Long lasting permethrin-impregnated (LLPI) clothing can retain permethrin and repel ticks for up to three months and without exceeding EPA-approved safe levels; however, little is known about longer term effects of wearing LLPI clothing. Here, permethrin content was measured in new forester pants soon after initial impregnation (Insect Shield) and again one year later after being repeatedly worn by foresters in the field. Urine samples were collected from foresters for biomonitoring of permethrin metabolites at multiple time intervals (pre-use, one-month, three-to-four-months, and one-year post-use). Lethality against nymphal Ixodes scapularisSay was measured in clothing after one year of wear by foresters. Furthermore, to test potential variability in permethrin impregnation of different batches of clothing, separate sets of clothing were anonymously sent to Insect Shield for permethrin treatment over a period of three months and permethrin was quantified. Results demonstrated 33% of participants’ pants had no measurable permethrin after one year of wear and permethrin content and tick mortality varied significantly between clothing. Only two of the participants’ clothing resulted in ≥ 30% tick mortality after one year of wear. Significant differences were observed in 3-PBA and trans-DCCA, but not cis-DCCA metabolites in participants over the four measured time points and were higher than general United States population levels. This study provides practical information on the safety (measured by urinary metabolites) over time of LLPI clothing. It also provides snapshots (pre-washing and after one year of wear) of effectiveness of LLPI clothing as personal protective equipment against ticks for outdoor workers.

Burgeoning cases of tick-borne disease present a significant public health problem in the United States. Passive tick surveillance gained traction as an effective way to collect epidemiologic data, and in particular, photograph-based tick surveillance can complement in-hand tick specimen identification to amass distribution data and related encounter demographics. We compared the Federal Information Processing Standards (FIPS) code of tick photos submitted to a free public identification service (TickSpotters) from 2014 to 2019 to published nationwide county reports for three tick species of medical concern: Ixodes scapularisSay (Ixodida: Ixodidae), Ixodes pacificusCooley and Kohls (Ixodida: Ixodidae), and Amblyomma americanumLinneaus (Ixodida: Ixodidae). We tallied the number of TickSpotters submissions for each tick species according to “Reported” or “Established” criteria per county, and found that TickSpotters submissions represented more than half of the reported counties of documented occurrence, and potentially identified hundreds of new counties with the occurrence of these species. We detected the largest number of new county reports of I. scapularispresence in Michigan, North Carolina, and Texas. Tick image submissions revealed potentially nine new counties of occurrence for I. pacificus, and we documented the largest increase in new county reports of A. americanumin Kentucky, Illinois, Indiana, and Ohio. These findings demonstrate the utility of crowdsourced photograph-based tick surveillance as a complement to other tick surveillance strategies in documenting tick distributions on a nationwide scale, its potential for identifying new foci, and its ability to highlight at-risk localities that might benefit from tick-bite prevention education.

Tick identification is critical for assessing disease risk from a tick bite and for determining requisite treatment. Data from the University of Rhode Island’s TickEncounter Resource Center’s photo-based surveillance system, TickSpotters, indicate that users incorrectly identified their submitted specimen 83% of the time. Of the top four most commonly submitted tick species, western blacklegged ticks (Ixodes pacificusCooley & Kohls [Ixodida: Ixodidae]) had the largest proportion of unidentified or misidentified submissions (87.7% incorrectly identified to species), followed by lone star ticks (Amblyomma americanumLinneaus [Ixodida: Ixodidae]; 86.8% incorrect), American dog ticks (Dermacentor variabilisSay [Ixodida: Ixodidae]; 80.7% incorrect), and blacklegged ticks (Ixodes scapularisSay [Ixodida: Ixodidae]; 77.1% incorrect). More than one quarter of participants (26.3%) submitted photographs of ticks that had been feeding for at least 2.5 d, suggesting heightened risk. Logistic regression generalized linear models suggested that participants were significantly more likely to misidentify nymph-stage ticks than adult ticks (odds ratio [OR] = 0.40, 95% confidence interval [CI]: 0.23, 0.68, P< 0.001). Ticks reported on pets were more likely to be identified correctly than those found on humans (OR = 1.07, 95% CI: 1.01–2.04, P< 0.001), and ticks feeding for 2.5 d or longer were more likely to be misidentified than those having fed for one day or less (OR = 0.43, 95% CI: 0.29–0.65, P< 0.001). State and region of residence and season of submission did not contribute significantly to the optimal model. These findings provide targets for future educational efforts and underscore the value of photograph-based tick surveillance to elucidate these knowledge gaps.

