Snails belonging to the genus Biomphalaria serve as obligatory intermediate hosts for the trematode Schistosoma mansoni, the causative agent for the most widespread form of schistosomiasis. The simpler nervous systems of gastropod molluscs, such as Biomphalaria, provide advantageous models for investigating neural responses to infection at the cellular and network levels. The present study examined neuropeptides related to cholecystokinin (CCK), a major multifunctional regulator of central nervous system (CNS) function in mammals. A neural transcriptome generated from the CNS of Biomphalaria alexandrina included a transcript encoding two CCK-related peptides, designated Balex-CCK1 (pEGEWSYDY (SO 3 H) GLGGGRF-NH 2 ) and Balex-CCK2 (NYGDY (SO 3 H) GIGGGRF-NH 2 ). Peptide expression was examined in Biomphalaria glabrata at the mRNA level using the hybridization chain reaction (HCR) protocol and at the protein level using an antibody against Balex-CCK1. Expression was detected in 60-70 neurons distributed throughout the CNS, as well as in profuse fiber systems connecting the ganglia and projecting to the periphery. CCK-like immunoreactive (CCK li ) fibers were also observed on organs associated with the cardiorespiratory (nephridium, mantle, gill) and male reproductive systems. A comparison of mRNA and peptide localization suggested that CCK expression could be regulated at the level of translation. Freshwater snails of the genus Biomphalaria serve as intermediate hosts for the digenetic trematode Schistosoma mansoni, the etiological agent for the most widespread form of intestinal schistosomiasis. As neuropeptide signaling in host snails can be altered by trematode infection, a neural transcriptomics approach was undertaken to identify peptide precursors in Biomphalaria glabrata, the major intermediate host for S. mansoni in the Western Hemisphere. Three transcripts that encode peptides belonging to the FMRF-NH 2 -related peptide (FaRP) family were identified in B. glabrata. One transcript encoded a precursor polypeptide (Bgl-FaRP1; 292 amino acids) that included eight copies of the tetrapeptide FMRF-NH 2 and single copies of FIRF-NH 2 , FLRF-NH 2 , and pQFYRI-NH 2 . The second transcript encoded a precursor (Bgl-FaRP2; 347 amino acids) that comprised 14 copies of the heptapeptide GDPFLRF-NH 2 and 1 copy of SKPYMRF-NH 2 . The precursor encoded by the third transcript (Bgl-FaRP3; 287 amino acids) recapitulated Bgl-FaRP2 but lacked the full SKPYMRF-NH 2 peptide. The three precursors shared a common signal peptide, suggesting a genomic organization described previously in gastropods. Immunohistochemical studies were performed on the nervous systems of B. glabrata and B. alexandrina, a major intermediate host for S. mansoni in Egypt. FMRF-NH 2 -like immunoreactive (FMRF-NH 2 -li) neurons were located in regions of the central nervous system associated with reproduction, feeding, and cardiorespiration. Antisera raised against non-FMRF-NH 2 peptides present in the tetrapeptide and heptapeptide precursors labeled independent subsets of the FMRF-NH 2 -li neurons. Within Biomphalaria, S. mansoni larvae multiply and transform into the cercariae form that can infect humans. Trematode development and proliferation is thought to be facilitated by modifications of host behavior and physiological processes, including a reduction of reproduction known as "parasitic castration." As neuropeptides participate in the control of reproduction across phylogeny, a neural transcriptomics approach was undertaken to identify peptides that could regulate Biomphalaria reproductive physiology. The present study identified a transcript in Biomphalaria alexandrina that encodes a peptide belonging to the gonadotropin-releasing hormone (GnRH) superfamily. The precursor and the predicted mature peptide, pQIHFTPDWGNN-NH 2 (designated Biom-GnRH), share features with peptides identified in other molluscan species, including panpulmonates, opisthobranchs, and cephalopods. An antibody generated against Biom-GnRH labeled neurons in the cerebral, pedal, and visceral ganglia of Biomphalaria glabrata. GnRH-like immunoreactive fiber systems projected to all central ganglia. In the periphery, immunoreactive material was detected in the ovotestis, oviduct, albumen gland, and nidamental gland. The simpler nervous systems of certain invertebrates provide opportunities to examine colocalized classical neurotransmitters in the context of identified neurons and well defined neural circuits. This study examined the distribution of γ-aminobutyric acid-like immunoreactivity (GABAli) in the nervous system of the panpulmonates Biomphalaria glabrata and Biomphalaria alexandrina, major intermediate hosts for intestinal schistosomiasis. GABAli neurons were localized in the cerebral, pedal, and buccal ganglia of each species. With the exception of a projection to the base of the tentacle, GABAli fibers were confined to the CNS. As GABAli was previously reported to be colocalized with markers for dopamine (DA) in five neurons in the feeding network of the euopisthobranch gastropod Aplysia californica (Díaz-Ríos, Oyola, & Miller, 2002), double-labeling protocols were used to compare the distribution of GABAli with tyrosine hydroxylase immunoreactivity (THli). As in Aplysia, GABAli-THli colocalization was limited to five neurons, all of which were located in the buccal ganglion. Five GABAli-THli cells were also observed in the buccal ganglia of two other intensively studied panpulmonate species, Lymnaea stagnalis and Helisoma trivolvis. These findings indicate that colocalization of the classical neurotransmitters GABA and DA in feeding central pattern generator (CPG) interneurons preceded the divergence of euopisthobranch and panpulmonate taxa. Planorbid snails of the genus Biomphalaria are major intermediate hosts for the digenetic trematode parasite Schistosoma mansoni. Evidence suggests that levels of the neurotransmitter dopamine (DA) are reduced during the course of S. mansoni multiplication and transformation within the snail. This investigation used immunohistochemical methods to localize tyrosine hydroxylase (TH), the rate-limiting enzyme in the biosynthesis of catecholamines, in the nervous system of Biomphalaria. The two species examined, Biomphalaria glabrata and Biomphalaria alexandrina, are the major intermediate hosts for S. mansoni in sub-Saharan Africa, where more than 90% of global cases of human intestinal schistosomiasis occur. TH-like immunoreactive (THli) neurons were distributed throughout the central nervous system (CNS) and labeled fibers were present in all commissures, connectives, and nerves. Some asymmetries were observed, including a large distinctive neuron (LPeD1) in the pedal ganglion described previously in several pulmonates. The majority of TH-like immunoreactive neurons were detected in the peripheral nervous system (PNS), especially in lip and foot regions of the anterior integument. Independent observations supporting the dopaminergic phenotype of THli neurons included 1) block of LPeD1 synaptic signaling by the D2/3 antagonist sulpiride, and 2) the similar localization of aqueous aldehyde (FaGlu)-induced fluorescence. The distribution of THli neurons indicates that, as in other gastropods, dopamine functions as a sensory neurotransmitter and in the regulation of feeding and reproductive behaviors in Biomphalaria. The digenetic trematode Schistosoma mansoni that causes the form of schistosomiasis found in the Western Hemisphere requires the freshwater snail Biomphalaria glabrata as its primary intermediate host. It has been proposed that the transition from the free-living S. mansoni miracidium to parasitic mother sporocyst depends on uptake of biogenic amines, e.g. serotonin, from the snail host. However, little is known about potential sources of serotonin in B. glabrata tissues. This investigation examined the localization of serotonin-like immunoreactivity (5HTli) in the central nervous system (CNS) and peripheral tissues of B. glabrata. Emphasis was placed on the cephalic and anterior pedal regions that are commonly the sites of S. mansoni miracidium penetration. The anterior foot and body wall were densely innervated by 5HTli fibers but no peripheral immunoreactive neuronal somata were detected. Within the CNS, clusters of 5HTli neurons were observed in the cerebral, pedal, left parietal, and visceral ganglia, suggesting that the peripheral serotonergic fibers originate from the CNS. Double-labeling experiments (biocytin backfill × serotonin immunoreactivity) of the tentacular nerve and the three major pedal nerves (Pd n. 10, Pd n. 11, and Pd n. 12) disclosed central neurons that project to the cephalopedal periphery. Overall, the central distribution of 5HTli neurons suggests that, as in other gastropods, serotonin regulates the locomotion, reproductive, and feeding systems of Biomphalaria. The projections to the foot and body wall indicate that serotonin may also participate in defensive, nociceptive, or inflammation responses. This study examined the catecholaminergic and serotonergic innervation of the foregut of Aplysia californica, a model system in which the control of feeding behaviors can be investigated at the cellular level. Similar numbers (15-25) of serotonin-like-immunoreactive (5HTli) and tyrosine hydroxylase-like-immunoreactive (THli) fibers were present in each (bilateral) esophageal nerve (En), the major source of pregastric neural innervation in this system. The majority of En 5HTli and THli fibers originated from the anterior branch (En(2)), which innervates the pharynx and the anterior esophagus. Fewer fibers were present in the posterior branch (En(1)), which innervates the majority of the esophagus and the crop. Backfills of the two En branches toward the central nervous system (CNS) labeled a single, centrifugally projecting serotonergic fiber, originating from the metacerebral cell (MCC). The MCC fiber projected only to En(2). No central THli neurons were found to project to the En. Surveys of the pharynx and esophagus revealed major differences between their patterns of catecholaminergic (CA) and serotonergic innervation. Whereas THli fibers and cell bodies were distributed throughout the foregut, 5HTli fibers were present in restricted plexi, and no 5HTli somata were detected. Double-labeling experiments in the periphery revealed THli neurons projecting toward the buccal ganglion via En(2). Other afferents received dense perisomatic serotonergic innervation. Finally, qualitative and quantitative differences were observed between the buccal motor programs (BMPs) produced by stimulation of the two En branches. These observations increase our understanding of aminergic contributions to the pregastric regulation of Aplysia feeding behaviors.

