The oesophagus of 18 minipiglets was exposed to endoscopic intraluminal irradiation with a 1320 nm Nd:YAG laser (10 W, 20 s) via a radial applicator with strictly radially symmetric light distribution. Immediately and at 2, 3, 4 and 8 wk after irradiation, the oesophagus was perfusion-fixed and filled with contrast solution. Radiographs were taken for evaluation by microradiometry. The specimens were subsequently prepared for light and transmission electron microscopy. The immediate reaction to irradiation was a morphological gradient of damage extending from the centre of the laser exposure where there was cellular thermonecrosis in all layers of the wall and condensation of the extracellular matrix, to a peripheral zone (at a distance of up to 8 mm from the region where the laser was centred) which showed only minor tissue damage manifested by intracellular vacuolation. In this zone dilatation of most vascular lumina was prominent. In the period between 2 and 8 wk after irradiation all phases of wound healing were observed and resulted in occlusion of the lumen of the oesophagus by early scar tissue after an interval of more than 3 wk at the former centre of laser exposure. Peripherally, epithelial regeneration resulted in a new luminal lining. Both the process of epithelial regeneration and that of fibrous repair indicated a good reparative capacity of laser-irradiated oesophageal wall tissues resembling the phases of normal wound healing. The immediate laser interaction with tissue indicated that the noncellular matrix components of tissues are more resistant to the photothermal effect than the cellular components.

The mucosal exudate from the gastrointestinal tract of six adult female New Zealand rabbits was studied using scanning electron microscopy and without any attempt being made to clean the luminal surface before screening. The exudate consisted of mucus, debris and bacteria. Qualitative assessment showed that the nature and distribution of exudate varied along the length of the gastrointestinal tract from the oesophagus to the anal canal, with little variation from animal to animal. Bacterial counts for rod-shaped bacteria were carried out on areas randomly selected from the upper, middle and lower oesophagus and the oesophageal-cardiac junction. The degree of bacterial colonisation was found to decrease along the length of the oesophagus from upper to lower parts, but it was increased at the oesophageal-cardiac junction. This assessment was not undertaken in the other regions of the gastrointestinal tract as the mucosal surface areas could not be easily measured owing to their undulating nature. The study indicates the variability of the mucosal exudate, which should be recognised as part of the true interface between ingested food and the cell surface along the gastrointestinal tract.

The stomach of the tammar may be divided into cardiac, "oesophageal", fundic and pyloric regions. In the cells of the cardiac region (which occupy 65% of the stomach) at least three types of mucous droplet are present. The stratified squamous "oesophageal" region occupies 20% of the stomach and the fundic and pyloric regions make up the remaining 15%. The cellular structure of the three last named regions seems to be similar to that described for other monogastric mammals. Endocrine cells were found in all glandular regions.

The oesophageal epithelium of the newborn opossum generally is two to three cells in depth and in some regions appears pseudostratified. By the 9th postnatal day the epithelium shows two distinct strata. Ciliated cells and occasional goblet cells also are observed within the epithelium during this stage and in subsequent stages. Cilia persist in the oesophagus of the adult opossum, but are restricted to the depths of the transverse folds found in the distal part of the organ. The epithelium covering the transverse folds of the adult likewise has an immature appearance. By 4-5 cm (ca. 20 days), the epithelium has assumed a more mature appearance and is of greater depth. This and later stages show three basic strata: a germinal layer, a spinous layer and, adjacent to the lumen, a flattened layer of cells that retain their nuclei. The epithelium throughout the postnatal period and in the adult does not undergo complete keratinization. The oesophageal glands begin as outgrowths from the epithelium just prior to 4-5 cm (ca. 20 days). The glands continue their development throughout the remainder of the postnatal period. The secretory units of the oesophageal glands of the the major portion of the secretory elements, and a light, rounded cell type which is less numerous and which occupies the terminal portions of the secretory units. Secretory material of the former appears complex, consisting of both neutral and acid glycoproteins. The secretory product of the light cell type is unknown at present. Both cell types are encompassed by myoepithelial cells. The relationship of the mitotic sequences to the observations made by microscopic examination of the developing oesophagus is discussed.

For the first time, at the ultrastructural level, the existence of free-ending, intraepithelial nerve fibres has been demonstrated in the oesophagus wall of adult cats and monkeys. Their form, the way they penetrate the epithelium, their location within the epithelium and their relationships with neighbouring cells have been established. A sensory function is suggested for this type of ending.

The volume density of mitochondria and the surface density of mitochondrial inner membranes were investigated in the stratified squamous epithelia of the oesophagus and forestomachs of sheep, and in the oesophagus and nonglandular part of the ventricle of rats. The higher values found for both parameters in all forestomach regions are thought to be correlated with the special metabolic activity known to take place in these epithelia.

The dendrite cells of Langerhans, first identified in the epidermis, have now been observed in the middle and superficial layers of the normal human oesophageal mucosa. They exhibit typical Langerhans granules, but no desmosomes and tonofilaments. They often have irregular indented nuclei, with a relatively pale cytoplasm contrasting with that of the adjacent squamous cells. These cells are sometimes difficult to distinguish from intra-epithelial lymphocytes, which are also encountered in the oesophageal mucosa and which share certain ultrastructural characteristics with Langerhans cells.

Normal squamous epithelium and submucosal glands of the human oesophagus have been studied in biopsy and resection specimens. The squamous epithelium has a structure similar to that seen in other histologically comparable sites, and contains Langerhans cells. Features of note within the squamous cells include microfibrillar intranuclear bodies and occasional intracytoplasmic desmosomes. The submucosal glands are comparable in many respects to the mucous-secreting minor salivary glands. Myoepithelial cells are observed within the acini and smaller ducts; occasional oncocytic cells are identified at the junction between duct and acinus. The innvervation of these glands appears to be indirect.
