Glyphosate is a nonselective systemic herbicide used in agriculture since 1974. It inhibits 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase, an enzyme in the shikimate pathway present in cells of plants and some microorganisms but not human or other animal cells. Glyphosate-tolerant crops have been commercialized for more than 20 yr using a transgene from a resistant bacterial EPSP synthase that renders the crops insensitive to glyphosate. Much of the forage or grain from these crops are consumed by farm animals. Glyphosate protects crop yields, lowers the cost of feed production, and reduces CO2 emissions attributable to agriculture by reducing tillage and fuel usage. Despite these benefits and even though global regulatory agencies continue to reaffirm its safety, the public hears conflicting information about glyphosate's safety. The U.S. Environmental Protection Agency determines for every agricultural chemical a maximum daily allowable human exposure (called the reference dose, RfD). The RfD is based on amounts that are 1/100th (for sensitive populations) to 1/1,000th (for children) the no observed adverse effects level (NOAEL) identified through a comprehensive battery of animal toxicology studies. Recent surveys for residues have indicated that amounts of glyphosate in food/feed are at or below established tolerances and actual intakes for humans or livestock are much lower than these conservative exposure limits. While the EPSP synthase of some bacteria is sensitive to glyphosate, in vivo or in vitro dynamic culture systems with mixed bacteria and media that resembles rumen digesta have not demonstrated an impact on microbial function from adding glyphosate. Moreover, one chemical characteristic of glyphosate cited as a reason for concern is that it is a tridentate chelating ligand for divalent and trivalent metals; however, other more potent chelators are ubiquitous in livestock diets, such as certain amino acids. Regulatory testing identifies potential hazards, but risks of these hazards need to be evaluated in the context of realistic exposures and conditions. Conclusions about safety should be based on empirical results within the limitations of model systems or experimental design. Three trials with 156 crossbred heifers were used to determine the effects of dietary energy and bovine somatotropin administration on subsequent heifer productivity. In Trial 1, heifer calves were weaned from their dams (n = 28; 113 +/- 13 d of age) and assigned to a 2 x 2 factorial arrangement of moderate (MDE) or high dietary energy (HDE) and injections of vehicle (VEH) or 250 mg of bovine somatotropin (bST) every 14 d. Heifer calves in Trial 2 (n = 28; 123 +/- 20 d of age) and Trial 3 (n = 100; 134 +/- 22 d of age) nursed their dams while grazing pasture and received no creep feed (MDE) or ad libitum access to creep feed (HDE) and received VEH or bST as in Trial 1. Treatments were administered for 112 d and heifers within trial were managed alike after that time. There were no dietary treatment x bST treatment interactions for any of the variables analyzed (P > .05). Dietary energy did not affect daily gain in Trial 1; however, heifers receiving HDE in Trials 2 and 3 had greater weight, hip height growth, and fat thickness (P < .01) by the end of the treatment period. Treatment with bST increased gain in Trials 1 and 2 (P < .01) but did not affect growth in Trial 3. There were trends in all trials for HDE to reduce age at puberty (P < .15), but there were no consistent effects of bST on reproductive measures. Mammary gland composition and milk production were unaffected by treatment in Trial 1. In Trial 2, HDE decreased subsequent milk production, calf weaning weight, and mammary dry fat free tissue and DNA (P < .05). Treatment with bST in Trial 3 tended (P = .08) to result in greater milk production and increased calf weaning weights (P < .05). We conclude that bST treatment may enhance growth with no effect on reproduction. Treatment with bST did not overcome deleterious effects of HDE on milk production but may play an important role in mammogenesis.

Insulin-like growth factors (IGF) are important mediators of growth, lactation, reproduction, and health. Considerable information on their role in ruminant animals has been learned in the past several years, but the precise mechanisms of their action are not known. The exact biological response of target cells is undoubtedly determined by the developmental state of the cell and synergism with other growth factors. Overall, somatomedins and their binding proteins seem to be major links between cellular developmental processes and nutrient supply. The mechanism by which nutrients control biological actions of somatomedins is not known but clearly involves the synthesis of IGF, as well as their binding proteins and receptors. In ruminants, severe feed restriction decreases circulating concentrations of IGF-I, whereas subtle alterations typical of those that occur in production systems have minimal effect. However, the responses of IGF to somatotropin are affected by modest alterations in nutritional status, including differences in nutritional status that are typically encountered in animal production systems.

To assess the potential of ytterbium chloride (Yb) as an indicator of fecal output, we dosed 13 mature beef cows with Yb and chromic oxide (Cr2O3) either once (0800) or twice (0800 and 1600 hr) daily. Fecal output and digestible dry matter were estimated from fecal marker concentration determined in grab samples collected at 0800, 1600 or 0800 and 1600 hr, and compared with values obtained by total fecal collection. With single daily dosings, fecal outputs estimated from Yb at all collection times did not differ from values obtained by total collection, whereas Cr2O3 underestimated (P greater than .05) fecal production at 0800 hr and in the composited (0800 and 1600 hr) collections. With twice-daily dosing, fecal outputs estimated from Yb differed to a greater extent from the total collection values for the 1600 hr collection; however, the Cr2O3 values more closely approximated those estimated by total fecal collection at 0800 hours. Relative marker concentration in the feces collected from grab samples at 4.hr intervals indicated that twice-daily dosing appeared to reduce diurnal variation. Diurnal fecal excretion patterns of Yb and Cr2O3 differed (P greater than .05); however, the variation observed for both markers within dosing schedule appeared to be as great for Yb as it was for Cr2O3. Although fecal output was accurately estimated from Yb with one simultaneous collection and dosing period, the response appeared to be due to time of collection rather than to an actual reduction in diurnal variation.
