African wormwood (Artemisia afra Jacq. ex Willd.) has been used traditionally in southern Africa to treat illnesses causing fever and was recently shown to possess anti-tuberculosis activity. As tuberculosis is an endemic cause of fever in southern Africa, this suggests that the anti-tubercular activity of A. afra may have contributed to its traditional medicinal use. Tuberculosis, caused by Mycobacterium tuberculosis (Mtb), is a deadly and debilitating disease globally affecting millions annually. Emerging drug-resistant Mtb strains endanger the efficacy of the current therapies employed to treat tuberculosis; therefore, there is an urgent need to develop novel drugs to combat this disease. Given the reported activity of A. afra against Mtb, we sought to determine the mechanisms by which A. afra inhibits and kills this bacterium. We used transcriptomics to investigate the impact of Artemisia spp. extracts on Mtb physiology. We then used chromatographic fractionation and biochemometric analyses to identify a bioactive fractions of A. afra extracts and identify an active compound. Transcriptomic analysis revealed that A. afra exerts different effects on Mtb compared to A. annua or artemisinin, suggesting that A. afra possesses other phytochemicals with unique modes of action. A biochemometric study of A. afra resulted in the isolation of an O-methylflavone (1), 5-hydroxy-7-methoxy-2-(4-methoxyphenyl)chromen-4-one, which displayed considerable activity against Mtb strain mc26230 in both log phase growth and metabolically downshifted hypoxic cultures. The present study demonstrated that an O-methylflavone constituent of Artemisia afra explains part of the activity of this plant against Mtb. This result contributes to a mechanistic understanding of the reported anti-tubercular activity of A. afra and highlights the need for further study of this traditional medicinal plant and its active compounds. Herbs of the genus Juniperus (family Cupressaceae) have been commonly used in ancestral folk medicine known as "Al'Araar" for treatment of rheumatism, diabetes, inflammation, pain, and fever. Bioassay-guided isolation of bioactives from medicinal plants is recognized as a potential approach for the discovery of novel drug candidates. In particular, non-addictive painkillers are of special interest among herbal phytochemicals. The current study aimed to assess the safety of J. thurifera , J. phoenicea , and J. oxycedrus aqueous extracts in oral treatments; validating the traditionally reported anti-inflammatory and analgesic effects. Further phytochemical investigations, especially for the most bioactive species, may lead to isolation of bioactive metabolites responsible for such bioactivities supported with in vitro enzyme inhibition assays. Firstly, the acute toxicity study was investigated following the OECD Guidelines. Then, the antinociceptive, and anti-inflammatory bioactivities were evaluated based on chemical and mechanical trauma assays and investigated their underlying mechanisms. The most active J. thurifera n -butanol fraction was subjected to chromatographic studies for isolating the major anti-inflammatory metabolites. Moreover, several enzymatic inhibition assays (e.g., 5-lipoxygenase, protease, elastase, collagenase, and tyrosinase) were assessed for the crude extracts and isolated compounds. The results showed that acute oral administration of the extracts (300–500 mg/kg, p. o.) inhibited both mechanically and chemically triggered inflammatory edema in mice (up to 70% in case of J. thurifera) with a dose-dependent antinociceptive (tail flick) and anti-inflammatory pain (formalin assay) activities. This effect was partially mediated by naloxone inhibition of the opioid receptor (2 mg/kg, i. p.). In addition, 3-methoxy gallic acid (1), quercetin (2), kaempferol (3), and ellagic acid (4) were successfully identified being involved most likely in J. thurifera extract bioactivities. Nevertheless, quercetin was found to be the most potent against 5-LOX, tyrosinase, and protease with IC 50 of 1.52 ± 0.01, 192.90 ± 6.20, and 399 ± 9.05 μM, respectively. J. thurifera extract with its major metabolites are prospective drug candidates for inflammatory pain supported with inhibition of inflammatory enzymes. Interestingly, antagonism of opioid and non-opioid receptors is potentially involved. Schumanniophyton magnificum is a medicinal plant used to manage many ailments including malaria, skin diseases, parasitic infections, male sexual dysfunctions, female infertility and typhoid fever. However, no scientific investigation has been made for its folkloric use by the "Baka" Pygmies of Cameroon as an aphrodisiac. To investigate the aphrodisiac and androgenic activities of the aqueous extract of the roots of Schumanniophyton magnificum in male rats and analyze the phytoconstituents by UHPLC/MS. Twenty-five male rats of 16-weeks old were divided into 5 groups and orally treated for 30 days with distilled water (10 ml/kg), or sildenafil citrate (5 mg/kg), or the aqueous extract of Schumanniophyton magnificum (43 mg/kg, 86 mg/kg and 172 mg/kg). The sexual behaviour parameters were monitored on day 1 and 30 by pairing male rats to receptive females. At the end of the experiment, rats were killed and the blood and reproductive organs were collected for histological sectioning, sperm analysis and biochemical analysis. The presence of phytoconstituents and their structures were revealed by UHPLC/MS. The plant extract significantly increased the mount, ejaculation and intromission frequencies in comparison to those in the normal control group; and significantly doubled the serum testosterone levels (2.15 ± 0.70 ng/ml) compared to the normal control group. UHPLC/MS of the aqueous extract of Schumanniophyton magnificum identified 7 major compounds such as Schumanniofioside A, Noreugenin and Rohitukine, with antioxidant and antibacterial activities. The plant extracts significantly increased the penile nitric oxide levels (P <0.05). These results were similar to those obtained after administration of sildenafil citrate. The aqueous extract of Schumanniophyton magnificum could be an alternative for erectile dysfunction management. Angola has an extraordinary plant diversity and a great ethnobotanical potential. However, there is a general lack of information about the first botanical explorations in the country and their contribution to the knowledge of the medicinal flora. The main aim of this study was to unveil the ethnobotanical legacy of José Maria Antunes and Eugène Dekindt, priests of the first Catholic mission in Huíla (Angola) and shed light on their contribution to the knowledge of medicinal wild plants of the country, including information on the uses, plant parts used, and preparation methods documented in the late 19th century. The findings are discussed considering recent ethnobotanical studies to offer a more comprehensive understanding of the historical and traditional uses of plants in Angola over the last two centuries. Based on the information available in manuscripts and on the study of botanical collections preserved in herbaria of Portugal and Angola, we extracted relevant information about the species used in traditional medicine by the rural population of Huíla, the health conditions treated, and the mode of preparation and application. Our results revealed that Antunes and Dekindt conducted the first ethnobotanical study in Huíla, and documented a large number of medicinal wild plants. From these, we report 191 medicinal species, including 25 endemic and four introduced species, belonging to 56 plant families and 146 genera. Fabaceae family presents the highest richness of medicinal plants (39 taxa), followed by Rubiaceae (13), Asteraceae (10), and Apocynaceae (9). The illnesses reported were classified into 15 different categories, with the highest number of species (49) corresponding to unspecific conditions, such as general pains, chills, and fever. Thirty-seven species were reported for respiratory diseases, 31 for musculoskeletal problems, and 30 for digestive issues. Leaves were the most used plant part for medicinal purposes (84 species). Infusion was the most frequently described preparation method (40 species), followed by maceration (24 species), and powdering (36 species). The legacy of Antunes and Dekindt's work improves our understanding of Angola's botanical richness and traditional uses of plant resources. Our findings highlight the presence of unique medicinal resources in Angola, especially among endemic species, which hold the potential to improve the quality of life of rural communities. In Senegal, upper and lower respiratory tract infections constitute a real health problem. To manage these disorders, most people rely on the use of local medicinal plants. This is particularly the case for species belonging to the botanical families, Combretaceae, Fabaceae, Myrtaceae and Rubiaceae, which are widely used to treat various respiratory problems such as colds, flu, rhinitis, sinusitis, otitis, angina, bronchitis, bronchiolitis and also pneumonia. The aim of this study was to identify medicinal plants traditionally used for the management of infectious diseases, in particular those of the respiratory tract. On the basis of these ethnopharmacological uses, this study made it possible to highlight the antibacterial, antiviral and cytotoxic activities of selected plant species. An ethnobotanical survey was conducted in Senegal among informants, including herbalists, traditional healers, and households, using medicinal plants in the management of infectious diseases, with a focus on respiratory tract infections. The most cited plant species were evaluated in vitro on a panel of 18 human pathogenic bacteria may be involved in respiratory infections and against the human coronavirus HCoV-229E in Huh-7 cells. The antiviral activity of the most active extracts against HCoV-229E was also evaluated on COVID-19 causing agent, SARS-CoV-2 in Vero-81 cells. In parallel, cytotoxic activities were evaluated on Huh-7 cells. A total of 127 informants, including 100 men (78.74%) and 27 women (21.26%) participated in this study. The ethnobotanical survey led to the inventory of 41 plant species belonging to 19 botanical families used by herbalists and/or traditional healers and some households to treat infectious diseases, with a specific focus on upper respiratory tract disorders. Among the 41 plant species, the most frequently mentioned in the survey were Guiera senegalensis J.F. Gmel. (95.2%), Combretum glutinosum Perr. Ex DC. (93.9%) and Eucalyptus spp. (82.8%). Combretaceae (30.2%) represented the most cited botanical family with six species, followed by Fabaceae (29.3%, 12 species). A total of 33 crude methanolic extracts of the 24 plant species selected for their number of citations were evaluated in vitro for their antimicrobial and cytotoxic activities. Guiera senegalensis , Combretum glutinosum , Vachellia nilotica subsp. tomentosa (Benth.) Kyal. & Boatwr, Eucalyptus camaldulensis Dehnh., and Terminalia avicennioides Guill. & Perr., showed antibacterial activities. The most active plants against HCoV-229E were: Ficus sycomorus L., Mitragyna inermis (Willd.) Kuntze, Pterocarpus erinaceus Poir., and Spermacoce verticillata L. One of these plants, Mitragyna inermis, was also active against SARS-CoV-2. This work confirmed the anti-infective properties of plant species traditionally used in Senegal. Overall, the most frequently cited plant species showed the best antibacterial activities. Moreover, some of the selected plant species could be considered as a potential source for the management of coronavirus infections. Djibouti was a country where malaria has been endemic for centuries. The local population use the plants as repellents or first aid for uncomplicated malaria. The aim was, for the first time, to collect and identify plants used by the local population to treat malaria and select the most interesting plants (those that are more commontly used, more available, and have fewer studies). These plants were evaluated for their antiplasmodial activity as well as their cytotoxicity on human cell lines for the most active ones. A semi-structured questionnaire was developed for this study to collect information about the use and identity of botanical drugs used to treat malaria. The use-reports (percentage) of each plant were recorded to determine their use importance. Also, the availability status of the plants was assessed; and those in critical condition were discarded excluded from further study. Fifteen plants, out of the 41 listed, were extracted with hydro alcohol, ethyl acetate, and dichloromethane for biological testing. Chloroquine-resistant strain FcB-1 of P. falciparum and a human diploid embryonic lung cell line were used for the antiplasmodial test, and to assess the cytotoxicity for human cells respectively. Preliminary analysis of extract constituents was carried out using thin layer chromatography (TLC). This study identifies 41 plant taxa belonging to 32 families and records their use against malaria. Balanites rodunfolia, belonging to the Zygophyllaceae family, was the most commonly used plant, representing 44 % of use-reports. It was followed by Cadaba rodunfolia (15 %) from the Capparaceae family, and then the three species of Aloe: Aloe djiboutiensis (8.2 %), Aloe ericahenriettae (3.4 %), and Aloe rigens (3.4 %) from the Asphodelaceae family. The leaves are the most commonly used part of the plants to treat malaria, accounting for 76 % of usage. The preparation methods were decoction (52 %), maceration (29 %), and boiling (19 %). The administration routes were by oral (80 %), inhalation 19 %), and bathing (1 %). The best antiplasmodial activities were observed in the dichloromethane extracts of Cymbopogon commutatus and the ethyl acetate extracts of Aloe rigens and Terminalia brownii, with IC 50 values of 9.8, 5, and 7.5 μg/mL, respectively. Their toxicity/activity levels were very favorable with selectivity indices of 5.6, 8.1, and 11.8 for C. commutatus, A. rigens , and T. Brownii, respectively. Forty-one species of botanical drugs were listed as being used to treat malaria in Djibouti. All fifteen selected species showed antiplasmodial activity (IC 50 < 50 μg/mL). Blood purification practices, also referred to as blood cleansing or detoxification, is an ancient concept which is widespread within African traditional medicine. There is, however, limited consolidated information about the selection of plant species, the purpose of use and how the preparation is made. An in-depth examination of how regional communities use medicinal plants for blood purification is needed to better understand this ethnopharmacological practice. To gather knowledge on the traditional use of medicinal plants used for the purpose of blood purification by laypersons in northern Maputaland, KwaZulu-Natal, South Africa. An ethnobotanical survey was conducted on medicinal plants used in the rural areas of northern Maputaland for blood purification. Interviews were conducted in 55 homesteads, where the most knowledgeable participant per household was interviewed using structured questionnaires. Information collected included knowledge of the concept of blood purification, purpose for medicating, plant selection, and mode of preparation. The most important species and families were identified through indices. Sixty-four plant species from 35 families were recorded as blood purifiers for the treatment of ailments linked to the need for blood purification. Terminalia sericea Burch. ex DC. was the most used plant species (mentioned by 39% of the people interviewed). The conditions that were mostly associated with blood purification were skin complaints, gastro-intestinal disorders, and overall improvement of general health. Blood purification is also commonly used for non-medical purposes such as granting good luck or as a love charm. Six new plant species and five new Zulu vernacular names had not previously been documented for blood purification. The frequently mentioned use of medicinal plants for blood purification by the rural community of northern Maputaland clearly emphasises the importance of this cultural practice. There is no doubt that when examining African traditional medicine, holistic terms such as "blood cleansing" cannot be ignored. Future studies may provide a better understanding of this often misunderstood and undocumented practice. The analyses of patterns of over-representation of southern African traditional medicinal plants at the genus and family level provide information about the differences in plant use among southern African countries and disease categories. 