Knockdown and residual activity of 10 minimal risk natural products (MRNPs), one experimental formulation of nootkatone, and two bifenthrin labels were evaluated against host-seeking nymphal Ixodes scapularisSay using a novel micro-plot product screening system placed in a landscape setting similar to a wooded residential property. The MRNPs evaluated included Tick Stop, EcoPCO EC-X, Met52 EC, CedarCide PCO Choice, EcoEXEMPT IC2, EcoSMART Organic Insecticide, Essentria IC3, privately labeled products 1 and 2 (based on EcoEXEMPT IC2and sold as a professional pest control application), and Tick Killz. Just the nootkatone and 4 of these 10 products tested (EcoPCO EC-X, Met52 EC, EcoEXEMPT IC2, and Essentria IC3) had statistically significant (P< 0.05) knockdown effects (killed ticks while active in the arenas) when compared to water-only controls, but only 2 of these, EcoPCO EC-X and nootkatone, displayed significant residual tick-killing activity after weathering naturally in the landscape for 2 wk prior to tick application/testing. Moreover, botanical oil-based products with the same active ingredients provided inconsistent results when tested multiple times across study years.

As tick vector ranges expand and the number of tickborne disease cases rise, physicians, veterinarians, and the public are faced with diagnostic, treatment, and prevention challenges. Traditional methods of active surveillance (e.g., flagging) can be time-consuming, spatially limited, and costly, while passive surveillance can broadly monitor tick distributions and infection rates. However, laboratory testing can require service fees in addition to mailing and processing time, which can put a tick-bite victim outside the window of potential prophylactic options or under unnecessary antibiotic administration. We performed a retrospective analysis of a national photograph-based crowdsourced tick surveillance system to determine the accuracy of identifying ticks by photograph when compared to those same ticks identified by microscopy and molecular methods at a tick testing laboratory. Ticks identified by photograph were correct to species with an overall accuracy of 96.7% (CI: 0.9522, 0.9781; P< 0.001), while identification accuracy for Ixodes scapularisSay (Ixodida: Ixodidae), Amblyomma americanumLinnaeus (Ixodida: Ixodidae), and Dermacentor variabilisSay (Ixodida: Ixodidae), three ticks of medical importance, was 98.2% (Cohen’s kappa [κ] = 0.9575; 95% CI: 0.9698, 0.9897), 98.8% (κ = 0.9466, 95% CI: 0.9776, 0.9941), and 98.8% (κ = 0.9515, 95% CI: 0.9776, 0.9941), respectively. Fitted generalized linear models revealed that tick species and stage were the most significant predictive factors that contributed to correct photograph-based tick identifications. Neither engorgement, season, nor location of submission affected identification ability. These results provide strong support for the utility of photograph-based tick surveillance as a tool for risk assessment and monitoring among commonly encountered ticks of medical concern.

Tick-borne diseases are a growing threat to public health in the United States, especially among outdoor workers who experience high occupational exposure to ticks. Long-lasting permethrin-impregnated clothing has demonstrated high initial protection against bites from blacklegged ticks, Ixodes scapularisSay (Acari: Ixodidae), in laboratory settings, and sustained protection against bites from the lone star tick, Amblyomma americanum(L.) (Acari: Ixodidae), in field tests. However, long-lasting permethrin impregnation of clothing has not been field tested among outdoor workers who are frequently exposed to blacklegged ticks. We conducted a 2-yr randomized, placebo-controlled, double-blinded trial among 82 outdoor workers in Rhode Island and southern Massachusetts. Participants in the treatment arm wore factory-impregnated permethrin clothing, and the control group wore sham-treated clothing. Outdoor working hours, tick encounters, and bites were recorded weekly to assess protective effectiveness of long-lasting permethrin-impregnated garments. Factory-impregnated clothing significantly reduced tick bites by 65% in the first study year and by 50% in the second year for a 2-yr protective effect of 58%. No significant difference in other tick bite prevention method utilization occurred between treatment and control groups, and no treatment-related adverse outcomes were reported. Factory permethrin impregnation of clothing is safe and effective for the prevention of tick bites among outdoor workers whose primary exposure is to blacklegged ticks in the northeastern United States.