A previous study reported that a peptide, sensorin-A, is expressed exclusively in mechanosensory neurons having somata in central ganglia of Aplysia. The present study utilized in situ hybridization, staining by nerve back-fill and soma injection, and electrophysiological methods to characterize the locations, numbers, and functions of sensorin-A-expressing neurons and to define the relationships between soma locations and the locations of peripheral axons and receptive fields. Approximately 1,000 cells express sensorin-A mRNA in young adult animals (10-30 g) and 1,200 cells in larger adults (100-300 g). All of the labeled somata are in the CNS, primarily in the abdominal LE, rLE, RE and RF, pleural VC, cerebral J and K, and buccal S clusters. Expression also occurs in a few sparsely distributed cells in most ganglia. Together, receptive fields of all these mechanosensory clusters cover the entire body surface. Each VC cluster forms a somatotopic map of the ipsilateral body, a "sensory aplunculus." Cells in the pleural and cerebral clusters have partially overlapping sensory fields and synaptic targets. Buccal S cells have receptive fields on the buccal mass and lips and display notable differences in electrophysiological properties from other sensorin-A-expressing neurons. Neurons in all of the clusters have relatively high mechanosensory thresholds, responding preferentially to threatening or noxious stimuli. Synaptic outputs to target cells having defensive functions support a nociceptive role, as does peripheral sensitization following noxious stimulation, although additional functions are likely in some clusters. Interesting questions arise from observations that mRNA for sensorin-A is present not only in the somata but also in synaptic regions, connectives, and peripheral fibers., (Copyright 2004 Wiley-Liss, Inc.)

