Studies were carried out in South America to assess the effect of maternally derived antibody (MDA) on the responsiveness of calves to FMD vaccination. It was found that calves with MDA did not merely fail to respond to vaccination, but that their serum titres were depressed. This depression was proportional to the level of pre-existing MDA at the time of vaccination and following primary vaccination it persisted for a least 60 days. High MDA titres interfered with both primary and secondary responses. Animals with relatively low MDA titres were able to respond to vaccination, or at least to be sensitized so that on revaccination they showed a satisfactory response. The half-life of MDA was shown to be approximately 22 days, suggesting that under field conditions significant MDA titres are likely to persist for 4–5 months. A trial carried out in Brazil in which the primary course of two inoculations, 4 weeks apart, was initiated when the calves were 5–6 months of age, resulted in the reduction of FMD in the calf population from 11% to 0·9% over a 12-month period. Serological evaluations of foot-and-mouth disease type SAT 2 viruses isolated in Kenya between 1979 and 1982 were performed using the two-dimensional microneutralization test. Nine field isolates of epizootiological significance were compared with four vaccine viruses. The results obtained identified Tan 5/68 as the most appropriate reference vaccine virus strain since it had the broadest serological spectrum. Potent Tan 5/68 vaccines would be expected to provide adequate protection against the contemporary SAT 2 field viruses. In the case of K 83/74, which also was shown to have a broad spectrum with viruses isolated in Kenya, the results show that the 1982 isolate from central Kenya was significantly divergent (r < 1·00 at P = 0·01) and warranted tactical revaccination for its control. The study highlighted the fact that strain R1215 which had been isolated from the oesophageal-pharyngeal swabs of asymptomatic carrier cattle had a narrow serological spectrum suggesting that such viruses could be unsuitable as vaccine for the national campaign. A study of the ability of 49 strains of FMD virus to replicate in BHK-21 monolayer cells maintained under a standard agar overlay containing 5·2 mM guanidine hydrochloride and to withstand heat inactivation at 54°C for 1 h showed that strains belonging to serotypes C, O and Asia 1 were generally more resistant to guanidine and heat stable than the SAT 1, 2 and 3 serotypes. The type A viruses as a whole occupied an intermediate position between these two groups.In vitro passage in BHK-21 cells influenced the guanidine sensitivity of 3(O, C and SAT 3) of the 7 FMD serotypes suggesting that this is not a stable genetic marker. Heat stability of the FMD viruses, however, did not change on passage, suggesting that this is a stable characteristic inherent in any homogeneous FMD virus population. [ABSTRACT FROM PUBLISHER]

A microneutralization test for serotyping of FMD viruses is described. It is based on earlier observations by Booth, Rweyemamu & Pay (1978) that dose-response relationships in quantal microneutralizations often deviated from linearity. The typing test described therefore utilizes undiluted virus preparations. In about 90% of samples a positive typing was obtained in contrast with about 50% for the complement fixation test. The test was also found to be susceptible to minimal quantities of heterotypic viral contamination.For strain differentiation the microneutralization test was carried out as a checkerboard test. When compared with the complement fixation test it was found to be more specific. The necessity to utilize virus-neutralization test systems for comparing (FMD) virus strains particularly for the purpose of vaccine selection is emphasized. Two-dimensional quantal microneutralization tests on foot-and-mouth disease viruses, in which neutralizing antibody activity was titrated against a serial range of virus doses, demonstrated a variety of dose-response curves some of which were rectilinear, others clearly curvilinear. Moreover, in the case of the non-linear responses obtained with some antisera, the shape of the curve was such that antibody titres recorded with doses of virus ranging from 103–105 TCD 50 were closely similar. Studies were carried out on the effect of varying the conditions of the test on the shape of the dose-response curve: significant differences were obtained after treatment of the antiserum–virus mixtures with anti-species globulin, and when the test was assayed in cells of differing susceptibility to infection. A comparison of homologous and heterologous rates of neutralization demonstrated that antigenic relationships of foot-and-mouth disease virus strains could be differentiated quantitatively by the kinetics of neutralization method described previously (Rweyemamu, Booth & Pay, 1977). Conventional formulae for r, R and D values could be applied to results obtained by this method. It was demonstrated that results analysed this way gave R values which were similar to those obtained with other neutralization test methods but which were generally smaller than those obtained with complement fixation test results. It was demonstrated that there were wide differences between the vaccine strains tested as demonstrated by R value relationships. An examination of r values, however, demonstrated that antisera to the Moz 1/70 strain were highly reactive with most of the virus strains from Central and Southern Africa. Deviations from first-order kinetics observed represented biphasic or parabolic and stepwise reactions. The initial rate was rapid and showed no lag phase or shoulder. The effects of deviations from linearity could be minimized by dilution of antiserum to a suitable extent. Treatment of virus–antibody mixtures with anti-species globulin resulted in enhancement of the rate of neutralization of homologous and heterologous reactions without significantly altering the relation between the two. This treatment also considerably reduced the amount of the persistent fraction.
