The Euwallacea fornicatus species complex (Coleoptera: Curculionidae: Scolytinae: Xyleborini) is a group of four cryptic ambrosia beetle species. Native to Asia, several members of the complex have invaded other continents, where they cause significant economic losses to agricultural crops (e.g., avocado) and natural ecosystems. We were primarily interested in developing management strategies by focusing on the flight behavior of the beetles. Thus, seasonal differences in flight activity were assessed using panel traps baited with a commercial quercivorol lure, placed in infested avocado orchards in Danei, Tainan, Taiwan. Same traps were used to investigate the flight activity of a natural enemy, an undescribed species of the Braconid genus Eucosmophorus sp. Shothole borer species were identified using a DNA-based, high resolution melting assay. Trap data were compared to the predictions of a simple degree-day model, incorporating developmental data and several environmental parameters known to influence flight. Such as the time period representing most of flight activity in a day and temperature-dependent flight propensity. In stark contrast to the degree-day model which predicted the highest emergence, and by extension flight, of shothole borers during spring and summer (May to November), flight activity was actually lowest during these months, and instead, peaked during the winter (October to March). Abundance of the parasitoid wasp closely mirrored flight activity of the shothole borers. The mismatch of trapping and modeling data can have many causes, heavy precipitation and possibly cooperative brood care may suppress the dispersal behavior of the shothole borers during the summer., (© The Author(s) 2022. Species belonging to the Euwallacea fornicatus Eichhoff (Coleoptera: Scolytinae) species complex have invaded the continental U.S. since at least 2003. Three species of this complex are known to have established, two in California (E. fornicatus; and Euwallacea kuroshio), and a third in Florida (Euwallacea perbrevis). Their native ranges are spread across southern and southeast Asia. In Taiwan, all three species occur in sympatry. They attack healthy trees of widely varied species and cause severe damage and death to the trees. The attractant quercivorol is commonly used to promote their detection by passive trapping. Recent studies in Florida have shown that trapping of E. perbrevis can be further improved by adding a synergist, α-copaene, alongside the quercivorol lure. Thus, we were interested in testing the effectiveness of α-copaene for trapping the other invasive members of the complex in California and in an area of Taiwan where all three species co-occur. We found that α-copaene marginally enhanced the trapping of E. perbrevis in Taiwan, but had no effect on the trapping of E. fornicatus or E. kuroshio in either California or Taiwan. We conclude that any enhancing effect of α-copaene is specific to E. perbrevis. This highlights the economic importance of accurate species identification in developing and implementing an efficient, and yet cost-effective, monitoring program for the management of E. fornicatus and E. kuroshio in California and elsewhere., (© The Author(s) 2021. Chrysoperla comanche (Banks) and its sibling species Chrysoperla rufilabris (Burmeister) are voracious predators of the Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), an invasive pest that vectors a bacterium responsible for the lethal and incurable citrus greening disease. The Comanche lacewing naturally occurs in Southern California, whereas C. rufilabris is currently one of only two commercially available green lacewing in the United States. These sister species can be separated by larval morphological traits, by differences in courtship songs, and possibly by three nuclear genes wingless, PepCK, ATPase, yet they are not distinguishable based on the mitochondrial barcode gene (COI). Releasing in a new area a biological control agent capable of hybridizing with a resident species may pose risks that range from local displacement to irreversible loss of genetic identity. Therefore, we performed no-choice laboratory crosses to assess pre- and postzygotic isolation. We show that fertile and viable hybrid progeny could be readily obtained in interspecific crosses and backcrosses and, although there is a trend toward lower hybrid fitness, postzygotic isolation is overall weak and might not prevent loss of genetic identity under natural conditions. It remains to be determined if differences in courtship songs will prevent hybridization in the wild, as shown for other green lacewings. Levels of armored scales (Hemiptera: Diaspididae) on Mexican Hass avocados imported into California over May 2008-June 2009 were monitored on 135 trucks entering the state via the Blythe border station, the entry point receiving the highest volume of fruit. Levels of live sessile scales were 3.9-fold higher than indicated in a previous survey (September 2007-April 2008) although levels of live eggs and crawlers were similar to previous levels. A survey of avocado fruit in California infested with armored scales detected four species known to be endemic but failed to find any of the seven exotic Diaspididae entering the state on Mexican fruit. Monitoring of Mexican armored scales on imported avocados from September 2007 to December 2010 recovered 10 species of parasitoids predominated by two species of Signiphora Ashmead (Hymenoptera: Signiphoridae). One of these species, Signiphora flavopalliata Ashmead, comprised 36% of all collected Mexican parasitoids and is a known hyperparasitoid. A survey of armored scale parasitoids present on commercial California avocados detected 17 genetic signatures, with only four of these in common with those detected on imported Mexican fruit. A modular system for studying the biology of Scolytinae using 3D printed emergence traps is presented. This system consists of traps that can be used for the introduction of beetles into trees as well as for the determination of the number of offspring emerging from trees and branches, and their production of frass and sawdust. Open-source files used for printing these traps have been deposited as supplementary material and can be downloaded for trap production by students of Scolytinae with access to a 3D printer. The cost in material for the production of a single emergence trap on the printer used here is approximately $1.30, with a print time of less than 90 min.

