Five d-amino acids have been used for the first time to synthesize chiral derivatizing reagents (as variants of Marfey's reagent) by nucleophilic displacement of one of the fluorine atoms in 1,5-difluoro-2,4-dinitrobenzene as against the literature reports on application of only l-amino acids or their amides as chiral auxiliaries in dinitrobenzene (DNB) moiety. Five other DNB based reagents were also prepared by nucleophilic substitution of fluorine atom with the set of the same amino acids in l-configuration, as chiral auxiliaries. These reagents were characterized and used for synthesis of diastereomers of (RS)-Mexiletine spiked in human plasma. Diastereomers were synthesized employing microwave irradiation and were separated on reversed-phase C18 column. Performance of the two types of chiral derivatizing reagents was compared. The reagents containing d-amino acids provided enhanced separation of diastereomers than those containing l-amino acids. The best resolution was obtained using mobile phase consisting of acetonitrile and 0.1% trifluoroacetic acid in gradient mode. The detection was carried out at 340nm. The method so developed was validated for linearity, accuracy and precision. The limit of quantitation was found to be approximately 25.2ngmL(-1) in human plasma., (Copyright © 2014 Elsevier B.V. (S)-Naproxen-benzotriazole was synthesized by the reaction of (S)-naproxen with 1H-benzotriazole using coupling reagent dicyclohexyl carbodiimide and 4-dimethylamino pyridine (DCC/DMAP). It was used as chiral derivatizing reagent for microwave irradiated synthesis of diastereomers of penicillamine, cysteine and homocysteine. The diastereomers were separated by reversed phase high performance liquid chromatography using gradient elution of triethylammonium phosphate (pH 3.5)-acetonitrile (30-65% within 30 min). A new series of chiral derivatizing reagents (CDRs) consisting of four dichloro-s-triazine reagents was synthesized by nucleophilic substitution of one chlorine atom in trichloro-s-triazine with amino acids, namely L-Leu, D-Phg, L-Val and L-Ala as chiral auxiliaries. Two other sets of CDRs consisting of four dichloro-s-triazine (DCT) and six monochloro-s-triazine (MCT) reagents were also prepared by nucleophilic substitution of chlorine atom(s) with different amino acid amides as chiral auxiliaries in trichloro-s-triazine and its 6-methoxy derivative, respectively. These 14 CDRs were used for the synthesis of diastereomers of (R,S)-mexiletine under microwave irradiation (i.e. 60s and 90 s at 85% power (of 800 W) using DCT and MCT reagents, respectively), which were resolved by reversed-phase high-performance liquid chromatography using C18 column and gradient eluting mixtures of methanol with aqueous trifluoroacetic acid (TFA) with UV detection at 230 nm. The resolution (R(s)), difference between retention times of resolved diastereomers (Δt) and retention factors (k) obtained for the three sets of diastereomers were compared among themselves and among the three groups. Explanations have been offered for longer retention times and better resolution of diastereomers prepared with DCT reagents in comparison of their MCT counterparts and, for the influence of hydrophobicity of the side chain R of the amino acid in the CDRs on retention times and resolution. The newly synthesized CDRs were observed to be superior as compared to their amide counterparts in terms of providing better resolution and cost effectiveness. Four dichloro-s-triazine (DCT) and five monochloro-s-triazine (MCT) chiral derivatizing reagents (CDRs) were synthesized by incorporating amino acid amide moieties as chiral auxiliaries in trichloro-s-triazine and its 6-methoxy derivative, respectively. Another MCT reagent was synthesized by substitution of two chlorine atoms with two different amino acid amides in trichloro-s-triazine. These reagents were used for synthesis of diastereomers of (R,S)-baclofen under microwave irradiation (i.e. 60 s at 85% power using DCT reagents and 90 s at 85% power using MCT reagents). The diastereomers were separated on a reversed-phase C18 column using mixtures of methanol with aqueous trifluoroacetic acid (TFA) with UV detection at 230 nm. The separation behavior in terms of retention times and resolutions obtained for the two sets of diastereomers prepared with DCT and MCT reagents were compared among themselves and among the two groups. Longer retention times and better resolutions were observed with DCT reagents as compared to MCT reagents. The calibration curves were linear for both (R)- and (S)-baclofen in the concentration range 50-500 μg/ml. The average regression was 0.999 for both (R)- and (S)-baclofen. The RSD for (R)-baclofen was 0.40-0.86% for intra-day precision and 0.60-1.40% for inter-day precision and these values for (S)-baclofen were 0.52-0.75% and 0.64-1.32%, respectively. The recovery was 97.2-98.9% for (R)- and 97.0-98.9% for (S)-baclofen. The limit of detection was 1.63 ng/ml and 1.52 ng/ml for (R)- and (S)-baclofen, respectively., (Copyright © 2010 Elsevier B.V. Taking into account the structural similarities of amino alcohols with amino acids and in order to reduce time for derivatization unconventional approaches viz. microwave irradiation, ultrasonication and ultra centrifugation were applied for synthesis of dinitrophenyl derivatives of nine amino alcohols and to work out a method of choice for sample preparation for direct enantioseparation. The enantiomeric dinitrophenyl derivatives so synthesized were separated on alpha(1)-acid glycoprotein and beta-cyclodextrin columns with detection at 230nm using photodiode array detection system. Derivatization methods and chromatographic parameters were optimized. beta-Cyclodextrin column was found better compared to AGP column for enantioseparation. Limit of detection, quantification, accuracy and precision were also determined. The developed method was successfully applied to determine enantiomeric purity of 1-amino-2-propanol obtained from vitamin B(12) hydrolysate.