The effects of prenatal ethanol exposure on the spatiotemporal expression of transforming growth factor-beta (TGFbeta) and its receptors in developing rat cerebral cortex in vivo were examined. Pregnant Long-Evans rats were fed ad libitum with a diet containing ethanol from gestational day (G) 6 through G21 or were pair fed an isocaloric nonalcoholic liquid diet. A quantitative immunoblotting study showed that expression of TGFbeta ligands was differentially affected by ethanol; ethanol decreased TGFbeta1 expression fetally and in the mature cortex and increased TGFbeta2 at most ages. A complementary immunohistochemical experiment generated similar results so far as the timing of ligand expression was concerned. In both control and ethanol-treated rats, TGFbeta1 was expressed by cells in the two neocortical proliferative zones and neurons in the cortical plate. TGFbeta2 was expressed principally by radial glia and astrocytes in developing rats. In the adult, both ligands were expressed by glia and neurons. Ethanol virtually eliminated the TGFbeta1 expression in the perinatal subventricular zone. The TGFbeta2-positive radial glial labeling was transient and was lost earlier in ethanol-treated neonates than in controls. Concomitantly, the appearance of TGFbeta2-positive glia occurred earlier in the ethanol-treated rats. The expression of only one receptor (TGFbetaIr) was affected by ethanol; it was increased during the pre- and early postnatal periods. TGFbetaIr was expressed by glia perinatally and by all cell types in weanlings. As with TGFbeta2, ethanol exposure promoted the loss of TGFbetaIr expression in radial glia and the precocious expression among astrocytes. TGFbetaIIr was expressed primarily by neurons. Thus, TGFbeta ligands and receptors are strategically placed both in time and space to regulate cell proliferation and migration. Neurotrophins are critical for neuronal development, plasticity, and survival. Ethanol affects these processes. We tested the hypothesis that ethanol inhibits nerve growth factor (NGF)-stimulated gene expression. Dissociated cultures of fetal cortical neurons were treated with NGF and/or ethanol. NGF sustained cell viability and reduced the incidence of terminal uridylated nick-end labeling and pyknosis. Ethanol eliminated these effects and induced neuronal death. Differential display of mRNA showed that one gene fragment (245 bp) was expressed by cells treated with NGF alone; ethanol blocked its expression. This fragment, named neg (nerve growth factor-stimulated, ethanol-depressed gene), has high nucleotide identity with genes from human myeloid cells and murine lymphocytes. Ribonuclease protection assay and in situ hybridization verified NGF upregulation and ethanol antagonism. Functional consequences of neurotransmitter coexistence and cotransmission can be readily studied in certain experimentally favorable invertebrate motor systems. In this study, whole-mount histochemical methods were used to identify neurons in which gamma-aminobutyric acid (GABA)-like immunoreactivity (GABAli) was colocalized with catecholamine histofluorescence (CAh; FaGlu method) and tyrosine hydroxylase (TH)-like immunoreactivity (THli) in the feeding motor circuitry (buccal and cerebral ganglia) of the marine mollusc Aplysia californica. In agreement with previous reports, five neurons in the buccal ganglia were found to exhibit CAh. These included the paired B20 buccal-cerebral interneurons (BCIs), the paired B65 buccal interneurons, and an unpaired cell with projections to both cerebral-buccal connectives (CBCs). Experiments in which the FaGlu method was combined with the immunohistochemical detection of GABA revealed double labeling of all five of these neurons. An antibody generated against TH, the rate-limiting enzyme in the biosynthesis of catecholamines, was used to obtain an independent determination of GABA-CA colocalization. Biocytin backfills of the CBC performed in conjunction with TH immunohistochemistry revealed labeling of the rostral B20 cell pair and the unpaired CBI near the caudal surface of the right hemiganglion. THli was also present in a prominent bilateral pair of caudal neurons that were not stained with CBC backfills. On the basis of their position, size, shape, and lack of CBC projections, the lateral THli neurons were identified as B65. Double-labeling immunohistochemical experiments revealed GABAli in all five buccal THli neurons. Finally, GABAli was observed in individual B20 and B65 neurons that were identified using electrophysiological criteria and injected with a marker (neurobiotin). Similar methods were used to demonstrate that a previously identified catecholaminergic cerebral-buccal interneuron (CBI) designated CBI-1 contained THli but did not contain GABAli. Although numerous THli and GABAli neurons and fibers were present in the cerebral and buccal ganglia, additional instances of their colocalization were not observed. These findings indicate that GABA and a catecholamine (probably dopamine) are colocalized in a limited number of interneurons within the central pattern generator circuits that control feeding-related behaviors in Aplysia.

Crustacean muscles are unique in their innervation by both excitatory and inhibitory neurons; therefore, they exhibit polyneuronal and multiterminal innervation. Because excitatory motoneurons are broadly divided into phasic and tonic types, we hypothesized that inhibitory neurons would follow a similar dichotomy. The abdominal extensor muscles in crayfish are separated into parallel deep and superficial bundles; the former has fast muscle fibers innervated by phasic excitatory motoneurons, and the latter has slow fibers supplied by tonic excitatory motoneurons. Each muscle also is innervated by a single, separate inhibitory neuron that uses gamma-aminobutyric acid (GABA) as the inhibitory neurotransmitter. The pattern of axonal branching by the separate inhibitory axons in phasic and tonic abdominal extensor muscles was visualized with confocal microscopy in preparations labeled for GABA-like immunoreactivity. Initial observations indicated that the phasic muscle was covered by extensive GABAergic, filiform axon terminals, whereas innervation of the tonic muscle was comprised of more localized and varicose terminals. With quantitative analyses, we found that the phasic axon has a more highly branched nature than the tonic in first- and second-order branches. The phasic axon branches also were significantly longer than the tonic branches in the second- and third-order branches. Synaptic varicosities in the phasic branches were smaller and less frequent than those in the tonic branches. The fine structure of the inhibitory nerve terminals near synaptic contacts examined with thin-serial-section electron microscopy revealed distinct differences between the phasic system and the tonic system. The phasic terminals were smaller in cross-sectional area than the tonic terminals, and they had smaller synapses and fewer mitochondria. The presynaptic active zone dense bodies were similar in length and number between phasic and tonic synapses. However, their number per synaptic area was two-fold higher in phasic synapses compared with tonic synapses because of the smaller size of the phasic synapses. Neurotrophins, including nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), and neurotrophin-3 (NT-3), are critical for the maintenance and plasticity of central nervous system (CNS) neurons. We tested the hypothesis that cortical neurons participate in redundant autocrine/paracrine systems. Three sets of studies determined the distribution of NGF-, BDNF-, and NT-3-expressing neurons, the frequency of neurons coexpressing NGF and BDNF, and the frequency of neurons expressing a neurotrophin and its associated high-affinity receptor. The distribution of NGF-, BDNF, and NT-3-immunoreactive neurons was identical. Neurotrophin-positive cells were parceled throughout the cortex, although the labeling frequency was not the same in all layers. More than 30% of the neurons in layers II/III, V, and VI were labeled, whereas only 5-10% of the neurons in layer IV was immunopositive for a neurotrophin. Some glia were also neurotrophin positive, particularly BDNF-positive glia. About 70% of the neurons in layers II/III and V coexpressed NGF and BDNF or coexpressed NGF and NT-3. Ligand-receptor colabeling was also common among cortical neurons. For example, nearly 70% of the NGF-, BDNF-, and NT-3-positive neurons in layer V colabeled with their respective high-affinity receptors, i.e., trkA, trkB, and trkC, respectively. Thus, (a) neurons express multiple neurotrophins and (b) cortical neurons (e.g., layer V neurons) contain the components required for autocrine/paracrine and/or anterograde communication (e.g., neurons in layer II/III support layer V neurons). These systems mean that the cortex is capable of regulating itself autonomously.

Gamma-aminobutyric acid (GABA) is present in the central nervous system of Aplysia californica (Gastropoda, Opisthobranchia) where its role as a neurotransmitter is supported by pharmacological, biochemical, and anatomical investigations. In this study, the distribution of GABA-immunoreactive (GABAi) neurons and fiber systems in Aplysia was examined by using wholemount immunohistochemistry and nerve backfill methods. GABAi neurons were located in the buccal, cerebral, and pedal ganglia. Major commissural fiber systems were present in each of these ganglia, whereas more limited fiber systems were observed in the ganglionic connectives. Some of the interganglionic fibers were found to originate from two unpaired GABAi neurons, one in the buccal ganglion and one in the right pedal ganglion, each of which exhibited bilateral projections. No GABAi fibers were found in the nerves that innervate peripheral sensory, motor, or visceral organs. Although GABAi cells were not observed in the pleural or abdominal ganglia, these ganglia did receive limited projections of GABAi fibers originating from neurons in the pedal ganglia. The distribution of GABAi neurons suggests that this transmitter system may be primarily involved in coordinating certain bilateral central pattern generator (CPG) systems related to feeding and locomotion. In addition, the presence of specific interganglionic GABAi projections also suggests a role in the regulation or coordination of circuits that produce components of complex behaviors.