'Over-representation' refers to the phenomenon whereby the proportion of plants belonging to a taxonomic group is higher in ethnobotanical pharmacopoeia than in the total flora. This study aimed to use the Imprecise Dirichlet Model (IDM) to analyse data from ten southern African countries to establish how over-represented medicinal plant families compare with over-represented genera, how over-represented medicinal taxa differ among countries in this region of Africa, and how over-represented taxa differ among six major disease categories. Floral data for the total species composition of each country were obtained from online databases. Medicinal plant species lists were generated from published surveys, inventories, and books. IDM calculations were executed using the inverse of the cumulative beta probability density function in Microsoft Excel™. Python programming language source code was used to calculate Pearson correlation (r) values and Jaccard coefficients (J). Nine of forty-two over-represented medicinal plant families in southern Africa (group 1) do not have over-represented genera. Seven of the forty genera with the highest margins of over-representation belong to under-represented families. Nineteen of the forty-two over-represented families have margins of over-representation smaller than the cumulative margins of their over-represented genera. Groups of countries with similar overall flora (J ≥ 0.333) are Botswana and Namibia (group 2), Malawi, Mozambique, Zambia and Zimbabwe (group 3). The families and genera with the highest margins of over-representation are Loganiaceae and Albizia in group 1, Combretaceae and Vachellia in group 2, Dioscoreaceae and Senna in group 3, and Sapotaceae and Solanum in group 4 (South Africa). The families and genera with the highest margins of over-representation across disease categories are Ebenaceae and Albizia , Canellaceae and Dicoma , Combretaceae and Pterocelastrus , Ebenaceae and Bersama , Francoaceae and Erythrina , and Aristolochiaceae and Strychnos for plants used in the treatment of STIs, febrile and mosquito-vector diseases, microbial infections, pain, skin conditions, and female sexual/reproductive problems, respectively. Genus-level calculations are more efficient in generating taxonomic lists that can be used for ethnopharmacological investigations due to the exclusion of under-represented genera. Limiting the size of geographical areas from which medicinal plant lists are sampled and targeting plants used to treat specific types of disease prevents the underestimation of niche over-represented taxa. Solanum incanum L. is commonly used in traditional herbal medicine (THM) in Kenya for treating various ailments. Recent developments in disease treatment have introduced the concept of host-directed therapy (HDT). This approach involves targeting factors within the host cell that can impede the growth or replication of a pathogen. One such host factor is delta aminolevulinate dehydratase (δ-ALAD), the second enzyme in the heme biosynthesis pathway utilized by Plasmodium for growth. Studies using mice models have shown an increase in δ-ALAD expression during Plasmodium berghei infection. Another plant in the Solanum genus, S. guaranticum , has been found to inhibit δ-ALAD in red blood cells in vitro and in the brain in vivo. Is it possible that the bioactive compounds in S. incanum extracts could also be effective in HDT for malaria treatment? To better assess the effectiveness of S. incanum leaf extracts as a curative and prophylaxis in malaria parasite infection, and to test the plant's ability to decrease δ-ALAD expression. The leaves of S. incanum were collected, dried, and pulverized before being subjected to a successive extraction protocol to obtain crude, hexane, ethyl acetate, and aqueous extract fractions. Phytochemical analysis was conducted on all extract fractions, followed by GC-MS analysis of the fraction with the most potent antimalarial activity. An acute toxicity study was also performed on the extracted fractions. The potency of the extract fractions as curative and prophylactic antimalarial was then evaluated in THM using Plasmodium berghei -infected mice at a dose of 100 mg/kg. The extract fraction with the highest activity was further evaluated at varying doses and its effect on δ-ALAD was measured using RT-qPCR. The percentage of parasitemia and chemosuppression, and mean survival time were used as indices of activity. Phytochemical analysis revealed that the ethyl acetate and aqueous extract fractions contained high terpenoids, flavonoids, and phenols levels. However, alkaloids were only present in moderate quantities in the aqueous extract, and quinones were found in high levels only in the crude extract. Additionally, all extract fractions contained saponins in high levels but lacked tannins. While the plant extracts were found to be non-toxic, they did not exhibit curative antimalarial activity. However, all extract fractions showed prophylactic antimalarial activity, with the ethyl acetate extract having the highest percentage of chemosuppression even at doses of 250 and 1000 mg/kg. In the negative control, the expression of δ-ALAD was 5.4-fold, but this was significantly reduced to 2.3-fold when mice were treated with 250 mg/kg of the ethyl acetate fraction. GC-MS analysis of the ethyl acetate fraction revealed high percentages of 2-methyloctacosane, tetracosane, and decane. The fractions extracted from S. incanum leaves have been found to possess only antimalarial prophylactic properties, with the ethyl acetate extract fraction showing the most effective results. The activity of this fraction may be attributed to its ability to decrease the expression of δ-ALAD, as it contains an alkane compound implicated with enzyme-inhibitory activity. A comprehensive checklist and analysis of medicinal plant species and their uses in Mozambique, and a comparison with South Africa provided a more profound understanding of the broad concept of Traditional African Medicine as a healing culture and the need for a more informative classification system. This study was aimed at recording all medicinal plants reported in Mozambique according to literature and answer four main questions. 1. How many medicinal plant species have been recorded for Mozambique? Of those, how many are indigenous to Mozambique, and how many are introduced or cultivated exotics? 2. What are the main medicinal uses in Mozambique and which species are used for which ailments? 3. What are the similarities and differences between the medicinal floras of Mozambique and South Africa – are the same species used for the same ailments and is there evidence of cultural exchange? 4. Do the data provide new insights into Traditional African Medicine as one of the oldest medicine systems in the world? A literature survey was done to compile the medicinal checklist of Mozambique using 29 available literature sources, which were found from published books, journal articles, reports, unpublished theses, dissertations, and online databases. A total of 731 medicinal plant species and infraspecific taxa from 447 genera and 120 families was recorded for Mozambique. Of these, 590 (81%) are indigenous, and 87 are non-indigenous (50 naturalised and 37 cultivated exotics). Of the 731 medicinal plant species, 494 (68%) are also used medicinally in South Africa and 148 (30%) are used for the same or similar ailments. Many of the shared taxa and uses have been recorded in the adjoining KwaZulu-Natal Province of South Africa, which has similar vegetation types. The similarity between Mozambique and South Africa (and especially the KwaZulu-Natal Province) not only suggest an exchange of traditional knowledge but also indicates the existence of a hitherto unnamed medicinal system of the southern and eastern African Bantu-speaking cultures that is in need of detailed comparative studies. Thespesia garckeana (F. Hoffm.) Exell & Hillc. is called Gorontula (Kola of Tula) in Nigeria, Morojwa in Bostwana, and Thespesia garckeana in South Africa and is widely distributed across Africa. Its parts reportedly possess multiple medicinal properties and are employed for treating various diseases. In Tula, Gombe State, Nigeria, the ripe fruit decoction is taken as remedy for female infertility as documented by Ochokwu and co in the Journal of Biology, Agriculture and Healthcare in 2015. This research examined the effects of aqueous fruit extract of T. garckeana (F. Hoffm.) Exell & Hillc. (AFETG) on selected reproductive tissues and hormones in female rats and also evaluated the inhibitory potentials of its phytoconstituents against human 5-alpha reductase 2 (SRD5α2) using in silico approach. Twenty-five (25) sexually matured female rats were randomized into 5 groups (i.e. A - E). Oestrous in the rats was synchronized (subcutaneous oestradiol conjugate [10 μg/100 g BW]). Group A received distilled water (control). Group B received standard drug, clomiphene citrate (0.85 mg/kg BW), while groups C, D, and E received AFETG at 50, 100, and 200 mg/kg BW respectively. The animals were treated for five (5) days and sacrificed 24 h after. Their blood was collected and prepared for analysis of serum prolactin, oestradiol (E2), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) while ovaries, uteruses, and oviducts were studied for histological and histomorphometric changes. For the in silico study, the target protein, human steroid 5α-reductase 2 (SRD5α2) was prepared and its receptor grid was generated using Optimized Potential for Liquid Simulations-2005. The ligand 2D structures were prepared using LigPrep 2.4 software and docked using Glide. The binding energy of the ligands to the protein receptor was predicted using Molecular Mechanics/Generalized Born Surface Area (MM-GBSA) analysis. AFETG significantly increased serum levels of E2 but did not alter serum levels of prolactin, LH and FSH when compared with distilled water and clomiphene citrate. AFETG also significantly increased ovarian tertiary follicular diameter, oviductal epithelial height and serosa thickness as well as uterine endometrial epithelial height, endometrial thickness, and myometrial thickness when compared with control. Ovarian secondary follicular diameter and oviductal submucosa thickness and muscular thickness were significantly decreased by AFETG when compared with control. Two compounds in T. garckeana (F. Hoffm.) Exell & Hillc.; D-Melezitose (−12.55 kcal/mol XP GScore) and 1, 3, 4-trihydroxy-5-oxo cyclohexane-1-carboxylic acid (−9.136 kcal/mol XP GScore) exhibited higher binding affinities for SRD5α2 than the reference ligand, epristeride (−8.096 kcal/mol XP GScore). In conclusion, the ability of AFETG to increase serum E2 level, thickness of uterine endometrium and ovarian tertiary follicles size can be explored for the treatment of female infertility caused by thinning of the uterine endometrium and reduced follicular size. Two compounds in AFETG (i.e. D-Melezitose and 1, 3, 4-trihydroxy-5-oxo cyclohexane-1-carboxylic acid are potential inhibitors of SRD5α2, thus aiding the biosynthesis of E2. While access to antiretroviral therapy (ART) continues to improve worldwide, HIV infection and AIDS persist as serious health challenges, particularly in sub-Saharan Africa. Complementary and Alternative Medicines (CAM), as part of indigenous and pluralistic medical systems, are important contributors to primary health care worldwide. However, this knowledge remains relatively undocumented in many parts of sub-Saharan Africa such as the Tutume subdistrict of Central Botswana, where CAM is widely used including potentially for HIV/AIDS and HIV-associated conditions. To explore the extent to which CAM is used by the BaKalanga Peoples of the Tutume subdistrict, we performed an exploratory community-based project to record medicinal plant use from this relatively undocumented region, with a particular focus on species used for management of HIV/AIDS and HIV-associated conditions. Using the snowball sampling technique, we recruited 13 Traditional Health Practitioners (THPs) and conducted in-depth interviews to explore medicinal plant uses and treatment regimens. Plant specimens were collected and bio-authenticated. We documented 83 plant species used as CAM to treat or manage a variety of conditions including HIV/AIDS, HIV-associated conditions, and other health conditions. Plants from the family Leguminosae were most frequently reported, comprising 21 species (25.3%), followed by 5 from