Intending to compare in vitro cell growth in different conditions, we established cell cultures using fin biopsies of two freshwater fishes, Astyanax bimaculatus and Geophagus proximus. Three different culture media (Leibovitz–L‐15, Dulbecco's Modified Eagle Medium [DMEM], and 199) were employed, with or without the addition of AmnioMax, maintaining a standard temperature of 29°C. Based on the results obtained, we standardized a cell growth protocol in which medium 199 was less efficient for both species. Notably, G. proximus cells exhibited superior proliferation in DMEM and L‐15 media, whereas A. bimaculatus cells demonstrated better parameters exclusively in the DMEM medium. Successful subculturing of cells with good proliferation index was observed, accompanied by preserved morphological characteristics. Although Astyanax bimaculatus is the most representative species of the genus in the Amazon region, there are no cytogenetic studies of A. bimaculatus species in Amazon region. Thus, we aimed to analyse the chromosome complements of specimens from this area using classic and molecular cytogenetic approaches. The results revealed the existence of a distinct cytotype and this is the first report of the occurrence of a B microchromosome in the species. Individuals of the same species may present different reproductive tactics depending on the environment in which they develop and mature. The present study aimed to define the gonadal development phases of males and females of Astyanax rivularis and to carry out a comparative analysis of the reproductive development of specimens captured in two isolated environments of the São Francisco River basin in Serra da Canastra, Brazil (Point 1: low vegetation and river showing calm and crystalline waters with small well formations; Point 2: current waters, and well‐established areas of arboreal vegetation). Thus, the gonads of A. rivularis specimens were collected, fixed and processed with techniques for light microscopy. Five maturation phases of the females' reproductive cycle were established: immature, developing, spawning capable, regressing and regenerating. Three maturation phases of the males' reproductive cycle were observed: spawning capable, regressing, and regenerating. There are differences in the phases of gonadal development of A. rivularis between the two sampling points so that, possibly, animals upstream of the waterfall demonstrate a delay in the reproductive cycle in relation to animals downstream. We explored patterns, rates and unexpected socio‐ecological consequences of tooth replacement in serrasalmids and characids of the Peruvian Amazon using microcomputed tomography. Of 24 specimens collected in February 2019, representing a mix of red‐bellied piranha Pygocentrus nattereri, redeye piranha Serrasalmus rhombeus, silver dollar fish Ctenobrycon hauxwellianus and mojara Astyanax abramis, six individuals possessed edentulous jaw quadrants. On average, 22.9% of fish collected per day from these species featured incomplete dentition, a value three to five times higher than anticipated based on replacement rates estimated from captive fish, differences that may be driven by ontogeny, seasonality or environmental quality. Animals evolve their sensory systems and foraging behaviours to adapt and colonize new and challenging habitats such as the dark cave environment. Vibration attraction behaviour (VAB) gives fish the ability to locate the source of a water disturbance in the darkness. VAB evolved in the blind Mexican cave tetra, Astyanax mexicanus. VAB is triggered in cavefish by vibration stimuli peaking at 35 Hz, which is within the main spectrum of water fluctuations produced by many prey crustaceans and insects. VAB has a genetic component and is correlated to an increased number of head mechanosensory neuromasts in the eye orbital region when compared to surface fish. Previous competitive prey capture assays have supported the advantage of VAB for foraging in the dark. Despite its putative adaptive function, VAB has been described as absent in some Astyanax cave populations (Tinaja and Molino) but present in others (Pachón, Piedras, Toro and Sabinos). Here we have tested the occurrence of VAB in the field and in multiple cave populations using a vibrating device in natural pools. Our results confirmed the presence of VAB in caves such as Pachón, Toro and Sabinos but showed that VAB is also present in the Tinaja and Molino cave populations, previously reported as VAB‐negative in laboratory experiments. Thus, VAB is available throughout the range of hypogean A. mexicanus. However, and most notably, within a given cave the levels of VAB were highly variable among different pools. Fish at one pool may express no VAB, while fish at another nearby pool of the same cave may actively show VAB. While a variety of environmental conditions may foster this diversity, we found that individuals inhabiting pools with a high abundance of organic matter have reduced expression of VAB. In contrast, in pools with little organic debris where fish probably depend more on hunting than on scavenging, VAB is enhanced. Our results suggest that expression of VAB is a plastic trait whose variability can depend on local conditions. Resource partitioning