug delivery systems, focusing on strategies for targeting microbial infections and contributing to the fight against ABR. Nanoparticles revolutionize antimicrobial approaches, emphasizing passive and active targeting. The role of various molecules, including small molecules, antimicrobial peptides, proteins, carbohydrates, and stimuli-responsive systems, is being explored in recent research works. The complex comprehension mechanisms of ABR and strategic use of nanotechnology present a promising avenue for advancing antimicrobial tactics, ensuring treatment efficacy, minimizing toxic effects, and mitigating development of ABR. Silk fibroin (SF) has received special attention from the scientific community due to its noteworthy properties. Its unique chemical structure results in an uncommon combination of macroscopically useful properties, yielding a strong, fine and flexible material which, in addition, presents good biodegradability and better biocompatibility. Therefore, silk fibroin in various formats, appears as an ideal candidate for supporting biomedical applications. In this review, we will focus on the hydrogels obtained from silk fibroin or in combination with it, paying special attention to the synthesis procedures, characterization methodologies and biomedical applications. The application of bone grafting materials in bone tissue engineering is paramount for treating severe bone defects. In this comprehensive review, we explore the significance and novelty of utilizing bioactive polymers as grafts for successful bone repair. Unlike metals and ceramics, polymers offer inherent biodegradability and biocompatibility, mimicking the native extracellular matrix of bone. While these polymeric micro-nano materials may face challenges such as mechanical strength, various fabrication techniques are available to overcome these shortcomings. As an abundant biopolymer, lignin gains interest owing to its renewable nature and polyphenolic structure. It possesses many biological activities such as antioxidant activity, antimicrobial activity, and biocompatibility. Studies are being carried out to relate the biological activities to the polyphenolic structures. These traits present lignin as a natural compound being used in biomedical field. Lignin nanoparticles (LNPs) are being investigated for safe use in drug and gene delivery, and lignin-based hydrogels are being explored as wound dressing materials, in tissue engineering and 3D printing. In addition, lignin and its derivatives have shown the potential to treat diabetic disease. This review summarizes latest research results on the biological activities of lignin and highlights potential applications exampled by selective studies. A series of carboxymethylcellulose (CMC) functionalized with glycidyl methacrylate (GMA) was successfully synthesized for producing of CMC-g-GMA copolymer. Water-soluble CMC-g-GMA copolymer was photo-crosslinked while Irgacure-2959 was used as a UV-photo-initiator at 365 nm. On the other hand, cellulose nanocrystals (CNCs) from sugarcane were graft-copolymerized in an aqueous solution utilizing cerium ammonium nitrate (CAN) as an initiator in a redox-initiated free-radical approach. CNCs were grafted with GMA to enhance their physicochemical and biological characteristics. Factors affecting hydrogel formation, e.g. CMC-g-GMA copolymer concentration, irradiation time and incorporation of different concentration of CNCs-g-GMA nano-filler, were discussed in dependance on the swelling degree and gel fraction of the produced hydrogels. Notably, the addition of CNCs-g-GMA nanofillers increased progressively thermal stability of the prepared hydrogel. CMC-g-GMA filled with CNCs-g-GMA composite hydrogel showed antimicrobial activity against multidrug resistance pathogens. The limited mechanical properties of biopolymer-based hydrogels have hindered their widespread applications in biomedicine and tissue engineering. In recent years, researchers have shown significant interest in developing novel approaches to enhance the mechanical performance of hydrogels. This review focuses on key strategies for enhancing mechanical properties of hydrogels, including dual-crosslinking, double networks, and nanocomposite hydrogels, with a comprehensive analysis of their underlying mechanisms, benefits, and limitations. It also introduces the classic application scenarios of biopolymer-based hydrogels and the direction of future research efforts, including wound dressings and tissue engineering based on 3D bioprinting. This review is expected to deepen the understanding of the structure-mechanical performance-function relationship of hydrogels and guide the further study of their biomedical applications. Chitosan (CS) composite gels have emerged as promising materials with diverse applications in biomedicine. This review provides a concise overview of recent advancements and key aspects in the development of CS composite gels. The unique properties of CS, such as biocompatibility, biodegradability, and antimicrobial activity, make it an attractive candidate for gel-based composites. Incorporating various additives, such as nanoparticles, polymers, and bioactive compounds, enhances the mechanical, thermal, and biological and other functional properties of CS gels. This review discusses the fabrication methods employed for CS composite gels, including blending and crosslinking, highlighting their influence on the final properties of the gels. Furthermore, the uses of CS composite gels in tissue engineering, wound healing, drug delivery, and 3D printing highlight their potential to overcome a number of the present issues with drug delivery. The biocompatibility, antimicrobial properties, electroactive, thermosensitive and pH responsive behavior and controlled release capabilities of these gels make them particularly suitable for biomedical applications. In conclusion, CS composite gels represent a versatile class of materials with significant potential for a wide range of applications. In this study, we developed a novel nanocomposite by synthesizing zinc (ZnNPs), copper (CuNPs), and silver (AgNPs) nanoparticles using olive leaf extract and incorporating them into a chitosan polymer. This approach combines the biocompatibility of chitosan with the antimicrobial