both Euphorbiaceae and Combretaceae families (6.0%). Four plants (4.8%) were used specifically to manage HIV (Lannea edulis (Sond.) Engl. root, Aloe zebrina Baker root, Myrothamnus flabellifolia Welw. whole plant, and Harpagophytum procumbens var. subulobatum (Engl.) tuber), while an additional 7 (8.4%) were reported specifically for treating combinations of HIV-related symptoms. Notably, 25 (30.1%) have not been reported previously as CAM and/or lack reported bioactivity data. To our knowledge, this is the first detailed ethnobotanical survey of CAM used by the BaKalanga Peoples of the Tutume subdistrict to manage HIV/AIDS and HIV-associated and other health conditions. Abstract Ethnopharmacological relevance Inflammation is a complex mechanism employed by the body to promote healing and restoration to normal function in the event of injury. Eleven plant species were selected in this study based on their use in traditional medicine against inflammation in South Africa. Methods Hexane, acetone, ethanol, methanol and water extracts of the powdered plants were prepared and a total of fifty-five extracts were tested for their anti-inflammatory and antioxidant activities. The anti-inflammatory activity of extracts was evaluated via the 15-lipoxygenase (15-LOX) inhibitory and the nitric oxide (NO) inhibition assays using lipopolysaccharide (LPS)-activated RAW 264.7 murine macrophages. Total flavonoid and total phenolic contents were determined. The antioxidant activity of the extracts was performed using radical scavenging DPPH (2, 2-diphenyl-1-picrylhydrazyl) and electron reducing ABTS (2, 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) assays. Results The hexane extract of Typha capensis (TC) had good lipoxygenase inhibitory activity with IC 50 of 4.65 µg/mL, significantly (p < 0.05) higher than that of the positive control quercetin (IC 50 = 24.60). The same extract also had good nitric oxide inhibitory activity with 86% NO inhibition and cell viability of 97% at 50 µg/mL. The TC acetone extract had the best antioxidant activity with IC 50 of 7.11 and 1.91 µg/mL respectively in the DPPH and ABTS assays. Following fractionation of the TC plant material, the ethyl acetate fraction had interesting antioxidant activity and the methanol/water (35%) and hexane fractions had good 15-LOX inhibitory activity. The antioxidant and anti-inflammatory activities therefore resided in both polar and more non-polar fractions. Conclusion The acetone extract of Typha capensis and its fractions had good anti-inflammatory and antioxidant activities, supporting the medicinal use of this species against inflammation. Other species including Ficus elastica, Carpobrotus edulis, Cotyledon orbiculata and Senna italica also had good activity worthy of further investigation. Abstract Ethnopharmacological relevance Piptadeniastrum africanum is widely used in treating oxidative stress related diseases. Oxidative stress, defined as the disturbance in the balance between the production of free radicals and antioxidant defenses, is the root cause of many pathophysiological conditions. Based on the dual properties of prooxidants as toxic and beneficial compounds, both prooxidants and antioxidants may be effective in the treatment of these conditions when the right dose is given to the right subject at the right time for the right duration. Aim of the study This study was aimed at investigating the in vitro and ex vivo anti- and pro-oxidative effects of P. africanum. Materials and methods Total phenolic and flavonoid contents of methanol and aqueous extracts of P. africanum stem back were quantified spectrophotometrically. The methanol extract, ascorbate radicals and reactive oxygen species in brain and liver homogenates of mice treated with the methanol stem bark extract were analyzed by electron paramagnetic resonance (EPR) spectroscopy. Free radical scavenging of DPPH was determined by spectrophotometric and EPR assays. Results The methanol extract was richer in both phenolic and flavonoid contents compared to the aqueous extracts and also showed better DPPH radical scavenging capacity. The EPR spectroscopy in vitro analysis exhibited high DPPH scavenging capacity before and after UV irradiation (99.5% and 98.76%) at 40 μg/ml extract. The ex vivo EPR spectroscopy studies demonstrated increased levels of ascorbate radicals (•Asc) in liver and brain homogenates of healthy mice treated with P. africanum in comparison with those of the non treated controls (0.6141 ± 0.026 vs 0.1800 ± 0.0073 arb. units for liver homogenates and 0.9605 ± 0.0492 vs 0.3375 ± 0.0062 arb. units for brain homogenates, correspondingly). Considerably, higher levels of reactive oxygen species (ROS) were measured in mice liver and brain homogenates after treatment with P. africanum extract compared to the control group, as well (1.9402 ± 0.1200 vs 0.6699 ± 0.062 arb. units for liver homogenates and 1.7325 ± 01503 vs 0.3167 ± 0.0403 arb.units, respectively). Conclusion Therefore, P. africanum exhibited antioxidant and pro-oxidant properties which may explain its broad spectrum use in a wide variety of ailments. Abstract Ethnopharmacological relevance Anthocleista vogelii Planch is a medicinal plant traditionally used in West Africa for the management and treatment of diabetes mellitus. Aim of the study To determine the antidiabetic activities of chloroform fraction (CF) of Anthocleista vogelii Planch root bark in rats with diet- and alloxan-induced obesity-diabetes. Materials and methods Inhibitory activities of CF against α-amylase and α-glucosidase activities were determined in vitro. Three weeks old rats were fed with high-fat diet for 9 weeks to induce obesity prior to further induction of diabetes using alloxan (150 mg/kg body weight, i.p.). Blood glucose levels and body weight were measured every 7 days throughout the experiment. Glucose tolerance was assessed in normal and CF-treated rats on day 21. Terminal blood samples were collected from sacrificed animals for the measurement of serum insulin levels. Pancreases were excised from treated and untreated animals for histopathological examination. Results LCMS/MS chromatographic profile of CF via positive and negative modes revealed 13 and 23 compounds respectively. Further analysis revealed quebrachitol (QCT), loganin, sweroside, oleoside 11-methyl ester and ferulic acid, which have been previously reported for their antidiabetic activities, as constituents of CF. CF inhibited activities of α-amylase (IC 50 = 51.60 ± 0.92 µg/ml) and α-glucosidase (IC 50 = 5.86 ± 0.97 µg/ml) in a dose-dependent manner. Treatment of animals with obesity-diabetes with 100 and 200 mg/kg CF significantly improved glucose tolerance (P < 0.001) and enhanced serum insulin levels (P < 0.05) compared to diabetic control rats. Conclusions Antidiabetic activities of CF might be mediated via inhibition of α-amylase and α-glucosidase activities, elevation of serum insulin concentration, and enhancement of insulin and leptin sensitivity in obesity-diabetes rats. This study further substantiates the traditional use of A. vogelii in the management and treatment of diabetes in Africa and encourages further studies to investigate its mechanism of action. Ethnopharmacological relevance According to previous ethno-medicinal reviews, Cocos nucifera , Elaeis guineensis and Phoenix dactylifera are among the main palms which are often used on the American and African continents to treat infections, infestations and disorders in the digestive, respiratory, genito-urinary, dermal, endocrine, cardiovascular, muscular-skeletal, mental and neural systems, as well as neoplasms, dental issues and metabolic and nutritional disorders. In addition, one or more species of the wild genera Acrocomia , Areca , Astrocaryum , Attalea , Bactris , Borassus , Calamus , Chamaedorea , Chamaerops , Euterpe , Hyphaene , Mauritia , Oenocarpus and Syagrus have a high number of records of these ethno-medicinal uses. The most used parts of the palm tree are the fruits, followed by roots, seeds, leaves and flower sap. Aim of the study This review discusses the phytochemical composition and the pharmacological properties of these important ethno-medicinal palms, aiming to provide a contribution to future research prospects. Materials and methods Significant information was compiled from an electronic search in widely used international scientific databases (Google Scholar, Science Direct, SciFinder, Web of Science, PubMed, Wiley on line Library, Scielo, ACS Publications), and additional information was obtained from dissertations, theses, books and other relevant websites. Results Palms, in general, are rich in oils, terpenoids and phenolic compounds. Fruits of many species are notable for their high content of healthy oils and fat-soluble bioactive compounds, mainly terpenoids, such as pigment carotenoids (and provitamin A), phytosterols, triterpene pentacyclics and tocols (and vitamin E), while other species stood out for their phenolic compounds derived from benzoic and cinnamic acids, along with flavan-3-ol, flavone, flavonol, and stilbene compounds or anthocyanin pigments. In addition to fruits, other parts of the plant such as seeds, leaves, palm heart, flowers and roots are also sources of many bioactive compounds. These compounds are linked to the ethno-medicinal use of many palms that improve human health against infections, infestations and disorders of human systems. Conclusions Palms have provided bioactive samples that validate their effectiveness in traditional medicine. However, the intensive study of all palm species related to ethno-medicinal use is needed, along with selection of the most appropriate palm accessions, ripe stage of the fruit and /or part of the plant. Furthermore, the complete profiles of all phytochemicals, their effects on animal models and human subjects, and toxicological and clinical trials are suggested, which, added to the incorporation of improved technological processes, should represent a significant advance for the implementation of new opportunities with wide benefits for human health. Ethnopharmacological relevance In West Africa, populations are used to taking traditional medicine as a first aid against common health problems. In this aspect, many plants are claimed to be effective in the treatment of Tuberculosis (TB), which according to the World Health Organization (WHO) remains one of the world’s deadliest communicable diseases. Aim of the study The main aim of this study was to identify plants used to treat TB-symptoms by the population of Senegal and to evaluate their possible concomitant use with clinically approved TB-drugs. This approach allowed the selection of plants effectively used in traditional medicine. In order to verify if the usage of some of these plants can be rationalized, the activity of their traditional preparations was assessed with both an intracellular and extracellular antimycobacterial host-pathogen assays. Materials and methods An ethnopharmacological survey conducted on 117 TB-patients and 30 healers in Senegal from March to May 2014. The questionnaires were focused on the use of medicinal plants to treat common TB -symptoms (cough longer than 2 weeks, fever, night sweats, weight loss and bloody sputum). Local plant names, utilized organs (herbal drugs) and traditional formulations of the plants were recorded. Extracts were prepared by mimicking the traditional decoction in boiling water and screened for their antimycobacterial activity using Mycobacterium marinum, as a validated TB surrogate, and an Acanthamoeba castellanii - M. marinum whole-cell based host-pathogen assay, to detect anti-infective activities. Results By the end of the survey, nearly 30 plants were cited and the 12 most cited herbal drugs were collected and their usage documented by extensive literature search. Extracts of the chosen herbs were screened with the described assays; with a main focus on traditional formulas (mainly herbal decoctions). Two of the water extracts from Combretum aculeatum and Guiera senegalensis showed significant antimycobacterial activities when compared to the positive control drug (rifampin). These extracts showed no observable toxicity against amoeba host cells ( Acanthamoeba castellanii ). Conclusions This study demonstrates that most of the patients do not concomitantly use plants and TB drugs (~90% of informants) but, instead, most are treated with medicinal plants before they are admitted to a hospital (41%). Interestingly, among the aqueous extracts assayed, two extracts ( Combretum aculeatum (Combretaceae) and Guiera senegalensis (Combretaceae)) collected within this survey demonstrate antimycobacterial activities on the validated whole-cell based host-pathogen assay. Both extracts showed significant activities against intracellular and extracellular - M. marinum growth presenting IC 50 lower than 0.5 mg/ml compared to the reference drug Rifampin (IC 50 of 0.4 and 7 µg/ml). Ethnopharmacoalogical relevance The genus Nauclea in Africa comprises seven species. Among them, N. latifolia , N. diderrichii and N. pobeguinii are widely used by the local population in traditional remedies. Preparation from various parts of plants (e.g. roots, bark, leaves) are indicated by traditional healers for a wide range of diseases including malaria, pain, digestive ailments or metabolic diseases. Materials and methods A literature search was conducted on African species of the genus Nauclea using scientific databases such as Google Scholar, Pubmed or SciFinder. Every document of ethnopharmacological, phytochemical or pharmacological relevance and written in English or French were analyzed. Results and discussion The Nauclea genus is used as ethnomedicine all along sub-Saharan Africa. Several local populations consider Nauclea species as a major source of remedies for malaria. In this regard, two improved traditional medicines are currently under development using extracts from N. latifolia and N. pobeguinii . Concerning the chemical composition of the Nauclea genus, indoloquinolizidines alkaloids could be considered as the major class of compounds as they are reported in every analyzed Nauclea species, with numerous structures identified. Based on traditional indications a considerable amount of pharmacological studies were conducted to ensure activity and attempt to link them to the presence of particular compounds in plant extracts. Conclusion Many experimental studies using plant extracts of the African species of the genus Nauclea validate traditional indications (e.g. malaria and pain). However, bioactive compounds are