Neutral molecular markers are gene sequences where variants are considered to confer no fitness advantage, such as microsatellites and mitochondrial haplotypes. Several types of neutral marker are easy to develop, cheap to use, and have found extensive application for addressing ecological questions. In biocontrol, these markers are used to simplify identification of cryptic species and of prey remains in predators. Here, we address the potential of neutral molecular markers for determining the relative performance of a "superior" strain of a species after release into an already established conspecific population. We used modeling to show that only under very limited conditions can traditional neutral markers be used to demonstrate that beneficial genetic variation was successfully introgressed into the existing population. However, new population genomic methods do make it possible to track alleles at a large number of loci and consequently make it possible to show if alleles from a superior strain spread in an already established conspecific population.

The genetic structure of 23 populations of Graphocephala atropunctata (Signoret) (Hemiptera: Cicadellidae: Cicadellinae), a vector of the plant pathogenic bacterium Xylellafastidiosa Wells et al., was investigated using ribosomal 28S and mitochondrial cytochrome C oxidase I gene sequences. The 28S sequences were identical across all G. atropunctata specimens and populations, but 16 mitochondrial haplotypes were detected and significant interpopulation differences were found in the distribution of these haplotypes. Pairwise estimates of Fst correlated positively with geographical distance between populations, a phenomenon known as genetic isolation by distance. Thus, despite potential for widespread movement of G. atropunctata through nursery and agricultural industries, isolated populations of G. atropunctata have remained genetically distinct, suggesting that negligible numbers of G. atropunctata are actually transported or population interbreeding rarely occurs. The phylogenetic relationship between G. atropunctata and two additional congeners, Graphocephala cythura Baker and Graphocephala flavovittata Metcalf, which have overlapping distributions with G. atropunctata, also was investigated. Although 28S sequences of G. flavovittata were strikingly similar to those of G. atropunctata, mitochondrial DNA (mtDNA) suggests that both species are genetically distinct from G. atropunctata.

Dispersal and colonization of new areas by armored scale insects (Hemiptera: Diaspididae) is achieved by mobile first-instar nymphs, called crawlers. Few studies have considered the actual mechanisms by which crawlers disperse, and although crawlers are capable of actively wandering over short distances (generally < 1 m), their dispersal over longer distances has been thought to be wind-mediated. Here, we present evidence of a potentially more important means of dispersal over longer distances (> 1 m). We first confirmed that crawlers of four species of Diaspididae [Abgrallaspis aguacatae Evans, Watson & Miller; Hemiberlesia lataniae (Signoret); Aspidiotus nerii Bouché; and Diaspidiotus perniciosus (Comstock)] have four hairs on the end of each of their legs and that each of these hairs ends in a suction cup-like structure, reminiscent of the attachment structures possessed by phoretic mites. In a controlled environment, using crawlers of A. nerii, we then showed that the crawlers use these structures to attach themselves to three different insect species [Musca domestica L., Cryptolaemus montrouzieri Mulsant and Linepithema humile (Mayr)] and can effectively be moved phoretically by these insects. Crawlers can remain attached to flying insects for considerable periods of time, suggesting that this may be an important means of dispersal for armored scale insects. The importance of phoresy for diaspidid dispersal in the field remains to be determined.

Over the past three decades, Western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), has become a major worldwide pest of many agricultural and horticultural crops. In response, much time, money, and effort have been put into pure and applied research focusing on the biology and control of this pest. Western flower thrips is native to Western North America and widespread in California. High levels of variation in basic biology, pest status, and resistance to insecticides bring into question the specific status of Western flower thrips. We used nuclear-mitochondrial barcoding to compare DNA sequences of nuclear and mitochondrial genes between Western flower thrips populations across California, looking for association between these unlinked loci. Sequences of D2 domain of 28S and cytochrome c oxidase I gene revealed the existence of two distinct but sympatric genetic entities, and we describe a simple polymerase chain reaction-based method for diagnosing these entities. The complete association of these nuclear and mitochondrial loci in areas of sympatry is indicative of reproductive isolation and the existence of two cryptic species, both of which key out to Western flower thrips by using morphological characters. The finding that Western flower thrips is a complex of two species has important implications for past, current, and most importantly future research on these pests.