and anticancer properties of metal nanoparticles, enhanced by the phytochemical richness of olive leaf extract. The significance of our research lies in its potential to offer a biodegradable and stable alternative to conventional antibiotics and cancer treatments, particularly in combating multidrug-resistant bacteria and various cancer types. Comprehensive characterization through Fourier Transform Infrared Spectroscopy (FTIR), X-Ray Diffraction (XRD), Thermogravimetric Analysis (TGA), Scanning Electron Microscopy (SEM), Energy Dispersive X-ray (EDX), and Transmission Electron Microscopy (TEM) confirmed the successful synthesis of the nanocomposites, with an average size of ~22.6 nm. Phytochemical analysis highlighted the antioxidant-rich composition of both the olive leaf extract and the nanoparticles themselves. Functionally, the synthesized nanoparticles exhibited potent antimicrobial activity against multidrug-resistant bacterial strains, outperforming traditional antibiotics by inhibiting key resistance genes (ermC, tetX3-q, blaZ, and Ery-msrA). In anticancer assessments, the nanoparticles showed selective cytotoxicity towards cancer cells in a concentration-dependent manner, with CuNPs and AgNPs showing particularly strong anticancer effects, while demonstrating minimal toxicity towards normal cells. ZnNPs were noted for their low cytotoxicity, highlighting the safety profile of these nanoparticles. Further, the nanoparticles induced apoptosis in cancer cells, as evidenced by the modulation of oncogenes (P21, P53, and BCL2), suggesting their therapeutic potential. The emergence of novel well-defined biological macromolecular architectures containing fluorine moieties displaying superior functionalities can satisfactorily address many biomedical challenges. In this research, ABA- and AB-type glucose-based biological macromolecules were synthesized using acryl-2,3,4,6- tetra - O -acetyl-D-glucopyranoside with pentafluorophenyl (FPM), pentafluorobenzyl (FBM), phenyl (PM) and benzyl (BM) methacrylate-based macro-RAFT agents following RAFT polymerization. The macro-RAFT agents and the corresponding copolymers were characterized by 19F, 1H, and 13C NMR and FTIR spectroscopic techniques to understand the chemical structure, molecular weight by size-exclusion chromatography, thermal analysis by TGA and DSC. Thermal stability (T d5%) of the FPM and FBM fluoro-based polymers was observed in the range of 219–267 °C, while the non-fluoro PM and BM polymers exhibited in the range of 216–264 °C. Among the macro-RAFT agents, PFPM (107 °C, ΔH: 0.613 J /g) and PPM (103 °C, ΔH: 0.455 J /g) showed higher T m values, while among the block copolymers, PFBM- b -PG (123 °C, ΔH: 0.412 J /g) and PG- b -PFPM- b -PG (126 °C, ΔH: 0.525 J /g) exhibited higher T m values. PFBMT and PPM macro-RAFT agents, PPM- b -PG and PG- b -PPM- b -PG copolymer spin-coated films showed the highest hydrophobicity (120°) among the synthesized polymers. The block copolymers exhibited self-assembled segregation by using relatively hydrophobic segments as the core and hydrophilic moieties as the corona. Synthesized biological macromolecules exhibit maximum antibacterial activity towards S. aureus than E. coli bacteria. Fluorophenyl (PFPM) and non-fluorobenzyl-based (PBMT) macro-RAFT agents exhibit low IC 50 values, suggesting high cytotoxicity. The demand for the functionalization of additive materials based on bacterial cellulose (BC) is currently high due to their potential applications across various sectors. The preparation of BC-based additive materials typically involves two approaches: in situ and ex situ. In situ modifications entail the incorporation of additive materials, such as soluble and dispersed substances, which are non-toxic and not essential for bacterial cell growth during the production process. However, these materials can impact the yield and self-assembly of BC. In contrast, ex situ modification occurs subsequent to the formation of BC, where the additive materials are not only adsorbed on the surface but also impregnated into the BC pellicle, while the BC slurry was homogenized with other additive materials and gelling agents to create composite films using the casting method. This review will primarily focus on the in situ and ex situ functionalization of BC then sheds light on the pivotal role of functionalized BC in advancing biomedical technologies, wound healing, tissue engineering, drug delivery, bone regeneration, and biosensors. Silk is a natural engineering material with a unique set of properties. The major constituent of silk is fibroin, a protein widely used in the biomedical field because of its mechanical strength, toughness and elasticity, as well as its biocompatibility and biodegradability. The domestication of silkworms allows large amounts of fibroin to be extracted inexpensively from silk cocoons. However, the industrial extraction process has drawbacks in terms of sustainability and the quality of the final medical product. The heterologous production of fibroin using recombinant DNA technology is a promising approach to address these issues, but the production of such recombinant proteins is challenging and further optimization is required due to the large size and repetitive structure of fibroin's DNA and amino acid sequence. In this review, we describe the structure–function relationship of fibroin, the current extraction process, and some insights into the sustainability of silk production for biomedical applications. We focus on recent advances in molecular biotechnology underpinning the production of recombinant fibroin, working toward a standardized, successful and sustainable process. One of the most effective and promising strategies to develop novel biomaterials with unique, tunable structure and physicochemical properties is by creating composite materials that combine synthetic polymers with natural proteins using ionic liquids. In this study, biodegradable poly(d , l -lactic acid) (PDLLA) was blended with silk fibroin (SF) to create biocompatible films using an ionic liquid-based binary solvent system (1-butyl-3-methylimidazolium chloride/ N , N -dimethylformamide), which can maintain the molecular weights of the proteins/polymers and encourage intermolecular interactions between the