Blood sample collection from the caudal vena cava at the site of uterine-ovarian drainage provides a more exact evaluation of the concentration and pattern of secretion of uterine or ovarian secreted products for studies of reproductive processes in cyclic and pregnant cattle compared with samples collected from general circulation. This paper describes a thorough and updated procedure for cannulating the coccygeal vein into the caudal vena cava for the collection of serial blood samples at or near the site of uterine-ovarian drainage. Concentrations of progesterone were quantified in cows of different reproductive tract sizes with an active corpus luteum to assess the distance for proper catheter placement compared with circulating concentrations collected from the jugular vein. This procedure has a low risk for side effects, can be used effectively in pregnant animals with no major consequence to the viability of the pregnancy, and provides means for frequent collections up to 12 d., (© The Author(s) 2021. The study of reproductive physiology in domestic ruminants has progressed from the whole animal to the molecular level in an amazingly short period of time. The volume of information on this subject is enormous; therefore, we have focused on domestic ruminants, with an emphasis on cattle. To date, artificial insemination (AI) is perhaps the most powerful technique that reproductive physiologists and geneticists have provided the livestock industry for genetic improvement. Early efforts to establish AI as a tool were initiated in Russia around 1899 and since that time major advances in methods of semen collection, evaluation of male fertility, cryopreservation of sperm, sex-sorted semen, and estrous cycle control have occurred. The preceding advances not only led to the widespread use of AI, but also contributed to our fundamental understanding of ovulation control, timing of insemination, gamete biology, and cryopreservation. In regards to anestrus, our understanding of the concept of neuroendocrine control of the pituitary gland and the role of steroid feedback led to the Gonadostat Theory, which proposes that onset of puberty is due to a decrease in the negative feedback of gonadal steroids over time. Subsequent studies in prepuberal and postpartum sheep and cattle established that a short luteal phase frequently precedes the first normal length cycle that is accompanied by estrous expression. This observation led to the common practice of treating prepuberal heifers and anestrous postpartum cows with a short-term progestin treatment (e.g., Controlled Internal Drug Release) to induce normal estrous cycles. In domestic ruminants, fertilization rate is high (85% to 95%); however, significant embryonic mortality before or around the time of maternal recognition of pregnancy (MRP) reduces the pregnancy rate to a single breeding. Significant effort has been directed at determining the time of MRP, the signal for MRP, as well as elucidating the physiological, cellular, and molecular dialogue between the conceptus and uterine environment. Advancements have now led us to the ability to edit the genome to alleviate disease and possibly improve production traits. In summary, major advancements in our understanding of reproductive biology have stemmed from efforts to establish the AI and embryo transfer technique and reduce the negative impact of anestrus and embryonic mortality in domestic ruminants.

Programs in animal science are particularly well suited for graduate education because students can receive comprehensive training in the laboratory as well as with the whole animal. Furthermore, graduate students in animal science have the opportunity to understand how their research relates to a real world problem. Graduate students need to take ownership of their education by identifying training goals, choosing a mentor who will help them achieve their goals, and becoming engaged in research as soon as possible. In my own graduate program, I emphasize concepts more than techniques and I believe that graduate course work should focus on the basic areas of science that underlie reproductive biology (e.g., endocrinology, biochemistry, physiology, immunology, and statistics). Based on the increase in technology available for scientific investigation and the diversity of expertise required to address important research problems, graduate students need to learn the importance of establishing productive collaborations and begin building a scientific network. Preparation for graduate school frequently begins early with a curiosity and passion for understanding how biology works. Undergraduate courses can facilitate scientific thinking by providing opportunities in lectures and laboratories for students to transition from passive learners to thinking of themselves as animal scientists. There is a profound difference between individuals who view themselves as practitioners of a discipline and those who are simply trying to complete a course requirement. Teachers of undergraduate courses should incorporate experiential learning exercises into their lectures and laboratories to provide undergraduate students the opportunity to function as animal scientists and to embrace their scientific education. Graduate training has been the most enjoyable aspect of my career and it has been a joy to witness the achievements of students following completion of their degree!

Two experiments were designed to test the hypothesis that pregnancy rates after fixed-time artificial insemination (FTAI) in beef heifers and cows may be improved by delaying insemination of females that have not expressed estrus before FTAI. In Exp. 1, estrus was synchronized for 931 heifers across 3 locations using the 14-d CIDR-PG protocol (controlled internal drug-release [CIDR] insert [1.38 gm progesterone] on d 0 with removal of CIDR insert on d 14; 25 mg PGF2α 16 d after CIDR insert removal on d 30; and 100 μg GnRH on d 33, 66 h after PGF2α). Estrous detection aids (Estrotect) were applied at PGF2α on d 30, and estrous expression was recorded at GnRH on d 33. Heifers within each location were randomly assigned to 1 of 2 treatments based on weight and reproductive tract score (RTS): 1) FTAI (concurrent with GnRH, 66 h after PGF2α) regardless of estrous expression or 2) FTAI for heifers expressing estrus and delayed AI (20 h after GnRH) for heifers failing to express estrus. Heifers assigned to treatment 2 achieved a higher AI pregnancy rate than heifers assigned to treatment 1 (54 versus 46%; P = 0.01). The observed increase in AI pregnancy rate is attributed to the delayed AI of non-estrous heifers in treatment 2, as AI pregnancy rates for non-estrous heifers were significantly higher for treatment 2 (49 versus 34%; P = 0.02), while AI pregnancy rates of estrous heifers did not differ by treatment (P = 0.24). In Exp. 2, estrus was synchronized for 951 mature, suckled cows across 9 locations using the 7-d CO-Synch + CIDR protocol (100 μg GnRH + CIDR insert [1.38 gm progesterone] on d 0; 25 mg PGF2α at CIDR insert removal on d 7; and 100 μg GnRH on d 10, 66 h after CIDR insert removal). Estrus detection aids (Estrotect) were applied at PGF2α and CIDR insert removal on d 7, and estrous expression was recorded at GnRH on d 10. Cows within each location were assigned to 1 of 2 treatments based on age, days postpartum, and BCS: 1) FTAI (concurrent with GnRH, 66 h after PGF2α) regardless of estrous expression or 2) FTAI for cows expressing estrus and delayed AI (20 h after GnRH) for cows failing to express estrus. No significant effect of treatment was found on AI pregnancy rate (P = 0.76). In summary, FTAI pregnancy rates in heifers can be improved through a strategy of "split-time" AI. However, a statistically significant increase was not observed in the pregnancy rates of mature suckled cows when using a similar strategy.

This experiment compared 2 long-term controlled internal drug release (CIDR)-based protocols to synchronize estrus before fixed-time artificial insemination (FTAI) in postpartum beef cows. Cows were assigned to treatments by age, BCS, and days postpartum. Cows assigned to the 14- to 19-d CIDR-PGF2α protocol (n = 196) received CIDR inserts (1.38 g progesterone [P4]) from d 0 to 14 and PGF2α (25 mg, i.m.) 19 d after CIDR removal on d 33. Cows assigned to the 14-to-16-d CIDR-PGF2α protocol (n = 195) received CIDR inserts from d 3 to 17 and PGF2α 16 d after CIDR removal on d 33. Cows were artificially inseminated on d 36, 72 h after PGF2α, with GnRH (100 μg, i.m.) at FTAI. Cows were exposed for natural service 14 d after FTAI for 75 d. Blood samples for P4 were collected at d -10 and 0 to determine pretreatment estrous cyclicity status and again at PGF2α. Blood samples for estradiol (E2) were collected at PGF2α and FTAI. HeatWatch estrus detection transmitters were used from CIDR removal until FTAI to determine onset of estrus after CIDR removal and PGF2α. Dominant follicle diameter was determined at PGF2α and FTAI. Pregnancy diagnosis was performed 70 d after FTAI and confirmed at d 140 of gestation. Estrous response after CIDR removal was similar between treatments. Cows in both treatments had similar size dominant follicles on d 33 at PGF2α and d 36 at FTAI. Progesterone at PGF2α was greater (P = 0.03) for 14-to-16-d compared to 14-to-19-d treated cows. Mean concentrations of E2 at PGF2α were similar between treatments but were greater (P = 0.01) at FTAI for 14-to-16-d compared to 14-to-19-d treated cows. Estrous response after PGF2α was greater (P < 0.01) for 14-to-19-d compared to 14-to-16-d treated cows (47.4 vs. 29.7%, respectively). Pregnancy rate resulting from FTAI was affected by the treatment × age group interaction (P = 0.08). Pregnancy rate after FTAI among cows ≥ 4 yr tended to be greater (P = 0.06) for 14-to-19-d compared to the 14-to-16-d treated cows, suggesting that the 14-to-19-d schedule works better for older age cows compared with the 14-to-16-d schedule. Final pregnancy rates were similar between the 2 treatments. In summary, these data indicate that a range in intervals from CIDR removal to PGF2α may be feasible when using long-term CIDR-based protocols in cows and raise questions that warrant further study regarding the benefits of extending this interval based on cow age.

This experiment was designed to test the hypothesis that delayed insemination of nonestrous cows would increase pregnancy rates when using sex-sorted semen in conjunction with fixed-time artificial insemination (FTAI). Estrus was synchronized for 656 suckled beef cows with the 7-d CO-Synch + controlled internal drug release (CIDR) protocol (100 μg GnRH + CIDR [1.38 g progesterone] on d 0, 25 mg PGF2α at CIDR removal on d 7, and 100 μg GnRH on d 10, 66 h after CIDR removal). Estrus detection aids (Estrotect) were applied at PGF2α and CIDR removal on d 7, and estrous expression was recorded at GnRH on d 10. Cows were assigned to 1 of 3 treatments: 1) FTAI (concurrent with GnRH, 66 h after CIDR removal) with conventional semen regardless of estrous expression, 2) FTAI with sex-sorted semen regardless of estrous expression, or 3) FTAI with sex-sorted semen for cows having expressed estrus and delayed AI 20 h after final GnRH for cows failing to express estrus. A treatment × estrous expression interaction was found (P < 0.0001). Higher pregnancy rates (P < 0.0001) were achieved with conventional semen (Treatment 1; 77%) than with sex-sorted semen (Treatments 2 and 3; 51 and 42%, respectively) among cows that expressed estrus. However, among cows that failed to express estrus, delayed insemination with sex-sorted semen yielded higher (P < 0.0001) pregnancy rates than with sex-sorted semen at the standard time (Treatments 2 and 3; 3 versus 36%, respectively). Furthermore, among cows that failed to express estrus, FTAI pregnancy rates when using sex-sorted semen at the delayed time (36%) were comparable (P = 0.9) to those achieved using conventional semen at the standard time (Treatment 1; 37%). These results indicate that delaying AI of nonestrous cows by 20 h from the standard FTAI improves pregnancy rates when sex-sorted semen is used with FTAI.

The objective was to examine the relationship between ovulatory follicle size and embryo and fetal survival by using circulating concentrations of bovine pregnancy associated glycoproteins (bPAG) to detect the presence of an embryo or fetus and monitor placental function. Before examining the relationship between bPAG, ovulatory follicle size, and embryo and fetal survival, the half-life of bPAG was determined in Exp. 1. The half-life of bPAG after PGF2α-induced abortion on d 32 to 36 postinsemination was 35.8 ± 21.9 h (mean ± SD; range 7.1 to 78.5 h). In Exp. 2, suckled beef cows (n = 91) were treated with the CO-Synch protocol (GnRH on d -9, PGF2α on d -2, and GnRH and AI 48 h later [d 0]) and classified into 1 of 2 ovulatory follicle size groups: 1) small follicle (<12.5 mm; n = 25) or 2) large follicle (≥ 12.5 mm; n = 66). The first increase (P < 0.0001) in serum bPAG occurred in pregnant cows on d 24 after insemination and circulating bPAG decreased before a decrease in progesterone in 3 of 4 cows that lost an embryo or fetus. Pattern of secretion of bPAG in serum from d 24 to 60 after insemination (d 0) was affected by day (P < 0.0001), but not ovulatory follicle size. In Exp. 3, suckled beef cows (n = 1164) were administered the CO-Synch protocol either with (donor cows; n = 810) or without (recipient cows; n = 354) AI on d 0. Single embryos (n = 394) or oocytes (n = 45) were recovered from the donor cows [d 7; embryo transfer (ET)] and all live embryos were transferred into recipients the same day. Cows were classified on d 0 as having a small (<12.5 mm) or large (≥ 12.5 mm) ovulatory follicle, and randomly chosen as donors or recipients to remove confounding effects of ovulatory follicle size on fertility. Serum concentration of bPAG at d 28 was not affected by ovulatory follicle size (P = 0.85), embryo stage at ET (P = 0.75), embryo quality at ET (P = 0.64), estradiol at GnRH2 (P = 0.62) or serum progesterone at ET (d7; P = 0.14). Compared with cows that maintained pregnancy (n = 176), cows that exhibited late embryonic or fetal mortality (n = 19) after d 28 had decreased (P < 0.05) concentrations of bPAG on d 28. In summary, there was no relationship between serum bPAG and ovulatory follicle size or embryo stage or quality at ET; however, cows that lost an embryo after d 28 had reduced concentrations of bPAG on d 28 compared with cows that maintained pregnancy.