Measuring rates of acquisition of the Lyme disease pathogen, Borrelia burgdorferi sensu latoJohnson, Schmid, Hyde, Steigerwalt & Brenner, by the larval stage of Ixodes scapularisSay is a useful tool for xenodiagnoses of B. burgdorferiin vertebrate hosts. In the nymphal and adult stages of I. scapularis, the duration of attachment to hosts has been shown to predict both body engorgement during blood feeding and the timing of infection with B. burgdorferi. However, these relationships have not been established for the larval stage of I. scapularis. We sought to establish the relationship between body size during engorgement of larval I. scapularisplaced on B. burgdorferi-infected, white-footed mice (Peromyscus leucopusRafinesque) and the presence or absence of infection in larvae sampled from hosts over time. Body size, time, and their interaction were the best predictors of larval infection with B. burgdorferi. We found that infected larvae showed significantly greater engorgement than uninfected larvae as early as 24 h after placement on a host. These findings may suggest that infection with B. burgdorferiaffects the larval feeding process. Alternatively, larvae that engorge more rapidly on hosts may acquire infections faster. Knowledge of these relationships can be applied to improve effective xenodiagnosis of B. burgdorferiin white-footed mice. Further, these findings shed light on vector–pathogen–host interactions during an understudied part of the Lyme disease transmission cycle.

The wetness and greenness indices created using Landsat Thematic Mapper (TM) data from June 1995 and 1997 and July 2002 were tested for their ability to predict the location of sites with different levels of nymphal blacklegged tick, Ixodes scapularisSay, abundance in Rhode Island. In 1995, there were statistically significant differences in the mean of greenness and wetness indices between sites classified as low and moderate tick abundance areas (P= 0.005 and P= 0.041, respectively). In 1997, there also were statistically significant differences in the mean of the greenness and wetness indices, but these differences were between the grouping of low/moderate tick abundance and the high tick abundance category (P= 0.023 and P= 0.015, respectively). The same indices from the 2002 image were not significant predictors of tick abundance. It may be that Landsat TM-derived indices can be used to predict nymphal blacklegged tick abundance in years (e.g., 1995 and 1997) when tick abundance is lower than average but not in years when it is higher (e.g., 2002). Thus, it seems unlikely that these remotely sensed indices will be very useful for modeling nonperidomestic Lyme disease risk over a large region in Rhode Island.

We compared the intrinsic vector competence of Ixodes dammini Spielman et al., Dermacentor variabilis (Say), and Amblyomma americanum (L.) for the Lyme disease spirochete (Borrelia burgdorferi Johnson et al.) on Prudence Island, Rhode Island, a Lyme disease-endemic site where all three ticks occur together. Natural and experimental spirochete infection rates were determined in those ticks and their degree of contact with white-footed mice (Peromyscus leucopus), the principal reservoir host, was compared. Host-seeking nymphal and adult I. dammini and A. americanum, collected by flagging, were nearly equally abundant, as were adult D. variabilis, but only I. dammini were infected with B. burgdorferi. Larvae and nymphs of both I. dammini and D. variabilis were found infesting mice, but A. americanum never were found on this host. Furthermore, although larvae of all three tick species became infected by ingesting spirochetes while feeding on experimentally infected mice, only I. dammini remained infected following the transstadial molt. These findings suggest that of these three tick species, only I. dammini is competent as a vector of the Lyme disease spirochete.