molecules. The effects of varying the ratio of PLA to SF were studied using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), water contact angle testing, and cytotoxicity analysis as well as enzymatic degradation. Results showed that the composite films were homogeneously blended on the macroscopic scale and exhibited typical fully miscible polymer blend characteristics. By increasing the SF content in the composites, the amounts of β- sheets in the films were significantly increased, allowing for SF to act as a physical crosslinker to maintain the stability of the protein-polymer network. Additionally, SF significantly improved the hydrophilicity and biocompatibility of the material and promoted the self-assembly of micelle structures in the biocomposites. Different topologies in the films also provided beneficial surface morphology for cell adhesion, growth, and proliferation. In the present study, chitosan (CS) reacted with vanillin through a Schiff base reaction forming the vanillin-CS (VACS) derivative. FTIR and 1H NMR spectra confirmed the derivatization of CS, the enhanced swelling behavior was long-established while XRD measurement stated the semicrystalline nature of the VACS derivative. In a further step, blends between CS and VACS were prepared in ratios CS/VACS 90/10 up to 10/90 w /w and the formation of hydrogen bonds was noticed through FTIR and XRD measurements. Structural optimizations were performed within the framework of density functional theory and interaction energies E int were calculated. Collectively, these results along with v iscosity measurements and SEM images pro v e the miscibility of CS/VACS blends. In the optimum CS/VACS ratios, inks for 3D printing application were prepared in different concentrations (3% w / v , 4%w/v, 5%w/v, 6% w / v). Natural polysaccharides and their designed structures are extremely valuable due to their intrinsic pharmacological properties and are also used as pharmaceutical aids. These naturally occurring polysaccharides (e.g., psyllium and alginate) are gaining popularity for their use in the preparation of interpenetrating polymer network (IPN) materials with improved swelling ability, biodegradability, stability, non-cytotoxic, biocompatibility, and cost-effectiveness. IPN is prepared sequentially or simultaneously by microwave irradiation, casting evaporation, emulsification cross-linking, miniemulsion/inverse miniemulsion technique, and radiation polymerization methods. In addition, the prepared IPNs have has been extensively characterized using various analytical and imaging techniques before sustainable deployment for multiple applications. Regardless of these multi-characteristic attributes, the current literature lacks a detailed overview of the biomedical aspects of psyllium, alginate, and their engineered IPN structures. Herein, we highlight the unique synthesis, structural, and biomedical considerations of psyllium, alginate, and engineered IPN structures. In this review, a wide range of biomedical applications, such as role as a drug carrier for sustain delivery, wound dressing, tissue engineering, and related miscellaneous application of psyllium, alginate, and their IPN structures described with appropriate examples. Further research will be carried out for the development of IPN using psyllium and alginate, which will be a smart and active carrier for drugs used in the treatment of life-threatening diseases due to their inherent pharmacological potential such as hypoglycemic, immunomodulatory, antineoplastic, and antimicrobial. Biosynthesis of bacterial cellulose (BC) in cylindrical oxygen permeable molds allows the production of hollow tubular structures of increasing interest for biomedical applications (artificial blood vessels, ureters, urethra, trachea, esophagus, etc.). In the current contribution a simple set-up is used to obtain BC tubes of predefined dimensions; and the effects of fermentation time on the water holding capacity, nanofibrils network architecture, specific surface area, chemical purity, thermal stability, mechanical properties, and cell adhesion, proliferation and migration of BC tubes are systematically analysed for the first time. The results reported highlight the role of culture time on key properties of the BC tubes produced, with significant differences arising from the denser and more compact fibril arrangements generated at longer fermentation intervals. World Health Organization estimates that 30–50% of cancers are preventable by healthy lifestyle choices, early detection and adequate therapy. When the conventional therapeutic strategies are still regulated by the lack of selectivity, multidrug resistance and severe toxic side effects, nanotechnology grants a new frontier for cancer management since it targets cancer cells and spares healthy tissues. This review highlights recent studies using biotin molecule combined with functional nanomaterials used in biomedical applications, with a particular attention on biotinylated chitosan-based nanosystems. Natural macromolecules have attracted increasing attention due to their biocompatibility, low toxicity, and biodegradability. Pectin is one of the few polysaccharides with biomedical activity, consequently a candidate in biomedical and drug delivery Applications. Rhamnogalacturonan-II, a smaller component in pectin, plays a major role in biomedical activities. The ubiquitous presence of hydroxyl and carboxyl groups in pectin contribute to their hydrophilicity and, hence, to the favorable biocompatibility, low toxicity, and biodegradability. However, pure pectin-based materials present undesirable swelling and corrosion properties. The hydrophilic groups, via coordination, electrophilic addition, esterification, transesterification reactions, can contribute to pectin's physicochemical properties. Here the properties, extraction, and modification of pectin, which are fundamental to biomedical and drug delivery applications, are reviewed. Moreover, the synthesis, properties, and performance of pectin-based hybrid materials, composite materials, and emulsions are elaborated. Polysaccharides are the most abundant naturally available carbohydrate polymers; composed of monosaccharide units covalently connected together. Chitosan is the most widely used polysaccharides