rarely identified and therefore, there is a clear need for further evaluations as well as for toxicity experiments. The sustainability of these plants, especially of N. diderrichii , a threatened species, should be kept in mind to adapt local uses and preparation modes of traditional remedies. Ethnopharmacological relevance According to Saharian traditional medicine, Anvillea radiata Coss. & Dur. (Asteraceae) has been valued for treating a variety of ailments such as gastro-intestinal, liver and pulmonary diseases, and has gained awareness for its beneficial effect on postprandial hyperglycemia. However, to best of our knowledge, no detailed study of the antidiabetic curative effects of this plant has been conducted yet. Aim of the study To determine the hypoglycemic and antidiabetic effect of dietary supplementation with Anvillea radiata extracts on high-fat-diet (HFD)-induced obesity and insulin resistance in C57BL/6 J mice in relation with antioxidant, anti-inflammatory, pancreatic beta-cells and skeletal muscle protection, and digestive enzyme inhibiting properties. Materials and methods Six extracts (water soluble and organic) from aerial parts of the plant were analyzed phytochemically (total phenolic and flavonoid content) and screened for in vitro superoxide (by chemiluminescence) and hydroxyl radical (by electron paramagnetic resonance spin-trapping) scavenging, antioxidant (DPPH, TRAP and ORAC assays), xanthine oxidase, metal chelating, α -amylase and α -glucosidase inhibitory property, and protective effects on copper-induced lipoprotein oxidation. Then selected hydroalcoholic and aqueous extracts were assessed for toxicity in normal human lung fibroblasts and A549 cancer cells using FMCA and MTT assays. Two water-soluble extracts having the best overall properties were assessed for their (i) protective effect at 1−15 µg/mL on metabolic activity of rat insulinoma-derived INS-1 cells exposed to hyperglycemic medium, and (ii) acute hypoglycemic effect on 16-weeks HFD-induced diabetic mice. Then diabetic mice were administered HFD supplemented by extracts (up to 150 mg/kg/day) for 12 additional weeks using standard diet as control and the antidiabetic drug, metformin (150 mg/kg), as positive control. Then the antidiabetic, anti-inflammatory and antioxidant activity of extracts were determined. Results Of the highly efficient polyphenolics-enriched hydroalcoholic and ethyl acetate extracts, the lyophilized aqueous (AQL) and butanol extracts were not toxic in cells (≤ 400 µg/mL) or when given orally in normal mice (≤ 2000 mg/kg), exerted a dose-dependent hypoglycemic action in diabetic mice, which was maximal at the dose of 150 mg/kg. Upon administering this dose for 12 weeks, both extracts significantly ameliorated body weight control capacity, recovery of plasma glucose and insulin level, reduced oxidative stress in blood, myocardial and skeletal muscles, and improved hyperlipidemic and inflammatory status. Moreover, diabetes-related complications were optimally ameliorated by oral therapy based on halved doses (75 mg/kg) of a mixture of AQL and metformin. Conclusions Current investigation supports the traditional medicinal usage of Anvillea radiata and suggests that both readily accessible and low-cost bio-extracts have the potency to develop an antihyperglycemic, antihyperlipidemic and protective agent against beta-cells and muscle dysfunction at doses compatible with the common practices of indigenous people for the management of metabolic disorders. Ethnopharmacological relevance Pittosporum viridiflorum Sims, a Pittosporaceae species, is used extensively in African traditional medicine (ATM) by various tribes. This review is an appraisal of the information concerning the description, distribution, conservation status, traditional uses, phytochemistry, pharmacology and toxicology of this species with the aim of reconciling it with its traditional use. Materials and methods A wide-ranging literature search was conducted using database platforms such as Scopus, Google Scholar, Web of Science, ScienceDirect, PubMed and books including local reports and thesis submissions. Results Ten categories to which P. viridiflorum finds use in traditional medicine (TM) were found, and they include well-being, wounds, treatment of veterinary ailments, gastrointestinal and sexually transmitted diseases, kidney, circulatory and inflammatory disorders, as well as diseases such as cancer, tuberculosis, and malaria. Pharmacological tests conducted include those investigating antimicrobial, antidiarrhoeal, antimalarial, anticancer, anti-inflammatory, antioxidant and acaricidal properties. Promising activity was shown in a number of assays. Toxicological effects have also been reported from this species. However, it is recommended to conduct a detailed toxicological study, including genotoxicity, as this has not yet been evaluated. Compound(s) with antimalarial, anticancer and acaricidal properties have been isolated from P. viridiflorum. Conclusions The collective pharmacological and phytochemical properties of P. viridiflorum gives credence to the use of this plant species against various diseases in ATM, thus steering significant interest towards in vivo studies and further research. Ethnopharmacological relevance Decoctions of the leaves of M. benthamianum Baill. are used by traditional healers in Guinea to treat malaria and this use was validated by a preliminary clinical assay. Aim of the study To evaluate the in vitro antiplasmodial activity and to identify active compounds from extracts of M. benthamianum leaves. Material and methods Antiplasmodial activity of extracts, fractions and pure compounds was evaluated in vitro against a chloroquine-sensitive strain of Plasmodium falciparum (3D7) using the measurement of the plasmodial lactate dehydrogenase activity. Selectivity of extracts and purified compounds for Plasmodium parasites was evaluated by using WST-1 test on HeLa human cells. Compounds were isolated using normal phase silica gel column chromatography and prepHPLC and their structures elucidated using extensive spectroscopic analysis. Results Hydroethanolic extracts (70% v/v) of M. benthamianum leaves showed a moderate in vitro activity against P. falciparum 3D7, with IC 50 in the range 22.5 – 32.6 µg/mL, depending on the batch; while a dark precipitate formed during ethanol evaporation showed higher activity (IC 50 =6.5 µg/mL). The fractionation was performed on this most active fraction and was followed by in vitro antiplasmodial assay. Active compounds (5, 7, 8) belong to several phytochemical classes, contributing together to the global antiplasmodial activity of the hydroethanolic extract against P. falciparum parasite. This study finally allowed the isolation of three diterpenes including two new compounds named Mezobenthamic acids A (1) and B (2) and neocaesalpin H (3), as well as quercetin (4), kaempferol (7), resveratrol (6), gallic acid (9) and its ethylester (5), β-sitosterol glucoside (10) and 13b-hydroxy-pheophorbide a (8). Conclusion This study gives some concrete evidence to support the ethnopharmacological use of Mezoneuron benthamianum leaves extract in the management of malaria. The active compounds can be further studied for their antiplasmodial potential, as well as their suitability to be used as quality markers for the standardization of this herbal drug from the Guinean traditional pharmacopeia. Ethnopharmacological relevance Sutherlandia frutescens is a traditional African medicinal plant used in the treatment of stress and anxiety, while also exhibiting anti-inflammatory properties. Aim of study The study aimed at linking anti-stress and anti-inflammatory properties of S. frutescens to its influence on glucocorticoid biosynthesis and the inflammatory response via steroid receptor interaction. Materials and methods The influence of S. frutescens extracts and sutherlandioside B (SUB),10 and 30 µM, on key steroidogenic enzymes was assayed in COS-1 cells. Effects were also assayed on basal and stimulated hormone levels in the adrenal H295R cell model. Agonist activity for transactivation and transrepression of the extract and SUB with the glucocorticoid- (GR) and mineralocorticoid receptor (MR) was subsequently investigated. Results Inhibitory effects of the extract towards progesterone conversion by CYP17A1 and CYP21A2 were significant. SUB inhibited CYP17A1 and 3β-HSD2, while not affecting CYP21A2. In H295R cells, SUB decreased cortisol and androgen precursors significantly. The extract decreased total steroid production (basal and stimulated) with cortisol and its precursor, deoxycortisol, together with mineralocorticoid metabolites significantly decreased under forskolin stimulated conditions. S. frutescens extracts and SUB repressed NF-κB-driven gene expression without activating GRE-driven gene expression and while neither activated MR mediated gene transcription, both antagonized the effects of aldosterone via the MR. Conclusion Data provide evidence linking anti-stress, anti-inflammatory and anti-hypertensive properties of S. frutescens to inhibition of steroidogenic enzymes and modulation of adrenal hormone biosynthesis. Findings suggesting S. frutescens and SUB exhibit dissociated glucocorticoid characteristics underline potential therapeutic applications in the treatment of inflammation and hypertension. Ethnopharmacological relevance Tetracera potatoria Afzel. Exg. Don (Dilleniaceae) is a medicinal plant used traditionally in Africa for the treatment of tuberculosis related ailments and respiratory infections. The antibacterial activity of the medium polar extracts of T. potatoria leaves and stem bark was recently reported against Mycobacterium smegmatis (MIC 25 µg/mL) and M. aurum (65 µg/mL), two fast-growing Mycobacterium strains used as model micro-organisms for the more pathogenic strain Mycobacterium tuberculosis ( Fomogne-Fodjo et al., 2014 ). The aim of this study was consequently to isolate the compounds possibly contributing to this activity, and which may therefore be promising precursors to be used for the development of novel anti-TB drugs. Materials and methods T. potatoria medium polar extract [MeOH/DCM (1:1, v/v)] was fractionated sequentially with petroleum ether to which EtOAC and MeOH were gradually added to increase the polarity. The examination of T. potatoria extract and its fractions was guided by bioassays for anti-mycobacterial activity against M. smegmatis (ATCC 23246) and M. aurum (NCTC 10437) using the minimum inhibitory concentration (MIC) method. All the isolated compounds were structurally elucidated using spectroscopic techniques and evaluated for their anti-mycobacterial activity. Results Two novel secondary metabolites ( 1 , 2 ) named tetraceranoate and N -hydroxy imidate-tetracerane, together with five known compounds [ β -stigmasterol ( 3 ), stigmast-5-en-3 β -yl acetate ( 4 ), betulinic acid ( 5 ), betulin ( 6 ) and lupeol ( 7 )] were isolated and identified. Tetraceranoate exhibited the best activity against M. smegmatis with a minimum inhibitory concentration (MIC) of 7.8 µg/mL, while β -stigmasterol, betulinic acid and betulin showed appreciable anti-mycobacterial activity against both strains (MIC 15 µg/mL). Conclusion Seven compounds were isolated from the medium polar extract [MeOH/DCM (1:1, v/v)] of T. potatoria stem bark. Only tetraceranoate one of the isolated compounds showed antibacterial activity against M. smegmatis having efficacy as high as rifampicin (one of a three drug regimen recommended in the initial phase short-course anti-tuberculosis therapy). Thus, tetraceranoate might be an interesting target for systematic testing of anti-TB treatment and management. Multiple plant species are used traditionally in southern Africa to treat fever. This review summarises the traditional use, highlights promising plant species and focuses on validation studies to highlight future areas of research. This study aimed to critically review the use of southern African plants to alleviate fever. Additionally, an examination of research into those plants was undertaken to highlight gaps in the literature with the aim of stimulating further research in this field. Ethnobotanical books, reviews and primary research studies were searched to identify plant species traditionally used to treat fever in southern Africa and to summarise the current knowledge and identify areas requiring further research. This study was non-biased and did not have any taxonomic preferences, nor did it favour any southern African healing systems over the others. The traditional therapeutic uses of 203 southern African plant species to alleviate fever were recorded. Leaves and roots/bulbs/rhizomes were the most commonly used plant part, and they were most commonly prepared as decoctions or infusions and consumed orally. Notably, only twenty-two species (∼11% of the identified species) have been scientifically evaluated for antipyretic properties, with substantially fewer mechanistic studies evident. Despite the availability of ethnobotanical records, the antipyretic properties of southern African medicinal plants are poorly reported. Ethnopharmacological relevance Crinum macowanii is a deciduous bulbous plant which grows in east, central and southern Africa. Crinum macowanii has been used as herbal medicine by the indigenous people of east and southern Africa has for several centuries. The bulb, leaves and roots of C. macowanii are reported to possess diverse medicinal properties and used to treat or manage various human and animal diseases and ailments throughout its distributional range. Crinum macowanii is used traditionally as a remedy for boils, diarrhoea, fever, inflammation, respiratory system problems, skin rashes, tuberculosis, wounds and urinary tract problems. Aim of the review The present review aims to summarize comprehensively the research that has been done on the ethnomedicinal uses, botany, phytochemistry and pharmacology of C. macowanii in different locations throughout its geographical range in east, central and southern African region so as to highlight research gaps and provide a foundation for further investigations on the plant species. Materials and methods A review of the literature was undertaken and an in-depth analysis of previous research on ethnobotany, therapeutic value, phytochemistry and pharmacology of C. macowanii throughout its distributional range in east, central and southern Africa. Literature sources included papers published in international journals, reports from international, regional and national organizations, conference