The effects of prenatal exposure to ethanol on the number of callosal axons were examined. Pregnant Macaca nemestrina were treated with ethanol (1.8 g/kg b.wt.) 1 day per week during the first 6 weeks (Et6) or full 24 weeks (Et24) of gestation. Control macaques were intubated with an isocaloric amount of sucrose water (Ct). The mid-sagittal area of the corpus callosum in 4- to 5-year-old offspring was examined in magnetic resonance (MR) images and in fixed brains. The number of callosal axons was determined by using electron microscopy. In both MR images and fixed brains of macaques treated with ethanol, the corpus callosum was 26% larger than in the controls. The rostral portion was particularly affected by ethanol; it was 55% larger in Et6- and Et24-treated macaques. Axonal size and myelin thickness were unaffected by ethanol, but ethanol-treated macaques had more callosal axons (13.7 x 10(7)) than did controls (9.4 x 10(7) axons). The increase in the rostral corpus callosum suggests that parietal and frontal cortices are particularly susceptible to ethanol. The altered callosal connectivity may be a component of the structural abnormalities that underlie executive processing problems associated with fetal alcohol syndrome., (Copyright 1999 Wiley-Liss, Inc.)

Two genes (L5-67 and LUQ-1) that encode neuropeptide precursors have recently been shown to be expressed in a distinct and non-overlapping manner in the five left upper quadrant (LUQ) cells of the abdominal ganglion of Aplysia (Landry et al. [1992]. J. Neurobiol 23:89-101). By using wholemount immunohistochemistry and radioimmunoassay (RIA), the pattern of expression of these two genes was assessed at the protein level throughout the central nervous system (CNS) and in peripheral tissues of Aplysia californica. The distribution of LUQ-1 precursor-like immunoreactivity was fairly limited, occurring in the ventral LUQ cell (L5) and in a total of approximately 20 additional neurons in the abdominal and cerebral ganglia. L5-67 precursor-like immunoreactive material was more prevalent, appearing in a total of approximately 100 neurons distributed among each of the central ganglia. Identified L5-67-immunoreactive neurons included the four dorsal LUQ cells (L2-4 and L6) and two giant neurons (R2 and LPI1). In one group of cells, the H cluster of the cerebral ganglion, L5-67 immunofluorescence was substantially more intense in larger versus smaller animals, suggesting that this peptide precursor is subject to developmental regulation in certain neurons. Immunoelectron microscopic examination of the subcellular localization of L5-67 immunoreactivity in LUQ cell somata and axons revealed its association with dense-core vesicles (approximately 114 nm in diameter). In the periphery, L5-67-immunoreactive fibers were detected in specific regions of the circulatory system (auricle, ventricle, cristae aorta, anterior aorta) and the reproductive system (genital ganglion, large hermaphroditie duct, small hermaphroditie duct, ovotestis). The kidney and the intestine, two tissues in which considerable secretion and absorption occur, contained material immunoreactive to both L5-67 and LUQ-1 antisera. The localization of the two peptide precursors in these tissues differed substantially, with L5-67 occurring in widely ramifying varicose fibers, whereas LUQ-1 was found in restricted foci of fibers and in small spherical cells that appeared to lack processes. These results support previous findings concerning the heterogeneity of neurotransmitter phenotypes in the LUQ cells. Furthermore, they are indicative of a fairly broad role for the L5-67-derived neuropeptides, and a more limited role for the LUQ-1-derived neuropeptides, in the regulation of the visceral organ systems of Aplysia.

The c-neu oncoprotein, p185c-neu, is a transmembrane tyrosine kinase that shares structural similarities with the receptor for epidermal growth factor (EGFr). We used immunoblots, immunoprecipitation, and immunohistochemistry 1) to test the hypothesis that p185c-neu and EGFr are coordinately expressed in central nervous system tissue and 2) to assess the spatiotemporal expression of both the c-neu oncoprotein and EGFr in the rostral cerebral cortex. In nondenaturing gels, anti-c-neu antibody identified high molecular weight proteins (about 300-400 kDa) that were reduced by EDTA to a molecular weight of 180-200 kDa. Sodium dodecylsulfate polyacrylamide gel electrophoresis broke down this protein into an array of smaller peptides, which were expressed prenatally, transiently during the first three postnatal weeks, or in the adult. Perinatally, c-neu immunoreactivity was evident in subplate neurons, ascending processes of neurons in the cortical plate, and ventricular zone cells. During the second postnatal week, cells throughout cortex expressed somatodendritic immunostaining, but, in the adult, c-neu immunoreactivity was expressed only by pyramidal neurons in layer V and by glia in the white matter and ependyma. EGFr-positive proteins behaved in the nondenaturing gels as did c-neu-positive oncoproteins, suggesting that both proteins naturally formed dimers. This contention was supported by the EGFr-or c-neu immunolabeling of tissue that was previously immunoprecipitated with anti-c-neu or anti-EGFr, respectively. The pattern of EGFr immunolabeling in the developing and mature cortex was virtually identical to that described for c-neu immunoreactivity. Cortical neurons express the c-neu oncoprotein and EGFr, probably as heterodimers. The specific immunolabeling of layer V neurons in the adult cortex with anti-c-neu and anti-EGFr suggests that the p185c-neu ligand and EGF regulate the activity of corticofugal systems. The expression of different c-neu- and EGFr-positive peptides is developmentally defined and may be related to specific ontogenetic events.

