High-resolution colposcopic optical coherence tomography (OCT) provides key anatomical measures, such as thickness and minor traumatic injury of vaginal epithelium, of the female reproductive tract noninvasively. This information can be helpful in both fundamental investigations in animal models and disease screenings in humans. We present a fiber-based visible-light OCT and two probe designs for colposcopic application. One probe conducts circular scanning using a DC motor, and the other probe is capable of three-dimensional imaging over a 4.6 x 4.6-mm area using a pair of galvo scanners. Using this colposcopic vis-OCT with both probes, we acquired high-resolution images from whole isolated macaque vaginal samples and identified biopsy lesions. [ABSTRACT FROM AUTHOR]

We investigated two-photon absorption-based photoacoustic generation and compared it with corresponding photoluminescence emission. Experimental results revealed expected quadratic dependences on the incident optical fluence in both photoacoustic and photoluminescence processes. We also investigated the influence of optical scattering on the generation of two-photon photoacoustic and photoluminescence signals and found that photoacoustic signals attenuated more slowly than photoluminescence signals when the optical scattering coefficient was increased, which was attributed to a weaker ultrasonic attenuation than that the optical attenuation in the scattering medium. Retinal hemoglobin oxygen saturation (sO2) level is believed to be associated with the pathophysiology of several leading blinding diseases. Methods to properly measure retinal sO2 have been investigated for decades; however, the accuracy of retinal oximetry is still considered to be limited. The Monte Carlo simulation of photon transport in retina to examine how the accuracy of retinal oximetry is affected by local parameters is discussed. Fundus photography was simulated in a multilayer retinal model, in which a single vessel segment with 0.7 sO2 was embedded, at six optical wavelengths. Then, 200 million photons were traced in each simulation to ensure statistically stable results. The optical reflectance and energy deposit were recorded to measure sO2 using both the reflection method (existing retinal oximetry) and a new absorption method, photoacoustic ophthalmoscopy (PAOM). By varying the vessel diameter and melanin concentration in the retinal pigment epithelium, the relative error of sO2 measurement in the reflection method increased with increasing vessel diameter and melanin concentration; in comparison, the sO2 measurement was insensitive to these two parameters in PAOM. We have developed an automatic algorithm to detect the skin profiles in the volumetric data acquired by photoacoustic microscopy for subcutaneous vasculature imaging. This algorithm analyzes the relationship between amplitudes of photoacoustic signals generated from the skin surface and underlying blood vessels to achieve a rough estimation of the skin profile. A better approximation of the skin profile is then acquired after nonparametric smoothing and Gaussian low-pass spatial filtering. An auto-fit scan mechanism is further developed based on the detected skin profile to achieve good ultrasonic focusing on the subcutaneous vessel layer when the skin contour variation is much larger than the ultrasonic focal zone. The importance of skin profile detection in calculating the maximum-amplitude-projection images and significantly improving the image quality by employing the auto-fit scan are demonstrated by in vivoexperimental results.