Two experiments were conducted to examine the necessity of adding a GnRH injection to a 14-d controlled internal drug release (CIDR)-based protocol for synchronization of estrus and ovulation in postpartum beef cows. The experiments were designed to characterize long-term CIDR-based protocols in cyclic and noncyclic postpartum beef cows on the basis of estrous response, follicular dynamics, and serum steroid hormone concentrations. In Exp. 1 and 2, crossbred lactating beef cows (n = 40 and 38, respectively) were randomly assigned to 1 of 2 treatments by age, days postpartum (DPP), BCS, and estrous cyclicity status: 1) cows received a CIDR from d 0 to 14 followed by GnRH 9 d after CIDR removal (d 23) and PGF2α on d 30 (CIDR Select) or 2) CIDR administration from d 0 to 14 followed by PGF2α 16 d later (d 30; Show-Me-Synch). Estrus detection was performed using HeatWatch transmitters applied from CIDR removal to AI. Cows in Exp. 1 were artificially inseminated based on detected estrus whereas cows in Exp. 2 were inseminated at a fixed time. In both experiments, follicle turnover on d 25 of treatment was greater among CIDR Select-treated cows (P < 0.001) compared with Show-Me-Synch-treated cows. In Exp. 1, CIDR Select-treated cows tended to have a reduced (P = 0.06) variance for the interval to estrus after PGF2α than Show-Me-Synch-treated cows. Also, cows assigned to the CIDR Select protocol had greater concentrations of progesterone (P < 0.05) on the day before PGF2α administration as well as greater concentrations of estradiol-17β (P < 0.01) 48 h after PGF2α administration. In Exp. 2, mean dominant follicle diameter on d 23 and at fixed-time AI (FTAI) did not differ between treatments (P > 0.10), but Show-Me-Synch-treated cows had larger follicles at d 28 (P < 0.001) and tended to have larger follicles at PGF2α (d 30; P = 0.06) compared with cows assigned to CIDR Select. In summary, the administration of GnRH on d 23 of a long-term CIDR-based estrus synchronization protocol increased follicle turnover; however, both long-term CIDR-based protocols yielded similar physiological outcomes among estrous-cycling and anestrous postpartum beef cows.

Reproductive failure in livestock can result from failure to fertilize the oocyte or embryonic loss during gestation. Although fertilization failure occurs, embryonic mortality represents a greater contribution to reproductive failure. Reproductive success varies among species and production goals but is measured as a binomial trait (i.e., pregnancy), derived by the success or failure of multiple biological steps. This review focuses primarily on follicular characteristics affecting oocyte quality, fertilization, and embryonic health that lead to pregnancy establishment in beef cattle. When estrous cycles are manipulated with assisted reproductive technologies and ovulation is induced, duration of proestrus (i.e., interval from induced luteolysis to induced ovulation), ovulatory follicle growth rate, and ovulatory follicle size are factors that affect the maturation of the follicle and oocyte at induced ovulation. The most critical maturational component of the ovulatory follicle is the production of sufficient estradiol to prepare follicular cells for luteinization and progesterone synthesis and prepare the uterus for pregnancy. The exact roles of estradiol in oocyte maturation remain unclear, but cows that have lesser serum concentrations of estradiol have decreased fertilization rates and decreased embryo survival on d 7 after induced ovulation. When length of proestrus is held constant, perhaps the most practical follicular measure of fertility is ovulatory follicle size because it is an easily measured attribute of the follicle that is highly associated with its ability to produce estradiol.

In postpartum beef cows, GnRH-induced ovulation of small dominant follicles decreased pregnancy rates and increased late embryonic/fetal mortality. In Exp. 1, single ovulation reciprocal embryo transfer (ET) was used to examine the relationship between preovulatory serum concentrations of estradiol at GnRH-induced ovulation in donor and recipient cows and establishment and maintenance of pregnancy. Suckled beef cows (n = 1,164) were administered GnRH (GnRH1, 100 μg) on d -9 (GnRH1), PGF(2α) on d -2, and GnRH2 (GnRH2, 100 μg) on d 0 (CO-Synch protocol) either with (donors; n = 810) or without (recipients; n = 354) AI. Single embryos (n = 394) or oocytes (n = 45) were recovered from the donor cows (d 7; ET) and all live embryos were transferred into recipients. Serum concentration of estradiol at GnRH2 was positively correlated with follicle size at GnRH2 (r = 0.45, P < 0.01) and progesterone at ET (r =0.34, P < 0.01). Donor cows with greater estradiol at GnRH2 were more likely to yield an embryo than an unfertilized oocyte (P < 0.01). Donor and recipient cows were retrospectively divided into 4 groups [low estradiol (<8.4 pg/mL) or high estradiol (≥8.4 pg/mL)] based on serum concentration of estradiol at GnRH2. Pregnancy rate at d 27 for low-low (n = 78), low-high (n = 80), high-low (n = 91), and high-high (n = 101) groups (donor-recipient, respectively) was 45, 65, 43, and 61% respectively (P < 0.02). Because recipient cows with greater estradiol concentration at GnRH2 had greater pregnancy rates in Exp. 1, the objective of Exp. 2 was to evaluate the effect of estradiol supplementation on pregnancy rate. Ovulation was synchronized in suckled beef cows (n = 600) using the CO-Synch protocol with the insertion of a controlled internal drug release (CIDR; intravaginal progesterone supplement) from d -9 until d -2. Approximately one-half of the cows (n = 297) received an injection of estradiol cypionate (ECP; 0.5 mg intramuscularly) 24 h before AI. Compared with the no treatment (Control) cows, ECP treatment increased (P < 0.01) pregnancy rates of cows induced to ovulate smaller dominant follicles (<12.2 mm). In conclusion, GnRH-induced ovulation of small dominant follicles was associated with reduced serum estradiol, fertilization rate (donor cows), and pregnancy establishment (recipient cows). Furthermore, ECP supplementation during the preovulatory period increased pregnancy rates in cows induced to ovulate smaller dominant follicles.

A single ovulation, reciprocal embryo transfer study was used to investigate effects of oocyte competence and maternal environment on pregnancy establishment and maintenance in beef cows. Estrous cycles were synchronized in suckled beef cows and embryo donors were inseminated on d 0 (n = 810). Cows were classified on d 0 as having a small (<12.5 mm) or large (≥12.5 mm) ovulatory follicle and randomly chosen as donors or recipients to remove confounding effects of ovulatory follicle size on fertility. Embryos (n = 393) or oocytes (n = 44) were recovered on d 7, and all viable embryos were transferred into recipients (n = 354). All statistical analyses were conducted using the GLM procedure of SAS. Path analysis (with significance set at P < 0.10) was used to examine potential cause-effect relationships among the measured variables. Greater donor cow BW, circulating estradiol concentration at insemination, postpartum interval, and ovulatory follicle size directly increased (P < 0.10) fertilization success. Greater donor cow age was the only factor that directly decreased (P < 0.10) fertilization success. Viability of d-7 embryos was directly inhibited (P < 0.10) by rapid follicular growth rate from d -2 to 0 and heavier BW. Direct beneficial effects to embryo viability were increased serum progesterone concentration on d -2 and ovulatory follicle size. Pregnancy maintenance from d 7 to 27 was enhanced (P < 0.10) by increased serum estradiol concentration on d 0 and progesterone concentration on d 7 in the recipient cow. Increased follicular diameter in the recipient cow on d 0 was detrimental to pregnancy maintenance from d 7 to 27. This manuscript defines the complex interplay and relative contributions of endocrine and physical factors both prior and subsequent to fertilization that influence both oocyte competence and maternal environment and their roles in establishment and maintenance of pregnancy.

Two experiments were conducted to compare pregnancy rates resulting from fixed-time AI (FTAI) after administration of 1 of 2 long-term controlled internal drug release (CIDR)-based protocols. Heifers were assigned to treatment by age, BW, and pubertal status. The CIDR Select-treated heifers (Exp. 1, n = 37; Exp. 2, n = 192) received a CIDR (1.38 g of progesterone) from d 0 to 14, followed by 100 µg of GnRH, intramuscularly (i.m.) 9 d after CIDR removal (d 23) and PGF(2α) (25 mg, i.m.) 7 d after GnRH treatment (d 30). Heifers assigned to the Show-Me-Synch protocol (Exp. 1, n = 40; Exp. 2, n = 200) received a CIDR from d 0 to 14, followed by PGF(2α) 16 d later (d 30). Artificial insemination was performed at 72 or 66 h after PGF(2α) treatment for the CIDR Select- and Show-Me-Synch-treated heifers, respectively, and each heifer was given GnRH (100 µg, i.m.) at the time of AI. In Exp. 1, ovaries of each heifer were examined by transrectal ultrasonography on d 23 and 30 to characterize follicular dynamics. Follicles ≥5 mm and the presence of corpora lutea were recorded. On d 25, ovaries of each heifer were examined to characterize the status of dominant follicles recorded on d 23. Heifers were fitted with HeatWatch (DDx Inc., Denver, CO) estrus-detection transmitters at PGF(2α) to characterize estrus distribution up to FTAI. The diameter of dominant follicles on d 23 at PGF(2α) and on d 30, and the estrous response after PGF(2α) treatment up to the point of FTAI did not differ between CIDR Select- and Show-Me-Synch-treated heifers. Concentrations of progesterone in serum at PGF(2α) were greater (P = 0.07) in Show-Me-Synch- than CIDR Select-treated heifers (6.0 vs. 4.8 ng/mL, respectively). Pregnancy rates of heifers resulting from FTAI did not differ (P = 0.33) between CIDR Select- and Show-Me-Synch-treated heifers (CIDR Select, 59%; Show-Me-Synch, 70%). In Exp. 2, FTAI pregnancy rates tended (P = 0.07) to be greater in Show-Me-Synch-treated (62%) than in CIDR Select-treated (51%) heifers. Pregnancy rates at the end of the breeding season did not differ (P = 0.72; CIDR Select, 85%; Show-Me-Synch, 83%) between treatments. In summary, pregnancy rates resulting from FTAI were comparable for heifers assigned to each of the 2 long-term progestin-based protocols. The reduced treatment cost and animal handling associated with administration of the Show-Me-Synch protocol offer distinct advantages over the CIDR Select protocol despite similarities in pregnancy rates resulting from FTAI.