The birth of a neuron initiates a series of ontogenetic events, e.g., neuronal migration and differentiation. The outcomes of these events are neurons that successfully integrate into the cortical circuitry and neurons that are unsuccessful and ultimately die. The present study determined whether there is a relationship between the generation and death of cortical neurons. The decrease in the density of postmigratory neurons (heavily labeled by a single injection of [3H]thymidine) during normal development was used as an index of neuronal death. The survival indices of neurons varied with their times of origin. Neurons born from gestational day (G) 15 to G18 had the highest rates of survival. In contrast, the earliest and latest generated neurons (i.e., those born on G12-G13 and those born on G19-G21, respectively) had the lowest survival rates. The role of neuronal death in the formation of cortical patterns was determined by assessing the survival of neurons in the barrels and septa of somatosensory cortex. No differences in the survival index were determined for neurons in the C-row barrels and adjacent septa with a particular time of origin. The survival rate of projection and local circuit neurons was determined with a double-labeling technique. One label, [3H]thymidine, was used to determine the time of origin of the neurons. The second label was used to identify the chemical or hodological characteristics of a neuron; projection neurons were labeled either by retrograde transport of horseradish peroxidase or by glutamate immunohistochemistry, and local circuit neurons were immunohistochemically identified with an antibody directed against gamma-aminobutyric acid (GABA) antibody.(ABSTRACT TRUNCATED AT 250 WORDS)

Postmitotic neurons migrate from a zone(s) near the ventricles to the neocortex. During this migration, neurons associate with radial glia. After serving their role as guides for neuronal migration, the radial glia transform into astrocytes. Prenatal exposure to ethanol causes abnormal neuronal migration. We examined the effects of gestational exposure to ethanol on radial glia and astrocytes. Radial glia were stained immunohistochemically with the antibody RAT-401, and astrocytes were labeled with an antibody directed against glial fibrillary acidic protein (GFAP). The subjects were the offspring of rats fed an ethanol-containing liquid diet (Et), pair-fed a liquid control diet (Ct), or fed chow and water (Ch). During the first postnatal week, radial glial fibers (in Et-treated rats and controls) stretched from the ventricular surface through the developing cerebral wall to the pial surface. In the Et-treated rats, the radial processes were less dense and more poorly fasciculated than they were in the Ch- and Ct-treated rats. Moreover, by postnatal day (P) 5, there was a significant reduction in RAT-401 immunostaining in the Et-treated rats, particularly in the superficial cortex. A similar reduction in control rats did not begin until P10. In all three treatment groups, GFAP-immunoreactive astrocytes were in the cortex throughout the period from P1 to P45. In neonates, GFAP-positive cells were distributed in the marginal zone (layer I) and the intermediate zone (the white matter). The number of GFAP-positive cells in the cortical plate increased steadily with time so that, by P26, GFAP-immunoreactive astrocytes were distributed evenly through all cortical laminae. Interestingly, between P5 and P12, the number of astrocytes was significantly greater in Et-treated rats than in controls. Thus prenatal exposure to ethanol induces the premature loss of RAT-401-positive processes and the precocious increase in GFAP immunostaining. These ethanol-induced changes in glial development indicate that ethanol accelerates the transformation of radial glia into astrocytes. Moreover, the ethanol-induced premature degradation of the network of radial glial fibers may underlie the migration of late-generated neurons to ectopic sites.

The effect of gestational ethanol exposure on stimulus-induced sensory activity in the trigeminal/somatosensory system was determined. The mature offspring of mothers fed an ethanol-containing diet (Et) or pair-fed a nutritionally matched control diet (Ct) were examined. The C-row mystacial whiskers were stimulated. Glucose utilization in the principal sensory nucleus of the trigeminal nerve (PSN), ventrobasal thalamus, and somatosensory cortex was determined with [14C]2-deoxyglucose autoradiography. In Ct- and Et-treated rats, whisker stimulation increased glucose utilization in C-row barrel(oid)s in the left PSN, the right ventrobasal thalamus, and the right somatosensory cortex. The rate of glucose utilization in the C-row barrel(oid)s and in nonstimulated regions was lower in the Et-treated rats than in controls. In the cortices of Ct-treated rats, the activity in the C-row barrels on the right side was greater than in the right nonbarrel somatosensory cortex. Et-treated rats also exhibited an increase in glucose utilization, albeit smaller than that in the Ct-treated rats. In contrast, the glucose utilization in the left B- and C-row barrels of Ct-treated rats was decreased. No such decrease was evident in the left cortices of Et-treated rats. Thus, stroking whiskers stimulates the activity of sites in the trigeminal/somatosensory system. In cortex, the definition of these sites is emphasized by depressed activity, i.e., "surround" inhibition, in sites connected via callosal or corticocortical projections. Prenatal exposure to ethanol depresses the metabolic activity regardless of the physiological state; however, the "surround" inhibition of cortical activity is eliminated by prenatal exposure to ethanol through an exuberant projection.

The early development of the principal sensory nucleus of the trigeminal nerve (PSN) was examined to determine whether spatiotemporal patterns of synaptogenesis coincide with patterns in neuronal generation, migration, and death. The morphogenesis of PSN neurons during the period from G16 to P14 was studied with a Golgi method. Prenatally, PSN neurons had dendrites that extended into the sensory tract of the trigeminal nerve (s5), and from as early as G18, these dendrites were studded with spines. The dendrites in the s5 degenerated or regressed in the early postnatal period so that the s5 was free of dendrites by P14. The development of anti-synapsin I immunoreactivity was traced from G14 to P10. Immunoreactive puncta (synaptic boutons) appeared in the medial third of the s5 transiently between G18 and P5. On the other hand, puncta in the PSN did not appear until G20, at which time they were confined to the lateral margin of the PSN. By P0, puncta were distributed throughout the PSN. Cytochrome oxidase activity in the PSN was low and unpatterned prenatally. Postnatally, cytochrome oxidase activity intensified and a segmented pattern of barreloids appeared in the ventral PSN on the day of birth. By P5, the complete pattern of barreloids, spanning the full width of the ventral PSN, was evident. The development of cytochrome oxidase activity in the PSN followed the lateral-to-medial gradient of synaptogenesis revealed by the development of synapsin 1 immunoreactivity. This gradient is opposite of that for neuronal generation, migration, and death. Moreover, the s5 serves as a transient synaptic field.

The overproduction and subsequent death of neurons is a common phenomenon in the developing vertebrate central nervous system (CNS). We tested the hypothesis that the survival of a neuronal subpopulation is related to its time of origin. Neuronal survival was examined in a well-defined CNS structure, the principal sensory nucleus of the trigeminal nerve (PSN) of the rat. The changes in the total number of PSN neurons and in the numbers of early- and late-generated neurons (i.e., neurons heavily labeled by a single injection of [3H] thymidine on G12 or G14, respectively), between gestational day (G) 16 and postnatal day (P) 10 were determined. The total number of neurons in the PSN rose prenatally to a maximum of 40,600 on G18.5. The increase in neuronal number correlates to the period of migration. More than half of the neurons that successfully migrated to the PSN were lost by P10. The patterns for the changes in the numbers of early- and late-generated neurons were similar; however, there were significant differences between the two subpopulations. The maximum number of early-generated neurons (4,250) was attained on G18.2 and subsequently 58.9% of these neurons were lost. In contrast, the maximum number of late-generated neurons (5,050) was attained on G20.0 and 66.6% of these neurons were lost by P10. Therefore, it appears that the survivability of early generated neurons is greater than for late-generated neurons. This enhanced survivability presumably results from a competitive advantage that early-generated neurons have for forming synapses or gaining access to trophic factor(s) that are in limited supply.