has an essential role in interspecific relations, especially in congener species, which share many morphological traits. In some places, small characids coexist through resource partitioning, which may reduce their interspecific competition. Astyanax species (e.g., Astyanax minor, Astyanax gymnodontus and Astyanax bifasciatus), for example, coexist in different water bodies from the Iguaçu River basin. These species have high phenotypic plasticity and many morphologic specializations that allow them to live in different habitats. Based on evidences that these species modified their feeding habits because of changes in resource availability in Iguaçu River, this study tested two hypotheses: (a) there are differences in head morphology, number of teeth and number of gill rakers among the species of Astyanax; and (b) there are differences in gill arch and gill raker morphology among the species of Astyanax, which may favour their trophic resource partitioning in Iguaçu River. The head morphological traits and quantitative morphological characters were summarized in a principal coordinates analysis (PCoA), and the analysis of similarities (ANOSIM) showed significant differences among species. Gill morphological measurements were analysed through analysis of covariance (ANCOVA), and it also showed significant differences in gill arch and gill raker morphology among species. Therefore, the analysis of ecomorphological traits related to trophic habits revealed some differences that may suggest a tendency of reducing competition for trophic resources in the Iguaçu River basin. Astyanax anai, a new species of characid fish, is described from the Sixaola River basin, eastern Costa Rica–western Panama, Central America. The new species can be distinguished from all other congeners by the following combination of characters: premaxillary teeth 4–5 at the inner series and 4–6 at the outer series; maxillary teeth tricuspid, 2–4; predorsal scale series irregular and incomplete, with an unscaled space behind tip of supraoccipital process and 12–14 scales; lateral line scales 34–39; humeral region with a conspicuous black and rounded to horizontally ovate spot and two diffuse brown and vertically elongate bars (the first through the rounded to horizontally ovate spot, the second 2–4 scales behind the first); body depth 36·6–42·3% of standard length (LS); midlateral stripe formed by a series of 10–14 anteriorly‐directed dermal herringbone, or chevron‐shaped, marks, most apparent in juveniles and in preserved specimens, extending above the lateral line from the black humeral spot or just behind it (from the second vertical bar) to the caudal peduncle; scale rows from lateral line to base of first dorsal‐fin ray 8–9; scale rows from lateral line to base of pelvic fin 7–8; pre‐anal distance 53·9–61·9% of LS; total anal‐fin elements 29–33; caudal spot elongated, rhomboid or rectangular, with its anterior margin surpassing the middle of the caudal peduncle, usually reaching the anal‐fin insertion, posteriorly covering 4–7 principal caudal‐fin rays and not extending onto the ventral and dorsal margins of the caudal peduncle, covering 3–5 horizontal scale rows. In order to test the phylogenetic relationships of the new taxon in relation to the other North and Central American species of the genus, a new phylogenetic hypothesis based on a reanalysis of the morphological matrix by Schmitter‐Soto (2016) is proposed. Environmental relevant concentrations of glyphosate‐based herbicide as 50 µg l−1, 300 µg l−1 and 1800 µg l−1 can affect sperm quality of yellowtail tetra fish Astyanax lacustris. Viability of sperm cells was impaired at 300 µg l−1, a concentration that is within legal limits in U.S.A. waterbodies, while motility was impaired at 50 µg l−1, which is the more stringent limit set in Brazilian law. A new species of Astyanax is described from the upper Rio Paraguai basin, Mato Grosso State, Brazil. The new species can be distinguished from congeners by having the body intensely yellowish in life ( v. silvery, reddish or lightly yellow) and by morphometric and meristics traits. Astyanax dolinae n. sp. cannot be assigned to any of the Astyanax species complex currently recognized for the genus. It is only known from the Dolina Água Milagrosa, a karstic sinkhole lake, entirely fed by groundwater, surrounded by Cerrado, the savannah-like vegetation of central South America. This study investigated whether the body morphology of the tetra fish Astyanax lacustris (previously Astyanax asuncionensis) varied between populations inhabiting one lagoon (a lentic, shallow environment, with great habitat complexity created by aquatic macrophytes) and an adjacent river (a deeper, lotic environment where aquatic macrophytes are scarce) in a seasonally flooded wetland, despite population mixing during the wet season. Morphological differences matched a priori predictions of the theory relating functional body morphology and swimming performance in fishes between lagoon and river habitats. The aim of this study was to estimate the genetic diversity and structure of the only known population of minnow Astyanax xavante, which inhabits a stretch of river including several waterfalls. The FST values among the samples were not