Several methods of extracting DNA from ticks were examined to improve the efficiency of polymerase chain reaction (PCR) detection of the human granulocytic ehrlichiosis (HGE) agent. DNA was extracted from laboratory-reared uninfected and HGE-infected ticks using 3 separate methods. In one treatment, unfed nymphs and engorged larvae of Ixodes scapularis Say, either individually or in pools of 3, were homogenized in 40 µl of 1x PCR buffer and boiled for 30 min. A 2nd group of ticks was extracted using the QiaAmp Tissue kit, a silica column separation method. A 3rd group was extracted with DNA-STAT, a guanidinium thiocynate method. Five microliters of each extract was used for PCR amplification. Pathogen-free tick DNA samples did not amplify a product. Laboratory-infected ticks extracted either with the QiaAmp kit or those homogenized and boiled in PCR buffer amplified product in 37.5% and 87.5% of the samples, respectively. Infected ticks extracted with DNA STAT-60 amplified a product in 100% of samples. No differences were observed in detection efficiency between ticks tested singly or in pools.

The spirochetal agent of Lyme disease, Borrelia burgdorferi, disseminated from the guts of infected Ixodes dammini ticks following host attachment and appeared in saliva before rapid engorgement commenced. In about half of the adult ticks studied, spirochetes disseminated to the hemocoel at 4 days after attachment. Of these, half produced saliva containing spirochetes. Infected nymphs correspondingly produced spirochete-containing saliva at 3 days postattachment, the time at which saliva was first collected. We conclude that contact between spirocheteinfected ticks and a host provides the stimulus that ultimately results in dissemination of infection and delivery via saliva. This salivary route of infection appears to be the route of Lyme disease transmission in nature.

The wasp Hunterellus hookeri Howard parasitizes several species of ixodid ticks including Ixodes dammini Spielman, Clifford, Piesman & Corwin, the vector of Lyme disease spirochetes, Borrelia burgdorferi, in the northeastern United States. We detected the occurrence and evaluated the distribution of this wasp parasitoid in populations of I. dammini in Rhode Island. There has been no intentional introduction of a tick parasitoid recorded anywhere in Rhode Island; yet, we found this wasp at one of the six study sites (Prudence Island), where it parasitized 21 (n = 243) and 17% (n = 284) of nymphal I. dammini collected during 1988 and 1989, respectively. The proportion of nymphs parasitized was greatest during May (46%) and was less in June (18%), July (18%), and August (11%). In Rhode Island, the wasp was only found parasitizing ticks at the site with the highest (by a factor of 2) tick population, confirming similar observations in Massachusetts and New York. It is suggested that establishment as well as the distribution of H. hookeri depends upon a superabundant deer tick population. The usefulness of this parasitoid as a biological control agent is yet unknown.

The risk for human infection with Lyme disease appears linked to the abundance of infected vector ticks, principally Ixodes dammini Spielman, Clifford, Piesman & Corwin, in the eastern United States. Habitat destruction by burning, although not well studied, has long been considered as an effective alternative to synthetic insecticides as a means of reducing tick populations. We evaluated the effect of a single spring burning of the woodland understory on the transmission risk of Lyme disease spirochetes (Borrelia burgdorferi Johnson, Schmid, Hyde, Steigerwalt & Brenner) on Shelter Island, Long Island, NY. Following a burn in early April 1991, the abundance of nymphal I. dammini was 49% lower in the burned portion of a woodlot compared with the unburned portion. However, risk of encountering nymphs infected with B. burgdorferi remained similar in both burned and unburned woods. It is suggested that burning vegetation may disproportionately kill deer-derived rather than rodent-derived nymphs, significantly reducing tick abundance without affecting transmission risk.

Seasonal variation in the capacity of bank voles, Clethrionomys glareolus (Schreber), to infect larval Ixodes ricinus (L.) ticks with Borrelia burgdorferi Johnson, Schmid, Hyde, Steigerwalt & Brenner was examined from May through October 1991 at Bogesund, near Stockholm in south-central Sweden. Although larval infestations of bank voles were greatest in June and July, nearly 70% of all larval ticks infected with spirochetes by bank voles at this site became infected during August and September. Seasonality of infectivity was related to the degree of earlier nymphal infestation on voles as well as to the age composition of the vole population. These factors may influence the infectivity of other rodent reservoirs of B. burgdorferi, both in Europe and North America. Moreover, in determining the reservoir potential of tick hosts, a host population's spirochetal infectivity should be determined for the entire period of larval infestation rather than just during the period of peak larval infestation.

