OBJECTIVE To evaluate the efficacy and safety of resveratrol combined with ablative fractional CO2 laser system (AFL) treating skin photoaging. METHODS Thirty-two subjects were assigned to the treatment group (TG) or the control group (CG), respectively, applied test product (resveratrol essence) or control product twice daily for 6 months. Each subject was given an AFL treatment or no laser treatment on left or right side of the face randomly. Subjective evaluations by investigators and subjects themselves were conducted after treatment. Melanin index, erythema index, and cuticle moisture content were conducted at baseline and after treatments. Adverse events (AEs) were evaluated during the study period. OBJECTIVE The main aim of this study was to optimize the formula of saffron compound essence (SCE) and investigate the protective effects and mechanisms of SCE against skin photoaging. MATERIALS AND METHODS The formula of SCE was optimized using single factor and orthogonal experiments. The optimized SCE was then applied on the back skin of mice under ultraviolet irradiation. The protective effect and possible mechanism of SCE on skin photoaging were investigated using macroscopic and histological evaluation, skin thickness tests, determination of skin microcirculation, and the skin content determination of type I and III collagen, matrix metalloproteinase-2 (MMP-2), and extracellular regulatory protein kinase (ERK1/2) on the skin of each group of photoaging mice model. RESULTS The SCE prepared using the optimal formula was golden yellow, uniform, fine, and viscous. The obtained results in the SCE high dose group indicated that SCE can inhibit the generation of wrinkles, decrease the skin thickness, reduce the microcirculation blood flow of the back skin of mice, alleviate the proliferation and abnormal accumulation of collagen fibers and elastic fibers, increase the content of skin type I and III collagen, up-regulate the expression MMP-2 protein, and down-regulate the expression ERK1/2 protein, when compared with the shave control group. CONCLUSION The results obtained in this study have indicated that the optimized SCE has the effect of preventing skin photoaging, and its mechanism may be associated with the ERK1/2 pathway. Therefore, SCE should be regarded as a potential therapeutic agent for preventing photoaging.

SummaryIntroduction Skin photoaging is the consequence of solar UV exposure, and DNA damage is an important part of this process. The objective of the current work was to demonstrate that in vitro skin models can be utilized to confirm that DNA damage protection is provided by sunscreens. Methods Skin equivalents were exposed to full-spectrum UV light administered with a standard research solar simulator with and without pre-application of sunscreen. Cyclopyrimidine dimer (CPD) and sunburn cell (SBC) formation as well as CPD quantitation were evaluated to determine DNA damage protection provided by the sunscreen. Results Marked decreases in both CPDs and SBCs were observed when sunscreen was applied prior to UV exposure. Conclusions Sunscreen application prior to full-spectrum solar UV exposure protects DNA from photodamage measured by CPD and SBC formation. This can be expected to lessen the risk of photoaging and malignant transformation.

Summary Background Retinaldehyde (RAL) was proven effective in treating photodamaged skin. Topical treatments with specific intermediate-size hyaluronate fragments (HAFi, 50–400 kDa) have been shown to stimulate keratinocytes proliferation and epidermal hyperplasia. The aim of this open, multicentric, international study was to assess the efficacy of the combination RAL–HAFi in the correction of skin photoaging. Patients/Methods Either RAL 0.05%–HAFi 0.5% (Eluage® cream; group 1) or RAL 0.05%–HAFi 1% (Eluage® antiwrinkle concentrate; group 2) or both products (group 3) were applied daily to the 1462 subjects during 90 days. Overall photoaging severity was evaluated in the three groups by the dermatologists at D0, D30, and D90 based on the Larnier’s scale. Wrinkles and/or furrows and clinical signs of aging were evaluated using a 4-point scale. Summary For years, cosmetic ingredients for anti-aging treatments have attracted consumers. Skin aging is accelerated by reactive oxygen species (ROS), generated by exposure to solar ultraviolet radiation (UVR), in a process known as photoaging. Because cutaneous iron catalyses ROS generation, it is thought to play a key role in photoaging. Iron is essential to almost all forms of life. However, excess iron is potentially toxic as its catalytic activity induces the generation of ROS. Iron-catalysed ROS generation is involved in numerous pathological conditions, including cutaneous damage. When skin is directly exposed to UVR, cutaneous intracellular catalytic iron levels increase because of the release of iron from iron-binding proteins such as ferritin. Consequently, the subsequent ROS generation may overwhelm cutaneous defense systems such as the cellular iron sequestration and ROS scavenging capacity. The harmful role of excess cutaneous iron implies that there may be a potential for topical iron chelator treatments. We now consider cutaneous photodamage skin photoaging as the result of iron-catalysed ROS generation and discuss preventative strategies based on iron chelators.