significant, except between two populations separated by a 30 m waterfall. Nevertheless, haplotype and nucleotide diversity increased in the downstream direction, indicating that gene flow is unidirectional, which indicates this genetic pattern as downstairs gene flow, as it has the effect of increasing genetic diversity in the downstream direction. Astyanax is among the most speciose genera in the Characidae. In this study, molecular markers were used to assess the extent to which some of the commonly recognized Astyanax species of the upper Iguaçu River correspond to natural groups. Constitutive heterochromatin makes up a substantial portion of the genome of eukaryotes and is composed mainly of satellite DNA repeating sequences in tandem. Some satellite DNAs may have been derived from transposable elements. These repetitive sequences represent a highly dynamic component of rapid evolution in genomes. Among the genus Astyanax, the As51 satellite DNA is found in species that have large distal heterochromatic blocks, which may be considered as derived from a transposable DNA element. In the present study, As51 satellite DNA was mapped through in situ fluorescent hybridization in the chromosomes of five species of the genus. The possible roles of this type of saltatory DNA type in the genome of the species are discussed, along with its use for the phylogenetic grouping of the genus Astyanax, together with other shared chromosomal characters. However, the number of As51 clusters is presented as a homoplastic characteristic, thereby indicating evident genomic diversification of species with this type of DNA. [ABSTRACT FROM AUTHOR]

Taxonomic studies of the genus Astyanax from the Iguaçu River (Brazil) indicate that they may be differentiated into 11 distinct species, some of which have not been formally described and named so for. This study focuses on three of these species, Astyanax sp. B, Astyanax sp. C and Astyanax sp. D from the Upper Iguaçu River Basin. Comparative cytogenetic analyses of C-banding, Ag-NORs (silver nitrate stained nucleolar organizer region) and 18S and 5S rDNA corroborate that they are distinct species. A diploid number of 50 chromosomes and similar karyotypic formulae were observed in the three taxa, with the exception of Astyanax sp. D that differs in the number of submetacentric and subtelocentric chromosomes. However, the NOR silver-staining pattern, the heterochromatic bands (C-bands) and the mapping of the 18S and 5S rDNA sites in the chromosomes showed divergences between all three species under study, supporting the occurrence of distinct evolutionary units. Cytogenetic data have provided important clues that the Astyanax fasciatus populations from the Upper Paraná River basin could be a part of a more diverse fish group, usually included on the same taxa. Samples collected in Cachoeira de Emas, SP, in Mogi-Guaçu River basin, show two major cytotypes presenting 2 n = 46 and 2 n = 48 chromosomes, with distinct karyotypic formula, despite the fact that the molecular data suggested some degree of gene flow between these cytotypes. Cytogenetic and morphometric analyses were performed in this species, aiming to contribute to the understanding of the natural history from such fish group. The influence of temperature on prey consumption and growth in mass of juvenile trahira Hoplias aff. malabaricus were investigated. Consumption of small-sized lambari Astyanax altiparanae (mean standard length, LS, 5·43 cm) varied from zero to 65 over a period of 30 days. Temperatures ranged from 14 to 34° C and the size of trahiras ranged from 17·5 to 24·7 cm LS. Prey consumption differed significantly among temperatures. Trahiras at 18° C consumed significantly less than those at 30° C. A linear multiple regression model including temperature, prey consumption and LS explained 89·4% of the variability in growth in mass. Some caution is suggested when inferring the impact of H. aff. malabaricus piscivory on assemblage structures in systems that, despite their location in tropical regions, are subjected to seasonal thermal variations. In a survey of epigean and cave streams in the São Domingos karst area, 38 species were present in the stream reaches, including 10 characiforms, 19 siluriforms, seven gymnotiforms and two perciforms. One species of Astyanax and the new armoured catfish species Parotocinclus were the most common epigean fishes in São Domingos. The most conspicuous non-troglomorphic cave fishes were Hoplerythrinus unitaeniatus, Astyanax sp., Brycon sp. and two species of Imparfinis, with I. hollandi being the most common fish in most caves. São Domingos karst area has the most diverse and abundant Brazilian cavefish fauna, not only in terms of troglobitic species but also in general fish richness, with 22 non-troglomorphic species recorded in caves in addition to five troglobitic ones. Most fishes examined for stomach contents had at least partially full stomachs. The studied fishes were carnivorous, feeding primarily on aquatic insects. Fishes with developed gonads recorded in caves were I. hollandi, Rhamdia quelen, Pseudocetopsis plumbeus, Hoplerythrinus unitaeniatus and Cichlasoma araguaiensis, indicating a potential for reproduction in the subterranean habitat.