because of its exceptional biocompatibility, mucoadhesion, and chemical versatility. However, it suffers from a few drawbacks, e.g. poor mechanical properties and antibacterial activity for biomedical applications. Blending chitosan with natural or synthetic polymers may not merely improve its physicochemical and mechanical properties, but may also improve its bioactivity-induced properties. This review paper summarizes progress in chitosan blends with biodegradable polymers and polysaccharides and their biomedical applications. Herein, a novel strategy for surface functionalization and drug loading of cellulose nanocrystals (CNCs) through formation of hydrazone bonds between functionalized CNCs and aldehyde group containing polyethylene glycol (CHO-PEG)/anticancer drug doxorubicin (DOX) was reported for the first time. DOX could be loaded on PEGylated CNCs with high capacity and released from drug complexes (P-CNCs-D) with pH dependent behavior. The biological evaluation results demonstrated that drug carriers (CNCs-EBO-NH) showed negative cytotoxicity while DOX could be transported into cells and exhibits desirable anticancer effects. As compared with other method, the method developed in this work is rather simple and effective and can be achieved for simultaneous for surface functionalization and drug loading in a one-pot route. This work will open a new avenue for fabrication of various multifunctional composites based on other carbohydrate polymers or materials and to explore their applications in biomedical fields. Electrospinning is the simplest technique to produce ultrathin nanofibers, which enables the use of nanotechnology in various applications. Nanofibrous materials produced through electrospinning have garnered significant attention in biomedical applications due to their unique properties and versatile potential. In recent years, there has been a growing emphasis on incorporating sustainability principles into material design and production. However, electrospun nanofibers, owing to their reliance on solvents associated with significant drawbacks like toxicity, flammability, and disposal challenges, frequently fall short of meeting environmentally friendly standards. Due to the limited solvent choices and heightened concerns for safety and hygiene in modern living, it becomes imperative to carefully assess the implications of employing electrospun nanofibers in diverse applications and consumer products. This systematic review aims to comprehensively assess the current state of research and development in the field of "green and natural" electrospun polymer nanofibers as well as more fascinating and eco-friendly commercial techniques, solvent preferences, and other green routes that respect social and legal restrictions tailored for biomedical applications. We explore the utilization of biocompatible and biodegradable polymers sourced from renewable feedstocks, eco-friendly processing techniques, and the evaluation of environmental impacts. Our review highlights the potential of green and natural electrospun nanofibers to address sustainability concerns while meeting the demanding requirements of various biomedical applications, including tissue engineering, drug delivery, wound healing, and diagnostic platforms. Recently, numerous studies have been conducted on renewable polymers derived from different natural sources, exploring their suitability for diverse biomedical applications. Lignin as one of the main components of lignocellulosic has garnered significant attention as a promising alternative to petroleum-based polymers. This interest is primarily due to its cost-effectiveness, biocompatibility, eco-friendly nature, as well as its antioxidant and antimicrobial properties. These characteristics could be more beneficial when incorporating lignin into the formulation of value-added products. Although lignin has a chemical structure that is suitable for various applications, these characteristics require modifications to guarantee that the resultant materials display the desired biological, chemical, and physical properties when applied in the creation of biodegradable hydrogels, particularly for biomedical purposes. This study delineates the recent modification approaches that have been employed in the creation of lignin-based hydrogels. These strategies encompass both chemical and physical interactions with other polymers. Additionally, this review encompasses an examination of the current applications of lignin hydrogels, spanning their use as scaffolds for tissue engineering, carriers for pharmaceuticals, materials for wound dressings and biosensors, and elements in flexible and wearable electronics. Finally, we delve into the challenges and constraints associated with these materials, discuss the necessary steps required to attain the appropriate properties for the development of innovative lignin-based hydrogels, and derive conclusions based on the presented findings. Novel biomedical materials provide a new horizon for the diagnosis/treatment of diseases and tissue repair in medical engineering. As the most abundant biomass polymer on earth, cellulose is characterized by natural biocompatibility, good mechanical properties, and structure-performance designability. Owing to these outstanding features, cellulose as a biomacromolecule can be designed as functional biomaterials via hydrogen bonding (H-bonding) interaction or chemical modification for human tissue repair, implantable tissue organs, and controlling drug release. Moreover, cellulose can also be used to construct medical sensors for monitoring human physiological signals. In this study, the structural characteristics, functionalization approaches, and advanced biomedical applications of cellulose are reviewed. The current status and application prospects of cellulose and its functional materials for wound dressings, drug delivery, tissue engineering, and electronic skin (e-skin) are discussed. DNA's programmable, predictable, and precise self-assembly properties enable structural DNA nanotechnology. DNA nanostructures have a wide range of applications in drug delivery, bioimaging, biosensing, and theranostics. However, physiological conditions, including low cationic ions and the presence of nucleases in biological