We have evaluated the in vitro low dose hepatotoxic effects of two flame-retardants (BDE-47 and BDE-32) in HepG2 cells. Both congeners declined the viability of cells in MTT and NRU cell viability assays. Higher level of intracellular reactive oxygen species (ROS) and dysfunction of mitochondrial membrane potential (ΔΨ m ) were observed in the treated cells. Comet assay data confirmed the DNA damaging potential of both congeners. BDE-47 exposure results in the appearance of subG 1 apoptotic peak (30.1%) at 100 nM, while BDE-32 arrested the cells in G 2 /M phase. Among the set of 84 genes, BDE-47 induces downregulation of majority of mRNA transcripts, whilst BDE-32 showed differential expression of transcripts in HepG2. The ultrastructural analysis revealed mitochondrial swelling and degeneration of cristae in BDE-47 and BDE-32 treated cells. Abstract: Nickel oxide nanoparticles (NiO-NPs) in the concentration range of 0.025–2.0mg/ml were examined for the induction of oxidative stress, mitochondrial dysfunction, apoptosis/necrosis in tomato seedling roots, as an in vivo model for nanotoxicity assessment in plants. Compared to the control, catalase (CAT), glutathione (GSH), superoxide dismutase (SOD) and lipid peroxidation (LPO) in 2.0mg/ml NiO-NPs treatments exhibited 6.8, 3.7, 1.7 and 2.6-fold higher activities of antioxidative enzymes. At 2.0mg/ml, 122% and 125.4% increase in intracellular reactive oxygen species (ROS) and mitochondrial membrane potential (ΔΨm) of seedling roots confirmed the oxidative stress and mitochondrial dysfunction. Comet assay exhibited a significant increase in the number of apoptotic (21.8%) and necrotic (24.0%) cells in 2.0mg/ml treatment groups vis-á-vis in control 7% apoptotic and 9.6% of necrotic cells were observed. Flow cytometric analysis revealed 65.7% of apoptotic/necrotic cell populations and 2.14-fold higher caspase-3 like protease activity were recorded in 2.0mg/ml treatment groups. Ultrastructure analysis revealed NiO-NPs translocation, nuclear condensation, abundance in peroxisomes and degenerated mitochondrial cristae. The dissolution of Ni ions from NiO-NPs signifies its potential to induce cell death presumably by Ni ions, triggering the mitochondrial dependent intrinsic apoptotic pathway. Abstract: Biodegradation of phenylurea herbicide isoproturon was studied in soil microcosm bioaugmented with a novel bacterial strain JS-11 isolated from wheat rhizosphere. The molecular characterization based on 16SrDNA sequence homology confirmed its identity as Pseudomonas aeruginosa strain JS-11. The herbicide was completely degraded within 20 days at ambient temperature with the rate constant of 0.08 day−1, following the first-order rate kinetics. In stationary phase, at a cell density of 6.5×109 CFUmL−1, the bacteria produced substantially increased amounts of indole acetic acid (IAA) in the presence of tryptophan as compared with the control. Also, the bacteria exhibited a time-dependent increase in the amount of tri-calcium phosphate solubilization in Pikovskaya''s medium. Further screening of the strain JS-11 for auxiliary activities revealed its remarkable capability of producing the siderophores and hydrogen cyanide (HCN), besides antifungal activity against a common phytopathogen Fusarium oxysporum. Thus, the versatile P. aeruginosa strain JS-11 with innate potential for multifarious biological activities is envisaged as a super-bioinoculant for exploitation in the integrated bioremediation, plant growth and disease management (IBPDM) in contaminated agricultural soils. Expanding applications of metal-based nanoparticles (NPs) in industry and agriculture have influenced agro-ecosystems. However, relatively little is known about the bioaccumulation, distribution, and phytotoxicity of ZnO-NPs, CuO-NPs, ZnO-bulk, CuO-bulk, Zn2+, or Cu2+ in maize. Plants were exposed to 0.05–2 mg ml−1 or g−1 of six tested materials in agar (7 days) in hydroponic medium (20 days), or sandy-clay-loam soil (20 or 40 days). Seed germination, emergence and lengths of plumules, principal and seminal roots were significantly inhibited by ZnO-NPs, CuO-NPs, Zn2+, and Cu2+. Toxicity was more pronounced in hydroponic culture than in soil, and perceptible alterations in biomolecules were evident. ICP-MS analysis exhibited progressive uptake of metals while morphological, elemental, and surface/deeper scanning showed translocation and distribution of NPs in tissues. Tested materials induced enhanced superoxide radical production, lipid peroxidation, and antioxidant enzymes and proline levels. Exposure significantly reduced P-accumulation, photosynthesis, and protein production. Zn2+ and Cu2+ were found to be more toxic than NPs. Compared to 20 days exposure in soil, toxicity slightly increased after 40 days. ZnO-NPs and CuO-NPs increased apoptotic sub-G1 population by 22.4% and 38%, respectively. These results provide a better understanding of the mechanistic aspects responsible for the nanotoxicities of ZnO- and CuO-NPs in maize.