As nuclei in the central nervous system develop, neurons actively migrate from their site of generation to their permanent residence. This study examines the spatiotemporal sequence of the migration of neurons to the principal sensory nucleus of the trigeminal nerve (PSN) of the rat. Tritiated thymidine autoradiography and bromodeoxyuridine immunohistochemistry were used to examine the spatiotemporal patterns of migration of PSN neurons born on gestational day (G) 12 (early-generated neurons) and of those born on G14 (late-generated neurons). The final residence of early- and late-generated neurons was determined by injecting a thymidine analog into a pregnant rat on G12 or G14 and sacrificing the pups on postnatal day (P) 30. Early- and late-generated neurons were distributed medially and laterally, respectively. The schedule of the migration of PSN neurons was also determined. A few pioneer neurons born on G12 reached the PSN by G14; however, the last of the neurons born on G12 arrived in the PSN by G18. The migration of neurons born on G14 was completed 2-6 days later than that of the early-generated neurons. The path followed by migrating neurons was delineated by radial glial fibers. These processes were identified in the developing metencephalon by RAT-401 immunohistochemistry. Radial glial fibers extended from the lateral part of the ventricular zone through the tegmentum and the PSN to the surface of the metencephalon external to the sensory tract of the trigeminal nerve. RAT-401-immunoreactive processes were detected during the period of neuronal migration, but disappeared by P5. Thus, the migration of PSN neurons follows an inside-to-outside sequence, which apparently is organized by radial glial fibers. The inside-to-outside sequence of neuronal migration directly opposes the outside-to-inside gradient of synaptogenesis.

The neuropeptide buccalin A was originally purified and sequenced from a nerve-muscle system used in feeding-related behaviors of Aplysia californica in which it has been proposed that it acts as a modulatory cotransmitter. The distribution of buccalin-like immunoreactivity in the central ganglia and in peripheral tissues of Aplysia californica was examined by whole mount immunohistochemical techniques. Immunoreactive material was located in specific cell bodies and clusters of neurons in each of the ganglia. Immunoreactive fibers were present in each of the connectives between ganglia, in tracts coursing through the ganglia, and in the majority of the peripheral nerves. Most fibers were smooth in contour, but some had regularly spaced swellings. Varicosities containing immunoreactive material were located on specific neuronal somata and on certain tissues associated with the feeding, circulatory, digestive, and reproductive systems. The specific and widespread distribution of buccalin-like immunoreactivity supports the hypothesis that members of the buccalin peptide family act as neuromodulators or neurotransmitters in a variety of central and peripheral circuits in Aplysia.

The distribution of myomodulin-like peptides in the nervous system of Aplysia californica was examined by using immunocytochemical techniques. Neurons and cell clusters containing immunoreactive material were located in each of the major central ganglia. Myomodulin-like immunoreactivity was also present in fibers in each of the connectives between the ganglia and in peripheral nerves. Varicosities containing immunoreactive material were located on specific regions of peripheral tissues associated with the feeding, digestive, cardiovascular, and reproductive systems. Double-labeling experiments were used to demonstrate myomodulin-like immunoreactivity in two identified neurons, the motor neuron B16 in the buccal ganglion and the widely acting interneuron L10 in the abdominal ganglion. Structures in the eye and cerebral ganglion that may correspond to the optic circadian pacemaker system were also stained. The central and peripheral distribution of myomodulin-like immunoreactivity indicates that this family of neuropeptides is present in specific efferent, afferent, and interneuronal elements that participate in a diversity of neural circuits in Aplysia.

Prenatal labelling with [3H]-thymidine was combined with retrograde tracing techniques in adult rats to determine the birthdates of the trigeminal (V) ganglion cells that contributed axons to the infraorbital nerve (ION) and the generation of the subsets of ION cells that innervated specific vibrissae follicles (C-1 and C-5). The V ganglion cells contributing axons to the ION are born between embryonic (E-, E-0 = the day of conception) days 9.5 and 14.5. The percentages (normalized so that they total 100%) of the total V ganglion population born on E-9.5 through E-14.5 were 5.8, 25.7, 19.8, 23.4, 21.0, and 4.4%, respectively. The distribution of birthdates for the V ganglion cells that were retrogradely labelled from the ION closely matched that for the ganglion as a whole. All of these neurons were also born on E-9.5 through E-14.5, and the percentages born on each day were 6.3, 23.6, 18.1, 24.0, 23.6, and 4.4%. Finally, a similar distribution of birthdates was obtained for the V ganglion cells that were retrogradely labelled after injection of retrograde tracers into either the C-1 or C-5 vibrissae follicles. We were unable to detect any distinctive spatial distributions for either all V ganglion or ION cells born on a specific embryonic day. Furthermore, neurons with a given birthdate and that innervated a given follicle were distributed throughout the entire region containing all of the ganglion cells supplying the follicle in question. Therefore, it appears that the V ganglion cells contributing axons to the ION are born over the entire period of ganglion neurogenesis and further that the organization of the ION's innervation of the periphery is not a function of cell birthdate.

The effects of prenatal exposure to ethanol on the structure and function of corticospinal neurons was investigated. The subjects were the 3-4-month-old offspring of hooded rats fed a nutritionally balanced liquid diet containing 6.7% (v/v) ethanol (Et), pair-fed a nutritionally matched isocaloric diet (Ct), or fed chow and water (Ch). Corticospinal neurons in primary somatosensory cortex were examined by intracellularly recording and filling cells that were driven by antidromic stimulation of the pyramidal decussation. In the control rats, corticospinal neurons comprised a homogeneous morphophysiological population. Morphologically, all of the antidromically driven cells examined were pyramidal neurons with cell bodies in layer Vb. The dendrites of these neurons were spinous and branched within layers I, IV, and V. Their axons arborized within layers IV, V, and VI and some collaterals extended laterally for distances up to 2.6 mm from the cell body. The mean conduction latency was 3.6 and 3.4 msec for Ch- and Ct-treated rats, respectively. In Et-treated rats, corticospinal neurons constituted a heterogeneous population. The laminar distribution of the corticospinal neurons in Et-treated rats was broad; the cell bodies of labeled neurons were in layers II, IV, V, and VI. The dendrites of layer Vb neurons were spinous; however, many of the spines appeared dysmorphic and the density of spines was significantly greater (32%) in Et-treated rats than in Ct-treated rats. Although the dendritic branching pattern for layer Vb neurons was similar to that described for the controls, a Sholl analysis showed that the complexity and extent of their dendritic trees were significantly greater in Et-treated rats. The axons of all layer Vb neurons in Et-treated rats had long horizontal processes that arborized in layers IV-VI, and some neurons also had an array of collaterals that ascended to layer I. The mean conduction latency for layer Vb neurons was 3.9 msec. The structure and function of ectopic neurons (those in layers II, IV, Va, Vc, and VI) in Et-treated rats differed markedly from those of the layer Vb neurons. Morphologically, the dendritic and axonal fields of these neurons were narrower than for the layer Vb neurons, and the ectopic neurons had a mean conduction latency of 7.1 msec. The heterogeneity of the population of corticospinal neurons in Et-treated rats may result from the effects of ethanol on early events in neuronal development such as neuronal generation and migration.