systems, can limit the efficacy of DNA nanostructures. Several strategies for stabilizing DNA nanostructures have been developed, including i) coating them with biomolecules or polymers, ii) chemical cross-linking of the DNA strands, and iii) modifications of the nucleotides and nucleic acids backbone. These methods significantly enhance the structural stability of DNA nanostructures and thus enable in vivo and in vitro applications. This study reviews the present perspective on the distinctive properties of the DNA molecule and explains various DNA nanostructures, their advantages, and their disadvantages. We provide a brief overview of the biomedical applications of DNA nanostructures and comprehensively discuss possible approaches to improve their biostability. The aim of the study is to explore the myriad of anti-activities of chitosan - deacylated derivative of chitin in biomedical applications. Chitosan consists of reactive residual amino groups, which can be modified chemically to obtain wide range of derivatives. Fatty acids-assisted superparamagnetic maghemite (γ-Fe 2 O 3) NPs was biologically synthesized using extract of polyherbal drug Liv52 (L52E). The NPs were characterized by UV–vis spectroscopy, FT-IR, SEM, TEM, EDX, XRD and VSM. The major biological molecules present in L52E analysed by GC–MS were saturated fatty acids (palmitic acid 21.95%; stearic acid 13.99%; myristic acid 1.14%), monounsaturated fatty acid (oleic acid 18.43%), polyunsaturated fatty acid (linoleic acid 20.45%), and aromatic phenol (cardanol monoene 11.92%) that could imply in bio-fabrication and stabilization of γ-Fe 2 O 3 NPs. The FT-IR spectra revealed involvement of carboxylic group of fatty acids, amide group of proteins and hydroxyl group of phenolic compounds that acts as reducing and capping agents. The synthesized NPs were used to investigate their antimicrobial, antibiofilm activity against P. aeruginosa , MRSA and C. albicans and anticancer activity on colon cancer cells (HCT-116) for biomedical applications. Further, molecular docking study was performed to explore the interaction of Fe 2 O 3 NPs with major cell wall components i.e., peptidoglycan and mannoproteins. The docking studies revealed that Fe 2 O 3 interacted efficiently with peptidoglycan and mannoproteins and Fe 2 O 3 get accommodated into catalytic cleft of mannoprotein. Due to magnetic property, the biological activity of γ-Fe 2 O 3 can be further enhanced by applying external magnetic field alone or in amalgamation with other therapeutics drugs. Natural polymer-based hybrid nanocomposites have been proposed as one of the most promising tools for biomedical applications, including disease treatment and diagnosis procedures. Xyloglucan nanocapsules can simultaneously load magnetic iron oxide nanoparticles and bioactive for a specific tissue, reducing the processes of degradation and metabolic inactivation of molecules with biological activity. In this work, magnetic nanocapsules of xyloglucan loaded with hydrophilic sulfated quercetin (MNXQ_SO 3) were successfully synthesized by inverse miniemulsion process through interfacial polymerization. The polymeric shell formation of nanocapsules was evidenced by Fourier Transform Infrared spectroscopy and Transmission Electron Microscopy. The ferrofluid (Fe 3 O 4 @PAAS) incorporated into the xyloglucan nanocapsules was synthesized by hydrothermal method, using polyacrylic acid sodium salt as coating. Dynamic Light Scattering technique confirmed the nanomeric dimensions (202.3 nm) and the good colloidal stability (−40.2 mV) of MNXQ_SO 3. The saturation magnetization analyses pointed out the superparamagnetic behavior of Fe 3 O 4 @PAAS (48 emu/g) and MNXQ_SO 3 (4.2 emu/g). MNXQ_SO 3 was able to modify the release profile of sulfated quercetin (67%) when compared to the free bioactive (100%), exhibiting a release profile compatible with the zero-order kinetic model. The results showed that the development of MNXQ_SO 3 presents a new perspective for biomedical applications, including studies of targeted drug delivery. Petrochemical plastics have become a cause of pollution for decades and finding alternative plastics that are environmental friendly. Polyhydroxyalkanoate (PHA), a biopolyester produced by microbial cells, has characteristics (biocompatible, biodegradable, non-toxic) that make it appropriate as a biodegradable plastic substance. The different forms of PHA make it suitable to a wide choice of products, from packaging materials to biomedical applications. The major challenge in commercialization of PHA is the cost of manufacturing. There are a lot of factors that could affect the efficiency of a development method. The development of new strategic parameters for better synthesis, including consumption of low cost carbon substrates, genetic modification of PHA-producing strains, and fermentational strategies are discussed. Recently, many efforts have been made to develop a method for the cost-effective production of PHAs. The isolation, analysis as well as characterization of PHAs are significant factors for any developmental process. Inspired from biological motors, cellulose nanocrystals (CNCs) are strategically modified to induce self-propulsion behavior with the capabilities to catalytically degrade pollutants along with magnetic hyperthermia to clean arterial plaques during its course of propulsion. CNCs derived from renewable biomass, are decorated with catalytically active, magneto-responsive nanomaterials (Fe 2 O 3 /Pd nanoparticles) through sustainable routes. CNC nanomotors show improved propulsion at lowered peroxide concentrations with remotely controlled trajectory through chemo-magnetic field gradients and ideal surface-wettability characteristics, overcoming the requirement of surfactants, as with traditional nanomotors. We observed that nanomotors undergo motion through heterogeneous bubble propulsion mechanism, with capability to in situ degrade pollutants and generate local heat through hyperthermia, enhancing the rate of degradation process in real time. As proof of concept, we demonstrate that the dynamics of nanomotors can be controlled in a microfluidic channel through site-directed magnetic field and induction of pH gradient, mimicking the chemotaxis in cell-like environment and as