Four chiral derivatizing reagents (CDRs) having enantiomerically pure amines and two CDRs namely, [N-succinimidyl-(S)-2-(6-methoxynaphth-2-yl)propionate], and [dinitrophenyl-L-Pro-N-hydroxysuccinimide ester, DNP-L-Pro-SU] were synthesized and were used to prepare diastereomers of (R,S)-mexiletine (MEX); these were separated by reversed-phase high-performance liquid chromatography (RP-HPLC). The method was validated for linearity, accuracy, limit of detection (LOD) and limit of quantification (LOQ).

Two sulfur-containing amino acids, DL-cysteine (Cys) and DL-penicillamine (PenA), were condensed with ninhydrin to form their spirothiazolidine derivatives. These were separated by HPLC using alpha-acid glycoprotein (AGP) and beta-cyclodextrin (beta-CD) columns. The resolution conditions were optimized and the results were compared. Since the method provided resolution greater than 2 it was also applied to preparative separation. After separation, each of them was detagged using Zn dust and 10% aqueous trifluoroacetic acid. For analytical purposes dinitrophenyl (DNP) derivatives of DL-Cys and DL-PenA were also prepared and were resolved on both the columns. The detection was carried out using photodiode array detection system at 231 nm. The limits of detection were found to be 0.01% and 0.004% for spirothiazolidine carboxylic acid and DNP derivatives, respectively.

Ten chiral derivatizing reagents (CDRs) were synthesized by replacing the L-Ala-NH(2) moiety in Marfey's reagent (MR) by seven L-amino acid amides and three L-amino acids employing microwave irradiation (MW) and were characterized. Ten racemic amino alcohols were derivatized with these CDRs under MW. The diastereomers were separated on a reversed-phase C(18) column using binary mixtures of acetonitrile with aqueous trifluoroacetic acid (TFA) and triethylammonium phosphate buffer (TEAP). In general, amino acid variants of MR provided better separation of diastereomers in comparison to amino acid amide variants. The method was also found successful for the separation of 20 diastereomers from a mixture.

Synthesis of N-succinimidyl-(S)-2-(6-methoxynaphth-2-yl) propionate was carried out by the reaction of (S)-naproxen with N-hydroxysuccinimide in the presence of dicyclohexyl carbodiimide. It was characterized and was used as a chiral derivatizing reagent, under mild conditions, to form diastereomers of dl-penicillamine which were resolved by reversed-phase high-performance liquid chromatography using triethyl ammonium phosphate buffer (pH 4.0, 5mM)-acetonitrile (linear gradient (30min) of acetonitrile from 30 to 70%). Excellent separation was achieved with gradient mobile phase. The detection limit was at pmol level.

Ten chiral dichloro- and monochloro-s-triazines were prepared by the nucleophilic displacement of chlorine atom(s) in s-triazine chloride and its 6-methoxy derivative with different amino acid amides. Dichloro-s-triazines (DCTs) were used as CDRs for derivatization of alpha-amino acids under basic conditions at room temperature (30 degrees C) while derivatization with monochloro-s-triazine (MCT) reagents was carried out at 80 degrees C. The resultant diastereomers were separated on a reversed-phase C(18) column using mixtures of acetonitrile and aqueous-trifluoroacetic acid (TFA). The separation results for the two were compared. One DCT reagent was optimized for derivatization kinetics with respect to the effects of pH, reagent excess, temperature and reaction time on derivatization yield. In most of the cases, DCT reagents provided better separation of diastereomers in comparison to MCT reagents. One DCT reagent was also validated for limit of detection, linearity, recovery and robustness. Effects of structural modifications in reagents on chromatographic properties were investigated. Separation mechanism of diastereomers was proposed in light of both MCT and DCT reagents.

Five new chiral derivatizing reagents 5-hydrazino-2,4-dinitrophenyl-l-alaninamide (HDNP-l-Ala-NH2), 5-hydrazino-2,4-dinitrophenyl-l-phenylalaninamide (HDNP-l-Phe-NH2), 5-hydrazino-2,4-dinitrophenyl-l-valinamide (HDNP-l-Val-NH2), 5-hydrazino-2,4-dinitrophenyl-l-leucinamide (HDNP-l-Leu-NH2) and 5-hydrazino-2,4-dinitrophenyl-l-phenylglycinamide (HDNP-d-Phg-NH2) were synthesized by straightforward two-step synthesis starting from 1,5-difluoro-2,4-dinitrobenzene. Nucleophilic substitution of one fluorine atom in DFDNB with different amino acid amides yielded Marfey's reagent (5-fluoro-2,4-dinitrophenyl-l-alaninamide) and its structural variants (5-fluoro-2,4-dinitrophenyl-l-phenylalaninamide, 5-fluoro-2,4-dinitrophenyl-l-valinamide, 5-fluoro-2,4-dinitrophenyl-l-leucinamide and 5-fluoro-2,4-dinitrophenyl-d-phenylglycinamide). Chiral hydrazine reagents were prepared by nucleophilic substitution of remaining fluorine atom in Marfey's reagent and its variants with hydrazine under basic conditions. These reagents react quantitatively with chiral carbonyl compounds under mild conditions (30 degrees C, 30 min) to form hydrazone diastereomers. The labeling reaction occurs only in the presence of acid which has a catalytic action and diastereomers have strong absorbance around 348 nm. The separation of diastereomers was tried on a reversed-phase C18 HPLC column using different binary solvent combinations. Excellent separation was achieved in case of cyclic ketones having substitution at alpha-position. Optimization for derivatization yield, limit of detection, limit of quantification, linearity, accuracy and precision was carried out with respect to HDNP-l-Val-NH2. Studies related to effects of structural modification in reagents and analytes on chromatographic behavior of diastereomers were also analyzed.