papers, books, theses, websites and other grey literature. Electronic search engines such as Google, Google scholar, publishing sites such as Elsevier, scienceDirect, BMC, PubMed and other scientific database sites such as ChemSpider, PubChem were used as well as searching the library collections of the National Herbarium and Botanic Gardens (SRGH), Harare, Zimbabwe and the University of Fort Hare, South Africa. Results A total of 32 ethnomedicinal uses of C. macowanii are documented in literature, which can be grouped into seven major ethnomedicinal general purpose usages, namely “fever”, “wounds, sores and skin rashes”, “boils and inflammation”, “respiratory system problems”, “blood cleansing”, “urinary tract problem” and “veterinary uses”. The chemical composition of C. macowanii is dominated by various isoquinoline alkaloids, which have been isolated from the bulbs, flowering stalks, leaves and roots. Major biological activities demonstrated by C. macowanii include antifungal, antiviral and antiplasmodial activities, cardiovascular effects as well as effects on the central nervous system. The population of C. macowanii is declining in the wild as the bulbs are over-collected for sale in medicinal ( muthi ) markets in southern Africa. Conclusion A literature search revealed that C. macowanii has a lot of potential as a possible source of pharmaceutical products for the treatment of a wide range of human and animal diseases and ailments. Some of the alkaloids isolated from C. macowanii have demonstrated various biological activities when investigated in in vitro assays. However, some of the ethnomedicinal uses of C. macowanii still require pharmacological investigations. Therefore, further studies are required to improve our knowledge about the mechanisms of action, efficacy, toxicity and clinical relevance of the plant species as well as its bioactive compounds. Ethnopharmacological relevance The aim of this study was to identify and document medicinal plants used to manage High Blood Pressure and Type 2 Diabetes Mellitus in Bitterfontein, Western Cape Province, South Africa. Methods One hundred and twelve (112) respondents were interviewed between August 2014 and September 2015 through semi-structured surveys to gather data on the percentage of people who had been diagnosed with High Blood Pressure and/or Type 2 Diabetes Mellitus and to determine the frequency of medicinal plant and allopathic medicine use. Twelve (12) key respondents were subsequently selected, using a non-probability snowball sampling method. They were interviewed in-depth concerning their plant practices and assisted with plant collection. Results Twenty-four plant (24) species belonging to 15 families were identified for the management of High Blood Pressure and Type 2 Diabetes Mellitus. The most frequently reported families were Asteraceae (20.8%), Lamiaceae (16.67%), Crassulaceae (8.33%) and Aizoaceae (8.33%). The remaining (45.54%) were evenly split over eleven families- Fabaceae, Amaryllidaceae, Anacardiaceae, Capparaceae, Geraniaceae, Apiaceae, Convolvulaceae, Apocynaceae, Rutaceae, Asphodelaceae and Thymelaeaceae. The most commonly used plant species overall was Lessertia frutescens (96.55%). The most frequently used plant parts included leaves (57.63%) roots/bulbs (15.25%) and stems (11.86%), mostly prepared as infusions or decoctions for oral administration. Conclusions Medicinal plants are widely used by High Blood Pressure and Type 2 Diabetes Mellitus sufferers. They employ diverse plant species to manage both conditions. In addition, some sufferers often use prescribed allopathic medication, as well as medicinal plants, but at different intervals. Background Malaria remains a major health problem worldwide especially in sub-Saharan Africa. In Kenya, 80% of the population is at risk of contracting the disease. Pregnant mothers and children under five years are the most affected by this disease. Antimalarial drug resistance poses a major threat in the fight against malaria necessitating continuous search for new antimalarial drugs. Due to inadequate and inaccessible health facilities, majority of people living in rural communities heavily depend on traditional medicine which involves the use of medicinal plants for the management of malaria. Most of these indigenous knowledge is undocumented and risks being lost yet such information could be useful in the search of new antimalarial agents. Aim of study An ethnobotanical survey was carried out among the Luhya community of Kakamega East sub-County, a malaria epidemic region, with the aim of documenting the plants used in the management of malaria. Materials and methods Semi-structured questionnaires were used to collect information from 21 informants who included traditional medicine practitioners and other caregivers who had experience in use of plants in management of malaria. These were drawn from 4 villages located in Kakamega East sub-county, within Kakamega County based on their differences in topography. Information recorded included plant names, parts used, mode of preparation and administration and the sources of plant materials. A literature search was conducted using PubMed and google scholar to identify the reported traditional uses of these plants and studied antiplasmodial activities. Results In this study, 57% of the informants were aged above 50 years and a total of 61% had either no formal education or had only attained primary school education. A total of 42 plant species belonging to 24 families were identified. Most plants used in the management of malaria in this community belonged to Lamiaceae (18%), Leguminosae (9%) and Compositae (9%) plant families. Plants mostly used included Melia azedarach L , Aloe spp, Ajuga integrifolia Buch. Ham, Vernonia amygdalina Del., Rotheca myricoides (Hochst.) Steane and Mabb, Fuerstia africana T.C.E.Fr., Zanthoxylum gilletii (De Wild.) P.G.Waterman and Leucas calostachys Oliv. Rumex steudelii Hochst.ex A. Rich and Phyllanthus sepialis Müll. Arg are reported for the first time in the management of malaria. Although Clerodendrum johnstonii Oliv. ( Jeruto et al., 2011 ) and Physalis peruviana L.(Ramadan et al., 2015) are reported in other studies for management of malaria, no studies have been carried out to demonstrate their antiplasmodial activity. The plant parts mostly used were the leaves (36%) and stem barks (26%). Majority of these plants were prepared as decoctions by boiling and allowed to cool before administration (66%) while infusions accounted for 28% of the preparations. The literature mined supports the use of these plants for the management of malaria since most of them have demonstrated in-vitro and in-vivo antiplasmodial activities. Conclusion Most of the reported plant species in this study have been investigated for antiplasmodial activity and are in agreement with the ethnomedical use. Two (2) plants are reported for the first time in the management of malaria. There is need for documentation and preservation of the rich ethnomedical knowledge within this community given that most of the practitioners are advanced in age and less educated. There is also the danger of over-exploitation of plant species as most of them are obtained from the wild, mainly Kakamega forest. Ethnopharmacological relevance Lippia multiflora is a plant with nutritional and pharmaco-therapeutic properties that is native to central and occidental Africa. The potential effects of plants on health are associated with their chemical composition. Therefore, the present study aimed to identify chemical variations in essential oils of Lippia multiflora as a function of geographic origin and time of annual harvest to determine optimal chemical profiles for ethno-pharmacotherapeutic applications. Materials and methods Experimental plants were cultivated at Abidjan (LPA), Toumodi (LPT) and Bondoukou (LPB). Natural Lippia multiflora seeds were sourced to produce standardized plants over a period of six months. Standard plants (n=40) were re-introduced into natural plots, cultivated for 12 months and leaves were sampled monthly in a standardized fashion. Essentials oils (n=36) were then extracted from these samples by hydro-distillation according to the European Pharmacopoeia and qualitatively and quantitatively analyzed using GC/FID and GC/MS. These data were then analyzed using Principal Component Analysis (PCA). Anti-inflammatory properties were also assessed against activated macrophages in vitro. Results The results indicated that chemical profiles and essential oil yields vary according to the location where the plants were cultivated. One essential oil chemotype corresponded to the LPA and LPT sites and one corresponded to the LPB site. Statistical analysis of the chemical profiles and monthly evolution of the three sites over a period of one year allowed assessment of variations in composition and the subsequent choice of the optimal harvest time for ethnopharmacological applications. Anti-inflammatory activity apparently correlated with chemical profiles of essential oils and the geographic origins of the plants. Ethnopharmacological relevance The genus Anogeissus (axlewood tree, ghatti tree, button tree and chewing stick tree) belongs to Combretaceae, includes eight species that are distributed in Asia and Africa. Plants are used as an ethnomedicine in Asia and Africa to treat various ailments like diabetes, fever, diarrhoea, dysentery, tuberculosis, wound healing, skin diseases (eczema, psoriasis), snake and scorpion venom. Based on the traditional knowledge, different phytochemical and pharmacological activities have been at the focus of research. The aim of this review is to provide updated, comprehensive and categorized information on the ethnobotany, phytochemistry, pharmacological research and toxicity of Anogeissus species in order to identify their therapeutic potential and directs future research opportunities. Materials and methods The relevant data was searched by using the keyword “Anogeissus” in “Scopus”, “Google Scholar”, “Web of Science”, “PubMed”, and “ScienceDirect” databases. Plant taxonomy was validated by the databases “The Plant List” and A.J. Scott, 1979. Results This review discusses the current knowledge of the ethnobotany, phytochemistry and in vitro as well as in vivo pharmacological evaluations carried out on the extracts and isolated main active constituents of Anogeissus genus. Among eight species, most of the phytochemical and pharmacological studies were performed on four species. About 55 secondary metabolites are isolated from the genus. Stem bark, leaf, seed, fruit, root of the plants are used for the treatment of several health disorders such as diabetes, fever, diarrhoea, dysentery, tuberculosis, wound healing, skin diseases (eczema, psoriasis), snake and scorpion venom. Gum ghatti obtained from Anogeissus latifolia is used after delivery as tonic and in spermatorrhoea. Many phytochemical investigations on this genus confirmed that it is rich in phenolic compounds. Modern pharmacology research has confirmed that the crude extracts or the isolated active compounds of the genus Anogeissus possess antioxidant, antimicrobial, wound healing, antiulcer, anti-inflammation, anti-diabetics, hepatoprotective, hypolipidemic, antiparasitic and neuroprotective effects. Conclusions This review confirms that some Anogeissus species have emerged as a good source of the traditional medicine for wound healing, inflammation, skin diseases, microbial infection and diabetes. Many traditional uses of Anogeissus species have now been validated by modern pharmacology research. Intensive investigations of all the species of Anogeissus regarding phytochemical and pharmacological properties, especially their mechanism of action, safety and efficacy could be the future research interests before starting clinical trials. Ethnopharmacological relevance Reproductive healthcare has been highlighted as a major challenge in Lesotho mainly due to the high prevalence of HIV/AIDS and sexually transmitted infections. As a result other reproductive ailments have not received much attention, particularly because healthcare facilities are already limited and many of them are inaccessible. For these reasons, medicinal plants play a major role in primary healthcare system in the country, in addition the plants are easily accessible, more affordable, and their use forms part of the cultural heritage. However, documentation of medicinal plants used for reproductive ailments is scattered, more importantly the biological and pharmacological properties, as well as toxicity of many of these plants are not yet known. Aim of the study To document the plants used by both male and female Basotho (residing in Lesotho) for the treatment of reproductive ailments, to explore their recorded biological and pharmacological effects as well as their toxicity, and to establish if these plants are used for similar purposes in other southern African cultures. Materials and methods The results stem from published findings of recent interviews of traditional medicinal practitioners in the Maseru District of Lesotho, first author's own experiences and observations from the Qacha's Nek District as well as comprehensive literature survey including numerous books and unpublished data. Electronic databases such as Google, Google Scholar, PubMed, and ScienceDirect were also used to search for the chemical compounds, pharmacological activity, and toxicity of the plants. Results A total of 87 plant species are reported to be used for the treatment of several reproductive problems such as infertility, complications associated with pregnancy (twelve plants are used to treat conditions such as colic, heartburn, nausea, and constipation), cleansing and/ or toning of the uterus (with a purpose either to induce pregnancy or to get rid of the placenta, for example Withania somnifera and Zantedeschia albomaculata ), difficult childbirth, as well treatment of breast and cervical cancer, cysts, fibroids, and testicular tumours (e.g. Hypoxis hemerocallidea ). For the toning of the uterus, it is common to use a combination of plants, e.g. Gunnera perpensa , Scabiosa columbaria , and Eriospermum ornithogaloides . Conclusions Of the 87 plants used for reproductive healthcare, the highest number (31) is used for the treatment of infertility (in both men and women). Ethnopharmocological relevance Members of ‘ Mycoplasma mycoides cluster’ are important ruminant pathogens in Africa. Diseases caused by these Mycoplasma negatively affect the agricultural sector especially in developing countries