Two experiments evaluated long-term progestin-based estrus-synchronization programs on the basis of potential for use in facilitating fixed-time AI in estrous cycling and prepubertal beef heifers. In Exp. 1, heifers were assigned to 1 of 2 treatments by age, BW, and estrous cyclicity status. Heifers assigned to the melengestrol acetate-PGF(2α) protocol (MGA-PG; n = 50) received MGA (0.5 mg·animal(-1)·d(-1)) in a 1.0-kg carrier from d 0 to 13 and were administered PGF(2α) (25 mg, intramuscularly) 19 d after MGA withdrawal (d 32). Heifers assigned to the Show-Me-Synch protocol (n = 49) received a controlled internal drug release (CIDR) insert (1.38 g of progesterone) from d 2 to 16 followed by PGF(2α) administration 16 d after CIDR removal (d 32). All heifers were fitted with HeatWatch estrus-detection transmitters at the time of progestin removal for continuous estrus detection through the synchronized period after PGF(2α). In Exp. 2, heifers (n = 396) were assigned to the same 2 treatments described in Exp. 1 by age, BW, and reproductive tract score. Heifers in Exp. 2, however, were fitted with HeatWatch estrus-detection transmitters at PGF(2α) to characterize estrus-distribution patterns during the synchronized period after PGF(2α). Heifers in both experiments were inseminated approximately 12 h after the onset of estrus. In Exp. 1, estrous response after PGF(2α) and mean interval to estrus after PGF(2α) did not differ between MGA-PG and Show-Me-Synch treatments (P = 0.97). The variance for interval to estrus after PGF(2α) tended (P = 0.06) to be reduced among MGA-PG-treated heifers compared with Show-Me-Synch-treated heifers. Conception to AI, AI pregnancy, and final pregnancy rates did not differ (P > 0.1) between treatments. In Exp. 2, estrous response after PGF(2α) was greater (P = 0.01) among Show-Me-Synch-treated heifers (92%) compared with MGA-PG-treated heifers (85%); however, mean interval to estrus after PGF(2α) did not differ (P = 0.74) between MGA-PG (57.4 ± 2.5 h) and Show-Me-Synch (56.2 ± 2.5 h) treatments. The variance for interval to estrus after PGF(2α) was reduced (P < 0.01) among Show-Me-Synch-treated vs. MGA-PG-treated heifers. Conception to AI, AI pregnancy, and final pregnancy rates did not differ (P > 0.1) between treatments. In summary, the Show-Me-Synch protocol compared favorably with the MGA-PG protocol on the basis of estrous response, synchrony of estrus, and resulting fertility after treatment administration.

Cows induced to ovulate small dominant follicles were reported to have reduced pregnancy rates compared with cows that ovulated large follicles. The reason for the presence of small dominant follicles at the time of GnRH-induced ovulation in timed AI protocols is unknown. The objectives of this experiment were to examine the role of day of the estrous cycle at initiation of treatment on ovulation after the first GnRH injection (GnRH1) and associated effects on growth rate and final size of the ovulatory follicle at the second GnRH injection (GnRH2), serum concentrations of estradiol at GnRH2, and subsequent luteal concentrations of progesterone in suckled beef cows. Estrous cycles of cows were manipulated to be at 1 of 5 specific days of the cycle (d 2, 5, 9, 13, and 18, d 0 = estrus; n = 12 per treatment group) at the beginning of the CO-Synch protocol (GnRH1 on d -9, PGF(2alpha) on d -2, and GnRH2 on d 0). Day of the estrous cycle at GnRH1 did not affect the size of the preovulatory follicle or the proportion of cows ovulating at GnRH2 (P = 0.65 and 0.21, respectively). When all cows were included in the analysis, cows that ovulated after GnRH1 had similar follicle size at GnRH2 compared with cows that did not ovulate after GnRH1 (11.4 and 10.4 mm, respectively; P = 0.23). When only cows that could ovulate after GnRH1 (excluding cows treated on d 2) were included in the analysis, cows that ovulated to GnRH1 had a larger follicle at GnRH2 than cows that did not ovulate after GnRH1 (11.4 and 9.5 mm, respectively; P = 0.04). Follicle growth from d -5 to 0 was similar between cows that ovulated after GnRH1 and cows that did not (1.01 vs. 0.89 mm/d, respectively; P = 0.75). There was a tendency for faster follicle growth rate in cows that ovulated a large follicle (>11 mm) compared with cows that ovulated a small follicle (< or = 11 mm; 1.01 vs. 0.86 mm/d, respectively; P = 0.07). Serum concentrations of estradiol at GnRH2 and progesterone after ovulation were reduced in cows that ovulated small follicles compared with cows that ovulated large follicles (P = 0.006 and 0.005, respectively). In summary, day of the estrous cycle at initiation of synchronization did not affect ovulatory follicle size, but follicle growth rates affected the size of the follicle at GnRH2. Cows that ovulated a small follicle had reduced serum concentrations of estradiol at GnRH2 and progesterone after ovulation.

There is large variation in dominant follicle diameter at the time of GnRH-induced ovulation in the CO-Synch protocol [a first GnRH injection on d -9 (GnRH1), followed by PGF(2alpha) on d -2, and a second GnRH injection (GnRH2) with timed AI on d 0], and the reason for the presence of small dominant follicles at GnRH2 is not known. Our hypothesis was that ovulatory response to GnRH1 and progesterone exposure [controlled intravaginal drug-releasing insert (CIDR; EAZI-Breed, Pfizer Animal Health, New York, NY)] would affect ovulatory follicle size at GnRH2 in anestrous cows. This study used a 2 x 2 factorial arrangement of treatments in which anestrous suckled beef cows (n = 55) either ovulated (Ov1+) or failed to ovulate (Ov1-) after GnRH1 and either received (CIDR+) or did not receive (CIDR-) a 7-d CIDR treatment (from GnRH1 to PGF(2alpha)), resulting in the following treatment groups: Ov1+CIDR+, Ov1-CIDR+, Ov1+CIDR-, and Ov1-CIDR- (n = 9, 17, 11, and 18, respectively). The Ov1+ cows had larger follicles at GnRH2 (12.3 vs. 11.0 mm; P = 0.04), a decreased proportion of small follicles within cows that ovulated to GnRH2 (2/16 vs. 14/23; P = 0.003), and a similar growth rate of the ovulatory follicle from d -5 to 0 (d 0 = GnRH2; 1.1 +/- 0.06 vs. 1.1 +/- 0.07 mm/d; P = 0.99) compared with Ov1- cows. Administration of a CIDR had no effect on follicle diameter at GnRH2 (11.8 vs. 11.2 mm; P = 0.3), proportion of small ovulatory follicles at GnRH2 (7/19 vs. 9/20; P = 0.6), and follicular growth rate from d -5 to 0 (d 0 = GnRH2; 1.2 +/- 0.07 vs. 1.1 +/- 0.07 mm/d; P = 0.76). Administration of a CIDR, but not ovulation to GnRH1, increased follicle growth from d -2 to 0 (d 0 = GnRH2; P = 0.03 and 0.9, respectively). Large follicles (>11 mm) had a similar growth rate from d -5 to 0 (d 0 = GnRH2; P = 0.44) compared with small follicles (1.1 +/- 0.07 vs. 1.2 +/- 0.07 mm/d), but the large ovulatory follicles were larger at d -5 compared with small ovulatory follicles (P < 0.001). Follicle diameter was positively correlated with serum concentrations of estradiol at GnRH2 (r = 0.622; P < 0.0001). In summary, ovulation to GnRH1, but not CIDR administration, resulted in increased dominant follicle diameter at GnRH2 in anestrous suckled beef cows. Large follicles were already larger 5 d before GnRH2 but grew at a rate similar to small follicles; follicle size was positively correlated with serum concentrations of estradiol at the time of GnRH-induced ovulation.

Two experiments evaluated short-term controlled internal drug release (CIDR) insert-based protocols to synchronize estrus and ovulation and compare differences in their potential to facilitate fixed-time AI (FTAI) in postpartum beef cows. Experiment 1 was designed to compare the 7- and 5-d Select Synch + CIDR protocols on the basis of timing and synchrony of estrus after treatment. Cows assigned to the 7-d protocol (n = 59) received GnRH [100 microg intramuscularly (i.m.)] and CIDR inserts (1.38 g of progesterone) on d 0 and PGF(2alpha) (25 mg i.m.) and CIDR removal on d 7. Cows assigned to the 5-d protocol (n = 58) received GnRH and CIDR inserts on d 2, PGF(2alpha) and CIDR removal on d 7, and a second injection of PGF(2alpha) 12 h after CIDR removal. Estrus detection and AI were performed for cows assigned to each protocol during the 144-h synchronized period. There was no difference in estrous response (P = 0.85), interval to estrus (P = 0.09), or variance for interval to estrus (P = 0.75) between treatments, nor were there differences in synchronized conception or pregnancy rates resulting from AI (P = 0.85, P = 0.91, respectively). Experiment 2 was designed to compare pregnancy rates resulting from FTAI after administration of the 7- and 5-d CO-Synch + CIDR protocols. Both treatments were administered the same as in Exp. 1; however, cows assigned to the 7-d protocol were inseminated 66 h after PGF(2alpha) and CIDR removal, and cows assigned to the 5-d protocol were inseminated 72 h after the first PGF(2alpha) injection. Cows assigned to both protocols were administered GnRH (100 microg i.m.) at AI. There was no effect of treatment (P = 0.85), technician (P = 0.20), or sire (P = 0.25) on pregnancy rates resulting from FTAI. Given these observations, the 5-d protocol provides an effective alternative to the 7-d protocol for use in facilitating FTAI; however, beef producers must carefully consider the increased labor and treatment costs associated with the 5-d protocol.

The objective of the experiment was to examine the necessity of adding a GnRH injection to a 14-d controlled internal drug release (CIDR)-based protocol for synchronization of estrus in beef heifers that were prepubertal or estrous-cycling at the initiation of treatment. The hypothesis tested was that the addition of GnRH in a CIDR-based estrus synchronization protocol would increase the synchrony of estrus after PGF(2alpha) (PG). Beef heifers (n = 285) were assigned to 1 of 2 treatments within reproductive tract scores (2 or 3 = prepubertal; 4 or 5 = estrous-cycling) by age and BW. Heifers assigned to CIDR Select received a CIDR insert (1.38 g of progesterone) from d 0 to 14 followed by GnRH (100 microg, intramuscularly) on d 23 and PG (25 mg intramuscularly) on d 30. Heifers assigned to CIDR-PG received a CIDR insert from d 0 to 14 and PG on d 30. Heifers were fitted with a HeatWatch estrus detection system transmitter at the time of PG administration for continuous estrus detection during the synchronized period (0 to 144 h after PG); AI was performed 12 h after estrus onset. Estrous response did not differ (P = 0.43) between treatments (94% CIDR Select, 98% CIDR-PG). Mean interval to estrus after PG was 7 h shorter (P = 0.01) and variance for interval to estrus was reduced (P < 0.01) among CIDR-PG-treated compared with CIDR Select-treated heifers. Conception rate to AI tended (P = 0.09) to be greater for CIDR-PG heifers (67%) compared with CIDR Select heifers (58%), and AI pregnancy rate was greater (P = 0.05) for CIDR-PG heifers (66%) compared with CIDR Select heifers (55%). Final pregnancy rate at the end of the breeding season was similar for the 2 treatments (81% for both; P = 0.94). We conclude that the administration of GnRH 9 d after CIDR removal in the CIDR Select protocol is not required to facilitate an improvement in the synchrony of estrus in beef heifers.