Stereological methods were used to examine the consequences of prenatal exposure to ethanol on the structure of area 3, primary somatosensory cortex, of the mature hooded rat. Pregnant rats were fed a liquid diet containing 6.7% (v/v) ethanol (Et), pair-fed an isocaloric liquid control diet (Ct), or fed a diet of chow and water (Ch). Cresyl violet-stained sections of 3-month-old pups were examined. The corrected mean size of the cell bodies of neurons in layers other than layer V was significantly smaller in the Et-treated rats; conversely, the mean somatic size of glia in each layer was significantly larger in the Et-treated rats. The laminar cell packing density for neurons and glia, however, was similar in rats from both treatment groups. The overall volume of area 3 and the volume of individual layers were about 33% smaller in Et-treated rats than in the pair-fed controls. Thus, the estimated total number of neurons in Et-treated rats (1.79 X 10(6] was significantly fewer than in Ch-treated rats (2.77 X 10(6] and in Ct-treated rats (2.66 X 10(6]. The total number of glia also was about 30% fewer in Et-treated rats than in the controls. Not all layers were affected equivalently. The space occupied by the neuropil was significantly greater in Et-treated rats, but only in layers II/III, IV, and VI; hence, the cell body/neuropil ratio in these layers was less in Et-treated rats than in the controls. Therefore, microcephaly caused by prenatal exposure to ethanol results not only from a miniaturization of the brain, but also from a permanent abnormal organization of cerebral cortex.

Clinical and experimental evidence shows that prenatal exposure to ethanol causes craniofacial malformations, microcephaly, and abnormal development of the central nervous system. This study describes the effects of ethanol on the development of the principal sensory nucleus of the trigeminal nerve (PSN). The offspring of two groups of rats were examined. Pregnant females in one group were fed a liquid diet containing 6.7% (v/v) ethanol (Et) and rats in the other group were fed an isocaloric liquid control diet (Ct). Each pregnant rat was administered [3H]thymidine on one day during the period from gestational day (G) 10 to G22. After pups grew to 30 days of age, they were killed and their brains were processed by an autoradiographic procedure. Qualitatively, the PSN of Ct- and Et-treated rats appeared similar; they were composed chiefly of small neurons and a few scattered large neurons. On the other hand, quantitative analyses revealed significant differences between both groups. Although the volume of the PSN of Et-treated rats was not significantly different (-3.2%) than that for Ct-treated rats, the PSN of Et-treated rats had significantly (P less than 0.01) fewer (30.0%) neurons than did the PSN of Ct-treated rats. The number of the small neurons, but not of the large neurons, was affected most by the ethanol exposure. Prenatal exposure to ethanol also altered the generation of PSN neurons. Most neurons in the PSN of Ct-treated rats were born between G12 and G15, the small neurons being generated before the large neurons. In Et-treated rats, too, small neurons were born before the large neurons; however, the time frame of neuronogenesis was delayed as it occurred between G13 and G16. Thus, prenatal exposure to ethanol produces profound developmental abnormalities that lead to permanent alterations in the structure of the mature central nervous system.

Prenatal exposure to ethanol alters the generation of neocortical neurons; ethanol reduces the numbers of neurons generated between gestational day (G) 12 and G19 but paradoxically increases neuronogenesis after G19. The present study used [3H]thymidine autoradiography to examine the effects of ethanol on the proliferation of cells in the two cortical germinal zones, the ventricular and subventricular zones. Pregnant rats were fed one of three diets: a liquid ethanol-containing (6.7% v/v) diet (Et), a nutritionally matched isocaloric liquid diet (Ct), or chow and water (Ch). Fetuses were administered with [3H]thymidine on G13, G17, or G20 and killed 45-60 minutes later. Autoradiographs were prepared to identify radiolabeled and mitotic cells. Additional brains from 13-23-day-old fetuses were processed for cytoarchitectonic analyses. The size of the lateral ventricles in the fetuses was not significantly affected by the prenatal exposure. Both the ventricular and subventricular zones were evident throughout the period from G13 to G23. In all three groups, the ventricular zone was thickest from G13 to G17, but from G15 on, the ventricular zone of Et-treated fetuses was significantly thinner than those of either controls. On G13, G17, and G20, the labeling index (the ratio of radiolabeled cells to the total population) was significantly less in Et-treated fetuses. The mitotic index was similar in Et-, Ch-, and Ct-treated fetuses. The subventricular zone of all fetuses was most prominent during later fetal ages and it was thicker in Et-treated fetuses than in controls. Moreover, the labeling and mitotic indices for the subventricular zone of Et-treated rats were significantly greater than for controls. Thus, ethanol had different effects on the two neocortical germinal zones: the proliferation of ventricular cells was inhibited, whereas the proliferation of subventricular cells was stimulated. These data suggest that ethanol-induced changes in neuronogenesis result from alterations in proliferative activity.

Each division of rat visual cortex, areas 17, 18a, and 18b, has connections with sensory, motor, and association cortices. These corticocortical connections were sampled using anterograde autoradiographic and retrograde horseradish peroxidase labeling techniques. Area 17 is connected via reciprocal pathways with each division of visual cortex, the posterior one-third of motor area 8, association area 7, and posteroventral area 36 of temporal cortex. It also receives projections from perirhinal areas 13 and 35. Area 18a has reciprocal connections with areas 17 and 18b, a patch in posterior somatosensory area 3, and dorsal auditory area 41. Like area 17, area 18a receives afferents from and projects to the posterior one-third of motor area 8. The connections of area 18a with association cortices are extensive; these regions include parietal areas 7, 39, 40, and 14, posteroventral and dorsal area 36, and perirhinal cortex. Area 18b is connected with areas 17 and 18a, a patch in medial area 3, and dorsal area 41. There are reciprocal projections between area 18b and posterior area 8. As for association cortex, area 18b projects to frontal area 11, area 7, posteroventral and dorsal area 36, and perirhinal cortex. In addition, area 18b receives input from and projects efferents to the dorsal claustrum. Most of the interconnections among areas 17, 18a, and 18b originate from neurons in layers II, III, and V and end in terminal fields in layers I-III and V. In contrast, projections of other sensory, motor, and association cortices to visual cortex originate mainly from neurons in layer V and to a lesser extent from layer II. The reciprocal pathways from visual cortex terminate predominantly in the supragranular layers. In conclusion, these corticocortical pathways provide the basis for cortical visuosensory and visuomotor integration that may aid the rat in the coordination of visually guided behaviors.