through losses in livestock productivity, mortality and international trade restrictions. There is therefore urgent need to develop antimicrobials from alternative sources such as medicinal plants to curb these diseases. In Kenya, smallholder farmers belonging to the Maasai, Kuria and Luo rely on traditional Kenyan herbals to treat respiratory symptoms in ruminants. In the current study extracts from some of these plants were tested against the growth of members of Mycoplasma mycoides cluster. Aim This study aimed at identifying plants that exhibit antimycoplasmal activities using an ethnobotanical approach. Materials and methods Kenyan farmers of Maasai, Luo and Kuria ethnic groups were interviewed for plant remedies given to livestock with respiratory syndromes. The plant materials were thereafter collected and crude extracts prepared using a mixture of 50% of methanol (MeOH) in dichloromethane (CH 2 Cl 2 ), neat methanol (MeOH), ethanol (EtOH) and water to yield four crude extracts per plant part. The extracts were tested in vitro against five strains of Mycoplasma mycoides subsp. capri , five strains of Mycoplasma mycoides subsp. mycoides and one strain of Mycoplasma capricolum subsp capricolum using broth micro-dilution assays with an initial concentration of 1 mg/ml. Minimum inhibitory concentration (MIC) of the most active extracts were determined by serial dilution. Results Extracts from five plants namely: Solanum aculeastrum , Albizia coriaria , Ekebergia capensis , Piliostigma thonningii and Euclea divinorum exhibited the highest activities against the Mycoplasma strains tested. Mycoplasma mycoides subsp. mycoides were more susceptible to these extracts than Mycoplasma mycoides subsp. capri and Mycoplasma capricolum susp. capricolum. The activities of the crude extracts varied with the solvent used for extraction. The MICs mean values of the active extracts varied from 0.02 to 0.6 mg/ml. Conclusions The results suggested that these plants could potentially contain antimicrobial compounds that might be useful for the treatment of respiratory diseases in ruminants. Future work should focus on the isolation and identification of the active compounds from the plant extracts that showed interesting activities and evaluation of their antimicrobial and cytotoxic potential. Ethnopharmacological relevance Syzygium guineense has been traditionally used in Mali in West Africa for the treatment of different diseases such as stomach problems, wounds, inflammations and various female disorders. Aims of the study (1) To perform an ethnopharmacological survey on the traditional use of S. guineense among Malian healers. (2) To isolate and identify chemical constituents from S. guineense leaves and to study their radical scavenging and enzyme inhibitory effects. Materials and methods In four different districts in Mali, 44 healers were interviewed about their medicinal use of S. guineense . A methanol extract of the leaves of this tree was prepared and further fractionated using different chromatographic methods. Isolated compounds were identified by 1D and 2D NMR spectroscopy. Extracts and isolated compounds were investigated as DPPH radical scavengers and as inhibitors of xanthine oxidase and 15-lipoxygenase, and the methanol extract was tested for toxicity towards Artemia salina nauplii. Results Major uses by Malian healers were against dermatosis, pain, malaria/fever and for wound healing. There was little consensus about the use in the different districts. Leaves were most commonly used. From the methanol leaf extract, the flavonoids gallocatechin ( 1 ), myricetin ( 2 ), myricetin-3- O -glucoside ( 3 ), myricetin-3- O -rhamnoside ( 4 ), myricetin-3- O -glucuronide ( 5 ) and myricetin-3- O -β-D-(6″-galloyl)galactoside ( 6 ), the gallotannins 1,2,3,6-tetra- O -galloyl-β-D-glucose ( 7 ) and 1,2,3,4,6-penta-O-galloyl-β-D-glucose ( 8 ), and the ellagitannins casuarictin ( 9 ) and casuarinin ( 10 ) were isolated. These ten polyphenols are all new for the species. The crude methanol extract was active as a radical scavenger and as an inhibitor of xanthine oxidase and 15-lipoxygenase. Among the isolated compounds, pentagalloylglucose was the best enzyme inhibitor (IC 50 25±4 μM for 15-lipoxygenase, 8±1 μM for xanthine oxidase), while casuarictin (IC 50 3.9±0.1 μM), casuarinin (IC 50 4.5±0.3 μM) and pentagalloylglucose (IC 50 5±1 μM) showed the highest radical scavenging activity. The methanol extract was non-toxic to Artemia salina nauplii. Conclusion S. guineense leaves are commonly used among Malian healers, however the traditional practice varies a lot between different regions. The leaves of S. guineense are rich in polyphenols; several are galloylated, either as galloylated flavonoids, gallotannins or ellagitannins. The high content of biologically active polyphenols might be important for medicinal effects of this plant and might give a rationale for the widespread usage of S. guineense in Mali. Malaria remains one of the most important pathogenic infectious diseases. Although Africa suffers the greatest brunt, a sizeable proportion of her population still relies on herbal medicines for reasons of cost as well as the belief etched in the minds of consumers that herbal medicines are safer and more efficacious than Modern medicines. Agbo-iba ; a concoction of two or more than two plants is commonly used for the management of malaria in Nigeria. This study assessed the safety and efficacy of a hepta-herbal Agbo-iba (HHA) antimalarial decoction used for the management of malaria in Benin city, Nigeria. Assessment was done against malaria parasite in culture as well as in vivo in pre-clinical murine model of malaria. HHA (IC 50 Pf 3D7 50 μg/ml) was moderately potent and only one of its constituent plants Annickia affinis (IC 50 Pf 3D7 1.49 μg/ml) was far more potent, while all others were moderately active to inactive against the parasite in vitro. HHA showed good selectivity in vitro and was safe at 2 g/kg in mice. However, at 100 mg/kg oral dose, while HHA suppressed parasite growth by 56.76%, the suppression caused by A.affinis was only 32.46% in mice malaria suggesting the existence of synergistic partner(s) in the herbal formula. LCMS revealed the presence of quaternary protoberberine alkaloids (QPAs) in A.affinis and HHA. Although QPAs have strong in vitro antiplasmodial activity, their in vivo antimalarial activity is undermined by being substrates of Permeability glycoprotein (Pgp) efflux pump. Our study suggests that inhibitor(s) of Pgp in HHA could improve the bioavailability of QPAs in mice fed the herbal combo. Further, molecules from other HHA constituent plants may also contribute to the better potency observed for the polyherbal in vivo. These possibilities were validated by the curative antimalarial study at 100 mg/kg, where A.affinis was inactive but the HHA suppressed parasite growth by 44.45%. African traditional medicine is one of the oldest and most diverse practices for treating ailments and numerous natural products have been recommended for gastric ulcer treatment. Helicobacter pylori is the main causative organism implicated in several diseases, most notably in causing inflammation and the onset of gastric ulcers. Current H. pylori treatment methods are losing efficacy as H. pylori rapidly gains resistance to antibiotics. Hence, a search into natural products and their historical traditional efficacy for the treatment of gastric ulcers is of interest. This review aimed to summarise the African use of natural products, including medicinal plants noted in ethnobotanical reviews, used traditionally to treat gastric ulcers, and highlights the investigations into the anti- H. pylori activity of medicinal plants and bee products found in Africa. A systematic review was carried out to identify natural products, including those used traditionally in Africa to treat gastric ulcers, and to correlate this with scientific investigations into the anti- H. pylori activity of natural products used in Africa. A total of 107 literature sources describing the traditional use of medicinal plants in gastric ulcer treatment were found, from which 360 medicinal plants were identified. Of the plants used traditionally for gastric ulcer treatment, 11% were investigated either in vitro or in vivo for anti-ulcer and anti- H. pylori activity. Of the 122 medicinal plants eliciting antimicrobial or anti-ulcer activity, Hibiscus sabdariffa L. calyx extract and Terminalia macroptera Guill. & Perr. root extract were found to have the most noteworthy antimicrobial activity, with minimum inhibitory concentrations (MICs) of 0.01 mg/mL and 0.03 mg/mL respectively. The essential oils of Piper longum L. and Pachira aquatica Aubl. displayed the most notable in vitro anti- H. pylori activity (MIC of 0.01 mg/mL and 0.02 mg/mL). Several in vivo studies found medicinal plant extracts effective in reducing the H. pylori load along the gastric mucosa. The South African honey variants, Pure Honey and Champagne Royal Train (common names given by supplier) were the most antimicrobially effective (MIC of 0.01–10.0%, 0.63–10.00% v/v) in inhibiting H. pylori when assessed in vitro. These results highlight the potential of natural products to inhibit H. pylori growth and serve as a possible stepping-stone in understanding the management of ulcers. Furthermore, effective natural product treatment or prophylactic use for preventing H. pylori growth may provide a more affordable option for African populations. Ethnopharmacological relevance Human Immunodeficiency Virus (HIV) strains resistant to licensed anti-retroviral drugs (ARVs) continue to emerge. On the African continent, uneven access to ARVs combined with occurrence of side-effects after prolonged ARV therapy have led to searches for traditional medicines as alternative or complementary remedies to conventional HIV/AIDS management. Aim of the study Here we characterize a specific three-step traditional HIV/AIDS treatment regimen consisting of Cassia sieberiana root, Vitex doniana root, and Croton megalobotrys bark by combining qualitative interviews of traditional medical knowledge users in Botswana with in vitro HIV replication studies. Materials and methods Crude extracts from a total of seven medicinal plants were tested for in vitro cytotoxicity and inhibition of wild-type (NL4.3) and ARV-resistant HIV-1 replication in an immortalized GFP-reporter CD4+ T-cell line. Results C. sieberiana root, V. doniana root, and C. megalobotrys bark extracts inhibited HIV-1 NL4.3 replication with dose-dependence and without concomitant cytotoxicity. C. sieberiana and V. doniana extracts inhibited HIV-1 replication by 50% at 84.8 µg/mL and at 25 µg/mL, respectively, while C. megalobotrys extracts inhibited HIV-1 replication by a maximum of 45% at concentrations as low as 0.05 µg/mL. Extracts did not interfere with antiviral activities of licensed ARVs when applied in combination and exhibited comparable efficacies against viruses harboring major resistance mutations to licensed protease, reverse-transcriptase, or integrase inhibitors. Conclusions We report for the first time a three-step traditional HIV/AIDS regimen, used alone or in combination with standard ARV regimens, where each step exhibited more potent ability to inhibit HIV replication in vitro. Ethnopharmacological relevance Alchornea cordifolia (Schumach. & Thonn.) Muell. Arg. (Euphorbiaceae) ( A. cordifolia ) is widely distributed throughout tropical Africa, where it is used extensively in traditional medicine. Conditions for which the plant has enjoyed wide use are: coughs, gonorrhoea, infertility, prostatitis, bacterial infections, diarrhoea, ulcers, pain, inflammation, fever and bronchial troubles. This review summarizes the achievements of the investigations in traditional uses, ethnobotany, phytochemistry, biological activities and toxicological profile of A. cordifolia ; this review also describes the shortcomings of studies on this herbal drug and thus serves as the basis of further scientific research and development of this traditional herbal drug. Materials and methods A. cordifolia -related information was collected from various resources including published articles in peer-reviewed journals, unpublished materials, textbooks, government survey reports and scientific databases such as Scifinder®, Pubmed, Science Direct, Wiley, Springer, ACS, Scielo, Web of Science and other web search instruments (Google, Yahoo), published on the subject from 1950 to 2016. ‘The Plant List’ ( www.theplantlist.org ) and ‘Kew Royal Botanic Gardens’ ( mpns.kew.org ) were used to validate the scientific name of the plant. Results The literature revealed several reports on traditional uses, biological activities, chemical constituents and toxicological evaluation of A. cordifolia . The phytochemical information indicates identification of 95 compounds including fatty acids, terpenoids, flavonoids, phenolic acids, alkaloids, which exhibited various pharmacological activities such as wound healing, anti-inflammation, anticancer, antioxidant, immunomodulation, antidiarrhoeal, antimicrobial, antidepressant, hepatoprotective, antiplasmodial and anxiolytic. However, there are still significant gaps in the completeness of our understanding of A. cordifolia bioactivity, therapeutic value, and roles played by each of the numerous phytoconstituents. Conclusions The present review indicated that A. cordifolia is a valuable medicinal plant with multiple pharmacological effects. However, further research on the pharmacological mechanism of action of this plant is recommended in order to unravel the pharmacokinetics, pharmacodynamics, clinical relevance and toxicity of its extracts as well as constituents. Ethnopharmacological relevance Benign prostatic hyperplasia (BPH) is a common urological disorder of men. The ethnomedicinal use of an African plant Prunus africana (Hook.f.) Kalkman ( Pygeum ) in treating men's problems made it a popular remedy all over the globe for the treatment of BPH and related disorders. However, rampant collections made from the wild in Africa have pushed the plant to Appendix II of CITES demanding conservation of the species. Aim of the study In the present study, the aim was to unearth the protective effect of bark of different species of Prunus against BPH. The five selected Indian plants of family Rosaceae viz. Prunus amygdalus Stokes , Prunus armeniaca L. , Prunus cerasoides Buch.