The objective of the experiment was to compare follicular dynamics, ovulatory response to GnRH, and synchrony of estrus and ovulation among estrous-cycling and prepubertal beef heifers synchronized with a controlled internal drug-release (CIDR)- based or GnRH-PGF(2alpha) (PG) protocol. Estrous-cycling beef heifers were randomly assigned to 1 of 4 treatments (C1, C2, C3, C4), and prepubertal beef heifers were randomly assigned to 1 of 2 treatments (P1, P2) by age and BW. Blood samples were taken 10 and 1 d before treatment to confirm estrous cyclicity status (progesterone > or =0.5 ng/mL estrous cycling). The CIDR Select (C1, n = 12; P1, n = 14)-treated heifers received a CIDR insert (1.38 g of progesterone) from d 0 to 14, GnRH (100 microg, i.m.) on d 23, and PG (25 mg, i.m.) on d 30. Select Synch + CIDR (C2, n = 12; P2, n = 11)-treated heifers received a CIDR insert and GnRH on d 23 and PG at CIDR removal on d 30. The CIDR-PG (C3, n = 12)-treated heifers received a CIDR insert on d 23 and PG at CIDR removal on d 30. Select Synch (C4, n = 12)-treated heifers received GnRH on d 23 and PG on d 30. HeatWatch transmitters were fitted at CIDR removal (C1, C2, C3, P1, and P2) or at GnRH administration (C4) for estrus detection. Ultrasound was used to determine the response to GnRH and the timing of ovulation after estrus. Among the estrous-cycling heifers, ovulatory response to GnRH and estrous response did not differ (P > 0.05). Among the prepubertal heifers, more (P = 0.02) P1 heifers responded to GnRH than P2 heifers, but estrous response did not differ (P > 0.05). Among the estrous-cycling heifers, variance for interval to estrus after PG was reduced (P < 0.05) for C1 compared with each of the other treatments, and C3 [corrected] was reduced (P < 0.05) compared with C2 [corrected] Variance for interval to ovulation after PG was reduced (P < 0.05) for C1 compared with each of the other treatments. Among the prepubertal heifers, there was no difference (P > 0.05) in variance for interval to estrus or ovulation. Results from C1 and P1 (T1) and C2 and P2 (T2) were combined to compare T1 and T2 among mixed groups of estrous-cycling and prepubertal heifers. Response to GnRH was greater (P < 0.01; 81% T1 and 39% T2), and variances for interval to estrus and ovulation for T1 were reduced (P < 0.01) compared with T2. In summary, CIDR Select improved (P < 0.01) the synchrony of estrus and ovulation compared with Select Synch + CIDR.

This experiment was designed to compare pregnancy rates in postpartum beef cows resulting from fixed-time AI (FTAI) at 54 or 66 h after administration of the CO-Synch + controlled internal drug-release (CIDR) protocol. Cows (n = 851) at 2 locations over 2 yr (yr 1, n = 218 and 206; and yr 2, n = 199 and 228 at the 2 locations, respectively) were stratified by age, BCS, and days postpartum to 1 of 2 FTAI intervals. Cows were administered GnRH (100 mug, i.m.) and were equipped with a CIDR insert (1.38 g of progesterone) on d 0. Controlled internal drug-release inserts were removed 7 d later at the time PGF(2alpha) (25 mg, i.m.) was administered (d 7). Continuous estrus detection was performed at location 2 by using the HeatWatch Estrus Detection System; the transmitters were fitted at the time of PGF(2alpha) and removed at the time of AI. Artificial insemination was performed at predetermined fixed times [54 h (FTAI 54; n = 424) or 66 h (FTAI 66; n = 427) after PGF(2alpha)] and all cows were administered GnRH (100 mug, i.m.) at AI. Two blood samples were collected on d -10 or -8 and immediately before treatment initiation to determine the pretreatment estrous cyclicity status of cows [progesterone >/=0.5 ng/mL (FTAI 54, 288/424 = 68%; FTAI 66, 312/427 = 73%; P = 0.07)]. Pregnancy rates were greater (P < 0.01) among cows that exhibited estrus than among those that did not (123/163 = 76% and 150/270 = 56%, respectively). There were no treatment x location interactions within year (P > 0.10) for age, days postpartum, or BCS; thus, the results were pooled for the respective treatments. Pregnancy rates were greater for FTAI 66 than FTAI 54 (P = 0.05; 286/426 = 67% vs. 257/424 = 61%, respectively). Pregnancy rates resulting from FTAI did not differ between year (P = 0.09), farm (P = 0.80), AI sire (P = 0.11), or technician (P = 0.64). There was no difference between pregnancy rates resulting from FTAI based on pretreatment cyclicity status (P = 0.30), and there was no difference between treatments in final pregnancy rates (P = 0.77). In summary, pregnancy rates resulting from FTAI following CO-Synch + CIDR at 66 h were greater than those resulting from FTAI at 54 h.

Induced ovulation of small dominant follicles (SF, < 12 mm; CO-Synch protocol) in postpartum beef cows resulted in formation of corpora lutea (CL) that exhibited a delayed rise in progesterone (P4) compared with CL from large dominant follicles (LF, > 12 mm). Experiment 1 characterized P4 concentrations from ovulation to subsequent estrus among GnRH-induced or spontaneously ovulated SF (or= 12 mm) to determine if P4 secretion by CL formed from GnRH-induced SF remains lower postovulation in nonlactating beef cows. Nonlactating beef cows were induced to ovulate 48 h after PGF(2alpha) (CO-Synch; GnRH on d - 9, PGF(2alpha) on d - 2, and GnRH on d 0) or exhibited estrus and spontaneously ovulated after PGF(2alpha). Follicle size was measured at the second GnRH in cows induced to ovulate or approximately 3 h after the onset of estrus for cows that ovulated spontaneously. Cows were classified into 1 of 4 groups: 1) GnRH-induced ovulation-SF (or= 12 mm; Ind-LF; n = 16); 3) spontaneous ovulation-SF (or= 12 mm; Spon-LF; n = 22). Serum concentrations of P4 from d 3 to 15 were reduced in the Ind-SF compared with the Ind-LF (P = 0.05), Spon-SF (P = 0.07), and Spon-LF (P = 0.03). Experiment 2 characterized P4 concentrations (0 to 60 d postAI) among GnRH-induced or spontaneously ovulated SF (or= 13 mm) to determine if P4 secretion by CL formed from GnRH-induced SF remained lower during early gestation. Ovulation was induced with GnRH 48 h after PGF(2) (CO-Synch) or occurred spontaneously, and ovulatory follicle size was measured at AI. Lactating cows were classified into 1 of 3 groups: 1) GnRH-induced ovulation-SF (or= 13 mm; Ind-LF; n = 43); or 3) spontaneous ovulation-LF (>or= 13 mm; Spon-LF; n = 27). The increase in P4 concentrations was greater (P = 0.06) in pregnant (d 2 to 12) compared with nonpregnant cows. Also, the increase in P4 from d 2 to 12 was greater (P = 0.01) in the Ind-LF compared with the Ind-SF groups, but there was no difference (P = 0.94) among groups in P4 from d 14 to 60 in pregnant cows. Follicle size at AI influenced the increase in P4 in cows that failed to conceive (P = 0.007), but not among cows that became pregnant (P = 0.32) to AI. In summary, P4 secretion after GnRH-induced ovulation of SF was decreased from d 2 to 12 compared with that of LF, but was similar among pregnant cows from d 14 to 60 postAI (d 0).

The COSynch protocol has been used to synchronize ovulation and facilitate fixed-time AI in beef cattle. Establishment and maintenance of pregnancy was negatively affected, in previous studies, by GnRH-induced ovulation of small dominant follicles (/=10 mm) and increased ovulatory response after GnRH 2.

The objective of the experiment was to compare pregnancy rates resulting from fixed-time AI after administration of either 1 of 2 controlled internal drug release (CIDR)-based protocols. Heifers at 3 locations (location 1, n = 78; location 2, n = 61; and location 3, n = 78) were assigned to 1 of 2 treatments within reproductive tract scores (1 = immature to 5 = cycling) by age and BW. Heifers assigned to CIDR Select received a CIDR insert (1.38 g of progesterone) from d 0 to 14 followed by GnRH (100 mug, i.m.) 9 d after CIDR removal (d 23) and PGF2alpha (PG, 25 mg, i.m.) 7 d after GnRH treatment (d 30). Heifers assigned to CO-Synch + CIDR were administered GnRH and received a CIDR insert on d 23 and PG and CIDR removal on d 30. Heifers at location 1 were fitted with a HeatWatch estrus detection system transmitter from the time of PG until 24 d after fixed-time AI to allow for continuous estrus detection. Artificial insemination was performed at predetermined fixed times for heifers in both treatments at 72 or 54 h after PG for the CIDR Select and CO-Synch + CIDR groups, respectively. All heifers were administered GnRH at the time of AI. Blood samples were collected 10 d before and immediately before treatment initiation (d 0) to determine pretreatment estrous cyclicity (progesterone > or = 0.5 ng/mL). At location 1, the estrous response during the synchronized period was greater (P = 0.06; 87 vs. 69%, respectively), and the variance for interval to estrus after PG was reduced among CIDR Select- (P < 0.01) compared with CO-Synch + CIDR-treated heifers. Fixed-time AI pregnancy rates were significantly greater (P = 0.02) after the CIDR Select protocol (62%) compared with the CO-Synch + CIDR protocol (47%). In summary, the CIDR Select protocol resulted in a greater and more synchronous estrous response and significantly greater fixed-time AI pregnancy rates compared with the CO-Synch + CIDR protocol.

This experiment was designed to compare pregnancy rates in postpartum beef cows resulting from fixed-time AI (FTAI) after treatment with 1 of 2 protocols to synchronize estrus and ovulation. Cross-bred, suckled beef cows (n = 650) at 4 locations (n = 210; n = 158; n = 88; and n = 194) were assigned within a location to 1 of 2 protocols within age group by days postpartum and BCS. Cows assigned to the melengestrol acetate (MGA) Select treatment (MGA Select; n = 327) were fed MGA (0.5 mg x head(-1) x d(-1)) for 14 d, GnRH (100 microg of Cystorelin i.m.) was injected on d 26, and prostaglandin F2alpha (PG; 25 mg of Lutalyse i.m.) was injected on d 33. Cows assigned to the CO-Synch + controlled internal drug release (CIDR) protocol (CO-Synch + CIDR; n = 323) were fed a carrier for 14 d, were injected with GnRH and equipped with an EAZI-BREED CIDR insert (1.38 g of progesterone, Pfizer Animal Health, New York, NY) 12 d after carrier removal, and PG (25 mg of Lutalyse i.m.) was injected and the CIDR were removed on d 33. Fixed-time AI was performed at 72 or 66 h after PG for the MGA Select or CO-Synch + CIDR groups, respectively. All cows were injected with GnRH (100 microg of Cystorelin i.m.) at the time of insemination. Blood samples were collected 8 and 1 d before the beginning of MGA or carrier to determine estrous cyclicity status of the cows (estrous cycling vs. anestrus) before treatment [progesterone > or = 0.5 ng/mL (MGA Select, 185/327, 57%; CO-Synch + CIDR, 177/323, 55%); P = 0.65]. There was no difference (P = 0.20) in pregnancy rate to FTAI between treatments (MGA Select, 201/327, 61%; CO-Synch + CIDR, 214/323, 66%). There was also no difference (P = 0.25) between treatments in final pregnancy rate at the end of the breeding period (MGA Select, 305/327, 93%; CO-Synch + CIDR, 308/323, 95%). These data indicate that pregnancy rates to FTAI were comparable after administration of the MGA Select or CO-Synch + CIDR protocols. Both protocols provide opportunities for beef producers to utilize AI and potentially eliminate the need to detect estrus.

Previous research indicated that the size of the ovulatory follicle at the time of insemination significantly influenced pregnancy rates and embryonic/fetal mortality after fixed-timed AI in postpartum cows, but no effect on pregnancy rates was detected when cows ovulated spontaneously. Our objective was to evaluate relationships of fertility and embryonic/fetal mortality with preovulatory follicle size and circulating concentrations of estradiol after induced or spontaneous ovulation in beef heifers. Heifers were inseminated in 1 of 2 breeding groups: (1) timed insemination after an estrous synchronization and induced ovulation protocol (TAI n = 98); or (2) AI approximately 12 h after detection in standing estrus by electronic mount detectors during a 23-d breeding season (spontaneous ovulation; n = 110). Ovulatory follicle size at time of AI and pregnancy status 27, 41, 55, and 68 d after timed AI (d 0) were determined by transrectal ultrasonography. Only 6 heifers experienced late embryonic or early fetal mortality. Interactions between breeding groups and follicle size did not affect pregnancy rate (P = 0.13). Pooled across breeding groups, logistic regression of pregnancy rate on follicle size was curvilinear (P < 0.01) and indicated a predicted maximum pregnancy rate of 68.0 +/- 4.9% at a follicle size of 12.8 mm. Ovulation of follicles < 10.7 mm or > 15.7 mm was less likely (P < 0.05) to support pregnancy than follicles that were 12.8 mm. Ovulatory follicles < 10.7 mm were more prevalent (28% of heifers) than ovulatory follicles > 15.7 mm (4%). Heifers exhibiting standing estrus within 24 h of timed AI had greater (P < 0.01) follicle diameter (12.2 +/- 0.2 mm vs. 11.1 +/- 0.3 mm) and concentrations of estradiol (9.9 +/- 0.6 vs. 6.6 +/- 0.7) and pregnancy rates (63% vs. 20%) than contemporaries that did not exhibit behavioral estrus. However, when differences in ovulatory follicle size were accounted for, pregnancy rates were independent of expression of behavioral estrus or circulating concentration of estradiol. Therefore, the effects of serum concentrations of estradiol and behavioral estrus on pregnancy rate appear to be mediated through ovulatory follicle size, and management practices that optimize ovulatory follicle size may improve fertility.