'Hass' avocado, Persea americana Mill., fruit imported into California from Mexico are infested with high levels of armored scale insects (Hemiptera: Diaspididae), constituting several species. The paucity and delicate nature of morphological characters traditionally used to diagnose armored scales often require careful preparation of slide-mounted specimens and expert knowledge of the group, for their accurate identification. Here, we present a simple, quick, and accurate means to identify armored scales on Mexican avocados, based on amplification of the internal transcribed spacer two of ribosomal DNA, by using the polymerase chain reaction (PCR). This region seems to show a high level of intraspecific conformity among scale specimens originating from different localities. A suite of species-specific reverse PCR primers are combined in a single reaction, with a universal forward primer, and produce a PCR product of a unique size, that after standard gel electrophoresis, allows the direct diagnosis of six diaspidid species: Abgrallaspis aguacatae Evans, Watson & Miller; Hemiberlesia lataniae (Signoret); Hemiberlesia sp. near latania; Hemiberlesia rapax (Comstock); Acutaspis albopicta (Cockerell); and Pinnaspis strachani (Cooley). Two additional species, Diaspis miranda (Cockerell) and Diaspis sp. near miranda, also are separated from the others by using this method and are subsequently diagnosed by secondary digestion of the PCR product with the restriction endonuclease smaI.

Between 1914 and 2007, a quarantine protected California avocado, Persea americana Mill., groves from pests that might be introduced into the state along with fresh, imported avocados. Soon after Mexican avocados were first allowed entry on 1 February 2007, live specimens of several species of armored scales (Hemiptera: Diaspididae) not believed to be present in California were detected on 'Hass' avocados entering the state from Mexico. Initially, the California Department of Food and Agriculture (CDFA) prevented avocados infested with these scales from entering the state or required that they be fumigated with an approved treatment such as methyl bromide. After a Science Advisory Panel meeting in May 2007, U.S. Department of Agriculture-Animal and Plant Health Inspection Service (USDA-APHIS) reaffirmed its position that armored scales on shipments of fruit for consumption (including avocados) pose a "low risk" for pest establishment. In compliance with APHIS protocols, as of 18 July 2007, CDFA altered its policy to allow shipments of scale-infested avocados into the state without treatment. Here, we report on sampling Mexican avocados over an 8-mo period, September 2007-April 2008. An estimated 67 million Mexican Hass avocados entered California over this period. Based on samples from 140 trucks containing approximately 15.6% of this volume of fruit, we estimate that approximately 47.6 million live, sessile armored scales and an additional 20.1 million live eggs and crawlers were imported. We found eight probable species of armored scales in the samples, seven of these are not believed to occur in California; 89.3% of the live scales were Abgrallaspis aguacatae Evans, Watson and Miller, a recently described species. In contrast to the USDA-APHIS opinion, we believe the volume of shipments and levels of live scales they contain present a significant risk to California's US$300 million avocado industry and to other crops that might become infested by one or more of these exotic species.

Effective plant quarantine and biological control initiatives require rapid and accurate identification of exotic and potentially invasive taxa that may cause high economic losses or environmental damage. The genus Scirtothrips Shull includes several species that are serious agricultural pests, and, because of their minute size and cryptic behavior, prone to undetected transport through international trade of plant material. Although assigning specimens to the genus Scirtothrips is straightforward using traditional taxonomic methods, identification of species is much more difficult and requires expert knowledge of the genus. Furthermore, the validity of some Scirtothrips species is questionable. Therefore, an easy, accurate, and highly reliable technique is desirable for Scirtothrips identification. Here, we provide a simple molecular key based on the internal transcribed spacer regions 1 and 2 (ITS1 and ITS2) of nuclear ribosomal DNA. Individual specimens can be identified by amplification of their ITS1 and ITS2 regions with general primers and determining the size of the products by using standard agarose gel electrophoresis, followed in some instances by DNA digestion with the restriction enzymes SacII or PspOM I. The advantage of this identification system is that nonspecialists are able to quickly and cheaply identify individual specimens. Material analyzed for this work was collected in the United States (California), India, South Africa, Kenya, Mexico, Guatemala, Honduras, Nicaragua, Costa Rica, Panama, Australia, New Zealand and Raiatea (Society Islands French Polynesia). We have identified seven pest species with the molecular-based methods described here. It is hoped that this system can be extended to other members of the genus as their ITS1 and ITS2 sequences become available. We also provide molecular confirmation for two new Scirtothrips species, one species from Honduras and one species from New Zealand.