-Ham. ex D. Don , Prunus domestica L. and Prunus persica (L.) Batsch were evaluated against P. africana (Hook.f.) Kalkman for a suitable comparison of efficacy as antiBPH agents. Materials and methods The antiBPH activity was evaluated in testosterone (2 mg/kg/day, s.c , 21 days) induced BPH in Wistar rats. The parameters studied were body weights; histopathological examination, immunohistochemistry (PCNA) and biochemical estimations of the prostate; supported by prostatic index, testicular index, creatinine, testosterone levels; antioxidant and anti-inflammatory evaluation. The study also included chemical profiling using three markers ( β -sitosterol, docosyl ferulate and ursolic acid) and estimation of β -sitosterol content through GC. Results The Prunus species showed the presence of all the three markers in their TLC fingerprint profile and maximum amount of β -sitosterol by GC was observed in P. domestica . Interestingly, all the species exhibited significant amelioration in testosterone induced parameters with P. domestica showing the most encouraging effect as indicated from histopathological examination, immunohistochemistry and biochemical studies. The Prunus species further showed remarkable anti-inflammatory and antioxidant activity signifying their role in interfering with various possible factors involved in BPH. Conclusions These findings are suggestive of a meaningful inhibitory effect of testosterone induced BPH by the bark of different species of Prunus in the order of P. domestica , P. persica , P. amygdalus , P. cerasoides and P. armeniaca with an efficacy of P. domestica comparable to P. africana and can be used as the potential backup of Pygeum for the management of BPH. Ethnopharmacological relevance Biophytum umbraculum Welw. (Oxalidaceae) is a highly valued African medicinal plant used for treatment of cerebral malaria, a critical complication of falciparum malaria. Aim of the study To provide additional information about traditional use of B. umbraculum and to test plant extracts and isolated compounds for in vitro activities related to cerebral malaria. Materials and methods The traditional practitioners were questioned about indication, mode of processing/application, dosage and local name of B. umbraculum . Organic extracts and some main constituents of the plant were investigated for anti-malaria, anti-complement activity and inhibition of NO secretion in a RAW 264.7 cell line. Results Treatment of cerebral malaria was the main use of B. umbraculum (fidelity level 56%). The ethyl acetate extract showed anti-complement activity (ICH 50 5.7±1.6 μg/ml), inhibition of macrophage activation (IC 50 16.4±1.3 μg/ml) and in vitro antiplasmodial activity (IC 50 K1 5.6±0.13 μg/ml, IC 50 NF54 6.7±0.03 μg/ml). The main constituents (flavone C -glycosides) did not contribute to the activity of the extract. Conclusion Inhibition of complement activation and anti-inflammatory activity of B. umbraculum observed in this study might be possible targets for adjunctive therapy in cerebral malaria together with its antiplasmodial activity. Ethnopharmacological relevance Pittosporum mannii (Pittosporaceae) is used in Africa traditional medicine to treat various ailments including pain and inflammation. Aim of the study The present work was undertaken to evaluate the antinociceptive effects of the aqueous (AEPM) and methanol (MEPM) extracts from the leaves of Pittosporum mannii . Methods High performance liquid chromatography coupled with mass spectrometry (LCMS) was used for the phytochemical analysis of AEPM prepared as decoction and MEPM prepared as cold maceration. The in vitro cytotoxicity of AEPM and MEPM were evaluated on Artemia salina larvae. AEPM and MEPM antinociceptive effects were evaluated at the doses of 35, 75, 150 and 300 mg/kg given orally, against pain induced by acetic acid, formalin, hot plate, capsaicin and glutamate. The rota rod test was also performed at the same doses. To determine the mechanism of action of these extracts, their antinociceptive effects were tested in animals pretreated with yohimbine (α 2 -adrenergic antagonist), atropine (muscarinic antagonist) or naloxone (an opioids antagonist). Result The LCMS analysis showed that both extracts contain pittovidoside and 1- O -rhamnopyranosyl-23-acetoxyimberbic acid 29-methyl ester, the aqueous extract being more concentrated. Oral administration of both extracts significantly reduced pain symptoms induced by acetic acid, formalin, capsaicin, glutamate and hot plate. The antinociceptive effect of AEPM was significantly inhibited by yohimbine, atropine and naloxone while these inhibitors tend to potentiate the activity of MEPM. Both extracts have no effect on Rota rod test. AEPM and MEPM showed respective LC 50 of 2.44 and 0.70 mg/ml on Artemia larvae and were therefore, considered non-toxic. Conclusion These results indicate that AEPM and MEPM possesses analgesic effects with different mechanism of action. Although effects of both extracts may involve TRPV1 receptors and glutamatergic pathway, AEPM may in addition, interact with alpha-adrenergic, muscarinic and opioidergic pathways that are not involve in the effects of MEPM. Ethnopharmacological relevance Ximenia caffra Sond. (Ximeniaceae), commonly known as “sour plum” is traditionally used, both topically and orally to treat a wide range of human diseases and ailments such as wounds, sexually transmitted infections (STIs), infertility, stomach ache, fever, eye problems, diarrhoea, bilharzia, menorrhagia, malaria, intestinal worms, impotence and coughs. The bark and fruits are used by small-scale farmers as ethnoveterinary medicine to treat dermatophilosis, foot rot, saddle sores and control ectoparasites. Oil from X. caffra seed is traditionally used as a moisturiser, soap and shampoo for dry, fragile and damaged hair. Aim of the review The aim of this study was to comprehensively summarize the research that has been done on the botany, ethnomedicinal uses, phytochemistry and biological activities of X. caffra in different locations throughout its geographical range in the sub-Saharan African region so as to understand its importance and potential in primary healthcare systems. Materials and methods This study was carried out using a comprehensive and systematic literature search on the ethnomedicinal uses, phytochemistry and biological activities of the species throughout its distributional range. Literature sources included papers published in international journals, reports from international, regional and national organizations, conference papers, books and theses. PubMed and Scopus, search engines such as Google Scholar and online collection ScienceDirect were used. Results This study showed that X. caffra is used as traditional medicine in 83.3% of the countries in tropical Africa where it is indigenous. A total of 65 human and animal ailments and diseases are recorded for X. caffra, with a high degree of consensus for wounds, sexually transmitted infections (STIs), infertility, stomach ache, fever, eye problems, diarrhoea, bilharzia, menorrhagia, malaria, intestinal worms and coughs. Phytochemical investigation of X. caffra revealed that the species has various compounds including flavonoids, phenols, phytosterols, tannins and fatty acids. Different plant parts, aqueous and organic extracts exhibited anti-amoebic, antibacterial, antifungal, anti-inflammatory, antioxidant, antiparasitic, antiproliferative, HIV-1 reverse transcriptase (RT) inhibitory, insecticidal, non-mutagenic and toxicity activities. Conclusion In this review, the ethnomedicinal uses, phytochemistry, biological activities and toxicity of different extracts and compounds of X. caffra have been summarized. Although many of the ethnomedicinal uses of X. caffra have been validated by phytochemical and pharmacological studies, there are still some gaps where current knowledge could be improved. There are very few to nil experimental animal studies, randomized clinical trials and target-organ toxicity studies involving X. caffra and its derivatives that have been carried out so far. At the present moment, there is not sufficient evidence to interpret the specific chemical mechanisms associated with some of the documented biological activities of the species. Therefore, future studies should identify the bioactive components, details of the molecular modes or mechanisms of action, pharmacokinetics and physiological pathways for specific bioactives of X. caffra . Ethnopharmacological relevance Extracts of the stem bark of Ficus platyphylla (FP) have been used in traditional the Nigerian medicine to treat psychoses, depression, epilepsy, pain and inflammation. Previous studies have revealed the analgesic and anti-inflammatory effects of FP in different assays including acetic acid-induced writhing, formalin-induced nociception, and albumin-induced oedema. Purpose/methods In this study, we assessed the effects of the standardised extract of FP on hot plate nociceptive threshold and vocalisation threshold in response to electrical stimulation of the tail root in order to confirm its acclaimed analgesic properties. We also investigated the molecular mechanisms underlying these effects, with the focus on opiate receptor binding and the key enzymes of eicosanoid biosynthesis, namely cyclooxygenase (COX) and 5-lipoxygenase (5-LO). Results FP (i) increased the hot plate nociceptive threshold and vocalisation threshold. The increase in hot plate nociceptive threshold was detectable over a period of 30 min whereas the increase in vocalisation threshold persisted over a period of 90 min. (ii) FP showed an affinity for µ opiate receptors but not for δ or κ opiate receptors, and (iii) FP inhibited the activities of COX-2 and 5-LO but not of COX-1. Conclusions We provided evidence supporting the use of FP in Nigerian folk medicine for the treatment of different types of pain, and identified opioid and non-opioid targets. It is interesting to note that the dual inhibition of COX-2 and 5-LO appears favourable in terms of both efficacy and side effect profile. [ABSTRACT FROM AUTHOR]

Ethnopharmacological relevance The rich flora of Guinea-Bissau, and the widespread use of medicinal plants for the treatment of various diseases, constitutes an important local healthcare resource with significant potential for research and development of phytomedicines. The goal of this study is to prepare a comprehensive documentation of Guinea-Bissau’s medicinal plants, including their distribution, local vernacular names and their therapeutic and other applications, based upon local notions of disease and illness. Materials and methods Ethnobotanical data was collected by means of field research in Guinea-Bissau, study of herbarium specimens, and a comprehensive review of published works. Relevant data were included from open interviews conducted with healers and from observations in the field during the last two decades. Results A total of 218 medicinal plants were documented, belonging to 63 families, of which 195 are native. Over half of these species are found in all regions of the country. The medicinal plants are used to treat 18 major diseases categories; the greatest number of species are used to treat intestinal disorders (67 species). More than thirty ethnic groups were identified within the Guinea-Bissau population; 40% of the medicinal plants have been recorded in the country’s principal ethnic languages (i.e. Fula and Balanta). Conclusions This multi-disciplinary, country-wide study identifies a great diversity of plants used by indigenous communities as medicinal, which constitute an important common reservoir of botanical species and therapeutic knowledge. The regional overlap of many indigenous species, the consensual nature of disease groups based upon local perceptions of health conditions, and the relevance of local vernacular including Guinean Creole are key factors specific to the country which enhance the potential for the circulation and transmission of ethno-botanical and therapeutic knowledge. Ethnopharmacological relevance Chromolaena odorata (L.) R.M. King & H. Rob. (Asteraceae) is a scrambling perennial shrub that originated in the Americas, but is now common in sub-Saharan Africa, Asia and Oceania, where it has become a serious weed. The species, particularly the biotype found in Asia and West Africa, has many ethnopharmacological uses, including treatment of malaria, wounds, diarrhoea, skin infection, toothache, dysentery, stomach ache, sore throat, convulsions, piles, coughs and colds. Furthermore, no attempt has been made to synthesise and review the available literature on the usefulness of the plant in the sub-Saharan African region, hence this paper examines the beneficial attributes of C. odorata in sub-Saharan Africa. Material and methods Published information on the species was gathered by the use of different database platforms, including Google Scholar, ScienceDirect, SciFinder and Scopus. Results Records indicate that two biotypes of C. odorata are present in sub-Saharan Africa viz. the more widespread Asian/West African C. odorata biotype (AWAB) and the southern African biotype (SAB). While the usefulness of the former is well elucidated in the literature, such information on the latter is still scarce. Although the importance of AWAB C. odorata as a fallow species and as a soil fertility improvement plant in the slash and burn rotation system of agriculture in West Africa is increasingly being recognised, its usage in traditional medicinal practice is far more appreciated. The species has a wide range of ethnopharmacological uses, possibly because of the presence of flavonoids, essential oils, phenolics, tannins and saponins. The plant is reported to have antibacterial, anti-inflammatory, antioxidant, anthelminthic, antifungal, cytotoxic, anticonvulsant, antiprotozoal, antispasmodic, antipyretic and analgesic properties. Conclusion While the results of this review suggest that the AWAB plant can be exploited as an alternative to other threatened plant species known to possess similar medicinal potential, the medicinal and pharmacological potential of the SAB plant remains to be established. Further studies on the phytochemistry and pharmacological properties of the SAB plants will not only advance our knowledge of ethnobotany and ethnomedicine, but may also improve the health and knowledge of the local people. Ethnopharmacological relevance Neurological diseases are rising all around the world. In a developing country such as Togo, although plant-based medicines are the only means, still very little is known regarding the nature and efficiency of medicinal plants used by indigenous people to manage central nervous system (CNS) disorders. Aim of the study This