Students in genetics and reproductive physiology, junior-/senior-level courses with enrollments averaging 76 and 41 students, respectively, were encouraged to read popular science books for extra credit. The objectives of the readings were to reinforce basic class concepts by forming ties to everyday life, and to expose students to a genre that can provide information to prepare them as citizens to engage in the debate over current issues. The books for genetics were The Double Helix, Genome, Voyage of the Beagle, and The Engineer in the Garden; and the books for reproductive physiology were Clone, Lives of a Cell, Life Before Birth and A Time to be Born, and The Second Creation: Dolly and The Age of Biological Control. To earn credit (3% of the course grade for each of up to three books), students had to demonstrate knowledge and understanding of each book during a 15-min discussion with the instructor. Discussions focused on questions designed to stimulate critical thinking about each book's content. For example: "There is approximately 98% homology in DNA sequence between humans and chimpanzees; in aspects ranging from basic physiology to behavior, what makes us similar to or different from the species that is our closest living relative?" The numbers of students reading three books in Years 1 and 2 were 2 and 12 in genetics, and 4 and 3 in reproductive physiology; those reading two books in Years 1 and 2 were 5 and 20 in genetics, and 0 and 2 in reproductive physiology; and those reading one book in Years 1 and 2 were 21 and 31 in genetics, and 7 and 8 in reproductive physiology. The numbers of students that read no books in Years 1 and 2 were 33 and 27 in genetics, and 45 and 20 in reproductive physiology. Participation in the reading project occurred independently of course grades. Sixteen genetics students from the second year's class, eight that had participated in the extra credit reading and eight that had not, volunteered to be reexamined on material from the class. The reexamination was conducted 3 mo after the end of the class. With first exam score fitted as a covariate (P < 0.10), participation in the extra credit reading assignment did not affect score on the reexamination; participants averaged 67% correct answers, nonparticipants averaged 59% (P = 0.37). Comments on course evaluations indicated broad satisfaction with the assignments. Although no improvement of retention could be documented from this project, our objective of encouraging students to engage in reading material supplemental to courses in genetics and reproductive physiology was satisfied.

The experimental objective was to compare pregnancy rates after fixed-time AI in postpartum suckled beef cows following administration of two progestin-based protocols to synchronize ovulation. Cows (n = 424) at three locations (n = 208, 122, and 92 per location) were stratified by age, BCS, and days postpartum (DPP) and assigned randomly to one of the two treatment protocols. The MGA Select-treated cows (MGA Select; n = 213) were fed melengestrol acetate (MGA, 0.5 mg x cow(-1) x d(-1)) for 14 d and carrier for 8 d, and then GnRH (100 microg i.m. Cystorelin; d 26) was injected 12 d after MGA withdrawal, and PG (25 mg i.m. Lutalyse) was administered 7 d after GnRH. Cows assigned to the 7-11 Synch protocol (7-11 Synch; n = 209) were fed carrier for 15 d and MGA for 7 d, and then injected with PG on d 22 (d 7 of MGA), GnRH on d 26, and PG again on d 33. Artificial insemination was performed at fixed times for cows in both treatments at 60 or 72 h after d 33 PG for 7-11 Synch and MGA Select groups, respectively. All cows were injected with GnRH (100 microg of i.m. Cystorelin) at AI. There was no treatment x location interaction for age (P = 0.90), BCS (P = 0.64), or DPP (P = 0.93), and the results were therefore pooled for the respective treatments (age [7-11 Synch, 5.5 +/- 0.2; MGA Select, 5.5 +/- 0.2], BCS [7-11 Synch, 5.7 +/- 0.1; MGA Select, 5.6 +/- 0.1], and DPP [7-11 Synch, 41.1 +/- 1.1; MGA Select, 42.1 +/- 1.1]). Blood samples were collected 8 and 1 d before MGA or carrier to determine pretreatment estrous cyclicity (progesterone >or=1 ng/mL; 7-11 Synch, 59/209 [28%]; MGA Select, 54/213 [25%]; P = 0.50) and again on d 33 PG to evaluate treatment response as a percentage of cows with progesterone concentrations in serum >or=1ng/mL (7-11 Synch, 184/209 [88%]; MGA Select, 177/213 [83%]; P = 0.15). Pregnancy rates resulting from fixed-time AI did not differ (P = 0.25) between treatments (7-11 Synch, 128/209 [61%]; MGA Select, 142/213 [67%]), nor did pregnancy rates (P = 0.77) at the end of the breeding season (7-11 Synch, 198/208 [95%]; MGA Select, 204/213 [96%]). These data indicate that pregnancy rates were comparable after fixed-time AI, following administration of the 7-11 Synch and MGA Select protocols. Both protocols provide opportunities for beef producers to use AI and eliminate the need to detect estrus.

The object of this study was to investigate the role of epidermal growth factor (EGF) and IGF-I in the regulation of preantral follicular growth, antrum formation, and granulosal cell proliferation/ apoptosis. Porcine preantral follicles were manually dissected and cultured for up to 8 d in Waymouth's (Exp. 1) or alpha-minimum Eagle's essential medium (Exp. 2 and 3) supplemented with 10 microg/mL of transferrin, 100 microg/mL of L-ascorbic acid, and 2 mU/mL of ovine FSH, in the presence (Exp. 1 and 3) or absence (Exp. 2) of 7.5% fetal calf serum. According to the experimental protocol, IGF-I (0, 1, 10, or 100 ng/mL; Exp. 1), or IGF-I (50 ng/mL), EGF (10 ng/mL) and EGF+IGF-I (Exp. 2 and 3) were added to the culture media. In Exp. 1, follicles exhibited a concentration-dependent response (P < 0.05) to IGF-I, with the highest rates of granulosal cell proliferation, follicular integrity, and recovery rate of cumulus cell-oocyte complexes and lowest incidence of apoptosis occurring at the highest IGF-I dose. In Exp. 2 serum-free medium, granulosal cell proliferation was low (1 to 5%), irrespective of whether EGF and/or IGF-I were present and cellular apoptosis was increased (P < 0.05) on d 4 and 8 in the EGF+IGF-I group compared with the addition of either factor alone. In Exp. 3, granulosal cell proliferation was high in all follicles cultured in serum-containing medium for the first 3 d, but fell sharply (P < 0.05) on d 4, except in media containing IGF-I. Collectively, EGF and IGF-I increased granulosal cell proliferation, decreased apoptosis, and promoted follicular antrum formation. These results may provide useful information for developing a preantral follicular culture system in which the oocytes are capable of fertilization and embryonic development.

Students in Reproductive Management (a senior-level course with approximately 20 to 50 students per semester) at the University of Missouri-Columbia are required to participate in a simulation exercise that is designed to improve reproductive efficiency in a beef herd. During a simulated 5-yr period, students must 1) improve reproductive efficiency in a beef cow-herd through implementation of reproductive management principles; 2) determine the economic impact of reproductive management decisions in a beef herd; and 3) evaluate the constraints of different geographical locations on approaches to reproductive management. Groups of three to four students are provided with the reproductive and economic records of a farm/ranch located in different parts of North America. Students create reproductive management plans consisting of 1) detailed discussion of farm/ranch environment (climate, terrain, forage and grain availability, and stocking rate; season for breeding and calving; and justification for choice of breed); 2) assessment of current level of reproductive performance; 3) identification and economic justification of specific (measurable) objectives; 4) discussion of alternatives for accomplishing specific objectives; 5) prediction of reproductive performance (pregnancy rate, quantity of calf weaned per cow exposed, and cost per quantity of calf weaned) in response to implementation of specific management practices; and 6) an annual and 5-yr reproductive and economic summary. Students obtain livestock marketing information for their assigned location via the Internet. Spreadsheets were developed to calculate the reproductive efficiency of postpartum cows and replacement heifers based on management decisions made by the groups and to calculate a yearly economic summary for each of the 5 yr. Management decisions are justified in a written report, and oral presentations are given to the class when the project is completed. Greater than 85% of students indicated that the exercise increased their understanding of how management decisions affect reproductive efficiency and profitability in a beef operation and also provided added confidence for students that applied for beef management positions.

Two progestin-based protocols for the synchronization of estrus in beef cows were compared. Cyclic, nonlactating, crossbred, beef cows were assigned by age and body condition score to one of two treatments. Cows assigned to the MGA Select protocol were fed melengestrol acetate (MGA; 0.5 mg x cow(-1) x (-1)) for 14 d, GnRH was administered (100 microg i.m. of Cystorelin) 12 d after MGA withdrawal, and PGF2alpha (25 mg of i.m. Lutalyse) was administered 7 d after GnRH. Cows assigned to the 7-11 Synch protocol were fed MGA for 7 d and were injected with PG on d 7 of MGA, GnRH on d 11, and PG on d 18. Transrectal ultrasonography was performed daily to monitor follicular dynamics from the beginning of MGA feeding through ovulation after the synchronized estrus. All cows exhibited estrus in response to PG. Mean interval to estrus was shorter (P < 0.01) for 7-11 Synch-treated cows (56 +/- 1.5 h) than for cows assigned to the MGA Select protocol (73 +/- 4.7 h). Mean interval from estrus to ovulation did not differ between treatments (P > 0.10). Variances for interval to estrus differed (P < 0.01) between treatments. Mean follicular diameter at GnRH injection, PG injection, and estrus did not differ (P > 0.10) between treatments. Relative to MGA Select, serum estradiol-17beta concentrations were higher (P < 0.01) for 7-11 Synch 2 d and 1 d before, on the day of GnRH injection, in addition to 4 d after GnRH, and 24 h after PG. Mean progesterone concentrations were greater (P < 0.01) for MGA Select cows from 4 d before to 7 d after GnRH. Forty-four percent of the variation in interval to estrus between treatments was explained by differences in estradiol-17beta concentrations 24 h after PG. This study suggests that follicular competence is likely related to steroidogenic capacity of the follicle and the endocrine environment under which growth and subsequent ovulation of the dominant follicle occurs.

Two progestin-based protocols for estrus synchronization in postpartum beef cows were compared following treatment administration on the basis of estrous response, interval to and synchrony of estrus, and pregnancy. Cows were assigned to one of the two treatment protocols by age, body condition score (BCS), and days postpartum (DPP). The MGA Select-treated cows (MGA Select; n = 109) were fed melengestrol acetate (MGA; 0.5mg x cow-1 x d(-1)) for 14 d, fed carrier for 8 d, GnRH (100 microg of Cystorelin) was injected i.m. 12 d after MGA withdrawal, and PG (25 mg of Lutalyse) was administered i.m. 7 d after GnRH. Cows assigned to the 7-11 Synch protocol (7-11 Synch; n = 111) were fed carrier for 15 d, fed MGA for 7 d, injected with PG on d 22 (d 7 of MGA), injected with GnRH on d 26, and injected with PG on d 33. Mean BCS (4.8 +/- 0.1, MGA Select; 4.7 +/- 0.1, 7-11 Synch) and DPP (40 +/- 1, MGA Select; 40 +/- 1, 7-11 Synch) did not differ between treatments. Blood samples were collected 8 d and 1 d before feeding of MGA or carrier to determine the pretreatment estrous cyclicity (progesterone > or = 1 ng/mL; 10/109 [9%], MGA Select; 12/111 [11%], 7-11 Synch), and again at PG on d 33 to evaluate treatment response (81/109 [74%], MGA Select; 84/111 (76%), 7-11 Synch). Serum concentrations of progesterone at PG on d 33 differed (P < 0.01) between treatments (3.3 +/- 0.3 ng/mL [MGA Select] vs. 1.7 +/- 0.1 ng/mL [7-11 Synch]). HeatWatch was used for 6 d after PG on d 33 to detect estrus, and AI was performed 12 h after the onset of estrus. Estrous response did not differ between treatments (100/109 [92%], MGA Select; 101/111 [91%], 7-11 Synch). Mean interval to estrus (65 +/- 2.7 h, MGA Select; 52 +/- 1.8 h, 7-11 Synch) and synchrony of estrus differed (P < 0.01) between treatments. Synchronized conception and pregnancy rates (61/100 [61%], 61/109 [56%], MGA Select; 71/101 [70%], 71/111 [64%], 7-11 Synch), and final pregnancy rates (94/109 [86%], MGA Select; 99/110 [90%], 7-11 Synch) did not differ between treatments. In summary, estrous response and fertility did not differ among cows assigned to the MGA Select or 7-11 Synch protocols. Synchrony of estrus, defined as the variance in the interval to estrus from PG, however, was improved following treatment with the 7-11 Synch protocol.