The distribution and the time of origin of corticospinal neurons were examined in rats prenatally exposed to ethanol and in control rats. The distribution of corticospinal neurons was determined by tracing the retrograde transport of horseradish peroxidase (HRP) from an injection site in the cervical spinal cord. In control rats, HRP-positive neurons were distributed in layer Vb throughout motor area 4, rostral motor area 6/8, dorsal somatosensory area 3, caudal somatosensory area 2, and various "association" regions including parietal areas 14, 39, and 40, occipital areas 18a and 18b, cingulate areas 24a and 24b, and prefrontal area 32. In ethanol-exposed rats, the distribution of retrogradely labeled neurons was similar to control animals with three notable exceptions: (1) HRP-positive neurons were evident throughout the rostrocaudal extent of area 6/8; (2) occasionally ectopic labeled neurons were identified in the supragranular layers, layers Va and Vc, and superficial layer VI; and (3) the density of HRP-labeled neurons and the ratio of labeled neurons to the total number of neurons in areas 4, 6/8, 3 and 2 were significantly greater (20-48%) in ethanol-exposed rats than in controls. There was, however, no intergroup difference in the area of the cell bodies of HRP-positive neurons. Taken together, these findings indicate that ethanol exposure resulted in an increased number of corticospinal neurons. The time of origin of corticospinal neurons was determined by using a technique that combined tritiated thymidine autoradiography and retrograde transport of HRP. In control animals, HRP-positive neurons were double labeled by an injection of tritiated thymidine on gestational day (GD) 15, 16, or 17. In ethanol-exposed rats, corticospinal neurons were generated on GD 16, 17, and 18, the late-generated ones being distributed in caudal area 6/8. These intergroup differences represent a persisting ethanol-induced alteration of cortical structure that may underlie motor dysfunction and mental retardation in fetal alcohol-affected offspring. Moreover, the increase in the number and the delay in the time of origin of corticospinal neurons suggest that the normal process of paring down exuberant corticospinal projections may be affected by prenatal exposure to ethanol.

The connections of rat cingulate cortex with visual, motor, and postsubicular cortices were investigated with retrograde and anterograde tracing techniques. In addition, connections between visual and the postsubicular (area 48) and parasubicular (area 49) cortices were evaluated with the same techniques. The following conclusions were drawn. Area 29 connections: Afferents to area 29 originate mainly from cingulate areas 24 and 25, visual cortex (primarily area 18b), motor cortex area 8, area 11 of frontal cortex, areas 48 and 49, and the subiculum. Efferent connections of area 29 within cingulate cortex and to visual areas differ for each cytoarchitectural subdivision of area 29. Thus, area 29c has limited projections both within cingulate cortex and to areas 48 and 49, while area 29d projects to these areas as well as to area 8, area 18b, and medial area 17. These visual cortex afferents originate mainly from layer V neurons of areas 29b and 29d, while areas 29a and 29c have virtually no projections to visual cortex. Area 24 connections: Afferents to area 24 originate primary from cingulate areas 25 and 29 and visual area 18b and medial area 17. Efferent projections of area 24a are distributed within cingulate cortex, while area 24b has more extensive projections to posterior cingulate and visual cortices. Area 24b is the cingulate subdivision which is both the primary recipient of visual cortex afferents as well as the source of most of the projections of anterior cingulate cortex to visual areas. Visual cortex has reciprocal connections with parts of the postsubicular and parasubicular cortices. Neurons of the internal pyramidal cell layer of both areas 48 and 49 project to areas 17 and 18b, while layers I and III of these parahippocampal areas receive projections from areas 17 and 8b. In conclusion, areas 29d have particularly extensive interconnections with visual cortex, while area 29d also maintains projections to area 8 of motor cortex. This connection scheme supports the view that cingulate cortex may have a role in feature extraction from the sensory environment, as well as in sensorimotor integration. Finally, the postsubiculum may be classified as a limbic association cortex in which extensive visual and cingulate efferents converge.

The generation, migration, and morphogenesis of atypically oriented pyramidal neurons in the rat visual cortex were examined. In the mature cortex, these neurons were distributed through layers II-VI. Moreover, the atypically oriented pyramidal neurons in a particular layer tended to be oriented in a specific way; atypically oriented pyramidal neurons in layer II, layers III-VIa, and layer VIb were obliquely, radially, and obliquely oriented, respectively. Ultrastructurally, the somata of atypically oriented pyramidal neurons contained large euchromatic ovoid nuclei and cytoplasm that was replete with rough endoplasmic reticulum and Golgi apparatus. These somata formed only symmetric axosomatic synapses. Many atypically oriented pyramidal neurons projected axons into the white matter as demonstrated by a Golgi method and by a retrograde tract-tracing technique; however, some of these pyramidal neurons in layers III-V had axons that ascended to layer I. By using a technique which combined retrograde tract tracing with [3H]thymidine autoradiography, it was determined that most atypically oriented pyramidal neurons in layers V and VIa, layer IV, and layer II were generated on gestational days (GD) 15-17, GD 17-19, and GD 20-21, respectively. Atypically oreinted pyramidal neurons were identified during the period from postnatal day 0 (day of birth) to day 30. On day 0, obliquely oriented pyramidal neurons were distributed in the deep cortical plate, i.e., immature layer VI. On day 3, the youngest atypically oriented pyramidal neurons were radially oriented and were located in layer IV. Some obliquely oriented pyramidal neurons were present in layer II on day 6, but the greatest number and the most severely canted pyramidal neurons in layer II were evident on day 9. The orientations of the cell body and the apical dendrite did not change appreciably after migration was complete, except for those in layers V and VI with obliquely oriented cell bodies and radially oriented apical dendrites. The second and third postnatal weeks were marked by substantial morphological differentiation of all pyramidal neurons as noted by the lengthening and branching of dendrites and by the appearance of dendritic spines. By the fourth postnatal week, atypically oriented pyramidal neurons achieved their mature morphology. The generation, migration, and morphogenesis of atypically oriented pyramidal neurons proceed by an inside-to-outside sequence. This development is similar and concurrent with that of typically oriented pyramidal neurons.
