Thus far, domino‐based reactions have received a great deal of attention as a step‐and atom‐economic route toward the synthesis of a plethora of heterocyclic molecules. In this work, we disclose a catalyst‐free domino procedure for rapid synthesis of 3‐substituted 6‐methyl‐1,2‐diarylpyrano[4,3‐b]pyrrol‐4(1H)‐one compound in water at 80°C, accompanied by multiple C‐H/C‐O bond cleavage and C‐C and C‐N bond formation. This approach is characterized by its mild reaction conditions, convenient separation, high tolerance of functional groups, and easily available starting materials, thereby offering a green and efficacious strategy for the design and synthesis of pyrano[4,3‐b]pyrrol‐4(1H)‐one derivatives by domino reaction. A series of 1,5‐dihydro‐4H‐pyrrolo[3,2‐c]pyridin‐4‐one derivatives with 3‐(4‐hydroxy‐2‐oxo‐2H‐pyran‐3‐yl) groups were synthesized via a one‐pot three‐component reaction of 4‐aminopyridin‐2(1H)‐ones, 2,2‐dihydroxy‐1‐arylethan‐1‐ones, and 4‐hydroxy‐2H‐pyran‐2‐ones in water. This strategy not only provides a valuable tool in the design and synthesis of new hydrogenated pyrrolo[3,2‐c]pyridines but also has the advantages of inexpensive starting materials, atom and step economy, environmental friendliness, good to excellent yields, and operational simplicity. A new three‐component domino reaction for the synthesis of 2‐amino‐6‐aryl‐5‐(phenylamino)‐3,7‐dihydro‐4H‐pyrrolo[2,3‐d]pyrimidin‐4‐one derivatives 4 using acetic acid as catalyst has been established. The reaction was performed in aqueous media using readily available and inexpensive 2,6‐diaminopyrimidin‐4(3H)‐one 1, 2,2‐dihydroxy‐1‐arylethan‐1‐one 2 and aniline 3 as substrates. An efficient four‐component approach for the synthesis poly‐substituted pyrano[3,2‐c]pyridones and spiro[indoline‐3,4′‐pyrano[3,2‐c]pyridine]‐2,5′(6′H)‐diones in water has been established. During the reaction, the products were readily achieved through one‐pot two‐step reaction using solid acid as catalyst. The advantages of atom and step economy, the recyclability of heterogeneous solid acid catalyst, easy workup procedure, and the wide scope of substrates make the reaction a powerful tool for assembling pyrano[3,2‐c]pyridone skeletons of chemical and medical interest. A series of heterocyclic compound derivatives, including 2,8‐diamino‐5‐aryl‐5,10‐dihydropyrido[2,3‐d:6,5‐d′]dipyrimidine‐4,6(3H,7H)‐diones and 2,2′‐diamino‐3H‐spiro [pyrimido[4,5‐b]quinoline‐5,5′‐pyrrolo[2,3‐d]pyrimidine]‐4,4′,6′(3′H,7′H,10H)‐triones, were synthesized via one‐pot method using Y (OTf)3 as catalyst. The reaction was performed in aqueous media using readily available and inexpensive 2,6‐diaminopyrimidin‐4(3H)‐one and aromatic aldehyde as substrates. The present methodology shows attractive characters, such as using water as green reaction media, high catalytic efficiency of catalyst, concise one‐pot method, easy work‐up/purification, and excellent yields. A variety of substituted benzo[ g]quinoline-5,10-dione derivatives were synthesized via one-pot three-component reaction of 2-hydroxynaphthalene-1,4-dione, aromatic aldehydes, and ( E)-3-aminobut-2-enoates using carbonaceous material as the heterocyclic catalyst. This procedure is simple and efficient owing to short reaction times, easy work-up, and recovery of heterogeneous catalyst.