Postpartum anestrous interval in beef cows is a major factor contributing to reproductive failure during a defined breeding season. Our objectives were to determine the ability of a controlled internal drug-releasing device (CIDR, 1.9 g of progesterone), a normal dose of melengestrol acetate (MGA, 0.5 mg x cow(-1) x d(-1)), or a high dose of MGA (4.0 mg x cow(-1) x d(-1)) to induce ovulation and to eliminate short estrous cycles. Multiparous beef cows (n = 100) were equally assigned to one of four treatments: CIDR, normal MGA, high MGA, or control by age, days postpartum, body condition, and body weight. All cows were fed carrier (0.9072 kg x cow(-1) x d(-1)) with (normal MGA, 0.55 mg/kg; high MGA, 4.41 mg/kg) or without MGA for 7 d (d -6 to 0). On d -6, CIDR were inserted and then removed on d 0. Estrous behavior was monitored continuously from d -6 until 29 using HeatWatch electronic mount detectors. Blood was collected on d -13, and three times weekly from d -6 to 29. Treatment influenced (P = 0.03) the percentage of cows that were detected in standing estrus. Beginning on d 2, more CIDR-treated cows had exhibited standing estrus compared with high MGA-treated or control cows, but CIDR- and normal MGA-treated cows did not differ. The percentage of CIDR-treated cows that had ovulated was greater (P < 0.05) than the percentage of normal MGA-treated, high MGA-treated, or control cows beginning on d 4. The percentage of cows that exhibited standing estrus before the first postpartum ovulation (CIDR = 65%, normal MGA = 57%, high MGA = 35%, control = 30%) did not differ (P = 0.09) among treatments. Luteal life span following the first ovulation postpartum and the percentage of cows with a normal luteal life span (i.e., progesterone > 1 ng/mL for > or = 10 d) was greater (P < 0.01) in CIDR-treated cows (14.0 +/- 0.8 d; 20/20, 100%) compared with normal MGA-treated (6.2 +/- 1.0 d; 3/13, 23%), high MGA-treated (9.6 +/- 1.0 d; 8/14, 57%), or control cows (6.1 +/- 0.9 d; 4/17, 24%), and greater (P < 0.03) in high MGA-treated cows than in normal MGA-treated or control cows. In the present study, treatment of early postpartum suckled beef cows with CIDR induced ovulation and initiated estrous cycles with a normal luteal life span in more cows than did treatment with MGA. Treatment with MGA (normal or high dose) did not induce ovulation earlier than in control cows, but a high dose of MGA increased the percentage of cows with normal luteal life spans following the first ovulation postpartum.

The purpose of the present study was to determine the effect of progesterone or progesterone + estradiol-17beta on oxytocin-induced prostaglandin F2alpha (PGF2alpha) secretion in postpartum beef cows. Thirty-four anestrous postpartum beef cows were ovariectomized (d 32 [Groups 1 to 3] or d 23 [Groups 4 to 6] postpartum [d 0 = parturition]) and allotted to six treatments (Group 1; negative control) to simulate short (Groups 2 through 5) or normal (Group 6) length estrous cycles. Steroid treatments for the respective groups were as follows: Group 1) no estradiol-17beta or progesterone treatment (n = 8; negative control); Group 2) progesterone (d 34 to 40; n = 6); Group 3) estradiol-17beta (d 32 to 33) and progesterone (d 34 to 40; n = 6); Group 4) progesterone (d 23 to 29), no estradiol-17beta (d 32 to 33), and progesterone (d 34 to 40; n = 5); Group 5) progesterone (d 23 to 29), estradiol-17beta (d 32 to 33), and progesterone (d 34 to 40; n = 5); and Group 6) progesterone (d 23 to 29), estradiol-17beta (d 32 to 33), and progesterone (d 34 to 50; n = 4; positive control). Oxytocin (100 IU) was injected (i.v.) at the end of each treatment to test the ability of the postpartum uterus to secrete PGF2alpha as measured by a stable metabolite of PGF2alpha, 15keto-13,14 dihydro-PGF2alpha (PGFM). Peak concentrations ofPGFM (P < 0.08) and total PGFM secreted (area under the curve; P < 0.05) were increased on d 6 following first (Group 2) or second (Group 4) exposure to progesterone and were similar to peak concentrations and total PGFM secreted 16 d following a simulated normal estrous cycle (Group 6). Administration of estradiol-17beta before first progesterone exposure (Group 3) did not reduce peak concentrations of PGFM or total PGFM secreted relative to the preceding groups. Peak concentrations of PGFM (P < 0.08) and total PGFM secreted (P < 0.05) were reduced following a second progesterone exposure, provided that cows were pretreated with estradiol-17beta (Group 5). In summary, oxytocin-induced release of PGFM was inhibited on d 6 following second exposure to progesterone only when cows were pretreated with estradiol-17beta. Therefore, estradiol-17beta and progesterone were both associated with the timing of PGF2, secretion in postpartum cows.

Treatment with melengestrol acetate (MGA), an oral progestin, prior to administration of gonadotropin-releasing hormone (GnRH) and prostaglandin F2alpha (PG) effectively synchronizes estrus and maintains high fertility in postpartum beef cows. The objective of this experiment was to determine whether treatment with MGA prior to a GnRH-PG-GnRH protocol would improve pregnancy rates resulting from fixed-time artificial insemination (AI). Multiparous crossbred beef cows at two University of Missouri-Columbia farms (n = 90 and n = 137) were assigned by age and days postpartum to one of two treatments. Cows were fed carrier (1.8 kg x animal(-1) x d(-1)) with or without MGA (0.278 mg x kg(-1)) for 14 d. All cows were administered GnRH (100 microg; intramuscularly) on d 12 after MGA or carrier withdrawal and 7 d before PG (25 mg; intramuscularly). All cows received a second injection of GnRH and AI 72 h after PG. Mean days postpartum for MGA and control cows at the initiation of treatment were 39.6 and 38.9 d for herd 1; and 51.9 and 50.9 d for herd 2, respectively (P > 0.70 within herds). Blood samples were collected from all cows at 10 and 1 d before the feeding of MGA or carrier began and at the times GnRH and PG were administered. Concentrations of progesterone in serum at the initiation of treatment were elevated (>1 ng/mL) in 0% of MGA and 7% of control cows in herd 1, and 54% of MGA and 49% of control cows in herd 2 (P > 0.05 within herds). Pregnancy rates to fixed-time AI were determined by transrectal ultrasonography 50 d after AI. Pregnancy rates in herd 1 were 58% (26/45) and 51% (23/45) for MGA-treated and control cows, respectively (P = 0.52), and 63% (44/70) and 45% (30/67) for MGA-treated and control cows in herd 2, respectively (P = 0.03). Differences in pregnancy rates to fixed-time AI were significant (P = 0.04) when data from the two herds were combined (with MGA = 70/115 [61%]; control = 53/112 [47%]). There was no difference (P > 0.20) in final pregnancy rates (timed AI plus 45 d exposure to bulls) between treatments, within herds, or when herds were combined. In summary, pregnancy rates resulting from fixed-time AI may be improved with treatment of MGA prior to a GnRH-PG-GnRH protocol.

Although treatment of cycling cows with low concentrations of melengesterol acetate (MGA) results in formation of persistent follicles, in the absence of corpora lutea, it is not known whether persistent follicles form in anestrous cows in response to a similar treatment. The objective of this experiment was to determine the effect of long-term MGA treatment (14 d) on follicular dynamics and the secretion of estradiol in anestrous postpartum beef cows. Treatment groups (replicated over 2 yr) included the following: anestrous control (AC; n = 11), anestrous MGA (AM; n = 16), and cycling MGA (CM; positive control; n = 16). Angus-crossbred cows were assigned to treatment by age, cow body condition, and days postpartum. Cows were fed carrier (AC group) or 0.5 mg MGA x animal(-1) x d(-1) (AM and CM groups) for 14 d beginning approximately 38 d postpartum. Cows allotted to the CM group were injected with PGF2alpha, on the first day of MGA treatment to induce luteolysis. The preceding treatment (CM) results in formation of persistent follicles and secretion of elevated concentrations of estradiol. Ovaries of each cow were examined daily by transrectal ultrasonography beginning 5 to 7 d preceding the initiation of feeding MGA or carrier and continued until ovulation or 7 d following MGA feeding. There was no difference among groups in the stage of follicular wave or diameter of the largest follicle at the start of carrier or MGA feeding. The length of the follicular wave present at the start of MGA feeding was greater (P < 0.01) for cows in the CM (14.5 d, yr 1; 18.3 d, yr 2) group compared to the AM (9.4 d, yr 1; 7.9 d, yr 2) or AC (9.7 d, yr 1; 10.7 d, yr 2) groups. Maximum follicular diameter over both years was greater (P < 0.01) for the CM (20.6 mm) group than the AM (15.1 mm) or AC (16.4 mm) groups. Circulating concentrations of estradiol were also increased (P < 0.05) in the CM group compared to the AM or AC groups. However, MGA appeared to have no effect (P > 0.05) on the number of follicles recruited, growth rate of the dominant follicle during the first 6 d oftreatment, or growth rate to the maximum follicular diameter. In summary, MGA treatment did not increase the duration ot the follicular wave, maximum follicular diameter, or secretion of estradiol in anestrous postpartum cows, nor did MGA affect the number of follicles recruited or growth rate of dominant follicles in cycling or anestrous animals.

In higher education, increasing emphasis is being placed on the use of new technologies in the classroom. However, the emphasis needs to be placed on methods that truly enhance understanding and knowledge retention. Class discussions help students understand and retain information previously presented in lecture format. Furthermore, if students are challenged to critically evaluate, communicate, and defend their ideas, knowledge retention and understanding will increase even more. Critical interactive thinking exercises (CITE) were employed at two different universities to enhance student knowledge retention and promote the development of critical thinking. Applicability of CITE to undergraduate learning was assessed over a 3-yr period in the undergraduate reproductive physiology courses at Michigan State University and the University of Missouri-Columbia. For each exercise, students were challenged to prepare a one-page, double-spaced composition addressing an incompletely understood phenomenon or problem-solving situation related to the reproductive system. In preparing their compositions, students were encouraged to use information previously presented in lecture plus outside information to develop their ideas. Students were required to formulate and defend a hypothesis or approach to the problem presented. At the subsequent class period, students were divided into groups of three to four, in which they interactively discussed their ideas. Each group member was challenged to defend his or her hypothesis and explanation and to persuade other group members to adopt their ideas. Each group then arrived at a consensual opinion that was presented during a discussion by the entire class. The class then debated the merits of each group's hypothesis or explanation and the supporting arguments presented. At first, the students were apprehensive about the CITE, particularly about communicating and defending ideas with their classmates. However, course evaluations showed that 131 out of 137 students considered the CITE a positive experience that enhanced learning. Additionally, 131 out of 137 students reported that the CITE enhanced their critical thinking skills. We feel that the use of CITE in teaching reproductive theory to undergraduate students fosters critical thinking skills, communication skills, and knowledge retention. The general concept can be readily applied to courses in other subject areas in the animal sciences.

