We developed a new method to detect Acidovorax avenae subsp. citrulli in watermelon seeds, by modifying the membrane filter immunostaining method, which was developed to detect Burkholderia glumae in rice seeds. The detection threshold of this new method for A. avenae subsp. citrulli was several cfu per 100 ml phosphate buffer using 1000 commercial watermelon seeds. Living cells of A. avenae subsp. citrulli could be obtained by stamping the membrane on selective medium plate before immunostaining.

極めて低濃度でしか存在しない農業環境中からイネもみ枯細菌病菌Burkholderia glumaeを検出するのは,選択培地を用いた塗沫法のような従来の方法では困難である.これを解決するために,メンブレンフィルターによる濾過法と増菌後にPCRを行う増菌PCR法とを組み合わせた新たな方法を考案した. PCRプライマーは, B. glumaeの16S-23S rDNAスペーサー領域を特異的に増幅するよう設計したもの(PGF1, 5'-TGTCTGACACGGAACACCTGGGTAG-3'; PPR1, 5'-AGGTTGAGTTCTCGCATTTGTGCCG-3')を用いた.本法により, 100 cfuのB. glumaeで人工的に汚染した1000mlの農業用水からの検出が可能であった.本法を用いて,水田地域における水圏環境から採取した大容量試料について調査を行った結果, 5月から11月にかけて田面水,農業用水,河川水および湖沼水からB. glumaeが検出されたが, 12月から4月の間は全く検出されなかった.また,もみ枯症が発生しB. glumaeが検出された水田の内外に自生する植物34株について検出を試みた結果,コナギから本細菌が検出された.

A selective medium (designated as NSVC-In medium) was developed to isolate of Erwinia ananas, the causal agent of bacterial palea browning of rice. Medium NSVC-In contains per 1000ml of distilled water: 10g inositol, 10g peptone, 50g NaCl, 100mg vancomycin, 100mg cycloheximide, and 15g agar. The medium is adjusted to pH 6.8 with NaOH before autoclaving. Plant pathogenic E. ananas grew well on this medium to develop characteristic colonies, but the growth of nonpathogenic E. ananas and Erwinia herbicola was inhibited. Plating efficiency of this medium for pathogenic E. ananas was the same as that of nutrient agar medium. Pathogenic E. ananas could be selectively detected from dead leaves of rice plants.

The causal bacteria of bacterial black node of barley and wheat, Pseudomonas syringae pv. japonica, invaded the leaf blade, leaf sheath, internode, node, awn, palea and lemma through the stomata, multiplied in the intercellular spaces of parenchyma, but never invaded the vascular bundles of infected plants. Brownish black lesions spread widely in spite of limited location of the bacterial multiplications in the parenchyma. In case of the palea, stomata existed on both sides of them, and they were connected with each others through the intercellular space of parenchyma. Thus, in the palea tissue, bacteria are able to move from one side to another side. This infection process is regarded as a main cause of the seed-borne mechanism of this disease. The causal bacteria infected the palea and lemma at first, then invaded the caryopsis through the funiculus, and multiplied in the intercellular spaces of parenchyma of caryopsis, but no bacteria were observed in the vascular bundles.

1985年8月から,沖縄県平良市(宮古島)で,マンゴー(品種:アーウィン)の葉および果実に黒褐色の隆起斑点を生ずる細菌性の病害が発生している。罹病組織からはコロニーの外観が均一な細菌が分離され,これらはマンゴー(品種:アーウィン)葉への接種により原病徴と同一の症状が発生した。また病原細菌は,約70項目の細菌学的性質の調査結果から,白∼クリーム色のXanthomonas campestrisであることが明らかになった。これらの結果とマンゴーbacterial cankerあるいはbacterial black spotに関する既往の報告から,分離細菌はXanthomonas campestris pv. mangiferaeindicaeと同定された。わが国における本細菌によるマンゴーの病害発生は,今回初めて確認された。病名はその病徴からマンゴーかいよう病(bacterial canker)とすることを提案した。

The causal bacterium of rice grain rot entered to the lemmata and paleae through the stomata, and multiplied in the intercellular spaces of the parenchyma. Stomata are mainly opened on the inner surfaces of lemmata and paleae, a few on outer surfaces of lemmata and they are connected each others through the intercellular space of parenchyma. The bacteria multiplied on the surface of albumen and embryo, but never invaded them.

1985年8月に高知県下でスターチスが萎ちょうする病気が大発生した。本病は葉が生気を失って萎ちょうし,葉脈が顕著に赤変する特徴がある。また,病植物の根の維管束が褐変し,細菌泥の漏出が観察される。8月下旬の高温期に定植するとすばやく発病するが,気温の低下を待って定植すると発病は遅れる。病勢は11月から停滞し,翌年の4月以降に再び活発となる。 分離細菌25株は均質な細菌学的性質を有し, Type strainを含むカーネーション由来のP. caryophylliの性質と一致した。両者は根部への穿刺接種あるいは断根後に菌液を灌注する方法でスターチスに自然発病の病徴を再現し,カーネーションに萎ちょう細菌病を起こした。したがって,分離細菌をPseudomonas caryophylli (Burkholder 1942) Starr and Burkholder 1942と同定した。 病名は,カーネーション萎ちょう細菌病の例にならい,スターチス萎ちょう細菌病(bacterial wilt of statice)を提案した。

The causal bacterium entered lemmata and paleae through stomata, and multiplied in the intercellular space of the parenchyma. Stomata are mainly open on the inner surface of lemmata and paleae, few on outer surface of lemmata, and they are connected each other through the intercellular space of parenchyma. This infection site does not seem specific phenomenon with regard as in case of Erwinia herbicola. The bacterial grain rot caused by Pseudomonas glumae and the bacterial seedling blight caused by P. plantarii followed this infection pattern to the rice grain. It is suggested that the seed-borne bacterial disease of rice generally follow this course of life cycle. Browning occurred only on the inner surface and the parenchyma of the palea affected with E. herbicola, and is suspected to be one of the defensive reaction of the palea against E. herbicola.