swarm of nano-surgeons removes plaques from clogged arteries. Our study shows that strategic modification of CNCs results in fabrication of nanomotors with efficient propulsion system infused with multi-functional characteristics of high catalytic activity and magnetic hyperthermia which opens up new avenues for utilization of bio-based nanomotors derived from lignocellulose for myriad applications. Lectins are ubiquitous carbohydrate-binding proteins that interact with sugar moieties in a highly specific manner. H-type lectins represent a new group of lectins that were identified in invertebrates. These lectins share structural homology and bind mainly to N- acetylgalactosamine (GalNAc). Recent structural studies on the H-type lectins provided a detailed description of the GalNAc-lectin interaction that is already exploited in a number of biomedical applications. Two members of the H-type lectin family, Helix pomatia agglutinin (HPA) and Helix aspersa agglutinin (HAA), have already been extensively used in many diagnostic tests due their ability to specifically recognize GalNAc. This ability is especially important because aberrant glycosylation patterns of proteins expressed by cancer cells contain GalNAc. In addition, H-type lectins were utilized in diagnostics of other non-cancer diseases and represent great potential as components of drug delivery systems. Marine biome exhibits an immense essence of excellence and enriched with high-value bioactive compounds of therapeutic and biomedical value. During the past several years, an array of biologically active molecules has been extracted/isolated and purified from numerous sources of marine origin with the aid of distinct techniques and methodologies for newer applications. The growing demand for bioactive molecules with unique functionalities in various industrial divisions, such as therapeutic sectors and biomedical, has endorsed the necessity for highly suitable and standardized strategies to extract these bioactive components using a state-of-the-art and inexpensive measures. This is also because many in practice conventional extraction methodologies suffer from processing limitations and low-yield issues. Besides that, other major issues include (i) decrease efficacy, (ii) excessive energy cost, (iii) low yield, (iv) lower cost-effective ratio, (v) minimal selectivity, (vi) low activity, and (vii) stability, etc. In this context, there is an urgent need for new and robust extraction strategies. The synergies of modern extraction techniques with efficient and novel pretreatment approaches, such as the integration of enzymes, accompanied by conventional extraction processes, should be the utmost goal of current research and development studies. The typical effectivity of the extraction techniques mostly relies on these points, i.e., (i) know-how about the source nature and type, (ii) understanding the structural and compositional profile, (iii) influence of the processing factors, (iv) interplay between the extraction conditions and the end-product, (v) understanding the available functional entities, (vi) reaction chemistry of the extract bioactive compounds, and (vii) effective exploitation of the end-product in the marketplace. Marine biome, among numerous naturally occurring sources, has been appeared an immense essence of excellence to isolate an array of biologically active constituents with medicinal values and related point-of-care applications. Herein, we reviewed the salient information covering various therapeutic potential and biomedical perspectives. Following a brief introduction and marine pharmacognosy, an array of high-value biomolecules of marine origin are discussed with suitable examples. From the robust extraction strategies viewpoint, a part of the review focuses on three techniques, i.e., (1) enzyme-assisted extraction (EAE), (2) supercritical-fluid extraction (SFE), and (3) microwave-assisted extraction (MAE). Each technique is further enriched with processing and workflow environment. The later part of the review is mainly focused on the therapeutic and biomedical perspectives of under-reviewed bio-active compounds or biomolecules. The previous and latest research on the anticancer, skin curative, cardio-protective, immunomodulatory and UV-protectant potentialities of marine-derived biologically active entities have been summarized with suitable examples and related pathways illustrations. This study explores the modification of bacterial nanocellulose (BNC), produced in fermented tea medium with bio-extracts from herbal plants, to produce entirely natural antibacterial nanocellulose films. The antibacterial property was imparted by impregnating the synthesized BNC into bio-extracts of Terminalia arjuna (arjuna), Azadirachta indica (neem), Withania somnifera (ashwagandha), Tinospora cordifolia (giloy), and Murraya koenigii (curry leaves). FE-SEM analysis of modified BNC films revealed the presence of cloudy layer of bio-extracts over the BNC nanofibrous network. The modified BNC production was confirmed by ATR-FTIR. The modified BNC showed tremendous antibacterial activity against Escherichia coli (E. coli) and Aerococcus viridians (A. viridans). T. arjuna modified BNC showed the highest antibacterial activity against E. coli and A. viridans with inhibition zone of 27.08 mm and 26.34 mm, respectively, while M. koeniggi modified BNC showed the lowest antibacterial activity for both E. coli and A. viridans with inhibition zone of 14 mm and 14.2 mm, respectively. The water retention, moisture content and porosity values reflect highly hydrophilic nature of BNC films and their well suitability for varied biomedical applications like antibacterial wound dressings, herbal biomasks, scaffoldings, etc. The modification of BNC films with the bio-extracts used in this study has not been reported previously. During the last decades, the ever-increasing incidence of various diseases, like cancer, has led to a high rate of death worldwide. On the other hand, conventional modalities (such as chemotherapy and radiotherapy) have not indicated enough efficiency in the diagnosis and treatment of diseases. Thus, potential novel approaches should be taken into consideration to pave the way for the suppression of diseases. Among novel