Zoonotic prevalence of Babesia microti Franca piroplasms infecting white-footed mice, Peromyscus leucopus Rafinesque, was determined at 34 sites in Rhode Island where nymphal blacklegged tick, Ixodes scapularis Say, densities ranged from low to hyperabundant (1.7–525.3 nymphs per hour of flagging). Babesia was only detected at sites where tick abundance was moderate to high (>20 nymphs per hour of flagging) and appeared to exhibit a clumped distribution. Where B. microti was detected, the mean number of nymphal ticks collected per hour of flagging was 229.2 compared with a mean of 40.1 at sites where Babesia was not detected. By combining the spatial occurrence of Babesia with a tick density database in a geographic information system, it may be possible to predict the pattern of zoonotic and human infection with B. microti.

Lyme disease is a tick-transmitted borreliosis of humans and domestic animals emerging as one of the most significant threats to public health in north temperate regions of the world. However, despite a myriad of studies into symptomology, causes, and treatment of the disease, few researchers have addressed the spatial aspects of Lyme disease transmission. Using statewide data collected in Rhode Island (United States) as a test case, we demonstrated that exposure to deer ticks and the risk of contracting Lyme disease occurs mostly in the peridomestic environment. A Geographic Information System model was developed indicating a strong association among Lyme disease in humans, the degree of nymphal blacklegged tick, Ixodes scapularis Say, abundance in the environment, and prevalence of Borrelia hurgdorferi infection in ticks. In contrast, occurrence of plant communities suitable for sustaining I. scapularis populations (forests) was not predictive of Lyme disease risk. Instead, we observed a highly significant spatial trend for decreasing number of ticks and incident cases of Lyme disease with increasing latitude. Geostatistics were employed for modeling spatial autocorrelation of tick densities. These findings were combined to create a model that predicts Lyme disease transmission risk, thereby demonstrating the utility of incorporating geospatial modeling techniques in studying the epidemiology of Lyme disease.

Activity of gravid Aedes triseriatus (Diptera: Culicidae) was monitored throughout the summer of 1981 in southwestern Wisconsin, USA, using ovitraps in open terrain such as fields between woodlots. Ovitraps were placed at various distances from the woods in various field types, including corn, alfalfa, pasture, mixed plantings, fallow old fields, and an open yard. Weekly ovitrap checks showed that Ae. triseriatus readily dispersed into all types of open areas; however, field type influenced the amount of oviposition activity observed. Dispersion of gravid females was lowest into alfalfa and corn fields, with only 4.3 and 7.6% of ovitrap observations positive for eggs, respectively. Dispersion was greater into wet meadow, old field, and open yard, as 17.9, 33.9, and 40% of ovitrap observations were positive for eggs, respectively. Distance between woodlots did not appear to influence dispersion rates, as a total of 14.5% of observations from all open-terrain ovitraps more than 100 m from woods were positive for eggs. In 1 pastureland site, ovitraps placed 273 m from the nearest woods were positive for eggs in 27.5% of trap observations. Ovitraps placed in fencerows or on isolated trees in open areas were more attractive to ovipositing mosquitoes than traps placed on wooden stakes in all open terrain habitats. Adult male and female Ae. triseriatus were observed in open terrain on a number of occasions. The effect of open-terrain dispersion by Ae. triseriatus on the epidemiology of La Crosse encephalitis virus is discussed.

To determine whether larval Ixodes dammini are infected transovarially by Lyme disease spirochetes (Borrelia burgdorferi), we swept larval I. dammini from vegetation in 4 regions of Massachusetts, USA, where the spirochetes are endemic. Larvae were permitted to feed on noninfected hamsters. After molting to the nymphal stage, 274 were examined for spirochetes. Two nymphs were found to be infected. The rarity of transovarial infection (0.7%) suggests that transovarial passage is of limited importance in maintaining B. burgdorferi in nature, but that it may provide a mechanism for transporting spirochetes to new sites.