The hypothesis that ovulation in response to short-term (48 h) calf removal (CR) is dependent on the developmental stage of the dominant follicle was tested in two studies. The objective of Exp. 1 was to characterize the fate of a dominant follicle following 48-h CR on d 2, 4, or 8 of a postpartum follicular wave. Ovaries of 61 beef cows were examined daily by transrectal ultrasonography starting at d 20 to 21 postpartum. Treatments were no CR (n = 14) and CR on d 2 (n = 12), 4 (n = 16), or 8 (n = 10) of first detected follicular wave. Percentage of cows that ovulated a dominant follicle following treatment was not different among groups (P = 0.62). Maximum size of dominant follicles was larger in cows that ovulated (P = 0.002) than in cows that did not ovulate. The objectives of Exp. 2 were 1) to determine whether a follicular wave could be synchronized in anestrous cows following injection of 1 mg of estradiol benzoate (EB) and 200 mg of progesterone (P4; EB + P4); 2) to characterize the fate of dominant follicles following 48-h CR at three stages of a synchronized follicular wave; and 3) to determine whether estrous cycles of normal length followed ovulation in cows pretreated with EB + P4. Ovaries of 50 anestrous beef cows were examined daily as in Exp. 1. Treatments were sesame oil (SO) injected (i.m.) on d 25 postpartum and no CR (n = 9); EB + P4 and no CR (n = 9); EB + P4 and CR on 6 (n = 12), 8 (n = 9), or 12 (n = 11) d after injection. The EB and P4 injections were given on d 25 postpartum. Variability in day of emergence of subsequent follicular waves was lower in cows receiving EB + P4 than in SO-injected cows (P < 0.05). The percentage of cows that ovulated was not different (P = 0.16), but CR increased the percentage of cows that ovulated when groups that received EB + P4 were compared to the EB + P4 group that did not have CR (53.1 vs 11.1%, respectively; P < 0.05). Maximum diameter of dominant follicles was larger (P = 0.05) in ovulatory follicles. The luteal phase was longer (P < 0.03) in cows receiving EB + P4 injection (10.6 +/- 1.2 d) than in cows receiving SO (4.4 +/- 2.2 d). In summary, the maximum size of ovulatory follicles was greater than that of nonovulatory follicles, the ovulatory response of postpartum anestrous cows was maintained through d 8 of a follicular wave, synchronization of follicular waves was accomplished in postpartum cows using EB + P4, and the subsequent luteal phase length was increased in animals that were administered EB + P4.

The objective of this experiment was to determine the effect of a GnRH injection within a melengestrol acetate (MGA)-PGF2alpha (PGF) estrus synchronization protocol on follicular dynamics and synchronization of estrus. Pubertal crossbred beef heifers (n = 34) were randomly assigned to one of two treatments. Both treatment groups were fed MGA (0.5 mg x hd(-1) x d(-1)) for 14 d and injected (i.m.) with PGF (25 mg of Lutalyse) 19 d after MGA withdrawal. Melengestrol acetate was delivered in a feed supplement of 1.8 kg x hd(-1) x d(-1). Seventeen heifers received an injection of GnRH (100 microg Cystorelin) 12 d after MGA withdrawal and 7 d before PGF. The control group (n = 17) received only MGA-PGF. Estrus was detected four times/d for 7 d beginning on the day PGF was injected. Transrectal ultrasonography was performed daily on eight heifers from each treatment to monitor ovarian activity and characterize changes in follicular dynamics after MGA withdrawal and until ovulation after PGF. Each of the GnRH-treated heifers either ovulated or had a luteinized dominant follicle following GnRH and subsequently initiated a new follicular wave (8/8, 100%). All GnRH-treated heifers (17/17, 100%) and 94% of controls (16/17) exhibited estrus after PGF. Estrus was exhibited over a 132-h period (12 to 144 h) for control heifers compared with 60 h (48 to 108 h) for GnRH-treated heifers. The peak synchronized period for both treatments was between 48 and 72 h after PGF, during which time 76% (13/17) of the GnRH-treated heifers exhibited estrus compared with 63% (10/16) for controls. Seventy-one percent (12/17) of the GnRH-treated heifers exhibited estrus from 48 to 60 h after PGF, compared with 38% (6/16) for controls (P < 0.05). In summary, injection of GnRH within a 14- to 19-d MGA-PGF protocol increased the synchrony of estrus during the synchronized period and concentrated the period of detected estrus. This protocol may offer potential for the fixed-time insemination of replacement beef heifers.

The objectives of this study were to evaluate the effects of pre- and postpartum undegraded intake protein (UIP) supplementation on body condition score (BCS), BW, calf weight, milk production, serum IGF-I concentrations, and postpartum interval in primiparous beef heifers (n = 44). Heifers were maintained on endophyte-free stockpiled tall fescue (11.7% CP, 38% ADF) and individually fed supplement daily beginning 60 d prepartum. Pre- and postpartum supplements provided 19.3% CP, 83.4% TDN (UIP); 14.1% CP, 84.1% TDN (Control); 21.5% CP, 81.5% TDN (UIP); and 14.6% CP, 81.4% TDN (Control); respectively. Blood meal (146 g/d) was the source of UIP. Six heifers were removed from the study due to calf loss unrelated to treatment; therefore, postpartum measurements are based on 19 animals per treatment. Statistical analyses using ANOVA and a split-plot design revealed no effects of treatment (P > 0.2) on BCS, BW, calf weight, milk production, or postpartum interval. There tended to be a treatment x time interaction on BCS (P < 0.09) with UIP heifers having higher BCS than Control at wk 5, 7, and 9 postpartum. There was a treatment x time interaction on serum IGF-I (P < 0.06) during the first 35 d postpartum. In UIP heifers, serum IGF-I was greater at calving compared with Control heifers (117.5 vs 92.4 ng/mL, respectively); however, these differences were not related to changes in BCS or BW. Although serum IGF-I concentrations were increased at calving in heifers receiving UIP, there were no treatment effects on postpartum interval (P > 0.7). During the first 30 d postpartum, IGF-I differed (P < 0.01) among heifers with postpartum intervals defined as short, < 50 d (128.9 ng/mL); medium, 51 to 65 d (115.2 ng/mL); and long, 66 to 130 d (52.9 ng/mL). When analyzed as a regression, a 1 ng/mL increase in IGF-I (UIP and Control heifers) at calving (P < 0.05) and throughout the postpartum period (P < 0.01) corresponded to a decrease in postpartum interval of 0.13 d. Based on the results of this study, the inclusion of UIP in diets for primiparous heifers and its effects on postpartum interval warrant further evaluation.

An estrus synchronization protocol (7-11 Synch) was developed to synchronize the first follicular wave and timing of ovulation in postpartum beef cows. In Exp. 1, follicular development and timing of ovulation in response to the following protocol were evaluated. Beef heifers (n = 12) and cows (n = 6), at random stages of the estrous cycle, were fed melengestrol acetate (MGA; .5 mg x animal(-1) x d(-1)) for 7 d and injected with PGF2alpha (PG; 25 mg) on the last day of MGA. A second injection of PG was administered 11 d after cessation of MGA. After the second injection of PG, estrus was synchronized in 6/12 heifers and 3/6 cows. The interval to estrus in heifers and cows was 54 and 64 h, respectively (P > .10). All animals exhibiting estrus ovulated first-wave follicles. Animals that failed to respond to the second injection of PG were in estrus later than 6 d after cessation of MGA and had corpora lutea that were unresponsive to the injection of PG. Based on the variation in interval to estrus following the first PG injection on the last day of MGA feeding in Exp. 1, an injection of GnRH (100 microg) was added to the protocol 4 d after the cessation of MGA to ensure ovulation or luteinization of dominant follicles and synchronization of first-wave follicular development. This revised protocol was termed "7-11 Synch." In Exp. 2, two estrus synchronization protocols were compared. Multiparous beef cows were stratified by breed and postpartum interval and randomly assigned to the 7-11 Synch (n = 44) or Select Synch protocols (GnRH injection followed by PG injection 7 d later; n = 45). Timing of estrus after the last PG injection (0 h) ranged from 42 to 102 h in the 7-11 Synch group and -30 to 114 h in the Select Synch group. Eight cows (18%) in the Select Synch group exhibited estrus 30 h before to 18 h after PG. Synchronized estrus peaked between 42 and 66 h after the last PG injection, and a maximum number of cows were in estrus at 54 h for both treatment groups. Synchrony of estrus from 42 to 66 h was greater (P < .05) in 7-11 Synch (91%: 41/44) than in Select Synch cows (69%: 31/45). Artificial insemination pregnancy rate from 42 to 66 h was greater (P < .05) in the 7-11 Synch group (66%: 29/44) than in the Select Synch group (40%: 18/45). In summary, the 7-11 Synch protocol improved synchrony of estrus without reducing fertility. This protocol has potential future application for fixed-time AI in beef cattle production systems.

Heifers treated with ivermectin at weaning have been reported to reach puberty at a younger age and lighter weight than untreated heifers. We tested the hypothesis that heifers administered ivermectin would respond with earlier follicular development and a greater LH response to a 1-mg estradiol-17beta challenge (E2C) than untreated heifers. Fall-born Angus heifers (n = 32) were randomly assigned on 284 +/- 9 d of age (215.5 +/- 20.8 kg) to receive ivermectin (IVR) or albendazole (ALB), IVR + ALB, or to remain as untreated controls (CONT). Each group (n = 8) was housed separately in adjacent pens throughout the trial and managed to gain .8 kg/heifer on a ration containing 13.2% CP, 58.8% TDN, and 49.9% DM. The CONT heifers received an additional 2.27 kg/heifer of corn silage and 1.59 kg/heifer of corn daily to maintain ADG at comparable levels. Individual body weight was recorded weekly, and nematode eggs per gram (EPG) of feces were measured every 21 d. Ultrasonography was performed on alternate days starting 2 wk prior to E2C to characterize follicular wave patterns. Follicles were separated into classes (C1 [3 to 5 mm], C2 [6 to 9 mm], and C3 [10 mm]) and sizes (largest [LF], second [SLF], third [TLF], and fourth largest follicles [FLF]). The sizes of the regressing dominant follicle 1 (DF1) and the progressing dominant follicle 2 (DF2) were also determined. Serum concentrations of LH were determined from hourly jugular blood samples collected 8 to 24 h after injection of E2C. The IVR + ALB treatment group had more C3 follicles than ALB and CONT (P < .07). The IVR-treated heifers had larger TLF than ALB and CONT (P < .04). The IVR- and IVR + ALB-treated heifers had larger FLF and DF2 than ALB and CONT (P < .1). Least squares means for DF2 were 9.5 +/- .5, 8.0 +/- .4, 9.5 +/- .3 and 8.3 +/- .3 mm, for IVR, ALB, IVR + ALB and CONT, respectively (P = .02 for treatment effect). The E2C-induced serum LH concentration did not differ with respect to treatment. We conclude that heifers administered IVR display increased follicular development, supporting our earlier investigations regarding reduced age at puberty in heifers treated with IVR near weaning.

Tissue inhibitor of metalloproteinases (TIMP)-1 mRNA and protein localize within granulosal cells of post-gonadotropin-surge follicles and luteal tissue in ewes. Our objectives were to test the hypotheses that 1) follicular fluid concentration of TIMP-1 increases following a gonadotropin surge induced by LHRH agonist (Exp. 1) and 2) luteal status affects peripheral serum concentration of TIMP-1 (Exp. 2 and 3). In Exp. 1, the concentration of TIMP-1 within antral fluid from post-surge follicles (28.7 +/- 6.65 microg/mL) was greater (P < .02) than from pre-surge follicles (2.37 +/- 2.47 microg/mL). In Exp. 2, serum concentration of TIMP-1 did not differ among d 0 to 6 (1.27 +/- .55 microg/mL) of the estrous cycle or among periods of luteal maintenance (1.29 +/- .06 microg/mL), spontaneous luteal regression (1.19 +/- .09 microg/mL), or luteal development (1.22 +/- .08 microg/mL). However, serum concentration of TIMP-1 was greater ( P < .001) during the period of luteal maintenance (1.14 +/- .04 microg/mL) than during PGF2alpha-induced luteolysis (d 26; .85 +/- .06 microg/mL) and induced luteal absence (d 27 to 33; .95 +/- .05 microg/mL). In Exp. 3, ewes (n = 14) were bled daily from d 1 to 19 (d 0 = estrus) and at 12-min intervals for 6 h on d 3, 10, and 17. Although concentration of TIMP-1 varied considerably within and among ewes, mean concentration of TIMP-1 per ewe per day increased ( P < .05) from d 3 to 17. These data indicate that follicular fluid concentration of TIMP-1 increases following a gonadotropin surge, but the contribution of ovarian derived TIMP-1 to peripheral serum concentration is negligible.