approaches, biomaterials, like chitosan nanoparticles (CS NPs, N -acetyl-glucosamine and D-glucosamine), have been approved by the FDA for some efficient pharmaceutical applications. These NPs owing to their physicochemical properties, modification with different molecules, biocompatibility, serum stability, less immune response, suitable pharmacokinetics and pharmacodynamics, etc. have received deep attention among researchers and clinicians. More importantly, the impact of CS polysaccharide in the synthesis, preparation, and delivery of metallic NPs (like gold, silver, and magnetic NPs), and combination of CS with these metallic NPs can further facilitate the diagnosis and treatment of diseases. However, these waste contain cellulose acetate (CA) and can be recycled, transforming into raw materials for new products. In this study, cigarette filters waste was collected, recycled and used for fabricating the nanofibrous membrane of cellulose acetate nanofibers (CFCA) through electrospinning technique. Tetracycline hydrochloride (TC) was encapsulated in the nanofibers to prevent bacterial infections. Various analyses were conducted: Scanning Electron Microscope (SEM), Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction Analysis (XRD) and Thermogravimetric analysis (TGA). CA and CFCA exhibited high water uptake properties and exhibited similar breaking stress and strain values. Both CA and CFCA effectively acted as stable drug carriers, with sustained in vitro drug release. Antibacterial activity was demonstrated by the drug-loaded CA and CFCA nanofibers against, Gram-positive bacteria Staphylococcus aureus and Gram-negative bacteria Escherichia coli. Based on their cytotoxicity evaluations on mouse fibroblast cells (L929), CA and CFCA fibrous mats demonstrated no cytotoxicity and similar cell viability results. Consequently, the TC-loaded nanofibers made from CA and CFCA exhibited suitable properties for wound healing applications. Carbohydrate used in biomedical applications is influenced by numerous factors. One of the most appealing characteristic of carbohydrates is their ability to reproduce from natural resources which makes them ecologically friendly. Due to their abundance, biocompatibility, and no contamination by residual initiators, the desire for polysaccharides in medical uses is growing. Research on fiber-based materials, with a variety of medical applications including bio-functional scaffolds, continues to yield novel and intriguing findings. Almost all biopolymers of diverse structural compositions are electrospun to fulfill biomedical usage criteria, and the electrospinning technique is widely employed in biomedical technologies for both in-vivo and in-vitro therapies. Due to its biocompatibility and biodegradability, polycaprolactone (PCL) is employed in medical applications like tissue engineering and drug delivery. Although PCL nanofibers have established effects in vitro, more research is needed before their potential therapeutic application in the clinic. Cancer is the leading cause of death all over the world and chemotherapy is an important approach to fight cancer, however, there are many obstacles against successful cancer chemotherapy such as development of multidrug resistance, poor solubility of chemotherapeutic agents and adverse side effects to healthy tissues. An important strategy to overcome these obstacles, is the use of nanotechnology. In recent years, natural polymers such as cellulose and its nanoform structure, nanocrystalline cellulose (NCC), have attracted the interest of researchers in the field of nanotechnology and specially drug delivery systems, due to biocompatibility and biodegradability of NCC. Cellulose is the most abundant natural biopolymer and changes to NCC by several chemical and mechanical methods. In this review, we mainly focus on the methods for production of NCC, physicochemical properties and medical applications of NCC (e.g. regenerative medicine, replacement of vascular grafts, tissue engineering, anti-bacterial/anti-viral applications, diagnosis and biosensing) with a special emphasize on drug delivery systems. Ionic liquids (ILs), are considered as eco-friendly solvents due to their low vapor pressure and non-volatile properties compared to organic solvents and can be employed during processes for the extraction and/or conversion of lignin and chitin to value-added products which can improve the overall economics of the integrated biorefinery concept. However, relatively few studies have investigated their ability to solubilize and modify during solvation of lignin and chitin in terms of crystallinity, molecular weight, etc. , with a significant focus on polysaccharides like cellulose and hemicellulose. Similarly, the biomedical applications of lignin and chitin are not recognised so much as compared to polysaccharides. The aim of this review is survey the current literature based on the cutting-edge-knowledge regarding the solubilization of chitin and lignin in various classes of ILs and furthermore, the upgradation of these biopolymers and/or their derivatives for the biomedical applications. Moreover, this review provides a comprehensive summary of the latest progress on the solubilization of integrated biorefinery lignin and chitin with a wide range of ILs using experimental and theoretical studies. This work addresses the competence of PVA/starch/citric acid (PVA/St/CA) based composite films for wound dressing applications. Literature reported composition of PVA, starch and glycerol were adopted and the optimality of crosslinking temperature (40 to 60 °C) and citric acid concentration (0.5 to 3 g) have been investigated during solution casting based composite film fabrication. The prepared polymer films have been characterized with swelling index, solubility based biodegradability, gel fraction, mechanical strength, water vapor transmission rate and antimicrobial effectiveness. The optimized film (composition) were analyzed to possess excellent swelling (260.5 ± 2.9%) and in-vitro degradation (45.5 ± 1.8–59.8 ± 0.4% degradation, during 14 days) characteristics. Further, the film possesses enhanced combinations of water vapor transmission rate and antibacterial activity with Gram-negative (Escherichia coli) and Gram-positive (Listeria