Abstract

Biochemistry students can be exposed to many analytical techniques through the fluorescent measurement of 2-p-toludinylnapthalene-6-sulfonate (TNS) binding to calmodulin. TNS binds to calmodulin only in the presence of Ca(II) ions which are known to expose a hydrophobic enzyme-binding site when bound to calmodulin. A significant fluorescence increase is recording when TNS binds to calmodulin that can be analyzed using a hyperbolic saturation curve and a Job plot. Experiment procedures are included.