study, an ethnobotanical survey, aimed to report plant species used in traditional medicine (TM) for the management of various CNS disorders in Togo. Materials and methods 52 traditional actors (TA) including 33 traditional healers (TH) and 19 medicinal plant sellers (MPS) were interviewed, using a questionnaire mentioning informants' general data and uses of medicinal plants. Results The present study reports 44 medicinal plant species distributed into 26 families, mentioning scientific and common local names, plant organs used, preparation method, root of administration and putative applications. Conclusion It appears that there is a real knowledge on medicinal plants used for traditional treatment of CNS disorders in Togo and that the local flora abounds of potentially neuroactive plants which could be useful for the discovery of antipsychotic or neuroprotective molecules. Ethnopharmacological relevance Ethnobotanical knowledge and traditional medicinal practices from different parts of the world are of global importance and documentation of ethnobotanical, and ethnopharmacological data is a key prerequisite for further research in the area of herbal medicine and its implementation in clinical practice. Aims An attempt was made to evaluate the scientific output of research related to ethnobotany and herbal medicine in journals indexed in the subject area “Integrative and Complementary Medicine” in the period 2001–2013, in order to ascertain research trends in both subdisciplines. Materials and methods All articles related to ethnobotany and herbal medicine, extracted from journals included in the field of I&C Medicine and published in the period 2001–2013, have been analyzed for general bibliometric data, and specific data: ethnobotanical data (geographic, floristic, pharmacological, sociological and other relevant data) and phytotherapeutic data (type of applied herbal medicine, plant species studied, pharmacological activity of studied plant species and disease and disorder type studied on a particular model). Results In the studied period, the number of articles dealing with ethnobotany and herbal medicine increased 6.3-fold. Articles related to ethnobotanical studies documented medicinal flora from 81 countries, either giving an overview of overall medicinal flora, or presenting the ethnomedicinal aspect of the use of plants for the treatment of ailments typical to the studied area. Additionally, the authors provided significant information on the methods of use and herbal preparations. In herbal medicine studies, plants, traditional plant remedies, herbal medicinal products and active herbal compounds were tested for many of pharmacological activities (146), with the curative activity emerging as most frequently tested. Out of 39 model systems, most of the studies were carried out under controlled in vitro conditions (4589 articles), followed by rat in vivo (2320), human in vivo (1285), mouse in vivo (955), and on agents of pathogenic diseases (887); more than 800 medical disorders were treated. Conclusions The study revealed the regions most studied for new records of floristic and ethnomedicinal diversity, the most frequently studied plant species, and the most promising therapeutic indications for the integration of herbal remedies in the curative process, as ascertained from the selected bibliographic databases. Ethnopharmacological relevance In Togo, many persons still rely on plants for healing, however very little is known about the medicinal practices of the indigenous people. The present study aimed to document the medicinal plant utilization for the management of liver diseases in the Maritime region of the country. Methodology This was an ethnobotanical survey conducted in the Maritime region of Togo from June to August 2015. The data were gathered from 104 traditional healers (TH) by direct interviews using a semi-structured questionnaire. The calculated use values (UV) were used to analyze the importance of the cited plants. Results A total of 99 plant species belonging to 88 genera and 49 families were cited by the TH as curing the hepatic diseases. The most represented families were Caesalpiniaceae with 8 species, followed by Euphorbiaceae with 7 species, Apocynaceae and Asteraceae with 6 species each. The highest UV were recorded with Gomphrena celosioides (0.13), Xylopia ethiopica (0.12), Senna occidentalis (0.12), Bridelia ferruginea (0.12), Cymbopogon citratus (0.12), Kigellia Africana (0.09), Cassia sieberiana (0.08) and Sanseviera liberica (0.08), showing their importance in the management of liver dysfunction in the surveyed region. The main used parts were the leaves, followed by the roots, the whole plant, the rhizome and the bark, accounting for more than 10% each. The herbal medicines were mostly prepared in the form of decoction and administrated by oral route. Conclusion This study showed that Maritime region of Togo has an important plant biodiversity that is exploited by the indigenous TH. However, some plants cited by the TH have not been studied for their possible hepatoprotective effects. These plants are therefore a starting point for biological screenings. [ABSTRACT FROM AUTHOR]

Ethnopharmacological relevance Considerable medicinal plant research in Brazil has focused on indigenous and mixed-race (caboclo and caiçara) communities, but relatively few studies have examined the medicinal plants and associated healing traditions of the descendants of enslaved Africans. This study surveyed the medicinal plants employed by a relatively isolated maroon community of Afro-Brazilians in the Atlantic coastal rainforests of Bahia, Brazil, a global biodiversity hotspot. The studied community is exceptional in that the residents were defacto slaves until several years ago, with no access to western medicine. We examined the following questions: 1) What medicinal plants are used in this community? 2) What are the principal taxonomic groups, life forms, source habitats, and geographical origins? 3) What species stand out as measured by use value and frequency indices? and 4) Is the community's geographical isolation and African ancestry reflected in their medicinal uses of the local flora? Materials and methods The study was carried out in the Quilombo Salamina Putumuju maroon community in Bahia, Brazil. Data were collected from May to October 2014 from 74 individuals (37 men and 37 women) by means of semi-structured interviews, walk in the woods, and vouchering of identified species. We used the Cultural Value Index (CV), the Relative Frequency Index (RF), and the Use Value Index (UV) to determine the importance of medicinal plant resources. Continuity of African medicinal plant uses and traditions was determined through self-reporting and comparison with previously published works. Results We recorded 118 medicinal plant species distributed in 100 genera and 51 families. The best represented families were: Asteraceae, Fabaceae, Lamiaceae and Myrtaceae. Most plant medicines were used to treat respiratory, digestive systems, genitourinary, and skin problems. The most common medicinal life form was herbs (44%), followed by trees (28%) and shrubs (18%). Native species (55%) were used somewhat more than exotic species (45%), and non-cultivated species (51%) were slightly more numerous than cultivated species (49%). In spite of abundant nearby old-growth forests, trails and gardens were the most common collection sites. A mean of 13.2 medicinal plant species were cited per participant. The highest CV was recorded for Cymbopogon citratus (0.20) followed by Lippia alba (0.19) and Stryphnodendron cf. adstringens (0.17). The highest RF included C. citratus (0.69), L. alba (0.59), and Eugenia uniflora (0.55). The highest UV figures were recorded for S. cf. adstringens (1.68), followed by Sida cf. cordifolia (0.97) and C. citratus (0.93).Fifteen species (13%) of this maroon medicinal flora trace their ancestry to Africa or African-derived healing traditions. Conclusion The Salamina maroon community maintains considerable knowledge of the medicinal value of the local flora. However, little of this knowledge is derived from the surrounding old-growth tropical forests. Their pharmacopoeia is a hybrid mix of wild and cultivated species, natives and exotics. Ethnopharmacological relevance Medicinal plants play a major role in many communities across the world, in the treatment and prevention of disease and the promotion of general health. The aim of the study was to escalate documentation from an earlier study of medicinal plants, traditionally used to combat malaria by the Ameru community of Imenti Forest area and Gatunga in Eastern Region of Kenya, and validate their ethnopharmacological claims by evaluating their antiplasmodial efficacies. Materials and methods The study was carried out in Meru County at Imenti Forest Game Reserve and in Tharaka Nithi County at Gatunga. Traditional health practitioners (THP) were interviewed with a standard questionnaire to obtain information on medicinal plants traditionally used for management of malaria. Group interviews were also held among THPs and members of the community. The antiplasmodial activities of the crude extracts against chloroquine sensitive (D6) and resistant (W2) Plasmodium falciparum were determined using the semi-automated micro-dilution technique that measures the ability of the extracts to inhibit the incorporation of (G-3H) hypoxanthine into the malaria parasite. Results Ninety nine (99) species in eighty one (81) genera and forty five (45) families were documented and evaluated for in vitro antiplasmodial activity. Compositae, Fabaceae, Meliceae, Rubiaceae, Rutaceae and Verbenaceae had the highest number of species mentioned in treatment of malaria in Meru/Tharaka Nithi study area. Twenty four (24.2%) species showed antiplasmodial efficacy of IC 50 ≤5 µg/ml and were considered to have potential for isolation of antimalarial compounds. Eight plant (8) species with moderate antiplasmodial activity namely; Cordia africana , Commiphora africana , Elaeodendron buchananii , Gomphocarpus semilunatus, Tarena graveolens , Plectranthus igniarius , Acacia senegal and Ziziphus abyssinica were documented from this region for the first time for the treatment of malaria. The antiplasmodial activity of MeOH root bark extract of Maytenus obtusifolia was very promising (IC 50 <1.9 µg/ml) and this is the first report on traditional use of M. obtusifolia for treatment of malaria and antimalarial activity. Conclusions The results seem to indicate that ethnopharmacological inquiry used in search for new herbal remedies as predictive and could be used as the basis for search of new active principles. Eight plant (8) species are documented from this region for the first time for the treatment of malaria. Aim of the study Pastoralist communities such as the Maasai are heavily reliant on traditional foods and medicines. This survey sought to identify traditional foods and/or medicinal plants of the Ilkisonko Maasai community living in Kenya. Materials and methods Ethnobotanical knowledge of traditional plants used as food and human/veterinary medicine was obtained using structured and semi-structured questionnaires administered through face to face interviews of key informants. Results A total of 30 species from 21 families and 25 genera were reportedly used as food and/or medicine by 48 respondents. The most commonly encountered genus was the Fabaceae . The growth forms encountered were tree (47%), shrub (33%) and herb (20%). Plants that were commonly mentioned by respondents were Salvadora persica (85%), Grewia villosa (52%), Ximenia americana (52%), Albizia anthelmintica (50%), Acacia robusta (46%) and Acacia nilotica (42%). The root/root bark was the most commonly used plant part (35%), followed by the stem/stem bark (30%), fruit (15%), leaves (11%) and whole plant (9%). Common ailments treated were stomach aches, constipation, back aches, joint aches, body pains and sexually transmitted infections. The plants were also used as tonics, digestives, and restoratives. Conclusion It was evident that traditional medicine was the preferred health care system for the Ilkisonko Maasai community. Ethnopharmacological relevance Malaria remains a major global health threat, with the heaviest burden of disease in sub-Saharan Africa. Effective treatment is not available in many affected areas, and the Plasmodium falciparum parasite is becoming resistant to existing drugs. Alternative therapies are necessary to overcome these challenges. Borassus aethiopum is the third most used palm species in traditional medicines in Africa. Yet, there is only limited information substantiating medicinal properties of the palm. The objective of this study was to document medicinal uses of B. aethiopum and investigate anti-plasmodial activity of the palm extracts used in traditional medicine to treat malaria. Materials and methods The fieldwork and collection of samples was done in Ghana in the Greater Accra, Brong Ahafo, and Volta regions. Our ethnomedicinal survey did not specifically focus on medicinal uses against malaria; any medicinal application of B. aethiopum was of interest. Data collection employed a structured questionnaire, open-ended questions, and group discussion. The experimental extraction of samples was carried out using three common solvents: distilled water, absolute ethanol, and dichloromethane (CH 2 Cl 2 ). Anti-plasmodial activity of compounds was determined against erythrocytic stages of the FCR3 strain of P. falciparum by a [3H]-hypoxanthine incorporation assay. Results A total of 37 use records were documented regarding the medicinal uses of B. aethiopum for the management of 24 different disorders. The highest medicinal use value was recorded for the use of B. aethiopum against malaria, and a subsequent laboratory investigation focused on evaluating anti-plasmodial activity of the palm. Several root and leaf extracts displayed anti-plasmodial activity, with the highest (78% at 50 μg/mL) elicited by one of the dichloromethane root extracts. Conclusion Our results demonstrate the value of integrating ethnobotanical and pharmacological research in the study of beneficial effects of palm products on human health. While the high inhibitory activity found in dichloromethane extracts cannot validate the ethnomedicinal use, the anti-plasmodial effect observed cannot be nullified. We brought preliminary evidence that this palm is a promising source of alternative medicines that could contribute to improving health conditions in malaria endemic areas of sub-Saharan Africa.