monocytogenes) bacteria. With all these promising characteristics, the low temperature (50 °C) and long duration (12 h) fabricated PVA-St-CA films have been inferred to be a potential candidate for wound dressing applications. • PVA-St-CA films: optimality of drying time and crosslinking temperature. • Composition optimality: 35 wt% CA • Superb absorption ability, 260%; up to 400% • Increasing CA amount dominate it's crosslinking effect via plasticizing effect. Pelvic floor dysfunction diseases (PFD) become more prevalent with the increase of elderly population, and complications of pelvic floor reconstructive surgery (e.g. infection and exposure of mesh) have been troubling to patients and gynecologists. In this study, the nanofibrous mats were prepared by alternately depositing chitosan (CS) and silk fibroin (SF) on Nylon6 (N6) nanofibrous mats via layer-by-layer (LBL) technique. The as-prepared mats were characterized. The results showed that CS and SF molecules were successfully assembled on the nanofibers. Additionally, after LBL modification, the hydrophilicity of the nanofibrous mats was reduced and the mechanical properties were improved. Furthermore, the antibacterial activity of the LBL-structured mats reached >95% inhibiting Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The in vitro cell co-culture experiments indicated that LBL-structured mats had smaller toxic effects and more excellent biocompatibility to L929 fibroblasts, especially the mats with 15 bilayers coated films. Abstract Hydroxyapatite (HAp) is a bioactive and biocompatible material possessing osteoconductive properties used widely in the biomedical sector. In the present study, synthesis of hydroxyapatite (HAp) using a Klebsiella pneumoniae SM24 (phosphate solubilizing bacteria) isolated from the slaughterhouse. HAp synthesized using biological source showed efficient and positive enzymatic activity in the National Botanical Research Institute Phosphate Medium (NBRIP). Characterization of HAp using FTIR revealed the presence of phosphate group hydroxyapatite and XRD spectra showed polycrystalline nature. The morphological characterization of HAp using FESEM revealed the mesoporous structure and EDX spectrum indicated presence of Ca and P as the major components. In addition, a new bone composite was prepared using the synthesized HAp, Gelatine (G), Chitosan (C), Fibrin (F) and Bone ash (HApGCF) using Simulated Body Fluid (SBF) solution. The confirmation of chemical and structural characteristics of HApGCF bone composite was achieved using FTIR, XRD and SEM analyses. The HApGCF bone composite was tested over osteoblast MG-63 cells showing effective biocompatibility and osteoblast attachment on the composite surface. Therefore, the present report proposes the in vitro application of HApGCF bone composite as a replacement for major bone damage and injury in a biocompatible and non-toxic way. Abstract PHAs (polyhydroxyalkanoates) have emerged as biodegradable plastics more strongly in the 20th century. A wide range of bacterial species along with fungi, plants, oilseed crops and carbon sources have been used extensively to synthesize PHA on large scales. Alteration of PHA monomers in their structures and composition has led to the development of biodegradable and biocompatible polymers with highly specific mechanical properties. This leads to the incorporation of PHA in numerous biomedical applications within the previous decade. PHAs have been fabricated in various forms to perform tissue engineering to repair liver, bone, cartilage, heart tissues, cardiovascular tissues, bone marrow, and to act as drug delivery system and nerve conduits. A large number of animal trials have been carried out to assess the biomedical properties of PHA monomers, which also confirms the high compatibility of PHA family for this field. Smart thermoresponsive polymers have long attracted attention as materials of a great potential for biomedical applications, mainly for drug delivery, tissue engineering and wound dressing, with a special interest to injectable hydrogels. Poly-N-isopropylacrylamide (PNIPAM) is the most important synthetic thermoresponsive polymer due to its physiologically relevant transition temperature. However, the use of unmodified PNIPAM encounters such problems as low biodegradability, low drug loading capacity, slow response to thermal stimuli, and insufficient mechanical robustness. The use of natural polysaccharides and proteins in combinations with PNIPAM, in the form of grafted copolymers, IPNs, microgels and physical mixtures, is aimed at overcoming these drawbacks and creating dual-functional materials with both synthetic and natural polymers' properties. When developing such compositions, special attention should be paid to preserving their key property, thermoresponsiveness. Addition of hydrophobic and hydrophilic fragments to PNIPAM is known to affect its transition temperature. This review covers various classes of natural polymers - polysaccharides, fibrous and non-fibrous proteins, DNA - used in combination with PNIPAM for the prospective biomedical purposes, with a focus on their phase transition temperatures and its relation to the natural polymer's structure. Chitosan is a biodegradable, non-toxic, and antimicrobial polymer. Chitosan films can be used as a dressing because they promote the healing of cutaneous ulcers. In this study, the mechanical, physical, and microbiological properties films of pure chitosan and films formulated with a glycerol-honey mixture were characterized. The films were smooth, homogenous, transparent, and porous, with no fractures or cracks. Additionally, it was found that all were resistant to breaking, that the tearing force was directly related to the chitosan concentration, and that the addition of honey and glycerol improved the elongation percentage. When evaluating the antimicrobial activity of the films against Pseudomonas aeruginosa and Klebsiella pneumoniae , the films were found to have an effect only by direct contact. In the films formulated with honey, the area of contact increased to 44%. The excellent color, structural, antimicrobial, and surface morphological properties of the newly developed films make them a promising alternative for use as a dressing for the healing of skin ulcers.
