Objective To prepare a phase⁃transforming multimodal molecular probe that carries the thrombophilic peptide glutamic acid⁃tryptophan⁃valine⁃aspartate⁃valine (EWVDV), and to study its phase change law，in vitro multimodality imaging and targeting of thrombus. Methods Nanoparticles carrying perfluorohexane（PFH），indian ink and Fe3O4 were prepared by a three⁃emulsification method using polylactic acid/glycolic acid（PLGA）as a carrier.The EGVDV⁃targeted nanoparticles modified with gallic acid（GA）were prepared by the carbodiimide method to characterize their physicochemical properties.Its phase transformation laws，in vitro multimodality imaging，and targeting of extracorporeal thrombi were studied. Results The prepared EWVDV⁃PFH⁃Ink⁃Fe nanoparticles were round，uniform in size，well dispersed，with an average particle diameter of 370.7 nm，and the EWVDV connection ratio was 62.5%.In the phase transition caused by heat，the phase transition of nano⁃particles tended to be obvious as the heating time increases and the temperature increases.In the photoinduced phase transition，after irradiation with a laser beam，when the temperature of the nanoparticles reached 60 to 80 ℃，white，uniform⁃sized phase change microbubbles were visible above the gel model，and the B⁃mode and contrast modes all probed the hy⁃perechoic ultrasound signals. At the same time，nanoparticles could also realize photoacoustic and nuclear magnetic resonance imaging.In thrombotic cryosections，a large number of fluorescein nanoparticles were connected to the thrombus surface in the EWVDV⁃PFH⁃Ink⁃Fe group compared to the non⁃targeted group（PFH⁃Ink⁃Fe）. Conclusion The EWVDV⁃PFH⁃Ink⁃Fe nanoparticles prepared in this experiment can successfully achieve phase transitions and are expected to achieve controlled phase transitions. It also has ultrasound，photoacoustic and MR imaging capabilities. With the help of EWVDV peptides， nanoparticles have good targeting properties for thrombosis，which provides the potential for multimodal imaging of thrombi and precise diagnosis at the molecular level.