The transition of a preovulatory follicle into a corpus luteum is a complex process involving mechanisms similar to wound healing and tumor formation. The objective of this review is to focus on mechanisms associated with corpus luteum development with specific attention to the follicular lineage of luteal cells, mechanisms associated with luteinization, and neovascular changes during luteal development. Corpora lutea are a continuation of follicular maturation and form from granulosal and theca interna cells. There is morphological and immunological evidence in ruminant species for the differentiation of granulosal and theca interna cells into large and small steroidogenic luteal cells, respectively. Different morphological, physiological, and biochemical characteristics of large and small luteal cells may reflect different follicular lineages with separate embryological origins. Following the preovulatory gonadotropin surge, follicular cells begin morphological, endocrinological, and biochemical changes associated with luteinization. Luteinization involves the transition of a preovulatory follicle into a highly vascular corpus luteum capable of secreting large quantities of progesterone. In addition, various cell types undergo hyperplasia, hypertrophy, and(or) migration during corpus luteum formation. An essential component of corpus luteum development is the recruitment of a blood supply. The development of a new microcirculatory bed involves breakdown of the follicular basement membrane, endothelial cell migration, endothelial cell proliferation, and development of capillary lumina. This process is regulated by the interaction of angiogenic and antiangiogenic substances. Further clarification of the preceding mechanisms may result in the development of improved methodologies for controlling the time of ovulation and(or) increasing pregnancy rates.

The objectives of the first experiment were to characterize the pattern of protein secretion by 1) porcine follicles before and after the preovulatory gonadotropin surge and 2) corpora lutea on d 4 and 11 after estrus (d 0 = estrus). Gilts were ovariectomized 1) on d 20 after estrus and before the preovulatory gonadotropin surge (pre-estrus group; n = 3), 2) 18 to 36 h after the onset of estrus and after the preovulatory gonadotropin surge (estrus group; n = 3), 3) on d 4 after estrus (n = 3), and 4) on d 11 after estrus (n = 4). Changes in pattern of protein secretion were determined by densitometric scanning of fluorographs. In the pre-estrus group, the major proteins secreted by follicular shells (FS) and granulosa cells (GC) had relative molecular masses (M(r)) of approximately 40,000, 46,000, and 55,000. In the estrus group (FS and GC), secretion of a M(r) 40,000 protein was decreased (P < .05) and secretion of a M(r) 30,000 protein was increased (P < .05) relative to the pre-estrus group. A predominant M(r) 30,000 protein was also secreted by corpora lutea on d 4 and 11. The objective of Exp. 2 was to determine whether synthesis of the M(r) 30,000 protein could be increased during the follicular phase by treatment with hCG, suggesting a role for the preovulatory gonadotropin surge in altering the pattern of protein secretion. Most estrus synchronization schemes facilitate the use of artificial insemination; however, combining estrus synchronization with natural service also offers several benefits to beef producers. This experiment was designed to study the effectiveness of one injection of prostaglandin F2 alpha (PGF2 alpha) 96 h after introduction of intact bulls on estrus synchronization and calving distribution of beef cows and heifers. Three hundred sixty beef cows and 45 beef heifers in seven trials were allotted by age, breed, and prior calving date to one of two treatments. Within each subclass, one group was injected with an appropriate dose of PGF2 alpha and the second group at each location received 5 ml of saline (SAL). Cows were classified as estrual or anestrual at the start of breeding based on serum progesterone levels and were observed for behavioral estrus regularly during the first 25 d of the breeding season in two of the trials (d 0 = day before bull turn-in). Pregnancy data and calving dates were recorded in each of the studies. All cows were palpated per rectum for pregnancy approximately 70 d after the introduction of bulls. During d 5 to 9 after treatment, the percentage of cows exhibiting behavioral estrus (P = .02) and the pregnancy rate (P = .02) obtained was greater for PGF2 alpha-treated cows than for SAL-treated cows. There were 6.5% more (P = .05) calves born during the overall calving period from animals treated with PGF2 alpha compared with those in the SAL group. In summary, treatment of cows and heifers with PGF2 alpha 96 h after bull turn-in was effective in synchronizing behavioral estrus of beef cows and heifers and increasing the calving rate during the subsequent calving season.

Mechanisms associated with subnormal luteal function are reviewed with particular reference to sheep and cattle. Since the corpus luteum is a continuation of follicular maturation, mechanisms during both the follicular and luteal phase have been considered. During the follicular phase, extrafollicular and intrafollicular events are important for adequately preparing follicular cells for luteinization and secretion of progesterone (P4). Mechanisms which alter the microenvironment of the preovulatory follicle may impair the ability of follicular cells to subsequently secrete P4 and result in subnormal luteal function. During the luteal phase, secretion of P4 is probably dependent upon a balance between luteotropic and luteolytic stimuli. Subnormal luteal function may be due to an inadequate luteotropic stimulus and(or) an inability of luteal tissue to respond to a luteotropic stimulus. Additionally, a premature luteolytic stimulus may be involved in reduced luteal function. Specifically, premature luteolysis may result from an increased or premature release of a luteolysin or perhaps an increased responsiveness of luteal tissue to a luteolysin.

The objective of the present study was to determine the temporal relationships among luteal adenylate cyclase activity, luteal phosphodiesterase activity, luteal progesterone concentration and plasma progesterone concentration during prostaglandin F2 alpha (PGF2 alpha)-induced luteolysis in ewes. Corpora lutea (CL) were removed from cycling ewes on d 9 (d 0 = first day of estrus) at 0, 2, 4, 6, 12 and 24 h (seven to eight ewes/group) after PGF2 alpha administration (im). Jugular blood samples were collected at the time of enucleation of CL and analyzed for progesterone. Plasma and luteal progesterone concentrations were decreased (P less than .05) by 4 and 12 h after PGF2 alpha injection, respectively. Basal adenylate cyclase, luteinizing hormone (LH)-activated adenylate cyclase, guanylylimidodiphosphate [Gpp(NH)p]-activated adenylate cyclase and LH plus Gpp(NH)p-activated adenylate cyclase activities were decreased (P less than .05) by 2 h after PGF2 alpha injection. The decrease in adenylate cyclase activity paralleled the decrease in plasma progesterone concentration over time. Luteinizing hormone stimulated (P less than .05) adenylate cyclase activity relative to basal activity at 0, 2, 12 and 24 h post-PGF2 alpha; whereas, Gpp(NH)p stimulated (P less than .01) adenylate cyclase activity relative to basal activity at each time point. In contrast to the decrease in adenylate cyclase activity, phosphodiesterase activity was increased (P less than .05) at 2 and 4 h post-PGF2 alpha. These results suggest that a decrease in adenylate cyclase activity coupled with an increase in phosphodiesterase activity may decrease the intracellular adenosine 3',5' cyclic monophosphate (cAMP) concentration.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of the uterus on luteal lifespan and pattern of secretion of progesterone following early weaning of calves from anestrous beef cows was studied. Calves were weaned from 15 anestrous beef cows 23 to 33 d postpartum, and cows were allotted to a control (sham surgery, n = 8) or a hysterectomy (n = 7) group, with surgery performed at weaning. Cows in the hysterectomy group were injected (im) with 25 mg prostaglandin F2 alpha (PGF2 alpha) approximately 20 d after first estrus (d 0). The interval from weaning to estrus was longer (P less than .05) for the hysterectomy group (10.4 +/- 1.6 d) than the control group (6.2 +/- .5 d). In the control group, the first estrous cycle (8.8 +/- .3 d) was shorter (P less than .01) than the second estrous cycle (20.2 +/- .5 d). Following first estrus in the hysterectomy group, cows were not detected in estrus until after injection of PGF2 alpha and did not return to estrus. From d 0 to 5, mean concentrations of plasma progesterone were similar (P greater than .05) between groups for both estrous cycles; after d 5 of estrous cycle 1, concentrations of plasma progesterone decreased in the control group. Within the hysterectomy group, the pattern of secretion of progesterone from d 0 to 16 was similar after the first and second estrus. Furthermore, there was no difference in the pattern of secretion of progesterone from d 0 to 16 between hysterectomy (first or second estrous cycles) and control (second estrous cycle) groups.(ABSTRACT TRUNCATED AT 250 WORDS)

An experiment was conducted to determine whether short estrous cycles following abortion of heifers between 70 and 75 d of gestation are due to factors associated with the previous presence of a conceptus or long-term exposure of the uterus and(or) ovaries to a progestogen. Fifty crossbred heifers were randomly allotted at estrus (d 0) to five groups: control (n = 10), pregnant (Preg.; n = 14), progestogen (norgestomet) implant (Norg.; n = 9), progesterone-releasing intravaginal device (PRID; n = 9), or hysterectomy (Hyst.; n = 8). Control heifers were injected during the mid-luteal phase of an estrous cycle with 25 mg prostaglandin F2 alpha (PGF2 alpha) and length of the subsequent estrous cycle was determined. Beginning 6 to 8 d after estrus, heifers in the Norg. or PRID groups were given norgestomet ear implants or intravaginal coils, respectively, every 10 d for 70 d. Heifers were hysterectomized 5 to 8 d after estrus. Seventy to 75 d after conception, progestogen treatment or hysterectomy, heifers were injected (i.m.) with 25 mg PGF2 alpha and the last norgestomet ear implants or PRIDs were removed. Interval from PGF2 alpha injection to first estrus (means +/- SE) ranged from 2.5 +/- .2 to 4.4 +/- .7 d (P greater than .05). Length of the first estrous cycle means +/- SE) following PGF2 alpha-induced luteolysis or progestogen withdrawal was shorter (P less than .01) for the Preg. group (8.2 +/- .4 d) than for the control, Norg. and PRID groups (21.5 +/- .6 d; 19.3 +/- 1.4 d; and 18.2 +/- 1.3 d, respectively).(ABSTRACT TRUNCATED AT 250 WORDS)

Early weaning of calves from anestrous cows results in formation of short-lived corpora lutea (CL) unless the animals are pretreated with a progestagen (norgestomet). This study was conducted to investigate the relationship between pre- and post-ovulatory gonadotropin secretion and luteal lifespan. Postpartum beef cows were assigned randomly into two groups, control (n = 5) and norgestomet (implant given at weaning for 9 d; n = 7). Calves from all cows were weaned 30 to 33 d postpartum. Coccygeal artery cannulas were placed into cows in the control group 1 d prior to weaning and 2 d before implant removal in cows in the norgestomet group. Plasma for determination of luteinizing hormone (LH), follicle stimulating hormone (FSH), estradiol-17 beta (E) and progesterone (P) was collected daily at 10-min intervals for 6 h from weaning (control) or the day prior to implant removal (norgestomet) to estrus (d 0) and on d 2, 4 and 6 following estrus. Average interval (X +/- SE; P less than .05) from weaning to estrus or implant removal was 4.2 +/- .8 and 2.3 +/- .2 d for the control and norgestomet groups, respectively. Estrous cycle length for the control group was 12.4 +/- 1.8 d compared with 20.4 +/- .3 d for the norgestomet group (P less than .05). Four of five control cows had an estrous cycle length of 7 to 14 d; all cows in the norgestomet group and the remaining control cow had an estrous cycle of normal length (16 to 21 d).2+ estrus.(ABSTRACT TRUNCATED AT 250 WORDS)
